One of the main aims of inquiry is to engage students as active, not passive, participants in science. The purpose of this study is to describe science educators’ and students’ views about inquiry-based instruction in order to better understand and improve implementation of evidence-based teaching strategies. Inquiry-based techniques have been shown to improve student understanding of scientific concepts, yet, there continue to be challenges in implementing these techniques. 2,5-diaminoimidazolone (Iz) is an important product of a 4-electron oxidation of guanine. The present research focuses on the mechanisms of formation of Iz via pathways initiated by guanine oxidation by one-electron oxidants (OEOs) generated by X-ray radiolysis in aqueous solutions. The kinetics of formation and yields of Iz in reactions of native highly polymerized DNA with different OEOs have been compared using an HPLC-based quantitative analysis of low-molecular products generated from the reaction of DNA-bound Iz with primary amines. Pseudomonas aeruginosa is an important opportunistic pathogen. Many P. aeruginosa virulence factors are regulated by the AlgZ/R two component system. AlgZ is the sensor histidine kinase which phosphorylates AlgR, the response regulator. AlgR activates transcription of different gene targets based upon its phosphorylation state. The genes that encode AlgZ and AlgR are transcribed in an operon. While regulation of algR expression has been well studied, regulation of algZ expression has not. Using a pilW mutant in concert with algZTF-lacZ transcriptional fusion, we conducted a transposon mutagenesis to identify algZ regulators. We identified an unknown autoregulatory loop. The type …

The present study used a mixed methodology approach to understand factors that predict and promote health related quality of life (HRQOL) for parents of youth with Autism Spectrum Disorders (ASD). The first component of the study used quantitative analyses to determine the extent to which study variables predict HRQOL for parents of youth with ASD (N = 365). Findings show that HRQOL is positively impacted by increased total annual household income, increased age of the child with ASD, male gender, and tangible support. Atherosclerosis is a chronic inflammatory disorder of medium and large vessels. Immune signaling and dyslipidemia are two of several processes which influence lesion development in atherosclerosis. Cannabinoids, such as those found in marijuana, exert their effects through two cannabinoid receptors, CB1 and CB2. Recent studies using CB2 knockout mice and CB2-selective ligands have shed light on a protective role of CB2 in early stages of atherosclerosis. The purpose of this quantitative study was to determine if a relationship existed between the quality of school-community partnerships and the average score of each ACT subtest for rural Tennessee high school students. Specifically, the researcher examined the following school-community partnerships: business partnerships, university partnerships, service learning partnerships, school-linked service integration, and faith-based partnerships (Sanders, 2006). Administrators from 62 rural Tennessee high schools rated the partnerships present in their schools using the Improving Community Partnership Quality rating scale developed by Sanders (2006). Abstract

Wildlife biodiversity was estimated amongst three loblolly pine (Pinus taeda) intra-plantation edge types of contrasting rotational phases in a two-year case study. Three structurally varied rotational phases that occur throughout plantation rotations combine to make three different edges: A= pre-thinned - post-thinned, B= pre-thinned - pre-clearcut, and C= post-thinned - pre-clearcut. Although black bears (Ursus americanus, Ursus americanus luteolus) were once found throughout the south-central United States, unregulated harvest and habitat loss resulted in severe range retractions and by the beginning of the twentieth century populations in Oklahoma, Louisiana, Texas and Arkansas were nearing extirpation. In response to these losses, translocation programs were initiated in Arkansas (1958-1968 & 2000-2006) and Louisiana (1964-1967 & 2001-2009). These programs successfully restored bears to portions of Louisiana and Arkansas, and, as populations in Arkansas began dispersing, to Oklahoma. In contrast, east Texas remains unoccupied despite the existence of suitable habitat in the region.

