Understanding The Mechanism Of Hard Metal (Wc-Co) Toxicity: In Vitro Studies And In Vivo Exploration, Andrea Lynn Armstead
Understanding The Mechanism Of Hard Metal (Wc-Co) Toxicity: In Vitro Studies And In Vivo Exploration, Andrea Lynn Armstead
Graduate Theses, Dissertations, and Problem Reports (ETD)
Hard metals, such as tungsten carbide cobalt (WC-Co), are frequently used for a number of industrial applications such as surface coatings for heavy machinery and tools. In particular, WC-Co coatings are prevalent in mining and drilling applications where extensive, repetitive use of these tools causes wear over time. In enclosed environments, WC-Co wear particles become airborne and present an occupational inhalation hazard. It is known that inhalation of WC-Co "dusts", composed of nano- and micro-sized WC-Co particles, contributes to the development of hard metal lung disease and increased risk for lung cancer; however, the relationship between acute WC-Co toxicity and …
Temporal, Spatial, And Environmental Influences On The Demographics And Harvest Vulnerability Of American Black Bears (Ursus Americanus ) In Urban Habitats In New Jersey, Pennsylvania And West Virginia, Andrew Tri
Graduate Theses, Dissertations, and Problem Reports (ETD)
To date, no research studies in the eastern United States have addressed the spatial ecology of black bears (Ursus americanus) in urban and suburban habitats, and there is limited information regarding black bear space use, habitat selection, and harvest vulnerability. I assessed the harvest vulnerability, home range size, and spatial ecology of black bears in New Jersey, Pennsylvania, and West Virginia in collaboration with the New Jersey Division of Fish and Wildlife, Pennsylvania Game Commission, and the West Virginia Division of Natural Resources. The major objectives of my study were to identify and quantitatively assess: (1) if black bears shift …
Altered Hemodynamic Control In The Skeletal Muscle Microcirculation In The Metabolic Syndrome: The Emergence Of A New Attractor, Joshua Thomas Butcher
Altered Hemodynamic Control In The Skeletal Muscle Microcirculation In The Metabolic Syndrome: The Emergence Of A New Attractor, Joshua Thomas Butcher
Graduate Theses, Dissertations, and Problem Reports (ETD)
Peripheral vascular disease is a pathological disease state whereby the peripheral vascular system becomes progressively limited in its ability to adequately perfuse extremities despite increases in metabolic demand. One of the primary risk factors for development of peripheral disease is the Metabolic Syndrome. It is the presentation of three simultaneous comorbidities, with obesity being the most common denominator in most metabolic syndrome patients, but also including a pro-inflammatory state, pro-oxidant state, pro-thrombotic state, increased blood pressure, atherogenic dyslipidemia, or insulin resistance. Currently, the population of the United States is approximately 75% overweight, obese, or extremely obese and the prevalence of …
Effects Of Estrogen In An Aged Animal Model Of Stroke, Rachel L. Leon
Effects Of Estrogen In An Aged Animal Model Of Stroke, Rachel L. Leon
Graduate Theses, Dissertations, and Problem Reports (ETD)
In 2000, approximately 10 million women were taking hormone replacement therapy (HRT) for alleviation of menopausal symptoms. A number of prior animal studies also suggested that HRT may be neuroprotective and cardioprotective as well. Although estrogen is neuroprotective in young adult animal models of stroke, clinical trials demonstrate that estrogen increases the incidence and severity of stroke in aged women. We have previously shown that experimental stroke pathophysiology differs between young adult and aged rats. In the first study, we show that aged rats treated with 17beta-estradiol had significantly increased infarct volumes compared to placebo-treated aged rats. Young adult rats …
Red-Headed Woodpecker Habitat Selection And Breeding Ecology On Fort Drum Military Installation, New York, Jacob L. Berl
Red-Headed Woodpecker Habitat Selection And Breeding Ecology On Fort Drum Military Installation, New York, Jacob L. Berl
Graduate Theses, Dissertations, and Problem Reports (ETD)
Red-headed woodpecker (Melanerpes erythrocephalus) populations have declined range-wide over the past 40 years, although the causal mechanisms implicated in the observed declines remain poorly understood. Population decline has been most severe at the periphery of their range, particularly in the northeastern United States, coinciding with a region where there is little information regarding red-headed woodpecker habitat use or demography. A detailed knowledge of habitat requirements and population dynamics is prerequisite for the effective conservation and management of imperiled wildlife populations, and this knowledge gap likely precludes informed management decisions for red-headed woodpeckers in the northeastern portion of their range. In …

