This dissertation examined and provided basis for addressing factors related to HIV pre-exposure prophylaxis (PrEP) engagement, outreach, and uptake among high-risk African American youth groups in Louisville, KY. This qualitative dissertation study was designed through the lens of an interpretive framework of social constructivism which holds that multiple realities and interpretations exist and are socially constructed through the lived experiences of individuals and their interactions with others. Thus, lived experiences of various African American youth groups in Louisville as well as key informants of AIDS service organizations (ASOs) across the U.S. (including Louisville) were explored to develop a deeper understanding …

Cardiovascular disease (CVD) is the leading cause of death worldwide. Tobacco smoke is the most significant modifiable risk factor in CVD development and contains numerous toxic compounds, including aldehydes, which have been linked to CVD. Formaldehyde, acetaldehyde, acrolein, and crotonaldehyde have been identified as significant contributors to cigarette-induced health effects, but the mechanism of these effects is not well understood. These aldehydes are also found in aerosols produced by e-cigarettes (e-cigs). Lassa Virus (LASV) and Venezuelan Equine Encephalitis Virus (VEEV) are two single stranded RNA viruses belonging to the Arenavirus and Alphavirus families, respectively. Both are emerging pathogens without approved vaccines or treatments. VEEV is an important biothreat pathogen because it has been weaponized and is extremely infectious as an aerosol. VEEV causes biphasic febrile illness that can progress to a viral encephalitis with low mortality, but high morbidity. LASV causes a viral hemorrhagic fever, Lassa Fever (LF), which is endemic in West Africa, and responsible for between 100-500k annual infections with a 1-2% overall mortality rate. This dissertation explores and expands the existing knowledge on the role of plant volatiles in facilitating seed priming and providing direct defense against herbivore. Although the roles of plant volatiles in priming a plant’s defenses and providing direct defense against pests is well known, information regarding their effects on seed and the fitness of future plants and direct toxicity to herbivores is understudied. Background. Acute kidney injury (AKI) develops in 30% of patients who receive cisplatin, a widely used chemotherapeutic agent. We previously showed that NHERF1 loss resulted in increased susceptibility to cisplatin nephrotoxicity. The overarching goal of this dissertation was to elucidate mechanisms of susceptibility to cisplatin-induced AKI, specifically the effects of NHERF1 loss on tubule cell metabolism, tubule cell mitochondrial function, and alterations in oxidative state and/or renal handling of cisplatin. Lung cancer is among the most frequently diagnosed cancers and is the leading cause of cancer-related deaths worldwide. One of the hallmark events in lung cancer pathogenesis is deregulation of the cell cycle. The retinoblastoma protein (pRb) is a tumor suppressor that is deleted, mutated, or inactivated in most lung cancer cases. Canonically, pRb functions to regulate cell cycle progression by repressing the transcriptional activity of the E2F family of transcription factors, inhibiting S phase entry. Although the cell cycle functions of pRb have been well established, recent studies have highlighted a functional role for pRb in controlling cellular metabolism. …

Organizations that have benefitted from the commission of federal crimes committed by their employees may be sentenced to federal criminal penalties. Two of these potential penalties include probation and the requirement to implement a corporate monitor. The federal guidelines provide suggestions for sentencing probation that echo the theoretical focal concern of blameworthiness. This research used eight years of United States Sentencing Commission data covering 2011 to 2018 (n= 1,224) to examine if organizations were being sentenced to probation and monitoring consistent with the federal guidelines and focal concern of blameworthiness. In the design of stream restorations, boundary shear stress (shear stress) and velocity during high flow events are the key parameters in the assessment of the risk of morphological failure associated with channel bank and bed erosion and sediment transport. The use of two-dimensional hydrodynamic models (2D Models) is becoming more common to estimate critical shear stress and velocity for stream restoration projects. These models can give detailed distribution of shear stress and velocity over floodplain surfaces and channel. Over two-thirds of the United States population have overweight or obese (OW/OB) weight statuses due in large part to an obesogenic environment that encourages unhealthful weight related