The purpose of this study was to identify and analyze patterns of institutional strategies and processes that promote the sustainability of laboratory schools housed and managed by institutions of higher education. First, a comprehensive analysis of the development, growth, decline, and current status of the laboratory school movement was conducted by means of a review of relevant literature. Interactions between chemokines and their receptors are involved in organ- specific homing and propagation of metastatic breast cancer (BrCa) cells. BrCa cells express higher levels of CXCR4 and CXCR7 mRNA and cell surface protein, than normal human mammary epithelial cells (HMECs). CXCR4 is ubiquitously expressed by a multitude of cancerous and normal cell types; while CXCR7 is differentially expressed by T helper lymphocytes, cancer cells, and normal epithelium. Importantly, recent studies show CXCR7 is highly expressed by activated endothelial cells (i.e., neovasculature), fetal liver cells, and many tumors, but not by non-transformed (or normal) human tissues. O -linked ß-N-acetylglucosamine ( O -GlcNAc) is an inducible, dynamically cycling, and reversible post-translational modification of serine/threonine amino acid residues of nucleocytoplasmic and mitochondrial proteins. O -GlcNAc transferase (OGT) adds, while O -GlcNAcase (GCA) removes O -GlcNAc from proteins. Albeit being a recruitable stress-induced signal in other tissues, the role of O -GlcNAc in the heart is unknown. Therefore, we hypothesized that O -GlcNAc is recruited in the heart during acute stress, and enhanced O-GlcNAc is cardioprotective. Subjecting neonatal rat cardiac myocytes (NRCMs) to hypoxia, or mice to myocardial ischemia reduced O-GlcNAc signaling. Augmented O-GlcNAc signaling attenuated, while diminished O-GlcNAc …

The purpose of this study was to explore how mothers with an ADHD child living in the household perceive and manage family communication. In-depth interviews were conducted amongst nine married mothers who have an ADHD child. Three themes emerged from the interview data: relational management, time management, and internal and external support. Mothers communicate with their ADHD child differently than their non-ADHD children and must also manage communication about the ADHD diagnosis. Mothers are not always open about their child's ADHD with non-family members as they are afraid of the stigma that is attached to the condition. Mothers recognize that …

