Inhibition Of Chemotherapeutic-Induced Apoptosis In Esophageal Squamous Cancer Cells By The Keystone Periodontal Pathogen, Porphyromonas Gingivalis., Atul Kumar Agrawal

Electronic Theses and Dissertations

Recent evidence has shown that P. gingivalis, a Gram-negative, anaerobic oral bacterium, is negatively associated with the presence and outcome of esophageal squamous cell carcinoma (ESCC). While potential underlying mechanisms are yet to be established, P. gingivalis infection is known to inhibit apoptosis in gingival epithelial cells. Therefore, we hypothesized that P. gingivalis may also inhibit of the induction of apoptosis in human ESCC cells exposed to the commonly employed anti-ESCC chemotherapeutic agent, cisplatin. The capacity of P. gingivalis, at variant multiplicities of infection, to suppress cisplatin-induced necrosis and apoptosis in EC9706 ESCC cells was established by lactate …

Identifying The Signaling Mechanisms Of Egfr-Mediated Apoptosis., Nicole Marion Jackson May 2017

Identifying The Signaling Mechanisms Of Egfr-Mediated Apoptosis., Nicole Marion Jackson

Electronic Theses and Dissertations

The Epidermal Growth Factor Receptor (EGFR) is a 170-kilodalton transmembrane protein that belongs to the ErbB family of receptor tyrosine kinases. Upon ligand-mediated activation, the EGFR is responsible for cell growth, proliferation, and tissue homeostasis; however, the EGFR is overexpressed in many human malignancies, including MDA-MB-468 cells, a metastatic breast epithelial cell line. Studies within this cell line, and other cell lines characterized with high EGFR levels, have shown that EGF stimulation results in the induction of apoptosis. However, the mechanisms and signaling effectors implicated in this process have yet to be elucidated. The overarching research goal of this dissertation …

Examining The Effects Of Macrophage Populations On Cancerous Tumor Growth., Grace E. Mahlbacher

Electronic Theses and Dissertations

The most abundant immune cell types of the tumor microenvironment macrophages recruited there by tumor-eluted factors. The role of these immune cells in tumor progression, and the interplay between tumor and immune cells is an emerging field of research with potential for novel treatment strategies. Here, a TIE2 expressing macrophage (TEM) subtype is integrated into a virtual tumor model. Within the 2D microenvironment, the TEM will differentiate from an extravasated monocyte precursor, congregate around the abluminal side of the vasculature in response to a chemoattractant gradient, secrete cytokines which favor differentiation of a separate angiogenic macrophage subtype [1]. The effects …

Effect Of High Cationic Flocculant On Anaerobic Digestion Of Municipal Wastewater., Prathap D. John May 2017

Effect Of High Cationic Flocculant On Anaerobic Digestion Of Municipal Wastewater., Prathap D. John

Electronic Theses and Dissertations

Anaerobic digestion (AD) of municipal sludge is a widely used stabilization process at municipal wastewater treatment plants. It is highly effective in reducing sludge volume and produces methane gas which is used as fuel in the wastewater plant. Following digestion sludge is sent to a centrifuge to separate the solids from the liquids. Louisville MSD adds flocculent to material in the centrifuge to better coagulate the solids, thereby aiding in separation of the solids from the liquids. The potential for further AD, COD reduction, and methane generation of the liquid effluent from the centrifuge was unknown, particularly in regards to …

The Establishment And Development Of The Mockingbird As The Nightingale’S “American Rival”, Gabe Cameron May 2017

The Establishment And Development Of The Mockingbird As The Nightingale’S “American Rival”, Gabe Cameron

Electronic Theses and Dissertations

During the eighteenth and nineteenth centuries, many British poets attempted to establish a universal poetic image in the European nightingale, often viewing it as a muse or contemporary artist. This use of the songster became so prevalent that it was adopted, along with other conventions, for use in the United States. Yet, despite the efforts of both British and American poets, this imperialized songbird would ultimately fail in America, as the nightingale is not indigenous to the United States. The failure of this nightingale image, I contend, is reflective of the growing need to establish a national identity in nineteenth-century …

The current study utilizes data from the 2007-2008 School Survey on Crime and Safety to address this gap in the literature by utilizing a series of negative binomial regression models that seek to determine differences between predictive factors in urban and rural settings. Results indicate that disorganization has a similar effect within urban and rural …

Peptide Identification: Refining A Bayesian Stochastic Model, Theophilus Barnabas Kobina Acquah May 2017

Peptide Identification: Refining A Bayesian Stochastic Model, Theophilus Barnabas Kobina Acquah

