Oxidative Phosphorylation And The Realkalinization Of Intracellular Ph During Recovery From Anoxia In Artemia Franciscana Embryos, Kurt E. Kwast, Joseph I. Shapiro, Bernard B. Rees, Steven C. Hand

The contribution of mitochondrial oxidative phosphorylation to the realkalinization of intracellular pH (pHi) and resynthesis of purine nucleotides during recovery from anoxia was investigated in embryos of Artemia franciscana by assessing the sensitivity of mitochondrial respiration to pH, calculating proton consumption by oxidative phosphorylation, and measuring changes in pHi using 31P nuclear magnetic resonance. Following short-term anoxia, pHi increased from 6.7 to 7.7 during 20 min of aerobic recovery and was temporally correlated with a large increase in ATP. State 3 respiration rates of isolated mitochondria were not substantially compromised at the acidic pH corresponding to the pHi during anoxia …

An Historical Critique Of The Emergence And Evolution Of Ernesto Che Guevara's Foco Theory, Matt D. Childs

This article provides an analysis of Ernesto Che Guevara's theory of guerrilla warfare, the foco. The numerous changes to the originalfoco thesis, as presented in Guerrilla Warfare (1960), are examined in detail covering two dozen articles, speeches, essays, interviews and books authored by Guevara, Castro and Debray while stressing their relation to national and international politics. The author argues that there was an apparent discourse between Cuban politics and the numerous changes in Guevara's writings. Juxtaposing changes to the foco theory from 1960 to 1967, to Cuban historical events, reflects the political expedience of the 196os and the primary …

Murine hepatitis virus strain (MHV-3) produces a strain-dependent pattern of disease which has been used as a model for fulminant viral hepatitis. This study was undertaken to examine whether there was a correlation between macrophage activation and susceptibility or resistance to MHV-3 infection. Peritoneal macrophages were isolated from resistant A/J and susceptible BALB/cJ mice and, following stimulation with MHV-3 or lipopolysaccharide (LPS), analyzed for transcription of mRNA and production of interleukin-1 (IL-1), tumor necrosis factor alpha (TNF-alpha), transforming growth factor beta (TGF-beta), mouse fibrinogen-like protein (musfiblp), tissue factor (TF), leukotriene B4, and prostaglandin E2 (PGE2). To understand the mechanisms responsible for seasonal fluctuations in growth and N2 fixation in intertidal microbial mat communities, we quantified seasonal changes in mat community composition, related these changes to diel and seasonal N2 fixation rates, and evaluated community responses (growth, N2fixation, composition) to long-term (22 d) nutrient addition bioassays. A temperate intertidal cyanobacterial mat community, located in coastal North Carolina, USA, was sampled at monthly intervals for 1 yr (1993-94) to determine changes in community composition. The abundances of major phototrophic groups were quantified based on the relative concentrations of taxaspecific photopigments (chlorophylls and carotenoids). …

Exocytosis of transmitter at most synapses is a very fast process triggered by the entry of Ca2+ during an action potential. A reasonable expectation is that the fast step of exocytosis is followed by slow steps readying another vesicle for exocytosis but the identity and kinetics of these steps are presently unclear. By voltage clamping both pre- and postsynaptic neurons in an isolated pair of retinal amacrine cells, we have measured evoked synaptic currents and responses to single vesicles of transmitter (minis). From these currents, we have computed the rate of exocytosis during a sustained presynaptic depolarization. We show here …

ID elements comprise a rodent SINE (short Interspersed DNA repetitive element) family that has amplified by retroposition of a few master genes. In order to understand the Important factors of SINE amplification, we investigated the transcription of rat ID elements. Three different size classes of ID transcripts, BC1, BC2 and T3, have been detected In various rat tissues, including brain and testes. We have analysed the nucleotlde sequences of testes- and brain-derived ID transcripts isolated by size-fractlonation, C-talling and RACE. We have purified two proteins from Drosophila that bind to the scs' boundary element of the hsp70 domain at locus 87A7. Their palindromic binding sites (CGATA-TATCG) symmetrically abut the previously mapped hypersensitive site of scs′. We have cloned a cDNA for one of these proteins, BEAF-32 (boundary element-associated factor of 32 kDa). It encodes a novel protein that is bound to scs' but not scs in vivo. Immunostaining localizes BEAF to hundreds of interbands and many puff boundaries on polytene chromosomes, suggesting that a chromosomal domain consists of a band (or puff) and part of the flanking interbands. Gross observation of testicular parenchyma of 1.5 to 2-yr-old horses reveals both light and dark regions. If this gross, differential shading reflects quantitative differences in the development of spermatogenesis and interstitial cell populations, the horse may prove to be a useful model for study of the paracrine relationships associated with initiation of spermatogenesis. The objective of this study was to characterize seminiferous tubules and interstitium of testes with gross, differential shading. Testes with both light and dark regions of parenchyma were obtained from horses 1.5- 2 yr old and compared to parenchyma of fetal, 2-yr old, or 5-yr-old horses. Stereology …