behaviors. The obesogenic environment appears to place a larger burden on women with low incomes as they experience OW/OB disproportionately compared to other groups. Studies seeking to understand the impact of the obesogenic environment on this population have been deficit focused, largely examining environmental risk factors for OW/OB and ignoring protective factors against it. Yersinia pestis has gained widespread infamy due to the historic outbreak during the middle ages, referred to as The Black Death. Infection with Y. pestis typically begins with deposition of Y. pestis into the dermis (bubonic plague) or respiratory tract (pneumonic plague). Tissue resident macrophages are the first innate immune cell encountered by Y. pestis. Advances in culture-independent techniques have facilitated the identification of new bacterial species in periodontal lesions, such as the Gram-positive anaerobe, Filifactor alocis. Cigarette smoke contains thousands of harmful substances and is one of the leading preventable causes of mortality and morbidity in the United States. Past studies examining tobacco smoke exposures on fecundability and pregnancy outcomes are inconsistent. NAT2 is an important enzyme in the metabolism of xenobiotic substances found within tobacco smoke. Avian facial plumage, bill coloration, and feather microstructure may serve one or more adaptive functions. Several researchers have proposed that dark eyestripes, bills, and facial masks aid in reducing glare, however, there have been relatively few tests of this hypothesis. Dark facial markings have been shown to have an adaptive glare-reduction function in recent field studies of a few species, but this hypothesis has never been tested in a broad multispecies analysis. Rising atmospheric carbon dioxide (CO2) may affect plant/pathogen interactions. This study focused on the effects of elevated CO2 on Root-Knot Nematode (Meloidogyne arenaria) and Tobacco Mosaic Virus (TMV) infection in genetically resistant versus susceptible tomatoes (Solanum lycopersicum). Both resistant and susceptible tomatoes were grown in chambers with either ambient CO2 or CO2 elevated to 750 ppm and infected with M. arenaria or TMV. Post-exercise hypotension (PEH) is believed to play a major role in the antihypertensive health benefits of exercise. While it has been shown that resistance exercise is effective at eliciting PEH, commonly consumed sports supplements may attenuate or completely eliminate that response. Caffeine, a popular stimulant, is often consumed prior to exercise. The concentrations of metals (Ba, Cd, Co, Cr, Cu, Mn, Ni, Pb, Sr, V, Zn) were investigated in soil and water samples collected over four different sites in the southern part of Lake Nacogdoches over one year from August 2018 through June 2019. Water samples were analyzed using the USEPA method 200.8 part 11.2 for total recoverable metals. Soil samples were analyzed using the USEPA method 3050B for total recoverable metals. Although the total concentration gives some indication of the level of contamination, it is not enough to give information on the bioavailability or mobility of the element. A modified …

This study focuses on the quality of life of suicide attempt survivors and the trajectories of their lives after their last attempt in the past two to ten years. Employing both a quantitative and qualitative approach but focusing largely on the qualitative data, I collected demographic data, gathered responses on an abbreviated Reasons for Living Inventory, and conducted open-ended phone interviews with 26 participants. The primary life course finding is that participants’ suicide attempts are often built on years of dealing with mental illness. Novel designed metal ligand complexes as prodrugs are useful in the effort to find more effective and selective anti-cancer treatments. Carbon monoxide, CO, is known to induce mitochondrial collapse in cancer cells. Herein is described the synthesis of seven novel bipyridine flavonolate palladium(II) and platinum(II) complexes with the property of releasing carbon monoxide, and their characterization with FTIR, UV-Vis, fluorescence, NMR, and ESI mass spectra. Invasive species are a prevalent problem all over the world. Controlling and eradicating an invasive species is an even more diffcult problem. The Trojan Y Chromosome (TYC) eradication strategy is one control method. This method alters the female to male sex ratio by introducing sex reversed males called supermales. These sex reversed males can only produce male progeny. Mathematical models of this strategy have shown that a population can be driven to extinction with a continuous supply of these sex reversed males. The purpose of the following descriptive pre and post study was to collect and analyze data to determine if there are benefits to alternative therapies on adult mental health. Three types of alternative therapies were studied: aromatherapy, recreation therapy, and yoga. A literature review was conducted and pre and post questionnaires were administered. Frequencies and averages were run which showed that participants were mostly Caucasian, with an average age of 31 years and most diagnosed with depression and anxiety. Participants showed decreased levels of anxiety and depression after attending both aromatherapy and yoga classes. Participants showed decreased anxiety levels after …