Since 1974, Luminant Mining Company, LLC (Luminant) has planted over 38.7 million trees on its reclaimed lignite surface mine operations in Texas. For decades, the use of improved reclamation techniques on Luminant’s mined lands have resulted in over 31,160 ha reclaimed to forests, wildlife habitat, and pastures with productivity levels similar to those found on unmined lands. The development of new reclamation methodologies offers opportunities to further improve productivity of planted trees at Luminant’s Martin Lake Oak Hill Mine in east Texas. The objective of this internet-based survey was to investigate the effects of positive religious or spiritual coping strategies on teachers’ chronic pain reports. Teachers in the United States may represent a vulnerable group due to a high prevalence of risk factors for chronic pain conditions. Teachers have been identified to experience high stress (Johnson, et al., 2005; Kyriacou, 2001) and report poor job satisfaction (Wang, Hall, & Rahimi, 2015), which are associated with development of chronic pain conditions (Kopec & Sayre, 2004). Although individuals may not engage in racism consciously due to social undesirability, they may have biases that manifest in more subtle and unconscious ways. Research has found that people of color experiencing different racial and ethnic microaggressions in their everyday lives are subject to pervasive and negative impacts on their mental health, though little is known about the influence of one’s ethnic identity formation, affective processes, and cognitive processes. Three experiments evaluated pinching and spacing on Helianthus annuus in East Texas to determine their effects on growth and development. Experiment 1 was designed to determine the effects of pinching nodes 1, 2, 3, or 4 on ‘Pro Cut Gold’ sunflowers. Results from experiment 1 showed non-pinched plants produced marketable stem lengths and flower sizes, while pinched plants’ stem length, stem diameter, flower diameter, and disk diameter decreased compared to the non-pinched plants. Phase-resolved ocean surface wave elevation maps provide important information for many scientific research areas (e.g., rogue waves, wave-current interactions, and wave evolution/growth) as well as for commercial and defense applications (e.g., naval and shipping operations). The goal of this project was to evaluate growth and nutritional characteristics of seven forages, including various warm season native grasses, grown under simulated partial shading (50%) typical of a loblolly pine silvopastoral system in East Texas. Primordial germ cells (PGCs) are hypothesized to deposit hematopoietic stem cells (HSCs) along their migration route through the embryo during the early stages of embryogenesis. PGCs also undergo global chromatin remodeling, including the erasure and reestablishment of genomic imprints, during this migration. While PGCs do not spontaneously form teratomas, their malignant development into germ cell tumors (GCTs) in vivo is often accompanied by the retention of hypomethylation at the IGF2-H19 imprinting control differentially methylated region (DMR). Gestational diabetes mellitus occurs in 4% of pregnancies and increases the risk of birth defects, pre-term birth, and miscarriage. Gingivitis during pregnancy also increases the risk for poor pregnancy outcome. Gingivitis is a bacterial-induced disease, and specific plaque pathogens have been associated with systemic sequelae to periodontal inflammation. Gold nanostructures exhibit technologically useful properties when they are polarized in an electric field. In two projects we explore instances where the polarized metal can be used in real world applications. The first project involves gold nanoparticles (GNP) for use in light actuated microelectromechanical systems (MEMS) applications. Although the GNPs were originally designed for volumetric heating in biomedical applications, we treat them as a thin film coating, opening the door for these particles to be used in MEMS applications. There are 5 Ubiquilin proteins (UBQLN1-4, UBQLN-L), which are evolutionarily conserved and structurally similar. UBQLN proteins have 3 functional domains: N-terminal ubiquitin-like domain (UBL), C-terminal ubiquitin-associated domain (UBA) and STI chaperone-like regions in the middle. Alterations in UBQLN1 gene have been detected in a variety of disorders including Alzheimer’s disease, Amyotropic Lateral Sclerosis and lung cancer. UBQLN1 has been largely studied in neurodegenerative disorders in the context of protein quality control. Signaling through Fas/FasL is critical to immune homeostasis and tolerance to self-antigens. SA-FasL is a chimeric protein of FasL and streptavidin. SA-FasL exists as oligomers with potent apoptotic function on Fas expressing immune cells and tightly binds to biotinylated surfaces. Islet grafts engineered to transiently display SA-FasL on their surface established tolerance in allogeneic recipients with a short course of rapamycin. Lung cancer is the most common malignant disease worldwide, and the rapid decline in functioning due to the often-later stage diagnosis can strongly impact a patient’s health-related quality of life (HRQOL). Palliative care, with the aim of improving later-stage HRQOL, is often a main goal of treatment. This dissertation examines how the field of public health addresses the impact of systemic racism on health and how that informs public health’s approach to youth violence prevention. Beginning with an overview of youth violence, it breaks down the concepts of race and racism and how they are addressed within the science. Fungi that can switch from budding to a filamentous infectious state have evolved mating type loci. Ustilago maydis, the maize pathogen, must mate with compatible partners possessing different alleles at two mating type loci for successful host infection. The a locus encodes pheromones and receptors, while the b locus encodes subunits of a heterodimeric transcription factor that regulates expression of virulence genes. Fibrinogen is the most abundant protein involved in blood coagulation and has been associated with many pathological implications in cardiovascular disease. At the final stages of blood clot formation, the transglutaminase Factor XIIIa introduces γ-glutamyl-ε-lysinyl covalent bonds between reactive glutamines and lysines in fibrin, which results in a tighter clot network that is resistant to fibrinolysis. Factor XIIIa crosslinks specific reactive glutamines on fibrinogen, selecting more reactive glutamines in the αC region of fibrinogen than any other chain. A Copy Number Variation (CNV) detection problem is considered using Circular Binary Segmentation (CBS) procedures, including newly developed procedures based on likelihood ratio tests with the parametric bootstrap for models based on discrete distributions for count data (Poisson and negative binomial) and a widely-used DNAcopy package. Results from the literature concerning maximum likelihood estimation for the negative binomial distribution are reviewed. Magnetic nanoparticles (MNPs) are used in a variety of applications, including as agents for magnetic resonance imaging, generation of local hyperthermia, and as platforms for drug delivery. Iron-based MNPs are often coated with a shell, such as silica or gold, to increase biocompatibility for drug delivery applications. The heat equation with a terminal condition problem is not well-posed in the sense of Hadamard so regularization is needed. In general, partial differential equations (PDE) with terminal conditions are those in which the solution depends uniquely but not continuously on the given condition. In this dissertation, we explore how to find an approximation problem for a nonlinear heat equation which is well-posed. Pseudouridine synthases (Ψ synthases) are the enzymes that catalyze the isomerization of uridine (U) to pseudouridine (Ψ), which is the most prevalent post-transcriptional modification of RNA. The Ψ synthases fall into six different families that share no significant global sequence similarity; however, they all involve a conserved aspartic acid residue which is absolutely essential for activity. Tyrosine is a conserved residue in the active site in five of the six families of Ψ synthases (phenylalanine in the TruD family) and was hypothesized as the general base for the isomerization reaction.
