This dissertation consists of a full-length novel for an adult audience as well as a substantial critical afterword to elucidate and complicate germane thematic concerns of the creative artifact. The novel, leaning on the rich traditions and many genre conventions of fictions of the American south, tells the story of an unlikely underground coal miner in Blue Banks, a fictional southern town set in the caverns and rolling hills of the western Kentucky coal fields. Periodontitis is an infection-driven inflammatory disease characterized by gingival inflammation and bone loss. The NF-ĸB signaling pathway is pivotal in osteoclastogenesis and infection-induced pro-inflammatory responses. The use of nanoparticles as a vehicle to deliver drug increases stability, loading capacity, and facilitates transmembrane transportation. The hypothesis was that a novel nanoparticle carrying therapeutic NBD inhibitory peptides (NBD-nanoparticles) will inhibit measures of periodontal disease. Urban heat island (UHI) is a term used to describe increased surface and atmospheric temperatures in an urban core relative to surrounding non-urbanized areas. To examine the variability introduced into derived estimates of the surface UHI, this study constructs and compares multiple remotely sensed indicators of the surface UHI for major metropolitan cities of a temperate eco-region of the United States. The Moderate Resolution Imaging Spectroradiometer (MODIS) 8-day, 500-meter product (MOD11A2) is the source data used to calculate six different RS-derived UHI indicators for the year 2002 to 2012. Verisimilitude, or the appearance of being true, is a concept I turn upside down; relating it to a guise I wear as a contemporary male in a society dictated by learned social behavior and gender norms. Cultural iconography and expected gender norms are tropes I confront within my artwork. Drawings of seemingly everyday objects act as meditations or a fetishized repetition of supposed unobtainable objects and ideals that deal with masculine societal norms. This dissertation aims to explore how the phenomenon of cosplay has been able to produce and sustain a diversity of gender expression due to its emergence from an activity-based community that emphasizes creative play. This creative energy is manifested through cosplay as an active, ritualized practice in which gender diversity is invited to be realized as a distinct possibility, resulting in a display of a full range of masculinities and femininities as well as crossplays and genderbend cosplays. Myeloid derived suppressor cells (MDSCs) are a heterogeneous group of immature myeloid cells that are significantly increased in cancer patients and correlate with higher stage and poor prognosis. MDSCs negatively modulate anti-tumor immunity, suppress T cell activity, promote angiogenesis and increase the risk of metastasis. In this study, we report that monocytic-MDSCs (M-MDSCs) but polymorphonuclear-MDSCs (PMN-MDSCs) over-express 6-phosphofructo-2-kinase/fructose-2, 6-bisphosphatases 3 (PFKFB3), an important regulator of glycolysis. Furthermore in the melanoma model, M-MDSCs but not PMN-MDSCs suppressed T cell function which correlated with PFKFB3 over-expression and increased rate of glycolysis. The perennial C4 grass Panicum virgatum (switchgrass) is a promising bioenergy feedstock. Switchgrass is symbiotic with arbuscular mycorrhizal fungi (AMF). I examined 12 varieties of switchgrass grouped into 2 ecotypes (upland, lowland) to see if they differed in AMF abundance and root colonization using the GLBRC Variety Trials experiment. There was higher root colonization of lowland ecotype. A greenhouse experiment evaluated the role of live soil organisms in seedling establishment of 11 varieties. Live soil increased seedling survival, growth and influenced root morphology. Ecotypes differed in root architecture. Lowland developed a higher specific root length (SRL), a trait associated …