The antigenic similarity between embryos and tumors has raised the idea of using embryonic materials as a preventative vaccine against neoplastic disease. Indeed, a previous study reported that a vaccine comprised of allogeneic murine embryonic stem cells (ESCs) and murine fibroblasts expressing immune-stimulatory granulocyte macrophage-colony stimulating factor (GM-CSF) successfully blocked the outgrowth of an implantable lung cancer (Lewis lung carcinoma; LLC) and lung tumors caused by a combination of a mutagen followed by chronic pulmonary inflammation. Salmonella appears in the bloodstream of mice in as little as 15 minutes after oral inoculation and establishes persistent colonies in the spleen and liver. While its pathway to blood is undetermined, this phenomenon is dependent on the activity of Salmonella pathogenicity island 2 (SPI-2) coded type III secretion system (T3SS) and CD18+ phagocytes. We hypothesize that dendritic cells associated with the basal face of the gut epithelium, that are naturally migratory and known to sample for luminal antigens directly transport Salmonella to the bloodstream. Historically, measures of coparenting have been reported by mothers. As a result, fathers’ perspectives and experiences have largely been excluded from this body of research. In response, this study relies on fathers own report of coparenting support. Emerging research has increasingly identified the detrimental effect of internalizing and externalizing comorbidity on the functioning of youth with Attention-Deficit/Hyperactivity Disorder (ADHD). Research in the broad child psychopathology literature has identified a variety of dispositional and developmental risk factors for psychopathology development in youth. However, a conceptual model of psychopathology development has yet to be developed and empirically evaluated in an ADHD sample. Background: Porphyromonas gingivalis adherence to Streptococcus gordonii may be important for P. gingivalis colonization in the oral cavity. Nanoparticles encapsulating synthetic peptide BAR (BAR-NPs) inhibit P. gingivalis adherence more potently than free BAR. However, BAR-NPs would exhibit low retention in an open flow environment. Hypothesis: Targeting BAR-NPs to the streptococcal surface using CafA protein will enhance their efficacy. Methods: CafA-modified NPs encapsulating BAR were synthesized using double emulsion approach. Surface binding and retention, and release kinetics of BAR from CafA-modified NPs was assessed. Using national dyadic data from the 2010 Married and Cohabiting Couples (MCC) survey, I examine the effects on relationship quality of union type: “direct marriage”, “marriage after cohabiting”, “cohabitation with plans to marry”, “cohabitation with no marriage plan”. In addition, I examine whether these effects are different for men and women. Consistent with prior research, I found that cohabitors without plans to marry report the lowest levels of relationship quality and those who married directly report the highest levels of relationship quality. This research examined the numerous fairness ordinances in Kentucky’s cities for repeated trends, patterns, and practices in their language. Through the use of a cumulative case study, cases were selected from the ordinances as well as state and federal law, judicial actions, and other relevant factors from within the state. Each of the ordinances were examined for generalizability and their ability to be applied to a single, unified state nondiscrimination law that conforms with existing state and federal law. Lung cancer patients experience high levels of distress, which commonly manifests as depressive symptoms. Importantly, depressive symptoms have demonstrated prognostic significance in cancer contexts, although the biological pathways by which depressive symptoms lead to poorer survival remain unclear and warrant greater attention. In addition to understanding the biological pathways by which depressive symptoms accelerate disease, identification of efficacious and effective psychological treatments for depressive symptoms are needed to improve both quality and quantity of life for cancer patients. Interventions that reduce depressive symptoms and improve downstream clinical outcomes are certainty needed; however, because many cancer patients face considerable burden, interventions …
