Paraspeckles are a relatively newly identified subnuclear body whose function is still largely unknown. They are membraneless organelles found within mammalian nuclei originally identified as being comprised of different paraspeckle proteins (PSPs), and later proven to also contain RNA. This study investigates the long non-coding RNA nuclear enriched abundant transcript 1 (NEAT1), which has been found to be indispensable in the formation of paraspeckles, with each containing up to 50 NEAT1 molecules. NEAT1 has two isoforms, the shorter NEAT1_1, and the longer NEAT1_2. The focus of this work will be on the NEAT1_2, which acts as a scaffold in the …

A network of site-specific single Cys-mutations coupled with CX-MS can be used to elucidate a more refined structure of GlyR and obtain a more definitive understanding of pentameric ligand-gated ion channel (pLGIC) allostery. Each Cys-mutant is introduced into an a1 homomeric Cys null background (C41S/C290A/C345S), or in the same background with F207G/A288G mutation that allows non-desensitizing GlyR activation by ivermectin (IVM). State-dependent crosslinking with methanethiosulfonate benzophenone (MTS-bzp) to a single thiol of purified, vesicle reconstituted GlyR are conducted after enriching the receptor in different allosteric states: resting (no ligand), open (F207G/A288G + IVM), or desensitized (excess glycine). Research has shown that people in the teaching profession have high levels of stress and often report increased levels of symptoms of anxiety and depression. Students’ academic progress may have adverse effects when their teachers have high-stress levels or report mental distress. Those who teach special education often face high levels of stress. However, there is little research considering special education teachers’ job-related health. This study collected data on 598 general and special education teachers in the United States. This research study aimed to understand how successful Texas educators who grew up in poverty understood and improved the educational experience of economically disadvantaged students. This study utilized a structural ecological theory and three theories of social relationships (Social Identity, Standpoint, and Cultural Capital). This phenomenological and qualitative study used a cross-sectional, descriptive, online case study design rooted in narrative nonfiction. Virtual interviews with six successful Texas educators that grew up in poverty were conducted. A narrative method of analysis was utilized to generate codes then organize them into themes, and to construct and compare the narrative findings. The results …

Striated Muscle UNC-45, also known as UNC-45b, is an important protein that acts as a chaperone for myosin in cardiac and skeletal muscles, binding to myosin at its C-terminal UCS domain and regulating its assembly into thick filaments and sarcomeric structures. The UCS domain contains a large loop that is believed to be the first point of interaction between myosin and UNC-45b. GATA-4 is an essential transcription factor that facilitates transcription of several genes in cardiac development, particularly alpha-heavy chain myosin in heart tissue. Native bees are an important part of terrestrial ecosystems due to their coevolution with flowering plants. This study catalogued the bee fauna of two sandyland sites in the Big Thicket National Preserve and assessed whether a community was impacted by a historic hurricane-induced flooding event. It was hypothesized that a change in diversity metrics would be evident following the flood. Datasets were analyzed for differences in species richness, abundance, evenness, and Shannon’s diversity. Despite the success of immunotherapy in many types of cancer, pancreatic adenocarcinoma (PDAC) has yet to benefit. Innate immune cells are critical to antitumor immunosurveillance and recent studies have revealed that these populations possess a form of memory, termed trained innate immunity, which occurs through transcriptomic, epigenetic, and metabolic reprograming. Though trained innate immunity has mostly been investigated in the context of infection, the induction of trained innate immunity could also protect against tumors, and specifically pancreatic tumors. Bacteriocins are narrow-spectrum antibiotics produced in nearly all lineages of bacteria, meaning that these antimicrobials target closely related individuals. The bacteriocins of Pseudomonas aeruginosa, called pyocins, are highly prevalent and diverse in populations of this species. Laboratory studies have shown that pyocins can function to mediate the outcome of interactions, often allowing for the coexistence of multiple strains, as no one pyocin genotype is competitively superior. Although this has been demonstrated