Improving Opportunities For Small Flock Egg Production And Proper Evaluation Of Glucanase For Commercial Poultry, Angela Elsie Lamp
Graduate Theses, Dissertations, and Problem Reports (ETD)
Consumers of pastured hen eggs have justified paying an associated premium price because they perceive animal welfare, sustainability, and nutrition are enhanced compared to conventionally produced eggs. The objective of Study 1 (Chapter 2) was to implement practical management strategies to increase eicosapentaenoic acid (EPA) and docosahexaenoic acid ( DHA) composition of eggs (Experiment 1) and to determine if the same diet formulation would produce an enhanced EPA and DHA egg composition and effect hen health when pasture access and hen breed varied (Experiment 2). For Experiment 1, four dietary treatments were utilized: 1) Basal, 2) Basal + 0.5% Sardine …
Grazing Impacts On Sediment Production And Runoff From Single And Mixed Species Grazing, Brittany Parks
Grazing Impacts On Sediment Production And Runoff From Single And Mixed Species Grazing, Brittany Parks
Graduate Theses, Dissertations, and Problem Reports (ETD)
With growing human populations, we need to produce more food. Food production requires growing crops and raising livestock with existing and suitable land resources, potentially causing nutrient (phosphorus (P) and nitrogen (N)) pollution. For purposes of this study, the runoff potential for the mixed and single species grassland grazing will be examined. Mixed species grazing refers to cattle and sheep grazing concurrently in the same area; while single species grazing refers to cattle grazing alone. Understanding the environmental effects of mixed grazing as compared to a single species grazing systems will help to improve farming and agricultural practices that increase …
Emotion Regulation And Cardiovascular Response To Emotion Provocation: Reappraisal Versus Suppression, Casey E. Cavanagh
Emotion Regulation And Cardiovascular Response To Emotion Provocation: Reappraisal Versus Suppression, Casey E. Cavanagh
Graduate Theses, Dissertations, and Problem Reports (ETD)
Emotion regulation is a process through which individuals alter the subjective, behavioral and physiological responses to emotional experiences. Within the emotion regulation model, reappraisal and suppression are two strategies that have been studied extensively. However, this research has largely ignored the effects of individual differences in emotion regulation when examining reappraisal and suppression. The purpose of this study was to examine individual differences in reappraisal and suppression (e.g., one's preferred emotion regulation strategy) as well as examining the effects of instructional use of reappraisal and suppression. In the present study, participants, classified as habitual reappraisers and habitual suppressors, engaged in …
Microbial Genome Evolution Due To Multifaceted Symbiosis Within The Tsetse Fly (Diptera: Glossinidae), Anna Kathleen Snyder
Microbial Genome Evolution Due To Multifaceted Symbiosis Within The Tsetse Fly (Diptera: Glossinidae), Anna Kathleen Snyder
Graduate Theses, Dissertations, and Problem Reports (ETD)
Microbes are capable of rapid genetic modification, enabling the habitation of a wide field of niches, including forming interdependent associations with macroscopic hosts. While ancient multipartite mutualisms have been shown to involve metabolic complementation, little is known concerning the early genomic adaptations leading towards co-residence within a novel host. The overall objective of this research is to gain insight on genome evolution resulting from symbiosis, particularly by examining bacteria with varying levels of host dependency and times of establishment. The tsetse fly (Diptera: Glossinidae) serves as a relatively simple model system to investigate evolutionary aspects of symbiosis, while also maintaining …

Stressors And Coping Behaviors Of Female Peer Leaders Participating In College Club Sports, Leigh A. Bryant
Graduate Theses, Dissertations, and Problem Reports (ETD)
Peer leadership positions on collegiate club sport teams can be valuable opportunities for students to develop leadership skills, maintain healthy habits, create friendships, and foster organizational and personal connections across the university setting. Nevertheless, involvement in club sports can create stress over and above the many stressors that college students may encounter during their undergraduate experience. This study focused on exploring female leaders' perceived stressors inside and outside of the sport setting, and the ways in which they coped with them. Participants ( N = 7) were interviewed based on nominations from their respective teammates. Interviews were conducted using a …
Development Of A 16s Rrna Gene Sequence As A Biomarker Specific To The Gastrointestinal Tract And Feces Of Swine Based On Suppressive Subtractive Hybridization, Autumn Sayre
Graduate Theses, Dissertations, and Problem Reports (ETD)
Fecal contamination of water bodies causes environmental issues and human health risks. Swine are a major contributor of fecal inputs to environmental waters due to their importance in American agriculture. Water quality currently is regulated using fecal coliforms, which have natural reservoirs in the environment and may not correlate with pathogens. In recent years there has been a shift towards microbial source tracking (MST). MST employs microbiological, genotypic, phenotypic and chemical methods to determine the source of fecal pollution in water bodies. Previous studies aimed at developing swine fecal biomarkers have had limited success. The overall objective of this research …

