Host-parasite interactions are ubiquitous and have negative risks and consequences on individual, population, and evolutionary scales. These risks and consequences include altered population densities, changes in interspecies interactions, and rapid changes in host phenotypes. In this thesis, I seek to understand the ecological and evolutionary ramifications of host-parasite interactions within ecosystems. To do this, I investigated the presence and environmental correlates of potential host-parasite interactions in a freshwater ecosystem and used a model ecosystem to explore how past host-parasite coevolution affects community assembly dynamics and future adaptation in the presence of new stressors. Leishmaniasis is a protozoan parasitic disease caused by the species Leishmania that affects millions of people around the world. It is a serious public health concern in the developing world with about 1 million cases and 70,000 fatalities occurring each year. It has several forms with visceral, and cutaneous leishmaniasis being the most common forms of the disease. Visceral leishmaniasis, caused by the Leishmania donovani occurs mainly in Brazil, Eastern Africa, and India, and is fatal if left untreated. This study aimed to formulate and evaluate Isoniazid-Pyridoxine-loaded gummies for use in a population ranging from pediatric to geriatric and to study the applicability of BIO X 3D printer in printing gummy formulations. Semisolid extrusion (SSE) 3D printing is an emerging technology in personalized medicine. To address clinical multi-dose requirements, SSE has been explored to manufacture new preparations. In this study, Isoniazid (INH) and Pyridoxine Hydrochloride (PDX) were used to make gummy formulations for use in the treatment of Tuberculosis. Infectious diseases are the leading cause of disability-adjusted life years worldwide. Glycans initiate host–pathogen interactions in two particular infectious diseases, severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and urinary tract infections (UTIs). In this work, recent advances of computational tools in drug design are utilized to target these glycanspecific host–pathogen interactions for the potential prevention of SARS-CoV-2 or UTIs.

In this study, we studied the role of IFITM3 in SVA replication in vitro by silencing the endogenous IFITM3 in NCI-H1299 cells. An average of 1.94-fold increase in SVA VP2 expression and an average of 4.4-fold increase in supernatant virus titer was observed after 24h SVA isolate SD15-26 infection. We have indeed observed an increase in viral mRNA copies by an average of 5-fold in IFITM3 silenced NCI-H1299 cells at 8 hpi indicating that IFITM3 restricts SVA replication post virus attachment and entry. A positive correlation between interferon-alpha induced IFITM3 upregulation and reduced SVA replication was observed. To further confirm …

