Synthesis of 2-alkenyl and 2-arylbenzimidazoles, having a variety of functional groups, has been accomplished via a novel palladium-catalyzed reductive N-heteroannulation of 2-nitro- N-alkyl or N-benzyl benzenamines. The cyclization precursors are readily prepared from commercially available compounds by a variety of methods. In addition, the first total synthesis of ht-13-A has been studied. The key steps of the synthesis are a Mitsunobu reaction, an intramolecular Heck reaction, and a reductive N-heteroannulation.

The Enhancer of split Complex (E(spl)C) in Drosophila melanogaster encodes seven bHLH repressors that serve as the final effectors of the highly conserved Notch signaling pathway during the process of cell fate specification. These E(spl) proteins have been considered to be functionally redundant because of their structural similarity, and the observation that over-expression of individual E(spl) members leads to neural hypoplasia, in general. And additional compounding factor is that loss-of function alleles in individual E(spl) members have been unavailable. However, individual E(spl) members exhibit inordinate conservation through Drosophila evolution and are differentially expressed, findings which challenge the premise of functional …

It is increasingly becoming apparent that dynamic control of phosphorylation and dephosphorylation underlies the proper spatial and temporal activities of many signaling circuits including Notch (N). Emerging evidence suggests that protein kinase CK2 mediates the effects of N during Drosophila eye and bristle development via phosphorylation of E(spl) repressors. Previous studies in our laboratory shown that reducing levels/activity of CK2 elicits rough eyes due to excess R8 founders and ectopic and split bristles due to excess SOP's, both indicative of impaired lateral inhibition and N signaling. Ultrasound extraction was used to compare the accelerated release rates of arsenic and selenium from pulverized rocks associated with mountain-top removal/valley fill method of coal mining. The kinetic studies demonstrated that the release rates are dependent on rock lithology as well as the total concentrations found in the rocks. Se (IV) was found to be stable under ultrasonic irradiation (200 W/cm2) but As (III) was quickly oxidized to As (V). Rocks such as BT700 were found to have more Se (IV) compared to Se (VI) and others such as BT571 were found to contain more Se (VI) than Se (IV).;Continuous …

According to the United States Environmental Protections Agency (USEPA), nearly one in four households in the United States depends on an individual septic system (commonly referred as an onsite system or a decentralized wastewater system) to treat and disperse wastewater. More than half of these systems are over 30 years old, and surveys indicate at least 10 to 20% might not be functioning properly. Cold shock domain proteins, or CSPs, are among the most ancient and conserved nucleic acid binding proteins. They have been well characterized in prokaryotic systems but little is known about their function in plants. The overall objective of this study was to understand the evolution and function of the cold shock domain in plant model systems. To pinpoint conserved areas within CSD an alignment was made of the cold shock domain protein sequences based on nucleotide sequence information found within EST databases. One alternative is hydrogen, which has the potential to replace conventional fuels such as gasoline and diesel in internal combustion engines. However, a key enabling factor for hydrogen to be become a viable alternative is the development of lubricating oils specifically formulated for hydrogen-fueled engines. Exchange Of The Feminine: Representations Of Multiple Marginality In Contemporary West Virginia Fiction., David E. Wilson

This study engages the short stories and novels of three contemporary fiction writers: acclaimed native West Virginians Denise Giardina and Breece D’J Pancake, and Laura Albert, a New York City native who, for a time, published under the pseudonym JT LeRoy. Using West Virginia as their settings, these authors have created Mountain State characters whose stratified marginality in heteronormative Appalachian society stems from their failure to meet traditional expectations of gender and sexuality. Those multiply marginal characters in West Virginia fiction who best establish a general accord do so through benign public personae, ancillary labor occupations and supportive kinship roles. …

Previous studies have shown that administration of fibroblast growth factor-19 (FGF-19) reverses diabetes, hepatic steatosis, hyperlipidemia, and adipose accretion in animal models of obesity. To investigate the mechanism for this effect, we determined whether FGF-19 modulated hepatic fatty acid synthesis, a key process controlling glucose tolerance and triacylglycerol accumulation in liver, blood, and adipose tissue. Incubating primary hepatocyte cultures with recombinant FGF-19 suppressed the ability of insulin to stimulate fatty acid synthesis. This effect was associated with a reduction in the expression of lipogenic enzymes. FGF-19 also suppressed the insulin-induced expression of sterol regulatory element-binding protein-1c (SREBP-1c), a key transcriptional …

In early spring, areas of soil directly under baylage, spread for winter consumption by steers, is void of vegetation even after reseeding. This study was conducted to determine if: 1) residual baylage leachate is allelopathic to orchardgrass and red clover, 2) soil microorganisms have an effect on the toxicity of the leachate, and 3) toxicity decreases over time. Petri dishes, as well as potted soil/sand mixtures, were used to test for allelopathy. Leachate reduced seed germination and seedling length as compared to distilled water. There is evidence that soil microorganisms did not affect leachate toxicity. Toxicity of leachate to both …

