Neutrophils respond to bacterial LPS through Toll-like receptor 4 (TLR4), which activates or potentiates immune defensive functions and prolongs cell survival. Activation of TLR4 signaling in neutrophils is beneficial for effective clearance of LPS-bearing Gram-negative bacteria, but may also drive aberrant inflammation if not stringently regulated. The regulatory processes by which neutrophil functions are calibrated to respond appropriately to different LPS-bearing bacteria are incompletely understood. Protein nuclear magnetic resonance spectroscopy (Protein NMR) is an invaluable analytical technique for studying protein structure, function, and dynamics. There are two major types of NMR spectroscopy that are used for investigation of protein structure – solution-state and solid-state NMR. Solution-based NMR spectroscopy is typically applied to proteins of small and medium size that are soluble in water. Solid-state NMR spectroscopy is amenable for proteins that are insoluble in water. “Articulating the New Normal(s): Mental Disability, Medical Discourse, and Rhetorical Action” studies the writing of people diagnosed with autism and post- traumatic stress disorder within online discussion boards related to mental health and outlines their unique rhetorical strategies for interacting with biomedical ideologies of psychiatry and activist discourses. This study furthers understanding of how trauma exposure may be connected to the development and maintenance of poor health by focusing on cytokines, tightly regulated proteins of the immune system. Cytokine responses to acute stress have been associated with the onset of poorer mental health in physically healthy women. The present study examined how two factors recently associated with cytokine reactivity –cumulative trauma and emotion reactivity– are associated with salivary cytokine reactivity among healthy women. Colorectal cancer (CRC) is the third leading cause of cancer-related deaths within the United States. Familial adenomatous polyposis (FAP) is an inherited disorder which if left untreated will develop into colon cancer. The family of plant-derived compounds, anthocyanins, show significant therapeutic potential against a variety of diseases, however, they are limited by their instability and poor bioavailability. In the first project, I apply a Bayesian hierarchical model to assess the stability of phosphorylated proteins under short-time cold ischemia. Traditionally, conceptual models of racial discrimination have characterized the reactive experiences of African Americans, particularly identifying how African Americans cognitively, emotionally, and behaviorally respond to racial stress. The current study extends beyond the reactive coping experience and identifies nuances in the anticipatory and preparatory coping processes associated with racial discrimination. Methods: 62 African American college students participated in a stress induction experiment that prompted anticipatory judgments of discrimination. Porphyromonas gingivalis, an obligate anaerobic bacterium associated with chronic periodontitis, utilizes various virulence factors to achieve pathogenicity, one of which is the Mfa1 fimbriae. As a surface structure comprising Mfa1 major subunit along with accessory fimbrial proteins Mfa2-5, the Mfa1 fimbriae has been shown to mediate the adherence of P. gingivalis to antecedent bacterial colonizers of the oral cavity to cause increased virulence. Earthen barriers such as CCLs and GCLs have been employed in geotechnical practices to provide a low permeability hydraulic barrier since long time ago. These types of barriers exhibited satisfactory performance for many applications such as landfills. The performance of low permeability barriers is based on the swelling potential of their component, which is mostly Na-bentonite, in contact with polar fluids such as water. The fungus, Ustilago maydis, a plant pathogen of Zea mays, is capable of a dimorphic switch, e.g., a transition between a yeast-like form and a filamentous form. This switch is activated when U. maydis cells are starved for their preferred nitrogen source - ammonium. U. maydis has two important genes that play a role in this switch, ump1 and ump2. Each gene is responsible for the uptake of ammonium and the latter is responsible for the filamentous response in the absence of ammonium. Spectral clustering is often used to partition a data set into a specified number of clusters. Both the unweighted and the vertex-weighted approaches use eigenvectors of the Laplacian matrix of a graph. Our focus is on using vertex-weighted methods to refine clustering of observations. An eigenvector corresponding with the second smallest eigenvalue of the Laplacian matrix of a graph is called a Fiedler vector. Coefficients of a Fiedler vector are used to partition vertices of a given graph into two clusters. The National Center of Education Statistics (2010) reported that cyberbullying incidents increased 73% between the years of 2007 and 2009. Sympathetic nerves are important for renal physiology and sepsis pathophysiology. A recent study showed sprouting of sympathetic nerves in spleen of septic mice. This study was done to test if renal sprouting of sympathetic nerves also happens and to investigate renal morphology in septic mice. Cecal ligation and puncture (CLP) was used to induce sepsis and kidneys were removed