Influence Of Weight Loss On Metabolic And Mitochondrial Function In The Mature Horse, Jennie L. Zambito
Influence Of Weight Loss On Metabolic And Mitochondrial Function In The Mature Horse, Jennie L. Zambito
Graduate Theses, Dissertations, and Problem Reports (ETD)
Obesity causes a multitude of metabolic issues in the horse, yet stepwise alterations in glucose and lipid metabolism, mitochondrial capacity and oxidant status during weight loss have yet to be analyzed. Skeletal muscle contains subsarcolemmal (SSM) and interfibrillar (IFM) mitochondria, which respond differently to physiological stimuli, impacting tissue and whole body oxidant status. We hypothesized that 8 light-type horses would display improvements in skeletal muscle mitochondrial subpopulation function and decreases in circulating oxidant status markers during weight loss from an obese (7 to 8) to moderate (5) body condition score (BCS). Additionally, we hypothesized improvements in morphometric measurements, circulating metabolic …

Exploring The Mechanism Of Shp2 And Egfr/Her2 Cooperation In Breast Cancer Cell Signaling, Zachary Raymond Hartman
Graduate Theses, Dissertations, and Problem Reports (ETD)
The Src homology 2-containing protein tyrosine phosphatase 2 (SHP2) has been established as a critical mediator of cancer-related cell signaling downstream of receptor tyrosine kinases like EGFR and HER2. As such, targeting of SHP2 is being recognized as a potentially viable therapeutic option in HER2-positive and triple-negative breast cancers. In this report, I employed molecular and cellular biology techniques to further elucidate the role of SHP2 downstream of receptor tyrosine kinases in breast cancer cell lines. In this way, I was able to contribute knowledge to how SHP2 controls triple-negative cell motility. In addition, the mechanism of SHP2 control of …

Firewood is the oldest source of energy for cooking and home heating and remains a primary heating source for half of the world's population, yet the industry remains relatively undocumented and immature both in developing and technologically advanced countries. Firewood is an environmentally friendly and renewable resource that is becoming popular once again in U.S. homes, 2.5 million of which utilize it as a primary heating source. However, firewood is also a proven vector for invasive insects and pathogens that are threatening the health of U.S. forests. In order to address the increasing trend of firewood use and prevent further …
Adjustment Of Initial- And Terminal-Link Response Patterns To Suddenly Equivalent Terminal Links, Shrinidhi Subramaniam
Adjustment Of Initial- And Terminal-Link Response Patterns To Suddenly Equivalent Terminal Links, Shrinidhi Subramaniam
Graduate Theses, Dissertations, and Problem Reports (ETD)
Three pigeons were trained on concurrent-chain schedules. Concurrent variable-interval initial links produced fixed-interval (FI) terminal links. Occasional `no-food' terminal links were interspersed pseudorandomly to obtain stop times, a single-trial measure of temporal discrimination. In rapid-acquisition conditions, terminal links were FI 10 s and FI 20 s. Across sessions, the location of the initial link leading to the shorter terminal link varied pseudorandomly. Both terminal links were FI 15 s in a uniform condition. In the rapid-acquisition conditions, initial-link response allocation tracked relative terminal-link immediacy and stop times tracked terminal-link FIs. Initial- and terminal-link pecking stabilized within the first half of …

Liquid Chromatography -- Tandem Mass Spectrometry Techniques For Drugs Of Abuse As Applied To Bacterial And Forensic Toxicology, Stephanie Marie Martindale
Graduate Theses, Dissertations, and Problem Reports (ETD)
The ability of bacterial species to transform complex chemical substances has been well documented. Some of these species are native to the human gastrointestinal tract and play an active role in the postmortem decomposition process. These species have potential to cause biotransformations that affect compound-to-metabolite ratios within the human body, especially after death. Postmortem changes such as these have rarely been evaluated or taken into consideration but they have potential to supply valuable information, especially concerning compound identification and confirmation.;The purpose of this research was to investigate the effects of Escherichia coli, Bacteroides fragilis, and Clostridium perfringens on drugs of …

Characterizing Peripheral Cellular And Humoral Immune Responses To Haemonchus Contortus In Sheep, Jesica Rae Jacobs
Graduate Theses, Dissertations, and Problem Reports (ETD)
Gastrointestinal nematode parasitism is the most widely reported health concern among sheep producers worldwide. The most concerning of which is Haemonchus contortus; a blood-sucking nematode found in the abomasum of sheep. Worldwide anthelminthic resistance has prompted exploration of alternative methods of parasite control. Many studies have shown St. Croix hair sheep posses an immunologically mediated resistance to H. contortus infection, however no studies have utilized a true parasite naive animal for immune characterization. In this study, development of peripheral immune response was characterized in parasite resistant St. Croix lambs and parasite susceptible Suffolk crossbred Lambs. Lambs were separated into three …