A solution to an optimization problem is developed that deals with minimizing a measure of difference between the values of observed and predicted variables at an open ocean boundary. Minimization is based on the change of the flux of energy through the open boundary. It is shown that many of the longwave radiation conditions that are commonly used in ocean modeling can be derived using this optimization criteria. However, the minimization process is seen to produce a modification of these radiation conditions in that they are multiplied by a coefficient, which allows the conditions to adapt to a change in …

Community respiration, net nutrient fluxes and heterotrophic bacterial production were investigated in the Mississippi River (USA) plume during May 1992 using dark bottle incubations of unfiltered water. Highest rates of community O2 consumption and dissolved inorganic carbon regeneration were observed at intermediate (10 to 27%0) plume salinities. Plume surface 02 consumption rates were 2- to 4-fold greater than rates reported previously during the summer and winter. Heterotrophic bacterial production ([3H]-leucine incorporation) was also highest at intermediate salinities and 2- to 4-fold greater than rates reported from other seasons. Many transportation problems can be formulated as a linearly-constrained convex programming problem whose objective function consists of entropy functions and other cost-related terms. In this paper, we propose an unconstrained convex programming dual approach to solving these problems. In particular, we focus on a class of linearly-constrained entropy maximization problem with quadratic cost, study its Lagrangian dual, and provide a globally convergent algorithm with a quadratic rate of convergence. Previously, we demonstrated induction of a unique macrophage prothrombinase during infection of BALB/cJ mice by mouse hepatitis virus strain 3 (MHV-3). By immunologic screening, a clone representing this prothrombinase was isolated from a cDNA library and sequenced. The sequence identified this clone as representing part of a gene, musfiblp, that encodes a fibrinogen-like protein. Six additional clones were isolated, and one clone, p11-3-1, encompassed the entire coding region of musfiblp. Murine macrophages did not constitutively express musfiblp but, when infected with MHV-3, synthesized musfiblp-specific mRNA. musfiblp mRNA induction was earlier and significantly greater in BALB/cJ than A/J …

We have investigated the regulation of accumulation of ribulose1,5-bisphosphate carboxylase/oxygenase activase and the periplasmic carbonic anhydrase (CA) in Chlamydomonas reinhardtii. In algae, the periplasmic CA is required for efficient CO2 fixation when the CO2 concentration is low. These two proteins are affected differently by the CO2 level in the environment. The steady-state level of the ribulose-1,5-bisphosphate carboxylase/oxygenase activase transcript was only slightly and transiently affected by a reduction in ambient CO2 concentration, whereas the CA transcript level was strongly induced by air containing ambient (350 parts per million) CO2 (low CO2) conditions. The capacity for anaerobic metabolism of endogenous and selected exogenous substrates in carbon- and energy-starved methanotrophic bacteria was examined. The methanotrophic isolate strain WP 12 survived extended starvation under anoxic conditions while metabolizing 10-fold less endogenous substrate than did parallel cultures starved under oxic conditions. During aerobic starvation, the cell biomass decreased by 25% and protein and lipids were the preferred endogenous substrates. Aerobic protein degradation (24% of total protein) took place almost exclusively during the initial 24 h of starvation. Metabolized carbon was recovered mainly as CO2 during aerobic starvation. Wheat forage was harvested at an early head stage of maturity and ensiled in 12 900-kg experimental silos at three percentages of DM (20.8% for direct-cut forage and 27.9 or 39.3% for wilted forage) either with or without application of a lactic acid bacterial inoculant. The objective was to test the efficacy of the inoculant to alter silage fermentation, preservation, and nutritive value of wheat forage ensiled at different moisture percentages because of wilting. Wilting enhanced DM preservation and decreased fermentation end products. Inoculation made the fermentation more homolactic but did not enhance DM preservation. Silage rations (80% DM as …

During the next few decades, the world demand for wood products is expected to rise sharply. To meet this growing demand, there will be an increasing need for mass production of improved-quality planting stock of many tree species. The conventional methods of tree improvement and selection offer only limited possibility of meeting the growing demands. Therefore, new and innovative techniques for the creation of new hybrids, early selection and testing of desirable genotypes, rapid vegetative propagation of selected genotypes, and improvement through genetic engineering, etc., must be developed to achieve these goals.

Native rat adipocytes and the mouse adipocyte cell line, 3T3-L1, possess transport vesicles of apparently uniform composition and size which translocate the tissue-specific glucose transporter isoform, GLUT4, from an intracellular pool to the cell surface in an insulin-sensitive fashion. Caveolin, the presumed structural protein of caveolae, has also been proposed to function in vesicular transport. Thus, we studied the expression and subcellular distribution of caveolin in adipocytes. We found that rat fat cells express the highest level of caveolin protein of any tissue studied, and caveolin is also expressed at high levels in cardiac muscle, another tissue possessing insulin responsive …
