Motility and chemotaxis have been considered as important candidates for the study of virulence in many pathogenic spirochetes, including B. burgdorferi. Little is known about the underlying mechanisms of motility and chemotaxis. In this study, we have carried out an extensive search for the motility and chemotaxis genes in B. burgdorferi. The unique technique of "semi-random PCR chromosome walking" was developed and used to clone several large motility and chemotaxis gene clusters. Together with the previously reported flaB and flgE genes, 38 motility and chemotaxis related genes (more than 36 kb) were identified. These genes demonstrated extensive homology with their …

Man and the Gulf of Mexico (MGM) was a federally-funded curriculum development project which produced a four-volume set of marine science curriculum materials for use by junior high and secondary school students in Alabama and Mississippi. Individual titles in the series are Marine and Estuarine Ecology, Marine Habitats, Diversity of Marine Animals, and Diversity of Marine Plants. This dissertation focused on two aspects of Nuclear Magnetic Resonance (NMR) techniques in the solid state: (i) the possibility of enhancing the NMR spectral resolution via utilization of single crystals (instead of powders) in the magic angle spinning experiments, and (ii) application of this methodology as a new probe in the studies of the microscopic mechanism of solid-solid phase transitions in hydrogen-bonded solids. This study examined differences in cognitive structure, content knowledge, navigational difficulties, and personal interviews between organizer and non-organizer students in a hypermedia environment. This information was used to ascertain the feasibility of using the pictorial graphic organizer as a tool to help alleviate navigational difficulties in a learner-controlled hypermedia environment without imposing structure on the student. Sixty-nine students from an undergraduate course completed a computer questionnaire as well as a pre content knowledge test and a Pathfinder network. 1,2,3-Butatrienes ( (3) cumulenes) were synthesized via 1,4-eliminations of hydroxytrimethylsilanes from 4-(trimethylsilyl)-2-butyn-1-ols. The precursors, 4-(trimethylsilyl)-2-butyn-1-ols, were easily prepared from a propargylic trimethylsilane and an aldehyde or a ketone. Treatment of the propargylic trimethylsilane in THF with n-butyllithium followed by condensation with an aldehyde or a ketone afforded 4-(trimethylsilyl)-2-butyn-1-ols in good yields. Sequential treatment of 4-(trimethylsilyl)-2-butyn-1-ols with n-butyllithium, methanesulfonyl chloride, and TBAF furnished the corresponding (3) cumulenes in good yields. (3) Cumulenes with diverse chemical structures were synthesized by selecting different combinations of aldehydes or ketones for condensation with propargylic trimethylsilanes to produce the cumulene precursors. To date, no standard dissolution methodology is recognized in the official compendia for protein-based injectable suspensions. Development of a sensitive dissolution method would have utility as an in vitro quality control tool and an indicator of associated in vivo product performance. Using the spin-filter as the dissolution device of choice, the objective of these studies was to develop a standard dissolution methodology for protein-based suspensions. To assist U.S. dental schools/colleges in meeting the mandates of the ADA, this study examined the key findings in relevant court cases, explored accommodations that are being made by dental programs, investigated barriers encountered when providing accommodations, and gathered data on admission policies, criteria and procedures. In 1995 a mail survey of all U.S. dental schools/colleges was conducted (N = 54). Forty-five completed questionnaires were returned, yielding 83 percent response. Six dental programs have adopted technical standards. The major skill areas addressed in each of these documents are communication; observation; intellectual-conceptual, integrative and quantitative; behavioral and emotional; and motor abilities. …

