Individuals at risk for developing Alzheimer's disease (AD) often exhibit neuronal network hyperexcitability, particularly in the CA3 region of the hippocampus, in the years preceding AD diagnosis, suggesting that aberrant network activity might contribute to the pathogenesis of AD. Recently, the microtubule-binding protein tau has been implicated in this hyperexcitability. Removal of tau in AD models overexpressing amyloid-beta (Abeta) or non-AD models of epilepsy decreases hyperexcitability and normalizes the excitation/inhibition imbalance. The exact mechanism by which tau produces hyperexcitability remains to be determined, but recent work suggests tau may mediate glutamatergic signaling. The 'click chemistry' which provides 1,4-disubstituted 1,2,3-triazoles has been reported in 2001. New method for highly funtionalized 1,2,3-triazole synthesis becomes very desirable along with the discovery of unique properties of this heterocyclic structure. Our group focused on the synthesis of functional 1,2,3-triazole for several years and new methodologies have been developed such as arylation, vinylation, propargylation and allenation. Shackelford Fluorescence in-situ hybridization (FISH) is a useful molecular cytogenetics technique for counting chromosomes and identifying specific chromosomal sequences of interest. FISH probe targets include centromeres, single loci, subtelomeres, and telomeres, using DNA or peptide nucleic acid (PNA) probes. FISH probes were used to determine chromosome number, copy loss or gain, and signal size in four studies involving acquired chromosomal changes in malignancy, mosaicism, and aneuploidy in pre- and postnatal constitutional abnormalities. The Arctic became a hot spot on the global map and discussion over the cold waters became an important issue in international relations. During the Cold War, as it is today, the Arctic was a separate area of rivalry between the superpowers. Despite the fact that today's debate is based on different issues, including sovereignty over the region, potential access to natural resources and the status of international waters, there is still a strong military component to the competing interests, as there was during the Cold War.

The purpose of this dissertation was twofold. The first purpose was to develop a valid and reliable measurement of health literacy appropriate for use in social science. The second purpose was to determine whether health literacy is a skill set that can be increased through the intervention efforts of communication studies scholars. With the loss of species worldwide due to anthropogenic factors, especially in forested ecosystems, it has become more urgent than ever to understand the biodiversity-ecosystem functioning relationship (BEFR). BEFR research in forested ecosystems is very limited and thus studies that incorporate greater geographic coverage and structural complexity are much needed. Here, I compiled ground-measured data from 436,177 Forest Inventory and Analysis (FIA) plots of every U.S. state except Hawaii to map current basal area, site productivity, and tree species diversity across the United States. Recent developments in analytical instrumentation have allowed scientists to investigate systems that were unattainable in the past. In the biomedical community this increase in investigative power has allowed the exploration of biological components like proteins, lipids, and DNA and their role in the many different cycles necessary for human life. By understanding the nuances of these processes, medical professionals can more easily recognize the products of mutations or malfunctions that can lead to more accurate prognosis and diagnosis of disease. Introduction: The unilateral posterior crossbite with a functional shift is one of the most common early adolescent malocclusions. It is caused by a transversely deficient maxilla, relative to the mandible, and results when the mandible shifts to one side to so the teeth can maximally interdigitate. This shift is thought to cause the contralateral condyle to move anteriorly, inferiorly, and medially within the TMJ. The three opioid receptor subtypes, mu (mu), delta (delta) and kappa (kappa) have long been associated with analgesia. Traditional opioid analgesics exert their effects through mu receptors located in the CNS. Recent studies suggest that the development of opioid analgesics displaying dual properties of mu agonism and delta antagonism could be of benefit by retaining potent analgesic properties while reducing the development of tolerance with chronic administration. Alzheimer's disease (AD) accounts for 60 to 80% of all dementias, affects over 5 million Americans, and incurs annual healthcare costs of over {dollar}200 billion (Alzheimer's Association, 2012). Hyperphosphorylation of tau, the main component of neurofibrillary tangles observed in AD, leads to deficits in cell structure and function. Many temperate forests in the Eastern US are young and have experienced decades of elevated inputs of acidic compounds from the atmosphere. As a result, I used the Fernow Experimental Forest Long Term Soil Productivity (LTSP) experiment near Parsons, WV to determine how ongoing acidic deposition may affect total ecosystem carbon (C) storage, stand level dynamics, and soil and foliar chemistry in a young regrowing temperate deciduous forest. The Transient Oscillatory Model (TOM) seeks to explain neural coding in the first processing center of the invertebrate olfactory system - the antennal lobe - by proposing that activated primary output neurons participate in the encoding ensemble only transiently, based on the coincidence of their spiking relative to local field potential (LFP) oscillation phase. Hydrogen-deuterium exchange (HDX) coupled with electrospray ionization (ESI) in mass spectrometry (MS) has become an important method in the study of protein structure and dynamics. It provides an opportunity for obtaining conformational structures of protein motions in physiological conditions and offers unparalleled limits of detection. This method relies on the fact that the accessibility of exchangeable amide hydrogens of a protein to D2O induces H/D isotopic exchange. This dissertation documents the development of a flipped classroom teaching model for teaching college algebra. The model supports the use of multiple teaching strategies including video lectures for students to view as homework, outside of class. A design and development framework is used to describe the design decisions, model implementation, and evaluation of the model across three deliveries, or case studies, of a college algebra course from fall of 2012 through fall of 2013. This dissertation examines the eroticization of female children in film during the 1930s as a mechanism for concealing troubling realities of the Great Depression in the United States. The repressed sexuality embedded in the plots of Post Hays Code movies featuring Shirley Temple and characters such as Stella Dallas, Dorothy Gale, and Scarlett O'Hara serve to suture over a crisis of masculinity triggered by changing roles in gender, race, and class. Early childhood externalizing behaviors are a known risk factor for future problem behaviors (e.g., poor achievement, delinquency). The present study seeks to illuminate the pathway of early childhood externalizing behaviors to five adolescent delinquency types (i.e., violent offenses, property offenses, illicit drug use, licit drug use, and minor offenses), in addition to overall delinquency. Study data came from two waves of the Children of the National Longitudinal Survey of Youth-1979 (CNLSY-79; N = 855, 52.4% male, 24.1% Hispanic, 36.6% Black, and 39.1% white). Boys engaged in higher levels of violent and property crimes, and black youth were less likely to …