Every year, one-eighth of all North American bison (Bison bison) are translocated, often across large climatic gradients. Over the last 16 years bison have been translocated from Wind Cave National Park in South Dakota to establish six herds across the United States central grasslands in Iowa, Missouri, Illinois, Indiana, and Kansas. Influence of climatic gradients on herd-level thermoregulation, body size, and helmintic infection were evaluated. Overall, 1174 thermal images and 1050 fecal samples were collected between 2021 and 2023. In recent years, research into air pollution has shown that exposure to certain components in air pollution, primarily PM2.5 can accelerate biological aging and thereby lead to increased susceptibility to multiple diseases. We hypothesize that prolonged exposure to air pollutants can result in premature aging leading to extensive tissue dysfunction and susceptibility to diseases. To examine this, we exposed mice to PM2.5 for 9, 15, and 21 days, then measured the telomere lengths, cellular senescence, and histone methylation patterns of multiple cell types. Porphyromonas gingivalis is a keystone oral pathogen in the progression of periodontal disease and has been implicated in the etiology of many other systemic human diseases. P. gingivalis produces primary virulence factors termed the gingipains, a group of cysteine proteases. There is a high demand for high purity gingipains for downstream in vitro studies where P.g. is implicated, both oral and systemic. This project explores the role of rhetoric in crisis—how rhetoric can contribute to both the stabilization and destabilization of a worldwide health emergency. Specifically, I utilize the COVID-19 pandemic as a case study to investigate how institutional rhetorics exacerbated the ongoing burnout epidemic amongst healthcare workers. We are facing an aging crisis, with 20% of the U.S. population projected to be geriatric (65+) by 2030 and live another 40+ years. Age-related diseases accompany a growing geriatric population, emphasizing the need to understand their etiology. Environmental pollutants compound this crisis by: 1) geriatrics are more susceptible, exacerbating age-related diseases and comorbidities, and 2) they accelerate biological aging, inducing age-related diseases at younger ages. The COVID-19 pandemic led to organizations rapidly transitioning their workforces to virtual work settings. This sudden shift brought about many challenges to organizations and their employees. In this study, a phenomenological research approach was used to describe the lived experiences of leaders who transitioned to managing a virtual team during the COVID-19 pandemic. The study also explored how leaders adapted their leadership practices and leveraged people and technology when transitioning to managing a virtual team. Many HIV-positive (HIV+) males on Highly Active Anti-Retroviral Therapy (HAART) experience metabolic abnormalities, including Non-Alcoholic Fatty Liver Disease (NAFLD) and lipodystrophy. The intestinal microbiota and short chain fatty acids (SCFA), participate in bidirectional communication with their host. Dysbiosis in HIV+ males on HAART demonstrate a Prevotella-rich enterotype shaped by multiple factors including, medications, adiposity, diet, intestinal permeability, and lifestyle; our objective was to investigate these factors. 19 HIV+ and 21 HIV- males were enrolled. Obstetric violence includes acts of abuse, coercion, or disrespect that occur during the labor process. The present study explores how obstetric violence impacts a person’s postpartum psychosocial adjustment. This study used a subset of data (N = 339) from a larger online study, which attempted to explore multiple facets of a person’s postpartum health. The psychological constructs examined are postpartum depression and postpartum anxiety. Additional constructs assessed were protective and risk factors: self-compassion and medical mistrust. Sepsis is a life-threatening condition characterized by overwhelming inflammation, resulting in organ system damage, leading to a high mortality rate. Care in the clinical setting is supportive, and there are no approved sepsis-specific treatments. In septic mice, activation of the cholinergic anti-inflammatory pathway decreases cytokine secretion by leukocytes and improves survival. The cholinergic anti-inflammatory pathway is a reflex of the parasympathetic nervous system, converging on the α7 nicotinic acetylcholine (α7nAChR) at the surface of macrophages. Preterm birth is the number one cause of death in neonates, accounting for 35% of neonatal mortality. The preterm birth rate in the U.S. in 2021 was 10.5%, disproportionally affected by race and ethnicity. Obese women have an increased risk of preterm pregnancy, and if delivered before 37 weeks of gestation, the offspring have higher rates of complications that extend from the neonatal period into life-long metabolic and immune adverse outcomes. The purpose of this phenomenological qualitative study was to examine the decision-making process of K-12 parents living in the Appalachian Highlands who chose to remove their children from traditional public schools to join learning pods (sometimes referred to as pandemic pods) during the COVID-19 pandemic. Finding a faster way of bacterial identification is necessary as it currently takes 1-4 days for a medical lab to culture and identify bacteria. Photoacoustic flow cytometry (PAFC) can be used as an alternative method resulting in swift identification within an hour (Edgar, 2019). Interleukin 1β (IL-1β), an important cytokine in both health and disease, functions as a major proinflammatory mediator of pathological challenges, including involvement in the cytokine storm associated with Macrophage Activation Syndrome, Hemophagocytic Lymphohistiocytosis and the severe secondary effects of SARS-CoV-2 infection. Therapeutic approaches to limiting the effects of IL-1β protein have focused either on specific protein neutralization before release from activated macrophages (by general metabolic inhibitors and inflammasome inhibition) or after release by ligand and receptor obstruction (e.g., Canakinumab, Anakinra and Rilonacept). A thorough understanding of receptor regulation is imperative to predict expression in varying contexts of disease or treatment. Lymphocyte surface receptors are often used as biomarkers and drug targets, making them particularly important for study. For receptors of debated functionality, such as the Fc receptor for IgM (FcMR), understanding regulation can also help to predict expression in vivo to supplement hypotheses of biological roles. Dryolestoids are extinct cladotherians mammals from the Jurassic and Cretaceous periods. I describe a collection of dryolestoid specimens from the Late Cretaceous localities of Cerro Tortuga (Allen Formation), Anfiteatro 1, and Shining (both La Colonia Formation) from Patagonia, Argentina. Using comparative morphology, I identify a new species of meridiolestidan dryolestoid based on eleven specimens across both formations. The new species’ recovery from La Colonia Formation represents the first dryolestoid connection between the two approximately contemporaneous formations. The species’ morphology may represent an ecological shift within Meridiolestida from insectivory to herbivory, showing a transition in characters between the plesiomorphic sharp-toothed meridiolestidans …

The adaptive immune system relies on a diverse set of over one hundred immunoglobulin (IG) genes across three genomic loci that are variably combined to form antibodies (Ab). The IG Lambda locus is one of two loci which encodes the IG light chain. The complexity of the IGL locus severely limits the effective use of standard short-read sequencing, limiting our knowledge of population diversity in these loci. We leveraged single molecule real-time (SMRT) long-read sequencing in conjunction with IGL-targeted DNA capture to develop the method IG-Cap for accurate and high-throughput sequencing of the IGL locus. Sleep disturbances are prevalent among primary care patients, and psychological dysfunction, including stress, anxiety, and depression, are robust contributors to poor sleep health. Yet, the presence of potential protective characteristics, such as health self-efficacy and engaging in adaptive health behaviors, may mitigate such outcomes. Gratitude (i.e., recognition and appreciation of experiences, relationships, and surroundings), a positive psychological cognitive-emotional characteristic, may serve as a catalyst of these beneficial downstream effects, given its association with improved health functioning and sleep. Salmonella enterica serovar Typhimurium is one of the most prevalent food-borne pathogens, affecting millions around the world every year, making it a threat to global health. Salmonella possesses the ability to survive the normally lethal condition of desiccation, however, discovery of the genes and mechanisms behind this phenomenon are still ongoing. Using a transposon mutagenesis approach to construct a broad transposon library, this study aimed to uncover genes that may be contributing to changes in Salmonella’s survivability under desiccation, particularly when exposed to the antimicrobial molecule salicylic acid.
