This study focused on developing an effective and environmentally friendly method to measure ligustrazine in rat serum by using polymer monolith micro-extraction (PMME) technique. A poly (methacrylic acid-ethylene glycol dimethacrylate) material was used to extract ligustrazine through hydrophobic and ion-exchange interaction. Qualitative and quantitative analysis was performed by a liquid chromatography and tandem mass spectrometry. After optimization of several PMME conditions, the developed method exhibited excellent extraction performance to the ligustrazine. Good linearity was acquired ranging from 10 to 2,000 ng mL-1, and the limit of detection of the proposed method was 0.14 ng mL-1. The recoveries measured by spiking …

Envirotyping is an essential technique used to unfold the nongenetic drivers associated with the phenotypic adaptation of living organisms. Here, we introduce the EnvRtype R package, a novel toolkit developed to interplay large-scale envirotyping data (enviromics) into quantitative genomics. To start a user-friendly envirotyping pipeline, this package offers: (1) remote sensing tools for collecting (get_weather and extract_GIS functions) and processing ecophysiological variables (processWTH function) from raw environmental data at single locations or worldwide; (2) environmental characterization by typing environments and profiling descriptors of environmental quality (env_typing function), in addition to gathering environmental covariables as quantitative descriptors for predictive purposes (W_matrix …

The relevance of multidimensional and porous crystalline materials to nuclear waste reme-diation and storage applications has motivated exploratory research focused on materials discovery of compounds, such as actinide mixed-oxoanion phases, which exhibit rich structural chemistry. The novel phase K1.8 Na1.2 [(UO2)BSi4 O12 ] has been synthesized using hydrothermal methods, rep-resenting the first example of a uranyl borosilicate. The three-dimensional structure crystallizes in the orthorhombic space group Cmce with lattice parameters a = 15.5471(19) Å, b = 14.3403(17) Å, c = 11.7315(15) Å, and V = 2615.5(6) Å3, and is composed of UO6 octahedra linked by [BSi4 O12 ]5− chains to …

Nitrous oxide (N2O) is a potent greenhouse gas and an ozone destroying substance. Yet, clear step-by-step protocols to measure N2O transformation rates in freshwater and marine environments are still lacking, challenging inter-comparability efforts. Here we present detailed protocols currently used by leading experts in the field to measure water-column N2O production and consumption rates in both marine and other aquatic environments. We present example 15N-tracer incubation experiments in marine environments as well as templates to calculate both N2O production and consumption rates. Bacteria play an integral role in shaping plant growth and development. However, the genetic factors that facilitate plant-bacteria interactions remain largely unknown. Here, we demonstrated the importance of two bacterial genetic factors that facilitate the interactions between plant-growth-promoting (PGP) bacteria in the genus Caulobacter and the host plant Arabidopsis. Using homologous recombination, we disrupted the cytochrome ubiquinol oxidase (cyo) operon in both C. vibrioides CB13 and C. segnis TK0059 by knocking out the expression of cyoB (critical subunit of the cyo operon) and showed that the mutant strains were unable to enhance the growth of Arabidopsis. About 20% of the organic carbon produced in the sunlit surface ocean is transported into the ocean’s interior as dissolved, suspended and sinking particles to be mineralized and sequestered as dissolved inorganic carbon (DIC), sedimentary particulate organic carbon (POC) or “refractory” dissolved organic carbon (rDOC). Recently, the physical and biological mechanisms associated with the particle pumps have been revisited, suggesting that accepted fluxes might be severely underestimated (Boyd et al., 2019; Buesseler et al., 2020). Germline mutations in BRAF and other components of the MAPK pathway are associated with the congenital syndromes collectively known as RASopathies. Here, we report the association of Septo-Optic Dysplasia (SOD) including hypopituitarism and Cardio-Facio-Cutaneous (CFC) syndrome in patients harbouring mutations in BRAF. Phosphoproteomic analyses demonstrate that these genetic variants are gain-of-function mutations leading to activation of the MAPK pathway. Activation of the MAPK pathway by conditional expression of the BrafV600E/+ allele, or the knock-in BrafQ241R/+ allele (corresponding to the most frequent human CFC-causing mutation, BRAF p.Q257R), leads to abnormal cell lineage determination and terminal differentiation of …

Butanol seems to be an eligible fuel for compensating for the increasing fuel consumption. Biobutanol could be produced from local sources in the place of use. Its properties show similar results to gasoline, so biobutanol could be added as a biocomponent into fuels. Important properties, in the case of blending biobutanol into gasoline, are its fluid properties and their dependence on the temperature. Therefore, in this paper, the volumetric mass density and viscosity of the selected ratios between biobutanol and gasoline (0, 5, 10, 85, 100 vol.%) were tested over the temperature range from -10 °C up to 40 °C. …