This dissertation includes three quantitative papers using data from the Fragile Families and Child Wellbeing Study. Paper one, “For us or the children: Exploring the association between coparenting trajectories and parental commitment,” is situated in commitment theory and estimates latent growth curves. Findings suggest that unmarried parents’ coparental relationships are: 1) strong across the first five years of their children’s lives, and 2) influencing parental commitment at year 5. Paper two, “Maternal partnership transitions and coresidence with extended kin,” is situated in life course theory and estimates fixed and random effects models. Recent advancements in the field of genetic testing have made it easier to prenatally identify the presence of Down Syndrome in a fetus. The purpose of this study was to examine the opinions of parents who are currently raising a child with Down Syndrome towards genetic screening and selective abortion for the condition, as well as any potential effects of this process on their families. Soil bacteria play important roles in nutrient cycling and other ecosystem functions, and many biotic and abiotic factors can influence bacterial functional diversity. The goal of this field study was to examine differences among bacterial communities in sand dunes of Lake Michigan. I used Biolog Ecoplates™ to compare bacteria functional diversity associated with four different plant species: the native dune-building grass Ammophila breviligulata, invasive species Leymus arenarius and Gypsophila paniculata, and native legume Lathyrus japonicus across 13 sites in Michigan, representing a gradient in abiotic factors such as precipitation and temperature. Background: Although pregnancy is prone to induce more severe periodontal inflammation and tissue damage, its mechanism remains elusive. Periodontal diseases during pregnancy are associated with adverse pregnancy outcomes including preterm birth or low birth weight. Animal models are ideal for investigating immunological mechanisms in periodontal disease. With a newly modified ligature-induced disease model, we managed to develop periodontal disease in pregnant mice and investigate the immunological mechanism of more severe symptoms during pregnancy. The purpose of this study was to examine the role of the planetarium on students while learning astronomy. The main goals of this study were to evaluate changes in students’ attitudes towards astronomy, whether students learned and retained more knowledge due to planetarium-enriched instruction, and how the planetarium helped students think about astronomical concepts. Cardiac cell therapy using cardiac mesenchymal cells (CMC) significantly reduces ventricular dysfunction in patients with ischemic cardiomyopathy. Despite the improvement in function, a modest number of CMCs survive in the heart post-transplantation. In this study, we sought to improve the survival and retention of transplanted CMCs to prolong the therapeutic benefits afforded by cardiac cell therapy. To do this, we targeted the enzyme telomerase (TERT), known to be active in some highly proliferating cells (e.g. germ, stem). TERT is responsible for preventing telomere attrition, thereby allowing continued proliferation. TERT has also been shown to be protective, improve cell migration and …

This study examined whether intake of caffeinated beverages (coffee, tea, and cola) was associated with fecundability (time to pregnancy) in a prospective cohort study. Data from 470 women from the Mount Sinai Study of Women Office Workers (1990-1994) were analyzed. Intake of coffee, tea, cola, and other variables were recorded in daily diaries and calculated as menstrual cycle level means for up to 20 cycles. Pregnancy was assayed using hCG and confirmed by physician diagnosis. The associations of caffeinated beverages with the probability of becoming pregnant during a given cycle were determined using discrete survival analysis, adjusted for potential confounders. …

Background: Aggregatibacter actinomycetemcomitans (Aa) possesses a two-component system, QseBC, which is key in biofilm formation/virulence and is activated in the presence of iron-catecholamine complexes. Aa does not synthesize enterobactin, a catechol-based siderophore, yet possesses the machinery for recognition and uptake. We hypothesize that Aa is able to acquire iron by catecholamine mediation through the enterobactin receptor/transporter. Methods: By insertional mutation, we attempted to delete the enterobactin permease from the genome using suicide vector pJT1. A sample of 365,315 employed individuals between the ages of 25 and 35 from the American Community Survey (ACS) were used in combination with data from three occupational databases (O*NET, the Occupational Outlook Handbook, the NORC occupational dataset) to examine this topic. Although the prevalence of smoking has declined in the U.S., vulnerable populations continue to suffer from tobacco-related health disparities. An estimated 68.0-80.0% of homeless adults are current cigarette smokers compared to 15.1% in the general population. The large gap in smoking rates suggests that current tobacco cessation programs designed to reduce smoking have little impact among individuals experiencing homelessness. The purpose of this study was to explore the barriers and facilitators to smoking cessation among the homeless. This dissertation examines socio-cultural experiences of national disasters in the United States as they intersect with political rhetorics of democracy, to understand the forms and functions of state power as it is mediated through famous historical figures. Tracing the manifold possibilities from within and around the disruptive spacetimes of disaster, which sociologist Harvey Molotch describes in their ultimate function as societal “rationalizers” (371), I analyze catastrophes as contested spaces and histories that often get situated as variations on the “man vs. nature/god/man” binaries in public discourse. The use of coal combustion for electricity and the storage of coal combustion by-products known as coal ash occur across the United States and around the world. The most abundant type of coal ash, fly ash, contains small particles with metals, some of which are known neurotoxins. Fly ash is currently stored in open-air landfills and surface impoundments that allow fugitive dust to escape into surrounding communities, potentially exposing children to small neurotoxic particles. This dissertation uses preliminary data from a larger cross-sectional study to investigate the relationship between particulate matter exposure, metal exposure, and problems with self-regulation.