Electronic Theses and Dissertations

Notwithstanding the challenges associated with different methods of peptide identification, other methods have been explored over the years. The complexity, size and computational challenges of peptide-based data sets calls for more intrusion into this sphere. By relying on the prior information about the average relative abundances of bond cleavages and the prior probability of any specific amino acid sequence, we refine an already developed Bayesian approach in identifying peptides. The likelihood function is improved by adding additional ions to the model and its size is driven by two overall goodness of fit measures. In the face of the complexities associated …

Functionalized Silica Gel For Adsorption Of Cesium From Solution, Kenneth Marshall Seaton Iii May 2017

Functionalized Silica Gel For Adsorption Of Cesium From Solution, Kenneth Marshall Seaton Iii

Electronic Theses and Dissertations

Mesoporous silica gel containing embedded phosphotungstic acid (PTA) was synthesized by sol-gel co-condensation of tetraethyl orthosilicate with PTA in acidic media. The obtained material had high Brunauer-Emmett-Teller Theory (BET) surface area and pore volume. A characteristic band of the Keggin structure of PTA was present in its FT-IR spectrum while its X-ray diffraction patterns were absent. This proved the embedding of PTA on a sub-molecular level and not as a second phase. Acidic sites were determined by neutralization with base in aprotic solvent, followed by titration of the remaining base with an acid. The material demonstrated high adsorption capacity of …

Electronic Theses and Dissertations

This thesis examines martial race ideology in the British Army during the mid-nineteenth century. A “martial race” was a group of people that the British considered to excel in the art of warfare due to biological and cultural characteristics. This thesis examines perceived “martial” natures or lack thereof of the Highland Scots and the Irish during this era. Central to this analysis are the Crimean War (1853-1856) and the Indian Mutiny of 1857 which provided opportunities for soldiers to display their “martial” qualities. The Crimean War was the first war where the daily newspapers covered every aspect of the war …

Denoising Tandem Mass Spectrometry Data, Felix Offei May 2017

Denoising Tandem Mass Spectrometry Data, Felix Offei

Electronic Theses and Dissertations

Protein identification using tandem mass spectrometry (MS/MS) has proven to be an effective way to identify proteins in a biological sample. An observed spectrum is constructed from the data produced by the tandem mass spectrometer. A protein can be identified if the observed spectrum aligns with the theoretical spectrum. However, data generated by the tandem mass spectrometer are affected by errors thus making protein identification challenging in the field of proteomics. Some of these errors include wrong calibration of the instrument, instrument distortion and noise. In this thesis, we present a pre-processing method, which focuses on the removal of noisy …

Quantifying The Structure Of Misfolded Proteins Using Graph Theory, Walter G. Witt May 2017

Quantifying The Structure Of Misfolded Proteins Using Graph Theory, Walter G. Witt

Electronic Theses and Dissertations

The structure of a protein molecule is highly correlated to its function. Some diseases such as cystic fibrosis are the result of a change in the structure of a protein so that this change interferes or inhibits its function. Often these changes in structure are caused by a misfolding of the protein molecule. To assist computational biologists, there is a database of proteins together with their misfolded versions, called decoys, that can be used to test the accuracy of protein structure prediction algorithms. In our work we use a nested graph model to quantify a selected set of proteins that …

Differentiating Between A Protein And Its Decoy Using Nested Graph Models And Weighted Graph Theoretical Invariants, Hannah E. Green May 2017

Differentiating Between A Protein And Its Decoy Using Nested Graph Models And Weighted Graph Theoretical Invariants, Hannah E. Green

Electronic Theses and Dissertations

To determine the function of a protein, we must know its 3-dimensional structure, which can be difficult to ascertain. Currently, predictive models are used to determine the structure of a protein from its sequence, but these models do not always predict the correct structure. To this end we use a nested graph model along with weighted invariants to minimize the errors and improve the accuracy of a predictive model to determine if we have the correct structure for a protein.

Cranial Morphological Distinctiveness Between Ursus Arctos And U. Americanus, Benjamin James Hillesheim

Electronic Theses and Dissertations

Despite being separated by millions of years of evolution, black bears (Ursus americanus) and brown bears (Ursus arctos) can be difficult to distinguish based on skeletal and dental material alone. Complicating matters, some Late Pleistocene U. americanus are significantly larger in size than their modern relatives, obscuring the identification of the two bears. In the past, fossil bears have been identified based on differences in dental morphology or size. This study used geometric morphometrics to look at overall differences in cranial shape and used step-wise discriminant analysis to identify specific characters that distinguish cranial morphology between …