under laboratory conditions, the function of pyocins in natural settings and the ecological, evolutionary, and genetic mechanisms underlying pyocin diversity remains unclear. Although exposure to PM2.5 is an established risk factor for CVD, the contribution of other hazardous pollutant exposure to CVD is less clear. Overall, this work aimed to examine the effect of pollutants with lesser documented effects on cardiovascular disease using a multi-pronged approach to exposure assessment. Pathogens may enter surface waters as they are shed in human feces and potentially delivered to surface waters via sewer overflows, particularly in the eastern United States. This study examined of the presence of fecal indicators in two forks of Beargrass Creek in Louisville, Kentucky. Surface water grab samples (N=30), sediment samples, water quality, and a paired wastewater surveillance study were analyzed. SARS-CoV-2 was detected in a single stream sample despite ubiquitous presence of the virus within the area in wastewater and consistent evidence of a human fecal indicator. Butterfly populations are declining, and habitat degradation due to urbanization is a major contributing factor. Gardens represent a large proportion of land area in many cities, and thus may be important for conserving butterflies in urban environments. In this dissertation, I examine how garden features (ex: size, level of urbanization, plant diversity) affect adult butterfly diversity/abundance, behavior, and the predation risk faced by caterpillars. The frequency of harmful algal blooms (HABs) has been increasing across the globe, especially in river ecosystems. Although more common in lakes, more HABs have been reported in rivers in the past decade, particularly in the Ohio River. In 2019, a HAB stretched 200 to 300 miles throughout the Ohio River from September to October. This dissertation exposes one manifestation of the hidden curriculum of gender bias in the biological academe and explores the impacts of implicit essentialist perspectives on biology curricula and student understanding of an important evolutionary concept. Chapter one introduces the concept of gender essentialism and its relationship to the hidden curriculum of gender bias. I conclude the introductory chapter by reviewing tenets from queer curriculum theory and suggesting that applications of the theory may provide educators with the pedagogical tools required to counter implicit essentialist perspectives. Alphaviruses are positive sense, single strand, RNA viruses. These viruses occur on every populated continent. Alphaviruses are divided into two clades, the New-World and Old-World viruses. The New-World viruses include Eastern (EEEV), Western (WEEV), and Venezuelan equine encephalitis viruses (VEEV), and cause neuroinvasive disease. The Old-World viruses include Chikungunya (CHIKV) and Ross River viruses (RRV), and typically cause multijoint arthralgia. There are currently no approved antiviral therapeutics or vaccines for any alphavirus, making them a high priority for antiviral drug design and discovery. Breast cancer genetic susceptibility is modulated by high to low penetrance risk alleles. Population frequencies of high and moderate penetrant predisposing mutations are rare and responsible for a small percentage of population cases. Most genetic variation in breast cancer susceptibility is low penetrance common variants where causal genes and risk-associated mechanisms are largely unknown. Comparative genetics uses animal models to isolate and mechanistically evaluate genetic susceptibility to human disease. Rat strains have different susceptibility phenotypes to chemical-induced carcinogenesis. This research study examined the lived experiences of elementary educators working with traumatized students. Secondary trauma among other professions such as social workers and first responders is well-documented (Bride et al., 2007). Research on the impact it has on educators is still limited (Hill, 2011). This dissertation provides evidence that educators are directly impacted by the work that they do to support and educate their students. A phenomenological approach is used to explore the troubling personal challenges educators face while working with traumatized students. As the relentless dream of creating a true lab-on-a-chip device is closer to realization than ever before, which will be enabled through efficient and reliable sample characterization systems. Dielectrophoresis (DEP) is a term used to describe the motion of dielectric particles/ cells, by means of a non-uniform electric field (AC or DC). Cells of different dielectric properties (i.e., size, interior properties, and membrane properties) will act differently under the influence of dielectrophoretic force.