Microwave assisted liquefaction (MAL) of alkali lignin in ethylene glycol (EG) in the presence of hydrazine was carried out using response surface methodology. Dielectric properties of mixtures of lignin/EG at different hydrazine concentrations were determined to enhance the understanding of the MAL process. MAL parameters were hydrazine concentration (1–3%), temperature (100-180 °C), time (5-45 min) with the maximum microwave heating power limited to 750 W. Lignin/EG ratio was fixed at 1 g:10 ml. Dielectric properties were significantly influenced by hydrazine concentration and temperature. Freshwater (FW) flux between the Arctic Ocean and adjacent waterways, predominantly driven by wind and oceanic currents, influences halocline stability and annual sea ice variability which further impacts global circulation and climate. The Arctic recently experienced anomalous years of high and low sea ice extent in the summers of 2013/2014 and 2012/2016, respectively. Here we investigate the interannual variability of oceanic surface FW flux in relation to spatial and temporal variability in sea ice concentration (SIC), sea surface salinity (SSS), and sea surface temperature (SST), focusing on years with summer sea–ice extremes. Law schools often claim that they are teaching students “how to think like a lawyer.” What is less touted, however, is that students are learning how to look like a lawyer. They receive this message from multiple sources (faculty, alumni, peers, the career office) concerning a variety of situations: class, interviews, moot court, trial team, symposia and conferences. For law students who are first generation, these sources may be the only avenue (apart from the entertainment industry) of determining how to look like a lawyer. Purpose: This qualitative study explores how discursive power shapes South Carolina LGBTQIA+ communities' health information practices and how participants resist this power. Design/methodology/approach: Twenty-eight LGBTQIA+ community leaders from South Carolina engaged in semi-structured interviews and information worlds mapping – a participatory arts-based elicitation technique – to capture the context underlying how they and their communities create, seek, use, and share health information. We focus on the information worlds maps for this paper, employing situational analysis – a discourse analytic method for visual data – to analyze them. Short sleep has been linked to adolescent obesity risk, but questions remain regarding the dietary mechanisms by which this occurs. We tested whether mildly shortening sleep influences how rewarding and appealing healthy adolescents find several kinds of foods. Eighty-eight healthy adolescents completed a within-subjects crossover sleep experiment comparing 5 days of Short Sleep (6.5 hour sleep opportunity) vs. 5 days of Healthy Sleep (9.5 hour sleep opportunity). Following each condition, adolescents completed measures of food appeal and reinforcing value of food across five food types: sweets/desserts, fruits/vegetables, lean meats/eggs, fast food entrees, and processed snacks. We present twisted spatiotemporal optical vortex (STOV) beams, which are partially coherent light sources that possess a coherent optical vortex and a random twist coupling their space and time dimensions. These beams have controllable partial coherence and transverse orbital angular momentum (OAM), which distinguishes them from the more common spatial vortex and twisted beams (known to carry longitudinal OAM) in the literature and should ultimately make them useful in applications such as optical communications and optical tweezing. Granulins (GRN 1-7) are short (~6 kDa), cysteine-rich proteins that are generated upon the proteolytic processing of progranulin (PGRN). These peptides, along with their precursor, have been implicated in multiple pathophysiological roles, especially in neurodegenerative diseases. Previously we showed that GRN-3 and GRN-5 are fully disordered in the reduced form implicating redox sensitive attributes to the proteins. Redox-based modulations are often carried out by metalloproteins in mitigating oxidative stress and maintaining metal-homeostasis within cells. This study aimed to analyze counter-movement jump (CMJ) performance in time and frequency domains. Fortyfour Division I American football players participated in the study. Kinetic variables were collected from both dominant and non-dominant legs using two force plates. Normalized peak power, normalized net impulse, and normalized peak force significantly correlated with jump height (r =.960, r =.998, r =.725, respectively with p < .05). The mean frequency component was significantly correlated with CMJ performance (r =.355 with p < .05). The reliability of the frequency variables was higher than the time domain variables. Frequency domain variables showed weaker correlations with jump height compared with time domain variables. Frequency domain analysis provides frequency components, which represent the rate of energy transmission from the eccentric phase to the end of the push-off phase. Frequency component information may provide additional information for the analyses of CMJ performance for athletes.

