The purpose of this qualitative phenomenological study was to investigate the perceptions of classroom-level educators regarding the application of trauma-informed instructional practices. This was achieved by evaluating educators' understanding of the influence of trauma on students, their level of familiarity with trauma-informed instructional practices, and their assessments of the effectiveness of these practices. Trauma refers to an individual's response to a single traumatic incident, a succession of traumatic events, or extended exposure to a traumatic event (SAMHSA, 2014). Sepsis is a life-threatening condition characterized by overwhelming inflammation, resulting in organ system damage, leading to a high mortality rate. Care in the clinical setting is supportive, and there are no approved sepsis-specific treatments. In septic mice, activation of the cholinergic anti-inflammatory pathway decreases cytokine secretion by leukocytes and improves survival. The cholinergic anti-inflammatory pathway is a reflex of the parasympathetic nervous system, converging on the α7 nicotinic acetylcholine (α7nAChR) at the surface of macrophages. Signaling through the receptor blocks NF-kB activation, thus cytokine secretion. Receptor activation has other effects on macrophages, including modulating their migration to …

Plants synthesize and store oil, mostly as triacylglycerols (TAG), in seeds that is transcriptionally controlled by WRINKLED1 (WRI1), an APETALA2 (AP2) transcription factor. Among the four Arabidopsis WRI paralogs, WRI2 is nonfunctional, while the others are expressed in a tissue-specific manner. Additionally, two rate-limiting enzymes, diacylglycerol acyltransferase (DGAT) and phospholipid diacylglycerol acyltransferase (PDAT) catalyze the terminal step in TAG assembly and contribute to oil accumulation. Avocado (Persea americana) mesocarp, a non-seed tissue, accumulates significant amounts of TAG (~70% by dry weight) enriched with heart-healthy oleic acid. Alcohol-associated liver disease (ALD) is a broad spectrum of diseases ranging from steatosis to severe hepatic cirrhosis, none of which have any FDA approved therapies. Downregulation of hepatic Sirtuin 1 (Sirt1) activity by ethanol has been shown to lead to an upregulation of the pro-lipogenic gene targets under its jurisdiction, resulting in increased lipogenesis and triglyceride accumulation. It was hypothesized that ethanol acts on α4*-nicotinic acetylcholine receptors (α4*-nAChRs) in hepatocytes to decrease Sirt1 expression, which ultimately dysregulates lipid metabolic homeostasis. In recent years, research into air pollution has shown that exposure to certain components in air pollution, primarily PM2.5 can accelerate biological aging and thereby lead to increased susceptibility to multiple diseases. We hypothesize that prolonged exposure to air pollutants can result in premature aging leading to extensive tissue dysfunction and susceptibility to diseases. To examine this, we exposed mice to PM2.5 for 9, 15, and 21 days, then measured the telomere lengths, cellular senescence, and histone methylation patterns of multiple cell types. We reported pulmonary artery hypertension (PAH) patients had elevated levels of Cd in both blood and urine, therefore, this study tested whether Cd directly induces PAH or facilitates PAH pathogenesis in mouse models. C57/6J mice were initially exposed to drinking water with or without 5ppm Cd for 8 weeks. Then, half the mice in both control and Cd groups were given SU5416 and hypoxia (SuHx) for 4 weeks to induce PAH, resulting in 4 subgroups: Control, Cd, PAH, and Cd+PAH. Diastolic and systolic functions of the left and right ventricles (LV, RV) were examined with echocardiography before and after PAH. …