Alcoholic liver disease (ALD) is a serious concern for the world's population. It is one of the leading causes of death and is also a huge economic burden. The biochemical mechanisms responsible for ALD are incompletely understood, therefore there is no FDA approved therapy to treat or reverse liver damage caused by alcohol exposure. Friedrich Schiller's psychological theory of play, his hypothesis about human nature, is embedded in the Aesthetic Letters . Its trans-historical value owes much to the committed interest in life in the late Enlightenment, and the theory itself is an example of that period's enthusiastic study of living organisms. Researchers have long been interested in the relationship among the separate fields of psychology and religion. This dissertation seeks to explore the differences in the way protestant Christians with various religious orientations view mental illness and how these views relate to their attitudes toward seeking professional psychological help. A total of 540 individuals from church congregations representing ten different denominations completed measures of religiosity, beliefs about mental illness, and attitudes toward seeking professional psychological help. Every new nursing graduate is challenged to successfully transition from student to professional nurse. The stress involved in that transition can manifest itself as performance anxiety, a type of anxiety occurring when someone is the focus of attention and is fearful of interactions or of being humiliated or embarrassed. It occurs only in specific situations. The new graduate's performance is the focus of attention and evaluation. The cyclic nitrone 5,5-dimethyl-1-pyrroline-N-oxide (DMPO), and the lesser known linear phenyl-N-tert-butylnitrone (PBN) and its phosphorylated analogues have been used as spin traps for the investigation of free radicals in biological systems. Theoretical work on these molecules suggests that there are important differences in their properties between biological systems and isolated molecules in the gas phase, most likely resulting from intra and intermolecular hydrogen bonding. Most dielectric solvation models such as the polarized continuum model and COSMO are incapable of direct determination of solvent-spin trap chemical interactions. Much attention has been paid to immobilization of enzymes to improve enzyme stability and permit its reuse. Glucose oxidase entrapment in different kinds of silica sol-gel matrices was investigated. The enzyme showed stable activity for 11 uses in the sol-gel with tetramethyl orthosilicate (TMOS) as precursor, and at least 7 uses in the sol-gel induced electrochemically with tetraethyl orthosilicate (TEOS) as precursor. The sol-gel made with TEOS as precursor and HCl as catalyst showed stability in enzyme activity for 11 uses but the activity decreases on the same sol-gel when modified with silica nanoparticles. Because of growing concern of habitat fragmentation and its adverse effects on salamander communities in Appalachian region, sympatric populations of ambystomatid salamanders A. opacum and A. maculatum were studied in Northeast Tennessee to address a number of questions: i) the extent of sexual size dimorphism (SSD) in both species, ii) what traits influence the dorsal skin pigmentation and how, iii) whether gender differences in developmental stability occur, and iv) the extent of phenotypic variation within each species. The genus Tsuga is comprised of eight extant species found in North America and East Asia and four species represented by fossils from Europe and Japan. This study presents the first phylogenetic analysis based on structural, biochemical, and molecular sequence data. Characters obtained from published and unpublished literature were combined with new morphological characters from seeds, seedlings, and leaf cuticle material. Results from parsimony analyses of these characters differed from the published molecular based phylogeny. The purpose of the study was to examine the burnout potential of certified athletic trainers (ATCs) working in the regions of Southwest Virginia and Northeast Tennessee. To determine burnout potential, the Maslach Burnout Inventory-Human Services Survey was used. Permission to modify the inventory was obtained from CPP, Inc. Participants for the research study were identified by searching the NATA membership directory, state athletic training websites and state licensure websites. Sixty-seven ATCs were emailed invitations to participate. Fifty surveys were completed. Of these 50, five were eliminated from statistical analysis. Due to their high reactivity, they have been extensively used to carry out a variety of unique and effective chemical transformations including but not limited to ionic and free radical additions and transition metal catalyzed cyclizations. As the chemistry of this group of compounds is explored further and their applications expanded there is a need to develop alternative and cost effective methods for their synthesis. Capsaicinoids are a class of compounds responsible for the "heat" of hot peppers. Capsaicin and dihydrocapsaicin have the highest burning effect. The aim of this work is to separate and quantify the two major capsaicinoids in fruits harvested at different stages of development and at different seasons. Simple and rapid HPLC method involves 73:27% methanol water mobile phase with C18 stationary phase and UV-Vis detector set at 210 nm. The method showed good reproducibility with 1.74% - 4.72% relative standard deviations, a linear response within 0.65–45.5 and 0.25-17.5 μg/mL for capsaicin and dihydrocapsaicin, respectively. The two variable Fitzhugh Nagumo model behaves qualitatively like the four variable Hodgkin-Huxley space clamped system and is more mathematically tractable than the Hodgkin Huxley model, thus allowing the action potential and other properties of the Hodgkin Huxley system to be more readily be visualized. In this thesis, it is shown that the Carleman Embedding Technique can be applied to both the Fitzhugh Nagumo model and to Van der Pol's model of nonlinear oscillation, which are both finite nonlinear systems of differential equations. Luminescence techniques are sensitive, selective, and widely used in analysis. Luminescence intensity is attenuated by quenchers. This research has focused on the use of surfactants such as CTAB, SDS, and TX-100 singly or together with TiO2 nanoparticles to evaluate their individual and combined effects on some fluorescent aromatic compounds such as pyrene, fluoranthene, anthracene, phenanthrene, and carbazole. Rutile phase TiO2 was synthesized using the low temperature sol-gel method. Carbazole and phenanthrene were severely quenched by all surfactants singly or in combination with TiO2. It interacts with the enzyme alliinase when garlic cloves are crushed, cut, or chewed to produce allicin, an unstable thiosulfinate that is the main biologically active component of fresh crushed garlic. The objective of this study was to identify genes that are differentially regulated during diapause when compared with nondiapausing pupae in Sarcophaga crassipalpis. The results of a Suppression Subtractive Hybridization procedure was used to indentify genes that are differentially regulated in both diapause and nondiapausing states while suppressing genes that are common to both states. Randomly picked colonies from both subtractive libraries were isolated and the inserts sequenced. The purpose of this study was to determine if individuals who supported denying citizenship to children of foreign parentage and making English the sole language spoken in the United States held the opinion that a sentence length over 15 years was appropriate for non-U.S. individuals. Other purposes were to determine if individuals with high religiosity or who carried a lethal weapon also held the opinion that a sentence length over 15 years was appropriate for non-U.S. individuals.

Standardized assessment tools have been criticized for use with young children; however, there is limited empirical research supporting developmentally appropriate, play-based assessments. Forty children, with and without developmental disabilities, ages 10-73 months referred from the community of a large city received a transdisciplinary play based assessment (TPBA-2) and standardized assessment. The critical role of parent involvement has been endorsed by educators and educational policy in the United States. However, various definitions and approaches to assess parent involvement have yielded inconsistent conclusions regarding the impact of parent involvement on child development and failed to provide foundations for practitioners. This historic case study addressed the issue of the lack of citizen action toward environmentally responsible behavior. Although there have been studies regarding components of environmental responsible behavior [ERB], there has been little focus on historic models of exemplary figures of ERB. This study examined one of the first conservation courses in the United States, Wildlife Ecology 118, taught by Aldo Leopold (1887-1948) for 13 years at the University of Wisconsin. Here, I address the function of the carboxy-terminal hydrophobic helix of the pro-apoptotic protein Bax. There has been considerable controversy as to whether this sequence is required for the targeting and insertion of Bax into the mitochondrial outer membrane. The Bax carboxy-terminal tail is tucked into a hydrophobic pocket within the closed/inactive conformation of Bax. Apoptotic stimulation results in an opening of the Bax conformation, exposing a mitochondrial-targeting signal and subsequent insertion of Bax into the mitochondrial outer membrane. Circulating nucleated cell populations found in whole blood, including both white blood cells (leukocytes) and endothelial cells, provide an ideal platform for studies seeking to understand the disease processes for development of drugs and treatments. This thesis presents an automated microfluidic device developed for leukocyte enrichment from peripheral blood. Briefly, the device allows for complete lysis of red blood cells and comprehensive analysis of nucleated cell populations in terms of quantity and activation status. The microfluidic lysis device was used in two Sickle Cell disease (SCD) studies to understand the effect of leukocytes in the initiation of vasoocclusive crisis. Findings …