for evaluation. Bowman’s space was diminished with cortical bubble cells present suggestive of acute renal pathology, however, renal function was unchanged. The coexistence of species is probably one of the most interesting and complex phenomenon in nature. We constructed an agent based model to study the coexistence dynamics of prey - predator populations by varying productivity levels of producers in fragmented and connected habitats along with different levels of quality of predators. Our results indicated that productivity levels of producers in fragmented and connected habitats along with levels of predator quality are significantly responsible for overall predator - prey population size and survivorship. Arsenic is a naturally prevalent metalloid. Chronic arsenic ingestion through drinking water causes skin cancer. Arsenic-induced cancer mechanisms are not well defined. Epigenetic changes, including microRNA expression changes, might be playing a role. This dissertation investigates the impact of miRNA expression changes in arsenic-induced skin cancer. MiRNA expression was measure and compared using 3 different techniques, RTq-PCR, hybridization arrays and RNA-sequencing. MiRNAs differential expression in skin lesions was phenotype- and stage-related. Immortalized human keratinocytes (HaCaT) were transformed by chronic low arsenite exposure serving as a model for arsenic-induced skin carcinogenesis. The artist discusses the work for his Masters of Fine Arts exhibition, Mind under Matter, held at the Tipton Gallery in downtown Johnson City, Tennessee. Exhibition dates are from March 27th through April 5th 2017. ‘Matter under Mind’ explores the balance of control and non-control within the art-making process. This technique creates an automatic dialogue resulting in abstraction guided by the subconscious. The purpose of this quantitative study was to compare measures of teacher effectiveness between two different Tennessee teacher evaluation models. The two teacher evaluation models compared were the Tennessee Educator Acceleration Model (TEAM) and the Teacher Instructional Growth for Effectiveness and Results Model (TIGER). The measures of teacher effectiveness used were final observation scores and individual value-added growth (TVAAS) scores. The purpose of this study was to explore teacher and parent perceptions of homework effectiveness and homework purpose. Specifically, the researcher examined teacher and parent perceptions, perceptions of teachers in different grade level bands, and perceptions of teachers with different levels of experience. In this nonexperimental quantitative study, the researcher examined and analyzed the answers provided on an anonymous survey (Appendix A) provided electronically to educators and parents of students in five school systems in northeast Tennessee.

Toll-like receptor (TLR)-mediated pathways have been demonstrated to play a role in myocardial ischemia/reperfusion (I/R) injury. We and others have shown that PI3K/Akt signaling is involved in regulating cellular survival and protecting the myocardium from I/R induced injury. In this dissertation, we provide compelling evidence that miR-125b serves to “fine tune” TLR mediated NF-kB responses by repressing TNF-a and TRAF6 expression. We constructed lentiviral expressing miR-125b, delivered it into the myocardium. Predator cues can influence how aquatic organisms respond to anthropogenic contaminants. This study examined the effects of predator cues on behavior, metabolic rate, and sulfate (as Na2SO4) toxicity in amphipods (Gammarus minus). Predator cues included alarm cue (macerated conspecifics) and kairomone from mosquitofish (Gambusia affinis). Amphipods decreased activity and increased time in refuge when exposed to alarm cue, and increased time in refuge when exposed to kairomone. While median lethal concentrations (96-h LC50) were not influenced by predator cues, analysis of dose response curves indicated that kairomone exposure increased amphipod …

Groundwater tracing studies have shown a well-developed conduit system in the karst aquifer beneath the Buffalo Creek Watershed of Carter County, TN. Groundwater can quickly travel beneath topographic divides, transporting contaminants. Using GIS and field reconnaissance, two sites were selected for a two phase dye trace study. In May-June 2016 groundwater resurgences were located using activated carbon samplers, and in October-December 2016 flow velocities were measured using ISCO automatic water samplers. The study employed Geographical Information System (GIS) technology to investigate the vulnerability of groundwater to NO3 content in Buncombe County, North Carolina in two different approaches. In the first study, the spatial distribution of NO3 contamination was analyzed in a GIS environment using Kriging Interpolation. Cokriging interpolation was used to establish how NO3 relates to land cover types and depth to water table of wells in the county. This thesis will argue that the Beowulf Manuscript, which includes the poem Judith, Saxo Grammaticus’s Gesta Danorum, and the Old-Norse-Icelandic Laxdæla saga highlight and examine the tension between the female pagan characters and their Christian authors. These texts also demonstrate that Queenship grew fragile after the spread of Christianity, and women’s power waned in the shift between pre-Christian and Christian Europe.