The purpose of this study was to investigate gender equity in medical coverage and injury prevention of basketball programs at NCAA Division I colleges and universities. The population for this study were 100 randomly sampled head athletic trainers at NCAA Division I colleges and universities that compete in both men's and women's basketball. Each participant was mailed a survey packet containing a cover letter and a questionnaire. The total return rate was 69%. The data were analyzed using the Statistical Analysis Systems. Currently, the Acquired Immune Deficiency Syndrome (AIDS) epidemic is world-wide and threatens the entire human population. The causative agent of AIDS is Human Immunodeficiency Virus (HIV), a retrovirus. The HIV viral population exhibits tremendous variation, making the development of vaccines difficult and anti-viral drug therapy ineffective due to the emergence of drug-resistant HIV strains. In this study, the Transit Automata (TA) model, a Cellular Automata (CA) based spatial model, has been developed. It includes a set of cells, states, rules, events, and a system clock. In contrast to CA, a TA model fragments the problem space along restricted routes. All cells on the routes are treated as equal and controlled by rules. Like CA models, an initial configuration of a TA model can be embedded and can be dynamically changed during the runtime. For a number of years, it was assumed that the protection measured during a laboratory fit test was indicative of the protection provided by the respirator during actual use in the workplace. Beginning in the early 1980's, workplace protection factor studies were conducted to validate this assumption. These studies indicated that quantitative fit test results did not correlate with actual workplace protection factors. A fit test that indicates the degree of exposure is especially important when Mycobacterium tuberculosis (TB) exposures are involved. Human exposure to silica is of great occupational concern as it is marked by pulmonary inflammation and fibrosis. The present work was performed to investigate, characterize, and modulate the responses that occur as a result of silica exposure. The intratracheal instillation of silica to rats results in pulmonary fibrosis but the cellular processes involved in the development of the chronic disease have not been well characterized. To examine the relation between the intensity of systemic bodily events and content-specific fear conditioning to stimuli that are fear-relevant or prepared for such associations, the present study involved a direct comparison between the conditioning effects produced by 20% and 13% CO{dollar}\\sb2{dollar}-enriched air (UCSs) to three CS types: external fear-relevant (snake moving toward viewer), internal fear-irrelevant (human heart beating arrhythmically), or neutral control (flowers swaying in the wind). Selenium is a nutritionally essential trace element for mammals, and several studies have suggested that selenium may have an important function in the central nervous system. We hypothesized that selenium may play an important role in the differentiation of myelin producing cells, i.e., oligodendrocytes in developing brain. We employed an in vitro model system of glial cell cultures prepared from newborn rat brains to test this hypothesis. The objectives of this study were to determine in humans the relationship between sex chromosome loss and increased micronucleus frequencies with age, the establishment of sex chromosome loss frequencies unbiased by cellular survival factors, the involvement of kinetochore proteins in sex chromosome loss and the effect of smoking. The purposes of this study were to gain an understanding of the Gypsy culture and to discover the relationship, within the school domain, between the Gypsy children and the non-Gypsy teachers. The long-term filtration behavior of six nonwoven geotextiles with fly ash from two different power plants was investigated. Five specimen preparation techniques were evaluated: Dry Method, ASTM Method (D 5101-90), Modified ASTM Method, Slurry Method, and Vibro-Prep. ASTM Procedure was modified. A new strategy in devising two-terminal voltammetric sensors is described. In our approach, such detectors are obtained by using ionic forces to confine two different redox groups to the same surface. The one is analyte-sensitive, and acts as the sensing site, while the other is analyte-insensitive, and thus operates as the reference site. The layer-by-layer deposition technique enabled us to attach multilayer systems to the surface of a gold electrode. To these multilayers various redox couples can be bound by ionic forces. Competition and convection effects in reaction-diffusion fronts and controlling chaos are studied. In Chapter 3 we report theoretical studies of competition in which two self-replicating organic molecules compete for a common reactant. We found that the self-replicating organic molecule can give sustained reaction-diffusion fronts. This phenomenon is investigated by numerical solutions of partial differential equations that couple local nonlinear kinetics and diffusion. Reaction-diffusion fronts involving competing species are then initiated and the concentration profiles of the autocatalysts monitored. The results show that the more reactive species totally predominates over the less reactive species as its front moves ahead into the …

The chicken egg yolk protein genes, vitellogenin (VTG) I, II, III, and the very low density apolipoprotein II (apoVLDLII), are highly expressed in hepatic parenchymal cells in an estrogen-dependent manner. Compared to the other egg yolk protein genes which respond in approximately 2-4 hours after the primary administration of estrogen, the VTGI does not respond for about 20 hours. The mechanism of activation of the egg yolk protein genes is not completely known. Inhibition of the chitin biosynthetic enzyme, glucosamine synthase (EC.2.6.1.16, L-glutamine:D-Fructose-6-phosphate amidotransferase) has been shown to be a promising approach to the development of antifungal agents targeting the cell wall. The best known inhibitors of glucosamine synthase possess epoxy-ketones and {dollar}\\alpha,\\beta{dollar} unsaturated carbonyls. In vivo, these inherently reactive moieties are easily deactivated by nucleophiles like glutathione. Hence we decided to develop inhibitors lacking such highly reactive groups. In the novels Lady Oracle, The Handmaid's Tale, and Cat's Eye, I locate and name the sites of memory to show how Atwood's protagonists appropriate them as their loci of resistance. The localized sites though marginalized are within the realm of representation; however, the "space-offs," a term I borrow from film theory, are beyond "the ideological." In these sites women create and/or validate counter-discourses that enable them to set up "politics of oppositionality" and provide them with a distance necessary to question the hegemonic. The purpose of this study was to monitor physiological responses of children in grades 4 and 5 during incremental tests on slide board and bicycle exercise and to determine whether children enjoy the slide board as a fitness activity. Twenty healthy subjects (10M, 10F; 10.1 {dollar}\\pm{dollar}.72 yrs) completed maximum continuous tests in 3 minute stages on the slide board, beginning at 40 slides/min, increasing by 10 slides/min each stage; and on the bicycle beginning at 25 watts, increasing by 25 watts each stage. Heart rate (HR), blood pressure, VO{dollar}\\sb2{dollar}, and rating of perceived exertion (CERT) were measured at each stage. …

The objective of this study was to determine the dominant mechanism (OH-radical scavenging or non-OH-radical scavenging mechanisms) of radioprotection by the thiol scavenger dithiothreitol (DTT) making comparisons between a cellular and isolated plasmid DNA model systems. The protection mechanism of DTT was determined experimentally by comparing the thiol to known OH-radical scavenging compounds (dimethyl sulfoxide, glycerol and 1,3-dimethyluracil) measuring the relative differences in their protection factor for radiation-induced chromosome aberrations in cells and single-strand break induction in the plasmid DNA model. This dissertation explores the Counter Propagation Network (CPN) to implement an image feature finder for automatic image matching systems. This research concludes that the CPN paradigm does not appear to be capable of extracting distinctive features for matching complex images such as fingerprints. There are three possible factors explored in the document that contribute to this inability to match. These are: (1) Image Enhancement - Most of fingerprint images have certain levels of localized blur or smear, locally low contrast, or other noise.