Forested ecosystems are the largest terrestrial sinks for carbon dioxide (CO2) on earth and are important in mitigating the negative effects of human induced increases of atmospheric CO2. Trees fix CO2 into sugars by using the nitrogen demanding processes of photosynthesis. Modeling and empirical studies have shown stimulations in photosynthesis under elevated CO2 conditions but the response decreases as plant demand for N increases. Caveolin-1, the protein coat of caveolae membrane invaginations, was originally hypothesized to function solely as a tumor suppressor in human cancers. However, since the early 1990s, caveolin-1 has been found to be up-regulated in a number of metastatic cancers. This expression has been shown to correlate with disease progression and decreases in overall patient survival in some tumor types. The actin filament associated protein of 110 kDA (AFAP-110) is a well-characterized protein composed of amino-terminal binding motifs that function upstream of a carboxy-terminal actin binding domain. In vitro studies demonstrate that the amino-terminal pleckstrin homology 1 (PH1) domain interacts with activated PKCα, relaying signals that activate Src family kinases via SH3 and SH2 binding. Serving as an adaptor protein, AFAP-110 relays signals that result in the assembly of motility structures. These findings could be of physiological as well pathological significance as AFAP-110 colocalizes with SFKs in the developing brain as well as in the glial-derived tumor, glioblastoma multiforme. Over the years, azaindoles and diazaindoles have generated an increased interest due to their structural similarities with indoles and their potential as pharmacologically active compounds. A mild and efficient methodology has been developed for the synthesis of azaindoles and diazaindoles. This novel procedure includes a palladium-catalyzed reductive N-heteroannulation as the key step. This methodology was successfully applied to the synthesis of 4-azaindole, 2-phenyl-5-azaindole, 2-phenyl-6-azaindole, and 7-azaindole. In addition, this method was efficiently utilized to synthesize 2-phenyl-4,6-diazaindole and 2-phenyl-4,5-diazaindole. One approach to this issue has focused on genomic and proteomic profiling. While previous work has focused on genomic profiling, few studies have explored the correlation of proteomic profiles with drug sensitivity. Early pregnant ewes, 5/6 day post coitus (dpc) were used as a model to study early embryonic loss via gram-positive bacterial infections, such as mastitis. Ewes 5/6 dpc were injected with a gram-positive bacteria cell wall component, peptidoglycan-polysaccharide (PG-PS), to induce an innate immune system acute phase response (APR). The induction of the APR and its reactant molecules, such as cytokines and acute phase proteins (APP), were initiated to study their effects on embryonic loss. This research presents experimental results for the application of Machine Vision (MV) techniques to address the problem of target detection and tracking. The main objective is the design of a prototype "UAV" surveillance environment to emulate real-life conditions. The model environment for this experiment consists of a target simulated by a small electric train system, located at "ground" level, and a MV camera mounted on a motion-based apparatus located directly above the model setup. Cyclobutenediones have proven useful as starting materials for synthesis of highly functionalized biologically relevant molecules. The rearrangement of cyclobutendiones has a wide synthetic scope and can be utilized as a key step in the construction of quinones, benzo[o]isoxazoles, and 3-methylenoxoindoles having a variety of substitution patterns. Construction of these molecules involves thermolysis of 4-hydroxy-2,4-di-aryl-2-cyclobutenones derived from squaric acid ester. In each of the cases the mechanism proceeds through an initial ring opening of the cyclobutene to afford a ketene intermediate that subsequently undergoes a ring closure with various intermolecular nucleophiles. Diel fluctuations of pH, specific conductance and Fe species have been documented in Dillan Creek, a small coal mine-impacted tributary to Deckers Creek, WV. Samples were collected hourly over 16-20 hour periods and analyzed for dissolved Fe(II) and dissolved total Fe using a Hach DR2800 spectrophotometer. Fe(II) concentrations increased during the day and decreased at night during late winter sampling. Sampling conducted in the summer shows an opposite trend with Fe(II) increasing at night and decreasing during the day. Discrepancies between timing of cycles indicate a seasonal variation of controlling mechanisms. In winter, iron cycles are likely controlled by photoreduction …