Porphyromonas gingivalis is a keystone oral pathogen in the progression of periodontal disease and has been implicated in the etiology of many other systemic human diseases. P. gingivalis produces primary virulence factors termed the gingipains, a group of cysteine proteases. There is a high demand for high purity gingipains for downstream in vitro studies where P.g. is implicated, both oral and systemic. Gingipain production candidate mutant P.g. strains were generated to isolate each gingipain for large scale purification and the purified gingipains characterized. Hypertension (HT) and chronic kidney diseases (CKD) are complex conditions, disproportionally prevalent among African Americans (AA) and are influenced by multiple genetic as well as environmental factors. Genetic risk scores (GRS) enable the analysis of the combined effects of multiple single nucleotide polymorphisms (SNPs). This study investigates the association of blood pressure (BP) and CKD GRS with BP, hypertension (HT) and CKD; early-onset HT and CKD; and investigates interactions of the GRSs with environmental measures in AA. Zinc deficiency is associated with metabolic dysfunction-associated steatotic liver disease (MASLD). Previous studies show zinc supplementation improves steatosis, but its therapeutic effects in established MASLD remain unclear. We developed a model to characterize the effects of zinc supplementation on high-fat diet (HFD) induced MASLD and hypothesized established MASLD would be attenuated. Mice were fed control diet or HFD for 12 weeks and then grouped into normal or zinc-supplemented diets for 8 additional weeks. Steatotic liver disease is brought upon by differing etiologies and its global prevalence is expected to keep increasing over the next several decades. Particularly, alcohol-associated liver disease (ALD) manifests from excessive alcohol consumption where the liver retains lipids and progresses with metabolic dysfunction. Students of color in low socio-economic, high crime urban areas are exposed to a higher level of trauma than their White contemporaries. The trauma that they bring into school with them create challenges for their teachers working to eliminate barrier to students’ education. Male sexual dysfunction comprises various conditions, including erectile dysfunction (ED) and ejaculatory dysfunction. Testosterone deficiency (TD), which has been linked to some of these sexual dysfunctions, is characterized by low production of testosterone by the testes. These male reproductive outcomes may develop due to exposure to various risk factors, including environmental exposures. Glioblastoma (GBM) is a highly invasive, aggressive brain cancer that carries a median survival of 15 months. This poor prognosis is due, in part, to its resistance to standard therapeutic intervention. Recent studies have demonstrated that tumor heterogeneity plays a critical role in facilitation therapy resistance by mediating tumor adaptation through microenvironmental cues. A protection zone model represents a patchy environment with positive growth over the protection zone and strong Allee effect growth outside the protection zone. Generally, these models are considered through the corresponding eigenvalue problem, but that has certain limitations. In this thesis, a general protection zone model is considered. This model makes no assumption on the direction of the traveling wave solution over the Strong Allee effect patch. Heterocyclic amines (HCAs) are mutagens generated when cooking meat for prolonged periods of time or until well-done. Recent epidemiological studies reported significant associations between dietary HCA exposure and insulin resistance and type II diabetes. However, no previous studies have examined if HCAs, independent of meat consumption, contributes to pathogenesis of insulin resistance or metabolic disease. It is well known that HCAs require hepatic bioactivation by cytochrome P450 1A2 (CYP1A2) and N-acetyltransferase 2 (NAT2). This research focuses on crucial aspects of the development process, including fabrication techniques, miniaturization, and integration for a more efficient collaborative human-robot interface. The fabrication process of robot skin sensors, designed to mimic human skin, is explored both within and outside cleanroom environments. This thesis investigates protein markers linked to pulmonary embolism risk using proteomics and statistical methods, employing unsupervised and supervised machine learning techniques. The research analyzes existing datasets, identifies significant features, and observes gender differences through MANOVA. Principal Component Analysis reduces variables from 378 to 59, and Random Forest achieves 70% accuracy. These findings contribute to our understanding of pulmonary embolism and may lead to diagnostic biomarkers. MANOVA reveals significant gender differences, and applying proteomics holds promise for clinical practice and research.

Preterm birth is the number one cause of death in neonates, accounting for 35% of neonatal mortality. The preterm birth rate in the U.S. in 2021 was 10.5%, disproportionally affected by race and ethnicity. Obese women have an increased risk of preterm pregnancy, and if delivered before 37 weeks of gestation, the offspring have higher rates of complications that extend from the neonatal period into life-long metabolic and immune adverse outcomes. Abstract As teachers make their way back to the classroom for the 2023–2024 school year, teacher burnout is manifesting itself mentally, physically, and emotionally. This study examined the role of educational robotics in fostering computational thinking in elementary settings, both in classrooms and extracurricular programs. Among growing concerns over K–12 students’ computational thinking deficits, the research evaluated the impact of Lego EV3 and VEX IQ platforms. Data was sourced from lesson plans, student work surveys, and teacher interviews and then subjected to thematic analysis using a qualitative approach. The participants were Texas educators engaged in robotics instruction, even though specific robotics statistics are absent in the Texas Education Agency. This study was conducted between March 2023 and April 2023 and measured if women leaders in the oil and gas industry could lead authentically while dealing with gender biases. Women executives encounter macro-level challenges such as recruitment, retention, maintaining a career in oil and gas, feeling a sense of belonging, and gender prejudices. Women have faced organizational obstacles and challenges such as modifying their personalities, being underrepresented in their organization, unequal compensation, discriminatory stereotypes, and scarcity of resources.
