This research addresses the question of why some people become entrepreneurs whereas others do not. The debate has been going on for decades in entrepreneurship. In this dissertation, I address this question by decomposing it into two related questions. The first question is whether entrepreneurs make different decisions compared to non-entrepreneurs when they are facing the same opportunities under risk. The second question is whether these differences in decision-making (if any) are due to the natural proclivity of entrepreneurs themselves. Brain derived neurotropic factor (BDNF) is required for the gustatory neuron survival,target innervation, and taste bud maintenance during development. However, whether BDNF has any function in the adult gustatory system in normal conditions or after nerve injury is unclear. To address these issues, I inducibly removed BDNF in all cells expressing BDNF in adult mice. In the experimental animals, Bdnf expression decreased to 5% of control mice in the lingual epithelium and geniculate ganglion (p< 0.01) at both two weeks and ten weeks after tamoxifen administration. I found no effect on taste bud morphology at four weeks following Bdnf gene deletion. However, ten weeks following Bdnf gene deletion, P2X3-positive and TUJ1-positive gustatory innervation to individual taste buds was reduced by nearly half (each with p < 0.01) and both taste bud volume and taste cell number decreased 30% (each with p< 0.01). These experiments demonstrate that BDNF is required for maintenance of normal levels of taste innervation and normal taste bud morphology in adulthood. In addition, taste cells expressing PLCß2 (phospholipase C ß2), a marker for taste cells that respond to sweet, bitter and umami, did not decrease after Bdnf gene deletion in the adult. Thus, the missing taste cells are of another type. This indicates that taste cell loss is not uniform across the various taste cell types, even if nearly all taste cell types receive the P2X3 and TUJ1 innervation. Since BDNF is required for initial innervation of the taste system and supports taste bud innervation and size in adulthood, it could also be required for nerve reinnervation after injury. To determine if Bdnf is still expressed following nerve section, the chorda tympani nerve (taste nerve) was sectioned and Bdnf level was detected with Real Time RT-PCR. Bdnf continued to be expressed at normal levels from two days to two months post-surgery in both geniculate ganglion and tongue epithelium. Therefore, BDNF could be involved with chorda tympani regeneration. To determine if this was the case, the Bdnf gene was deleted in adult inducible transgenic mice (under the control of a Ubiquitin promoter) two weeks before chorda tympani nerve section. Taste bud number was reduced by half in all genotypes at two weeks post-surgery (p < 0.01). For the remaining taste buds, gustatory innervation was nearly gone with only a little innervation from the trigeminal nerve remaining in the taste bud (p < 0.01). Taste bud volume (p < 0.01) and taste cell number (p< 0.01) were reduced by half for both control and experimental genotypes. Eight weeks post-surgery, taste bud number recovered in mice without Bdnf gene deletion, but did not recover in mice following Bdnf gene deletion (p < 0.01). Gustatory nerve innervation returned in 70% of the taste buds in control mice (p< 0.01). For those reinnervated taste buds, both taste bud volume and taste cell number increased to normal levels. However, in mice lacking the Bdnf gene, gustatory fibers only reinnervated 7.8% of the taste buds (p < 0.01); for most uninnervated taste buds, both taste bud volume and taste cell number remained small. These experiments demonstrate that BDNF is crucial for promoting regeneration of gustatory nerve fibers in adulthood. Following gustatory nerve section, considerable adult plasticity has been observed on the contralateral side including enlarged taste buds with more cells (Guagliardo and Hill, 2007). To determine if this anatomical change was associated with alter Bdnf expression. I examined Bdnf level in the geniculate ganglion and tongue epithelium on the contralateral side following chorda tympani nerve section. Results showed Bdnf expression increased two fold at two weeks post-surgery in geniculate ganglion on the contralateral side (p < 0.05), indicating BDNF may involve with the observed plastic changes. To determine if the increase in taste bud size was associated with increased innervation and/or regulated by BDNF, the Bdnf gene was then deleted in inducible knockout mice before nerve surgery, and taste bud size and amount of innervation were measured on the contralateral side. The results showed taste bud volume, taste cell number and a marker for nerve fibers all increased on the contralateral side in mice without Bdnf gene deletion at eight weeks post-surgery. This indicates that larger taste buds could be supported by increased TUJ1 positive fibers from trigeminal nerve. In addition, in mice lacking Bdnf, taste bud volume, taste cell number and innervation did not increase on the contralateral side after surgery, which indicates that Bdnf may contribute to larger taste buds on the contralateral side following nerve section by supporting increased innervation to the larger taste buds.

