It is primarily found in Africa and is transmitted to humans through contact with sick animals, particularly rodents and monkeys, or through human-to-human transmission [2]. From the beginning of May 2022, cases of mpox have been recorded from non-endemic nations, and the illness has continued to be reported in other endemic nations. Underexplored environments such as soil samples continue to be an untapped source of bacterial strains with great potential to produce secondary metabolites for medicinal applications. As a result, these microorganisms represent a broad and yet unknown reservoir of new strains capable of producing these novel compounds. The current research primarily seeks to perform the isolation, purification, and characterization of secondary metabolites from a soil bacterium (Janthinobacterium lividum TAJX1901). Microsporidia are a distant fungal pathogen that have severe clinical consequences for the immunocompromised. Previous work identified a microsporidian pathogen protein termed Microsporidian ADAM or MADAM. This protein has close sequence homology to other ADAM proteins (A Disintegrin and Metalloproteinase) in two microsporidian species, Encephalitozoon intestinalis and E. cuniculi. ADAM proteins have a wide range of functions, including binding to integrins and host signaling. It is known that many pathogens manipulate integrins to invade host cells, and it is predicted that microsporidia are also exploiting this host target. This variation in overdose mortality can be attributed to the risk environment for overdose in rural Appalachia. Glioblastoma is the most commonly diagnosed central nervous system primary malignancy and it is considered a terminal diagnosis with few treatment options available. Glioblastoma tumors frequently develop treatment resistance due in part to their highly heterogenic nature. The heterogeneity of glioblastoma is partially attributed to the presence of glioma stem-like cells (GSC), which are highly invasive and resistant to chemotherapy and irradiation treatments. Signaling of the receptor tyrosine kinase MET is a known regulator of GSC. Glioblastoma patients have an increasingly poor prognosis that corresponds with increasing MET expression. Yersinia pestis is a Gram-negative re-emerging bacterial pathogen that is responsible for bubonic, septicemic, and pneumonic plague. Y. pestis and other bacteria require transition metals, such as iron, zinc, and manganese, to maintain intermediary metabolism, transcriptional regulation, and virulence. To inhibit infection, eukaryotic organisms have developed distinct mechanisms, called nutritional immunity, to sequester these important nutrients from invading bacteria. For pathogens to colonize the vertebrate host, they have evolved dedicated acquisition systems for transition metals. Principles of natural selection have proven valuable for explaining why pathogens cause the diseases that they do. In theory, the evolved level of host exploitation should reflect how dependent a pathogen is on host health for transmission. This dependency is shaped by transmission mode and transmission opportunity, which should therefore be predictors of disease manifestations. In this dissertation, I apply these principles to investigations of depression in Chlamydia trachomatis and virulence of SARS-COV-2 and Plasmodium species. This dissertation has five chapters. The study explored the role social support (as an external factor of resilience) and self-efficacy (as an internal factor of resilience) played in the resilience of Head Start teachers during the COVID-19 pandemic. This study employed a sequential explanatory mixed methods design involving two phases: a quantitative and a qualitative phase. This study used the Faucet Theory (Alexander et al., 2001) as a theoretical framework to determine the extent that the COVID-19 learning environment impacted the Southern Public Schools District’s African American, low-income, and high-income high school students’ proficiency level based on the Algebra I section of the 2020-2021 Louisiana Education Assessment Program (LEAP) assessment. The Central Appalachian region of the United States disproportionately experiences higher rates of cardiovascular diseases (CVD) and associated risk factors. Primary risk factors for CVD include hypertension, hypercholesterolemia, and tobacco use, all of which have the potential to be mitigated through lifestyle behavior changes. Dietary and physical activity practices are the two main driving forces for the development of hypertension and hypercholesterolemia, while tobacco use is a behavioral choice on its own. The study examined the frequent frames adopted by the CDC in the communication of the Covid-19 vaccine and how it impacts vaccine uptake through quantitative content analysis. The study builds on the framing theory and elaboration likelihood model. This study’s content collected 327 posts by the CDC on covid-19 vaccinations from December 2020- December 2022. A research randomizer was used to choose 200 posts as the final sample from the data that had been gathered. 122 posts out of the 200 total posts that the randomizer tallied were pertinent to the research questions for the study. Tick populations have been immigrating into northeast Tennessee from east Tennessee, Kentucky, Virginia, and North Carolina. Counties in states bordering northeast Tennessee harbor tick species associated with human illness. Human diseases transmitted by ticks include ehrlichiosis, spotted fever rickettsial group diseases, tularemia, anaplasmosis, babesiosis, Lyme disease, alpha-gal syndrome, Heartland virus, Powassan virus, and southern tick-associated rash illness (STARI). These diseases cause morbidity and mortality in human populations and may pose a high risk to individuals, wildlife, and livestock. 2-ω-carboxyethylpyrrole (CEP) is a product of docosahexaenoic acid (DHA) oxidation, which forms covalent adducts with different proteins. CEP-modified proteins can interact with macrophage receptor, integrin αDβ2. This study aims to compare αDβ2 binding to its physiological ligand, fibrinogen, and CEP-modified fibrinogen, which is formed during inflammation. We hypothesize that modification of fibrinogen changes its ligand-binding properties to integrin αDβ2 which can affect macrophage migration and retention. Recombinant αD I-domain and αDβ2-transfected HEK293 cells were used for the experiments. Regulation of the bacterial second messenger cyclic-di-GMP in Salmonella Typhimurium allows it to delicately alter phenotypes to optimize invasion and survive intracellularly in epithelial cells and macrophages to become virulent and cause infection. Cyclic-di-GMP concentration is regulated by the presence of external stimuli, sensory diguanylate cyclases (DGCs) and phosphodiesterases (PDEs), and cyclic-di-GMP binding effectors. Previous studies established that maintenance of low cyclic-di-GMP concentrations is required for survival in macrophages, and that deletion of 3 active PDEs reduces this survival. Leprosy was an endemic disease in colonial Southeastern Nigeria but became an epidemic between the 1920s and 1940s. Unlike malaria and sleeping sickness, its endemicity did not impede European penetration at the dawn of the twentieth century. However, its spread across Nigeria intensified during colonial rule, with acute prevalence in regions hitherto prone to the disease. Therefore, from the 1920s to 1960, Southeastern Nigeria became the epicenter of British policies tailored towards preventing, curing, and eradicating leprosy disease in Nigeria. Niger is one of the countries with the lowest exclusive breastfeeding (EBF) rates in the world. The purpose of this study was to determine the barriers to EBF in Niger and to determine the types of feedings that are most commonly practiced in Niger. Participants (n=31) were mothers with children aged 0 to 24 months, recruited on-site at a healthcare facility in Niamey, the capital of Niger.