We investigated changes in soil chemistry and foliar metabolism of Himalayan cedar [Cedrus deodara (Roxb. Ex Lamb.) G.Don] and Himalayan spruce [Picea smithiana (Wall.) Boiss] trees along a steep elevational gradient in the lower Himalayan Mountains at Kufri, Himachal Pradesh (HP), India. The foliar and soil samples were collected from four locations along a 300 m elevational gradient at ridge, high-, mid-, and low-elevation sites within the forested Shimla Water Catchment Wildlife Sanctuary that provides water for the city of Shimla, HP,. Foods and beverages provide nutrients and alter the gut microbiota, resulting in eubiosis or dysbiosis. Chronic consumption of a diet that is high in saturated or trans fats, meat proteins, reducing sugars, and salt and low in fiber induces dysbiosis. Dysbiosis, loss of redox homeostasis, mast cells, hypoxia, angiogenesis, the kynurenine pathway, transglutaminase 2, and/or the Janus kinase pathway are implicated in the pathogenesis and development of inflammatory bowel disease, celiac disease, and gastrointestinal malignancy. Silver nanoparticles (AgNPs) are widely used in medical applications due to their antibacterial and antiviral properties. Despite the extensive study of AgNPs, their toxicity and their effect on human health is poorly understood, as a result of issues such as poor control of NP properties and lack of proper characterization. The aim of this study was to investigate the combined characterization, bio-uptake, and toxicity of well-characterized polyvinylpyrrolidone (PVP)-coated AgNPs in exposure media during exposure time using primary human cells (peripheral blood mononuclear cells (PBMCs)). Eucalyptus plantations have replaced other (agro)ecosystems over 5.6 Mha in Brazil. While these plantations rapidly accumulate carbon (C) in their biomass, the C storage in living forest biomass is transient, and thus, longer-term sustainability relies on sustaining soil organic matter (SOM) stocks. A significant amount of harvest residues (HR) is generated every rotation and can yield SOM if retained in the field. Yet, there is little information on how managing eucalyptus HR changes SOM dynamics. Coastal ecosystems are experiencing degradation from compound impacts of climate change and multiple anthropogenic disturbances. These pressures often act synergistically and complicate designing effective conservation measures; consequently, large-scale coastal restoration actions become a wicked problem. The purpose of this study was to use two different food web models in a coordinated manner to inform resource managers in their assessment of the ecological effects of a large-scale marsh restoration project. Eutrophication is one of the most important anthropogenic pressures impacting coastal seas. In Europe, several legislations and management measures have been implemented to halt nutrient overloading in marine ecosystems. This study evaluates the impact of freshwater nutrient control measures on higher trophic levels (HTL) in European marine ecosystems following descriptors and criteria as defined by the Marine Strategy Framework Directive (MSFD). We used a novel pan-European marine modeling ensemble of fourteen HTL models, covering almost all the EU seas, under two nutrient management scenarios. Compound heterozygous (CH) variants occur when two recessive alleles are inherited and the variants are located at different loci within the same gene in a given individual. CH variants are important contributors to many different types of recessively inherited diseases. However, many studies overlook CH variants because identification of this type of variant requires knowing the parent of origin for each nucleotide. Using computational methods, haplotypes can be inferred using a process called “phasing,” which estimates the chromosomal origin of most nucleotides. Background
Beyond intakes of total energy and individual nutrient, eating patterns may influence health, and thereby the risk of adverse outcomes. How different diet measures relate to frailty—a general measure of increased vulnerability to unfavorable health outcomes—and mortality risk, and how this might vary across the life course, is not known. We investigated the associations of five dietary indices (Nutrition Index (NI), the energy-density Dietary Inflammatory Index (E-DII™), Healthy Eating Index-2015 (HEI-2015), Mediterranean Diet Score (MDS), and Dietary Approaches to Stop Hypertension (DASH)) with frailty and mortality.

Interactions between biological molecules in a cell are tightly coordinated and often highly dynamic. As a result of these varying signaling activities, changes in gene coexpression patterns could often be observed. The advancements in next-generation sequencing technologies bring new statistical challenges for studying these dynamic changes of gene coexpression. In recent years, methods have been developed to examine genomic information from individual cells. Many computerized tools for comparing phonetic transcriptions have been proposed and shared in the past; however, previous tools are relatively difficult to access and incorporate into clinical and research practice, or require users to learn additional phonetic symbol systems. The purpose of this project was to develop and test a readily available web-based application for quantitatively comparing phonetic transcriptions that are input using International Phonetic Alphabet (IPA) symbols. Dynamic thermogravimetric analysis under nitrogen environment was used to understand the thermal decomposition process of 3D printing filaments made of wood-filled polylactic acid (PLA)/starch blend. The characteristic temperatures and apparent activation energy (AAE) of the filaments with various starch contents were calculated with well-known kinetic models by Friedman, Flynn–Wall–Ozawa, Coats–Redfern, and Kissinger. With the increased starch content in the filament, the onset thermal decomposition temperatures of the filaments decreased gradually from 272.4 to 155.1°C. The thermal degradation degree became smaller, and the transitional temperature interval became larger with increased starch proportion. The AAE values of the three types of filaments …

Objective
This study examines the relationship between adverse childhood experiences (ACEs), internalized and externalized psychological distress, and six measures of parenting behavior among fathers in the United States.
Background
Prior research on ACEs and parenting has focused almost exclusively on mothers, specific types of childhood adversity, and the intergenerational transmission of abuse, neglect, and other traumatic experiences. This study extends the literature by considering ACEs in fathers, using a multidimensional measure of ACEs, and multiple measures of positive and negative fathering behavior.
