On The Roles Of Drosophila Notch Signaling And Atonal Autoregulation In The Specification Of R8 Photoreceptors, Adam T. Majot

Drosophila retinogenesis has long served as a paradigm of tissue patterning. Notch, an anciently conserved signaling pathway, is critical to both the formation of and proper spacing of founding R8 photoreceptors. Notch is intimately interwoven into the activity of Atonal (Ato), a proneural basic helix-loop-helix (bHLH) that is required for R8 specification. Ato is expressed through dual enhancers: ato-3' and 5'-ato. ato-3' is activated in response to Notch-independent signaling cues whereas 5'-ato exhibits complex dependence on Notch signaling. First, Notch is required for 5'-ato activation, eliciting formation of Ato intermediate groups (IGs, evenly spaced clusters of 10--20 cells). Subsequently, Notch …

Acid mine drainage (AMD) is an environmental concern that is well known and studied in West Virginia and other mining regions. This pollution threat can be prevented and mitigated with current mining permitting regulations; however some mine waters require treatment, which generally consists of the neutralization of affected waters, resulting in the precipitation of characteristic 'floc' material. Functional assessment of created wetlands is an important factor in monitoring the success of wetland mitigation projects. Determining the ability of a created wetland to replace lost wildlife habitat and to support productive wildlife populations should be a priority in the assessment of created wetland success. In response to criticism of traditional forensic methods of human head hair examination, the goal of this study was to develop and evaluate an objective method using texture-based image analysis. 120 individuals, including 20 from six different maternal groups, were sampled and their hairs evaluated using the developed method. Different variables were evaluated to develop the most efficient method to increase intra/inter ratio, as well as the percentage of well-classified hairs. Cerium dioxide nanoparticles (CeO2 NP), an engineered nanomaterial, have great potential from a consumer and therapeutic perspective. Currently, CeO2 NP are added to diesel fuel to decrease the soot emissions commonly associated with diesel engines. CeO2 NP also have therapeutic applications (e.g. improve outcomes following stroke and radiation treatments) due to their anti-oxidant capabilities. Rheumatoid arthritis (RA) is the most prevalent chronic inflammatory joint disease, affecting approximately 1% of the world population. This autoimmune disease is characterized by pain, stiffness, swelling, and breakdown of cartilage in synovial joints. Current RA analgesic treatments (i.e., pain reducing drugs) are ineffective or induce negative side effects, and so there are many concerted efforts to discover new RA treatments. Depression affects more than 350 million people and is the leading cause of disability worldwide. Major challenges in the management of depression remain the 4-12 weeks of delayed efficacy and 30% of patients who are resistant to the therapeutic effects of current pharmaceutical treatments. In an aqueous solution the phospholipids dimyristoyl-sn-glycero-3- phosphocholine (DMPC) and 1,2-dihexanoyl-sn-glycero-3-phosphocholine (DHPC) self-assemble to form thermo-responsive non-Newtonian fluids (i.e., pseudo-gels) in which small temperature changes of 5-6 °C decrease viscosity dramatically. This characteristic is useful for sieving-based electrophoretic separations (e.g., of DNA), as the high viscosity of linear sieving additives, such as linear polyacrylamide or polyethylene oxide, hinders the introduction and replacement of the sieving agent in microscale channels. Gonadal development is a dominant physiological process that alters growth and fat metabolism, thereby reducing fillet quality. Sterile, triploid (3N) female rainbow trout, generated by pressure-induced shock of zygotes 30 minutes after fertilization (3NP), are used to eliminate reduced growth and quality associated with gonadogenesis. However, this method does not produce 100% 3N offspring. Crossing a diploid (2N; fertile) female and a tetraploid (4N; fertile) male produces triploid offspring 100% of the time (3NC; sterile). The finely orchestrated development and maturation of the oocytes has been the focus of many studies in which oocyte-specific genes essential for folliculogenesis and early embryonic development have been identified. Studies using knockout mice models of Nobox, Sohlh1, Sohlh2, Figla and Lhx8 have revealed that these oocyte specific transcriptional factors form networks in which they regulate the expression of each other and downstream targets. A mixed-integer programming model was developed to optimize forest carbon sequestration considering carbon price, biomass price, harvest area restriction, and harvest method. The model was applied to examine the harvest scheduling strategies and carbon sequestration in a mixed central Appalachian hardwood forest. Sensitivity analyses were conducted over a range of carbon and biomass to timber price ratios, harvest area limitations and harvest methods. The results showed that the carbon sequestration rate of the central Appalachian hardwood forests could gradually increase as the carbon to timber price ratio changed from 0.0 to 1.0 with an average sequestration rate of 0.917 Mg·ha …

Multiple PDEs have been shown to play vital roles in the central nervous system, with involvement in mood, reward mechanisms, and learning and memory. There are several methods that are currently used to estimate a postmortem interval (PMI), including physiological, biochemical, entomological, and archeological approaches. However, these methods are not effective in estimating PMI's extending from several weeks to months. The study presented here was performed to create an independent, objective biological method to estimate PMI by analyzing the degradation of 18S ribosomal RNA (rRNA) from porcine and human dental pulp. They play important roles in cellular and developmental processes and are present in various species. Arbuscular mycorrhizal fungi (AMF) in the phylum Glomeromycota are globally distributed biotrophic fungi that form symbiotic associations with a majority of terrestrial plants. Their global beneficial role in plant and soil ecology warrants a better understanding of their evolutionary history and taxonomic relationships. This project focused on species relationships in two deeply rooted families Ambisporaceae and Archaeosporaceae. Little research has examined short-term change in self-assessed health (SAH), limiting what is known about the influence of daily life on these personal health evaluations. Investigating short-term change in SAH can add to our understanding of the ways in which SAH relates to future health outcomes. Therefore, the current study sought to examine change and stability in SAH across the day and across the week in an age-diverse community sample (n = 57, ranging in age from 25 - 71 years, 52.6% female). Burn injuries are a serious health concern for youth. In particular, adolescents are at risk for sustaining burn injuries, with recent estimates suggesting that adolescents make up nearly 30% of the burn injury cases treated in emergency departments in the United States. Despite the prevalence of burn injuries in adolescents, little research has examined possible correlates of adolescent fire-risk behavior (e.g., using accelerants to start a fire). The state of West Virginia requires stream mitigation to offset anthropogenic impacts to streams; consequently there is a high concentration of mitigation projects in the mountaintop mining/valley fill region. Projects are typically outside mine boundaries and sites are selected at the discretion of the West Virginia Department of Environmental Protection. This dissertation evaluated current in-stream, natural channel design (NCD) structures and prioritized future project locations. First, we assessed the Little Coal River, which is one of the largest physical restoration projects in the state. The Little Coal drains 994 km2 with extensive historic and contemporary coal mining. Metal contaminated soils are a problem throughout the world. Because metals accumulate in the food chain they have been linked to adverse human and ecosystem health. One promising approach to remediating metal contaminated soils is phytoremediation but success has been limited by an incomplete understanding of the factors that control metal uptake by plants, including the effects when more than one contaminant metal is present in the system.
