Biological Nitrogen Fixation is the process of conversion of atmospheric dinitrogen into ammonia which is performed by symbiotic as well as free-living diazotrophic bacteria. A previous project to isolate free-living nitrogen fixing bacteria from a natural prairie yielded Streptomyces which grew on nitrogen free medium. Genome sequencing of three isolates revealed the presence of a second 16S rRNA gene sequence aligning with Bacillus, however repeated streaking only yielded Streptomyces colonies. The aim of this research was to characterize the interaction of three Streptomyces isolates that grow under nitrogen-free conditions, and appeared to be associated with Bacillus. Mountain Sucker, Pantosteus jordani, is a cold-water species native to the Intermountain West. Fringe populations of Mountain Sucker have experienced declines in recent decades. The population of Mountain Sucker found in the Black Hills of South Dakota represents the eastern fringe of the species’ native range. Recognized as both an indicator of biologic health and as a species of greatest conservation need in South Dakota, recent studies have suggested significant declines in both distribution and abundance. Higher and stable yield is always a major objective of wheat genetic improvement programs. The SQUAMOSA-promoter binding protein-like (SPL) genes constitute a small family of plant-specific transcription factors with diverse functions in plant development and growth and have great potential in improving yield and other major agronomic traits. However, the functional characterization of the SPL gene family in wheat is far behind other cereal crops such as rice. Using phylogenetic analysis, we identified 56 wheat orthologues of rice SPL genes belonging to 19 homoeologous groups. Soybean cyst nematode (SCN) is a microscopic worm that infects and feeds from the soybean roots, hence robbing the plant of nutrients, and entry wounds may facilitate other pathogens to infect the roots. Current SCN management practices include cultural, chemical, and biological methods. Influence Of Mink Predation On Brown Trout Survival And Size-Structure In Rapid Creek, South Dakota, Austin Galinat

I compared movement, home range, habitat selection, and mortality among radio-tagged Brown Trout (Salmo trutta) at an American Mink (Neovison vison)-removal site, improved habitat site, and an undisturbed stream site in Rapid Creek, South Dakota. I observed high, post-surgery (> 26 days) survival rate (90%) of radio tagged Brown Trout. Average gross movement of Brown Trout was greater at the improved habitat site (127 m) compared to the mink removal (31 m) site, while average home range size (i.e., stream distance) among all three sites was not significantly different indicating strong site fidelity (125 to 200 m). Transposable elements, also called jumping genes, comprise almost 45% of the human genome. In contrast, only 1% of the human genome is protein-coding sequences. The function and advantages of maintaining such massive copies of transposable elements in the human genome are still unclear. Long interspersed element 1 (L1), the most substantial group and the only active autonomous transposable element in the human genome, has revealed its unique roles in many diseases. This work combines traditional microbiology with bioinformatic and synthetic biology approaches to study antibiotic tolerance. Antibiotic tolerance is a widespread phenomenon that facilitates antibiotic resistance and decreases the effectiveness of antibiotic treatment. Tolerance is distinct from antibiotic resistance, because tolerance is short term survival and typically results from phenotypic variations rather than genetic variation. The molecular mechanisms underlying tolerance are varied and debated in the literature. I have explored two intracellular processes related to tolerance, toxin-antitoxin (TA) systems (Chapter 2) and proteases (Chapter 4). This thesis seeks to understand how newlyweds discuss their plans for family expansion among each other and to others by applying the theory of communication privacy management. Ten newlywed couples were interviewed regarding their experiences navigating the topic of family expansion and reproduction. The purpose of this paper is to describe teachers’ perceptions of experiential learning and its effects on student learning, understanding, and engagement through agriculture education's three-circle model. This paper explores teachers’ perceptions of the value and importance of experiential learning in agriculture education instruction, FFA, and supervised agricultural experience. Further, I explore experiential learning, constructivism, and hands-on learning as the foundational instructional practices used in agricultural education. Agricultural education teachers from southwest Minnesota were surveyed to determine how they are using experiential learning in their agriculture education programs, including classroom instruction, FFA, and supervised agricultural experiences.

Rising temperatures in the Arctic may affect vegetation, which in turn can affect herbivores, such as caribou, that rely on these plants for forage. Several plant traits contribute to forage quality, including digestibility, nitrogen content, and antiherbivory secondary compounds, but the effect of temperature on these traits individually and combined is unclear. Glyceollin is a secondary metabolite produced under stress conditions by soybean to act as a plant defender to pathogen attack. The antimicrobial nature of glyceollin makes it a promising natural alternative to antibiotics if incorporated into livestock feed. However, production of glyceollin is highly variable, dependent on soybean variety, and fungal elicitor used as inoculum. Our study compares two fungal spore elicitors, Trichoderma reesei and Aspergillus sojae on soybeans of diverse maturity level, and fungal susceptibility to determine highest glyceollin titer. As a result of their extended shelf-life, dried dairy ingredients constitute a significant category produced for global markets. During the development of dairy ingredients, several drying trials are typically conducted to determine optimum drying conditions. The results of these trials can be critical in determining optimum dryer design. However, these trials can be expensive and time-consuming. An alternative that has recently been developed using a new technique called single droplet drying (SDD).