This dissertation offers and theorizes findings of a two-year mobile ethnography investigating the complexity of students’ movements within and among secondary and tertiary educational institutions and the labor market. This project investigated the role of miRNA-21 in the macrophage response to peritonitis; with the goal of understanding whether the associated mechanism has potential benefits in the clinical treatment of peritonitis. A novel therapeutic intervention in the treatment of peritonitis could modulate the host immune response to decrease an exaggerated pro-inflammatory innate response and then prevent or ameliorate the development of sepsis and organ failure. Carbohydrate binding agents that target viral envelope glycans are being studied for their potential use as microbicides and antiviral therapeutics. Griffithsin is a lectin originally identified in a red alga Griffithisia sp. Multiple studies have shown that GRFT inhibits HIV-1, Coronaviruses, Hepatitis C, influenza and Ebola virus replication in vitro. This antiviral activity suggests potential uses in chemoprophylaxis and disease treatment. However, safety of GRFT administration has not been extensively studied. This thesis examined whether water quality diminished as a result of increasing pollution downstream along the Middle Fork of Beargrass Creek, Louisville, Kentucky in response to urbanization. Samples of key water quality indicators/pollutants were collected for 12 weeks (November - January) along the Middle Fork of Beargrass Creek at three different stream gauges. A spatial and statistical analysis was performed to verify any significant correlations between the water quality indicators and pollutants, and whether basin wide landcover/sewer outlet numbers led to an increase in these indicators downstream. Mice lacking or pharmacologically inhibited for calcium/calmodulin-dependent protein kinase kinase 2 (CaMKK2) have enhanced bone mass and microarchitecture. This enhanced bone mass and architecture is due to changes in osteoblast and osteoclast numbers as well as activity. Whether the enhanced bone mass translated to increased bone quality and strength was further explored by developing a method to directly test the trabecular strength of the distal femur. Probiotics are viable nonpathogenic microbes that positively affect host health. Probiotics inhibit infection, activate immunity, and promote mucosal-barrier development. Many microbes have probiotic activity. Nonetheless, the selection of stable strains and their specific mechanism(s) of action are not fully elucidated. Bacteria from ‘Amabere amaruranu’ cultured milk from Kenya were isolated and identified by PCR sequence analysis of the 16S rRNA gene. Isolates were examined for stability to acid and bile, antimicrobial activity, mucin production, and degradation and sensitivity to antibiotics, hence their potential for probiotics. Salicylic Acid Binding Protein2 (SABP2) is an enzyme known to play important role in SA mediated pathway. SBIP-428 (SABP2 Interacting Protein-428), a SIR2 like deacetylase, has been found to interact with SABP2. We demonstrate that SBIP-428 functions as a Sirtuin deacetylase. We show that SBIP-428 itself is lysine acetylated. Interactions of a SBIP-428 with SABP2 also raised the possibility of SABP2 itself being lysine acetylated. The recombinant purified SABP2 or native partially purified SABP2 displayed no acetylation. In response to TMV infection, the expression of SBIP-428 was down regulated at 48 hpi. There is still a lack of mechanistic understanding of how the factors, such as temperature and food, coordinate the labor of ants. Streptomyces are known to produce a large variety of antibiotics and other bioactive compounds with remarkable industrial importance. Streptomyces sp. COUK1 was found as a contaminant on a plate in which Rhodococcus erythropolis was used as a test strain in a disk diffusion assay and produced a zone of inhibition against the cultured R. erythropolis. The identity of the contaminant was confirmed as Streptomyces through 16S rRNA sequencing. This Streptomyces produces a strong inhibitory compound in different growth media. Little is known about the distribution, abundance, or life histories of the Northern Saw-whet Owl (Aegolius acadicus) in the Southern Appalachian Mountains of Northeast Tennessee. This study relied upon the Pennsylvania Protocol of audio playback of a Northern Saw-whet Owl call to monitor for owl presence at various areas above 3,500 feet in elevation. Owls were found at multiple areas in Northeast Tennessee, including Roan Mountain, Unaka Mountain, Rocky Fork, and the Pond Mountain Area. Statistical analysis revealed that these owls were not limited by habitat, showing equal presence in Hardwood habitats along with Spruce and/or Fir habitats. …

The ability to measure biomechanical kinematics at the sub-mm scale is essential for obtaining valuable insight into pathologies, but small motions of the joints are difficult to quantify. Estimates of skeletal kinematics are commonly made from optical motion capture systems and markers placed on the skin. The discovery of the Ziegler Reservoir fossil site near Snowmass Village, Colorado presents an opportunity to examine subalpine ecosystem response, during a relatively unknown period, in which climate conditions were similar to present. Finegrained sediments at Ziegler Reservoir represent continuous deposition between ~140–55 ka (thousand years before present), spanning the close of the Bull Lake glacial period [marine isotope stage (MIS) 6], the Sangamon interglacial (MIS 5) and the early stages of the Pinedale (early Wisconsin) glacial period (MIS 4). Stress Corrosion Cracking (SCC) has proven to be one such example of conditions found in use in high voltage transmission line applications that leads to brittle fracture of polymer matrix composites.

Optimization of nitroxides as probes for EPR imaging requires detailed understanding of spectral properties such as spin lattice relaxation times, spin packet linewidths, and nuclear hyperfine splitting. Initial measurements of relaxation times for six low molecular weight nitroxides at X-band stimulated further measurement at frequencies between 250 MHz and 34 GHz. The impact of tumbling was studied with perdeuterated 2,2,6,6-tetramethyl-4-piperidinyl-1-oxyl (PDT) in five solvents with viscosities resulting in tumbling correlation times, τR, between 4 and 50 ps.