Past research indicates that men tend to be more financially risk tolerant than women, and declines in self-reported risk tolerance are common with age. A life-span sample of adult men and women were recruited to investigate these gender- and age-related patterns using an ecologically valid measure of financial risk tolerance. As previously found, men reported greater risk tolerance than women, and risk tolerance declined with age. Biological mechanisms, such as testosterone, are often implicated in men's tendency to be more financially risk tolerant than women. Despite the historic declines in the prevalence of cigarette smoking in the United States, tobacco use remains a major public health burden. The persistent use and susceptibility of tobacco arises from tobacco-related health disparities, which historically occur among vulnerable and underserved populations. Since rural/nonmetropolitan populations suffer disproportionately more tobacco-related health disparities than urban/metropolitan populations, the main goal of this project and its three component studies was to address rural tobacco-related health disparities. Microfluidic technologies have enabled in vitro studies to mimic in vivo microvessel environment with sufficient complexity. However, there are still existing knowledge gaps and lack of convincing evidence to demonstrate and quantify key features of a functional microvessel. In this dissertation, a physiologically realistic microvessel model was developed with a stable and mature endothelium for studying complex vascular phenomena, such as endothelial cell signaling and barrier functions with the microscopic resolution at individual cellular levels. Solid-state NMR spectroscopy is a powerful technique to study the structure of biological and inorganic systems, especially non-crystalline and insoluble samples that are difficult to study for X-ray diffraction and solution NMR spectroscopy methods. We used solid-state NMR spectroscopy to elucidate the structure and interactions of tri-peptides on gold nanoparticles. Tri-peptides CysAlaAla (CAA) and AlaAlaCys (AAC) are the focus of this work. These peptides are incorporated on gold surfaces through thiol-gold interactions and their structures on gold surfaces are studied using 1H and 13C solid-state NMR spectroscopy. We have discovered a practical general method for the preparation of diverse 2-halomethylene-4-cyclopentene-1,3-diones and/or 2-halo-1,4-benzoquinones. An in-depth study of the reactions of 4-(ethynyl)-4-hydroxy-2,3-substituted-2-cyclobuten-1-ones with N-bromo- or N-iodosuccinimide is described. This reaction was used in a short total synthesis of methyl linderone. Also, a series of facile, palladium catalyzed, direct, and regiospecific Wacker-type oxidations has been developed to yield various alpha-arylated ketones from aryl-substituted internal alkenes.

Parent-Child Interaction Therapy (PCIT) is a "probably efficacious" treatment for children with disruptive behavior disorders (Eyberg, Nelson, & Boggs, 2008). However, many of the characteristics of the efficacy studies (e.g., therapy conducted in university-based clinics, graduate student therapists, supervision and feedback on integrity to the model, recruited families, homogeneous samples) supporting PCIT may limit the generalization of the results to the community setting where most families receive treatment (e.g., Weisz, Jensen-Doss, & Hawley, 2006).
