The formation of a plasma membrane furrow is an essential process during development. Furrow formation is necessary for successful cell division and cytokinesis in addition to the ability to create multicellular tissues. Here, I will explore the role of the Golgi-associated Rab protein Rab39 in furrow formation during early Drosophila embryogenesis. Rab39 is one of eight Rab proteins that has been shown to localize to discrete puncta by live imaging in early Drosophila embryos, but its function and pathway have not been well characterized. Students who have contact with the juvenile justice system are a particularly vulnerable and unique school-aged population. The risk factors that plague the likelihood of justice-involvement are numerous and inter-connected. Early experiences of trauma and adversity, limited familial and financial capital, and challenges with mental health all contribute to increased likelihood of youth contact with juvenile justice systems. Despite said risk factors effects on young people overall, youth of color are particularly susceptible to become justice-involved. School and community discipline statistics are grossly, racially disproportionate.

This phenomenological study sought to examine the experiences of teachers in an urban K-8 school after a system-wide whole school implementation of trauma-informed practices. The practices teachers implemented in their classrooms that aligned with their personal perceptions of trauma-informed practices and its efficacy were explored. Additionally, the personal and professional barriers to implementation were also investigated. Identified practical strategies at both the elementary and middle school levels included establishing and maintaining relational trust and classroom community, actively teaching emotional regulation skills, and teaching and reinforcing rituals, routines and expectations throughout the school year. Large dense core vesicles (LDCVs) form at the trans-Golgi network (TGN) and mediate the regulated release of neuropeptides and peptide hormones. Despite their central role to physiology, the mechanisms controlling biogenesis and sorting to LDCVs is not well understood. Optimizing the retention using selective hooks (RUSH) method in neuroendocrine cells, we show it is possible to visualize sorting to the constitutive and regulated secretory pathways in real-time and that the bulk of transmembrane LDCV cargoes do not sort directly onto LDCVs, but exit the TGN into non-regulated vesicles to be incorporated to LDCVs at a later step. This qualitative study used a phenomenological approach to capture the lived experience of non-relational foster families navigating the Colorado public education system. While most research on children in foster care looks at all sub-populations of foster care combined, this is the first known study to focus solely on non-relational foster families navigating the education system. MftC is a radical S-adenosyl L-methionine (SAM) enzyme that catalyzes the first step in the production of mycofactocin. It is a ribosomally synthesized post-translationally modified (RiPP) redox cofactor shown to be essential for the survival of bacteria in the Mycobacterium genus in the presence of cholesterol as a carbon source and for the sequestration of ethanol. MftC catalyzes the C – terminal decarboxylation of tyrosine and the subsequent cross-linking of tyrosine to the penultimate valine on the precursor peptide MftA. Epithelial tubules form critical structures in various body tissues; how- ever, since they are difficult to access experimentally, their architecture and dynamics are not well understood. Here we examine the dynamic remodeling of epithelial tubes in vivo using a novel and uniquely accessible model system: the extracorporeal vasculature of Botryllus schlosseri (sea squirt). In Botryllus, massive retraction of blood vessels can be triggered without loss of barrier function, through (i) disrupting collagen crosslinking in the basement membrane using β-aminopropionitrile (BAPN); or (ii) disrupting the integrin pathway through inhibition of focal adhesion kinase (FAK). Rock glaciers are common landform features found in deglaciated alpine areas. They are commonly used in the study of climatic changes throughout the Holocene and the reconstruction of neoglacial chronologies. For this research, Schmidt hammer rebound values, weathering rind thicknesses, and the length of lichen thalli diameters found on rock glacier surfaces are used to investigate their effectiveness as field-based relative age determination techniques. In spite of efforts made to improve the accuracy of FER systems, existing methods still are not generalizable and accurate enough for use in real-world applications. School psychologists pay a critical role in providing assessment and intervention services within the realm of special education. Within this role, they are highly likely to interact with students with intellectual and developmental disabilities (IDD). Students with IDD are characterized by significantly lower than average cognitive and daily living skills that may be comorbid with difficulties with communication, social skills, or other domains. Purines are a class of nitrogenous bases and are essential small molecules to life. Purines are used within the cell as genetic information carriers, energy currency, signaling molecules, and cofactors for multiple processes. They are formed through de novo and salvage pathways found in cells across the phylogenetic tree. The substrates of enzymes within de novo purine synthesis are known to influence other processes within the cell, such as energy homeostasis. Students with emotional disabilities are disproportionately suspended and expelled in K-12 schools. Attribution theory suggests individuals are less likely to provide assistance to others if they believe the individuals are responsible for their own difficulties. To test attribution theory, this study created new measures of explicit attitudes and implicit associations of licensed 6-12th grade staff regarding students with depression as well as a helping behavior measure of staff toward students with depression. Stigma has been associated with physical and mental health conditions and their treatment long before formal care systems. Negative perceptions within the lay community, such as stigma and social distancing, negatively impact individuals with autism and their families. The purpose of this study was to explore factors associated with autism stigma among laypeople in the Southern United States. Apart from an evolutionary role, transposable elements have been implicated in animal development and also in pathophysiology. Non-LTR retrotransposons– LINE-1, Alu and SVA - are responsible for over 120 cases of human genetic diseases as heritable insertions, and are emerging as an important etiological factor for cancer and neurological disorders as somatic mutations. It is estimated that among the total number of 500,000 LINE-1s presents in the human genome, 80-100 LINE-1s remain competent for retrotransposition. Enteric bacteria that are resident in the hindgut of mammals are critical in immune development, digestion, and colonization resistance against pathogens. The microbial attachment and colonization on separation membranes lead to biofilm formation. Some selective isolates within the biofilm constitutive microflora acquire resistance and emerge predominant over prolonged use of the membrane. Thus, proving the cleaning and sanitization protocols to be ineffective in adequately removing resilient biofilms. The overall objective of this dissertation was to determine the impact of selected post weaning management strategies on beef carcass characteristics and meat quality. In Lake Oahe, a large mainstem Missouri river reservoir that spans 375 river kilometers in central North and South Dakota, naturalized Rainbow Smelt are the primary forage species for a regionally important Walleye Sander vitreus fishery. Walleye condition in Lake Oahe is highly correlated to Rainbow Smelt relative abundance through time, the latter species being typified by erratic abundance trends since accidental introduction in 1971. Cisco Coregonus artedii were introduced in the early 1990s to augment the cold-water prey base. This research aimed to understand the evolutionary mechanism of seed dormancy and flowering time and to provide basic knowledge for the manipulation of germination features in crop breeding. Flowering time and seed dormancy are two adaptive traits for flowering plants. Seed dormancy (SD) helps regulate the timing of germination to promote the survival of seed-bearing plants in adverse environments. Cereal crops and weedy/wild relatives, such as Asian cultivated rice and weedy rice (Oryza sativa L.), have weak and strong seed dormancy, respectively. Historically, pheasants (Phasianus colchicus) have thrived in South Dakota in conjunction with successful land retirement programs or early farming practices through the 1950s, which created interspersions of agriculture and native landscapes that were ideal for pheasants. Recently, the Prairie Pothole ecosystem has undergone rapid agroeconomic expansion, effectively reducing ideal interspersions of native prairie and cropland into agriculturally homogeneous landscapes. Indices of pheasant abundance have suggested persistent population declines since 2008, raising concerns regarding landscape suitability.