The Role Of Neurokinin B In The Regulation Of The Preovulatory Lh Surge In Ewes, Katrina L. Porter
The Role Of Neurokinin B In The Regulation Of The Preovulatory Lh Surge In Ewes, Katrina L. Porter
Graduate Theses, Dissertations, and Problem Reports (ETD)
Hypothalamic input is vital to the regulation of reproductive function. Gonadotropin-releasing hormone (GnRH) is secreted by hypothalamic GnRH neurons into the hypophysial portal circulation. When GnRH reaches the anterior pituitary, it stimulates gonadotropes to secrete luteinizing hormone (LH) into the systemic circulation, ultimately leading to ovulation. GnRH/LH secretion occurs in two different patterns: pulsatile or surge. Pulsatile GnRH release is governed by estradiol (E2) negative feedback while surge release is the result of E2 positive feedback. However, GnRH neurons lack the alpha isoform of the estrogen receptor, indicating that E2 feedback on GnRH release is indirect. Other hypothalamic neurons that …

Analog Signal Buffering And Reconstruction, Brandon M. Kelly
Analog Signal Buffering And Reconstruction, Brandon M. Kelly
Graduate Theses, Dissertations, and Problem Reports (ETD)
Wireless sensor networks (WSNs) are capable of a myriad of tasks, from monitoring critical infrastructure such as bridges to monitoring a person's vital signs in biomedical applications. However, their deployment is impractical for many applications due to their limited power budget. Sleep states are one method used to conserve power in resource-constrained systems, but they necessitate a wake-up circuit for detecting unpredictable events. In conventional wake-up-based systems, all information preceding a wake-up event will be forfeited. To avoid this data loss, it is necessary to include a buffer that can record prelude information without sacrificing the power savings garnered by …

Thrombospondin: An Emerging Keystone In Skeletal Muscle Structure And Function, Gerald Nelson Audet
Graduate Theses, Dissertations, and Problem Reports (ETD)
Our objective was to elucidate the effects of TSP-1 on skeletal muscle structure and function, and its potential role in the progression of skeletal muscle dysfunction. The central hypothesis was that TSP-1 is a critical negative regulator of skeletal muscle angiogenesis. In chapter 3, we used a TSP-1 mimetic delivered by mini osmotic pumps to show that TSP-1 can reduce capillarity across three distinct muscle types. We found that those given the mimetic had a 35% decrease in soleus capillarity, a 20% decrease in the gastrocnemius, and an 11% decrease in the plantaris. This confirmed the critical role that TSP-1 …

The Involvement Of Sigma Receptor Modulation In The Antidepressant Effects Of Ketamine And The Neurotoxic Actions Of Methamphetamine, Matthew J. Robson
Graduate Theses, Dissertations, and Problem Reports (ETD)
Depression is estimated to affect at least 30% of the world's population at some point during their lives. Currently marketed antidepressants require a period of at least 2 to 3 weeks to display any antidepressant effects in clinical populations and rates of clinical resistance to the antidepressant effects of these drugs are high. Ketamine is an N-methyl-D-aspartate (NMDA) antagonist and dissociative anesthetic that has been shown to display rapid acting and prolonged antidepressant activity in smallscale human clinical trials. Ketamine also binds to sigma receptors, which are believed to be protein targets for a potential new class of antidepressant medications. …
Involvement Of Cytochromep4504a In Adenosine A1 Receptor Mediated Regulation Of Vascular Tone, Swati S Kunduri
Involvement Of Cytochromep4504a In Adenosine A1 Receptor Mediated Regulation Of Vascular Tone, Swati S Kunduri
Graduate Theses, Dissertations, and Problem Reports (ETD)
Cardiovascular diseases are one of the leading causes of morbidity and mortality worldwide. The regulation of vascular tone plays an important role in normal cardiovascular function. Adenosine, an autacoid has several physiological and pathophysiological roles, apart from the regulation of vascular tone. Adenosine receptor (AR) contracts and relaxes blood vessels through all four subtypes (A1, A2A, A2B, and A3) linked to different signaling mechanisms. Deciphering complex tissue responses remains difficult because relationships of individual receptor subtypes and various end-effectors (e.g., ion channels) are yet to be identified. Apart from adenosine, 20-HETE, a cytochrome P4504a (Cyp4a) metabolite of arachidonic acid (AA) …