Caspases are proteolytic enzymes involved in committing apoptosis, but through the studies of knockout phenotypes in mice and Drosophila , it has been speculated that caspases might possess additional non-apoptotic functions.;We have found non-apoptotic functions for caspase-8 in both normal and tumor cell lines. Specifically, we found that caspase-8 promotes cell migration, adhesion, and Rac activation. Subsequently, we also found that caspase-8 interacts with the p85 subunit of PI3K. Accompanying stimulation of motility with epidermal growth factor the phosphorylation of caspase-8 on tyrosine-380 is induced and this phosphorylation allows for p85 interaction, cell migration, adhesion, and Rac activation. The purpose of the first study was to determine how age and ovarian hormones affect flow-induced vasodilation in the coronary microcirculation. Coronary arterioles were isolated from young, middle-aged, and old control, ovariectomized (OVX) and ovariectomized + estrogen replaced (OVE) female Fischer-344 rats to assess vasodilation to increases in flow. L-NAME, a nitric oxide synthase (NOS) inhibitor, was used to assess NO contribution to flow-induced dilation (FID). FID of coronary arterioles was impaired with advancing age. Ovariectomy reduced FID in young females only; however, estrogen-replacement restored or improved FID in all age groups. FID was reduced after pretreatment with L-NAME in …

Variable recovery of hypovirulent (HV) and virulent (V) isolates from repeatedly sampled, identical chestnut blight cankers, caused by Cryphonectria parasitica, prompted this study to monitor more closely the isolate recovery over time to determine hypovirulent or virulent status. To meet the objective, laboratory and field experiments were conducted between 1998 and 2000 to recover isolates from cultures and cankers challenged with HV inoculum. Cultural studies demonstrated (1) hypovirus transfer readily occurred to actively growing colony margins, and (2) that established, virulent mycelium one-to-six weeks-old at time was unable to acquire hypovirus. Non-specific gamma-secretase inhibitors are not ideal for intervention of familial Alzheimer's disease. The four essential include Presenilin (PS1 or PS-2), APH-1, Pen-2, nicastrin (Nct). Prevalence rates of non-suicidal self-injury among college students ranges from 17% to 38%. Research indicates that individuals with borderline personality disorder (BPD) who self-injure sometimes report an absence of pain during self-injury. Furthermore, self-injury in the absence of pain has been associated with more frequent suicide attempts. The present study examined pain thresholds and tolerance among 44 college students (11 self-injurious and 33 non-self-injurious). Pain thresholds and tolerance were measured using an algometer pressure device which has been used to produce pain previous laboratory research. Self-injurious participants had higher pain tolerances than those who do not engage in self-injury. In …

The goals of my research project were to clone the E(var)3-5 gene and characterize the lethality of E(var)3-5 mutations. The E(var)3-5 gene was identified by dominant mutations that enhanced PEV of wm4, thus implicating the gene product in promoting a euchromatic chromatin structure (Dorn et al. 1993). These mutations were found to be recessive lethal as well and the lethal phenotype was used to map the E(var)3-5 gene (Dorn et al. 1993; this work). I characterized the lethality of E(var)3-5 mutations and found that lethality in E(var)3-5 hemizygotes is manifested at the pupal stage. I mapped the lethality to a …

Current face recognition systems capture faces of cooperative individuals in controlled environment as part of the face recognition process. It is therefore possible to control lighting, pose, background, and quality of images. However, in a real world application, we have to deal with both ideal and imperfect data. Performance of current face recognition systems is affected for such non-ideal and challenging cases. This research focuses on designing algorithms to mitigate the effect of covariates in face recognition.;To address the challenge of facial aging, an age transformation algorithm is proposed that registers two face images and minimizes the aging variations. Little is known about the slow-to-warm-up infant temperament. The present study evaluated the usefulness of this temperament as conceptualized by Thomas and Chess in predicting child and maternal parenting behaviors, with a particular focus on its conceptual link to child inhibition. Participants were 1,072 mothers and their children in the NICHD Study of Early Child Care. Slow-to-warm-up temperament in infancy did predict later inhibition, p = .000, but not as well as the difficult temperament or the individual temperament subscales of mood and approach. Sphingosine 1-phosphate (S1P), a biologically active lipid, induces a myriad of cellular events including enhancement of the endothelial barrier and assembly of adherens junction proteins, VE-cadherin and catenins. The over all goal in this set of studies was to investigate what cellular mechanisms contribute to S1P-induced enhancement of the vascular endothelial barrier.;Since VE-cadherin contributes to the stabilization of the endothelial barrier, I determined in the first study if the rapid, barrier-enhancing activity of S1P requires VE-cadherin. This thesis involved two components: identification of the parasite Anguillicola crassus in American eels, Anguilla rostrata and PIT tag retention in eels in a laboratory setting. The recent spread of the swim bladder nematode parasite, Anguillicola crassus, in American eels has caused concern among biologists and fishery managers. A total of 244 yellow-phase American eels were collected at the Millville Dam eel ladder on the Shenandoah River, WV. Swim bladders were removed and examined for the presence of the nematode parasite. The number of parasites in each eel was recorded, and prevalence, intensity, and mean intensity were calculated.
