Human C-reactive protein (CRP) is the prototypic acute phase protein whose serum concentration increases rapidly during inflammation. CRP is also associated with atherosclerosis; it is deposited at lesion sites where it may interact with modified lipoproteins. There are 2 major questions regarding CRP: 1. How is the serum concentration of CRP regulated? 2. What are the functions of CRP in atherosclerosis?

Alcohol is the most socially accepted addictive drug, and it can cause alcoholic liver disease (ALD), which is a major cause of morbidity and mortality in the United States and worldwide. Animal and human studies demonstrate that chronic alcohol consumption causes a pro-oxidant environment in the liver and increases hepatic lipid peroxidation and the accumulation of by-products such as acrolein and 4-hydroxynonenal. Acrolein is the most reactive and toxic aldehyde generated through lipid peroxidation. Alcohol induced hepatic steatosis is a significant risk factor for progressive liver disease. Steatotic hepatocytes have increased sensitivity to injury produced by inflammatory cytokines, particularly TNF. Cyclic adenosine monophosphate (cAMP) has been shown to play a significant role in the regulation of both TNF production and lipid metabolism. However, the role of altered cAMP homeostasis in alcohol mediated hepatic steatosis and injury has not been studied. cAMP levels are tightly regulated by phosphodiesterase family of enzymes. This dissertation is a critical intervention into the question of student agency. An interdisciplinary project that draws upon philosophy and linguistics, it reviews four major tendencies that have animated composition pedagogy over the last several decades— process theory, social-constructivism, procedural rhetoric, and trans-lingual pedagogies— and identifies some of the key tensions that both motivate and problematize these approaches. First, it examines the debate between Peter Elbow and David Bartholomae, and the interplay between teachers’ authority and student agency. Lunasin is a 44 amino acid peptide derived from the soybean seed that has been shown to have cancer chemopreventive and chemotherapeutic properties. In this study, we investigated the potential utility of lunasin as a chemotherapeutic in a melanoma model. Initial studies showed that lunasin has little activity against established melanoma cell lines in vitro using adherent culture methods; however, lunasin’s in vitro activity was significantly higher in non-adherent colony-forming assays in soft agar and oncosphere assays. Salicylic acid binding protein 2 (SABP2) that converts methyl salicylate to salicylic acid (SA) plays an obligatory role in the SA-mediated disease resistance pathway in plants. SABP2 interacts with SBIP24 in a yeast two-hybrid screening. SBIP24 belongs to the stearoyl-acyl carrier protein-desaturase protein family. To biochemically characterize the SBIP24, it was cloned from tobacco leaves using RT-PCR and expressed in E. coli. Recombinant SBIP24 was affinity purified using Ni-NTA chromatography. Impulsivity, Venturesomeness, And Pride: Potential Moderators Of The Relationship Between Childhood Trauma, Substance Use, And Physical Aggression, Joshua P. Hatfield

Impulsivity, venturesomeness, and pride variables were examined as potential moderators of the associations between childhood trauma and physical aggression, alcohol use and physical aggression, and drug use and physical aggression. Participants (n = 457) were college students recruited from a university in the Southeast. It was hypothesized that childhood trauma, alcohol use, and drug use would be associated with increased scores of physical aggression. In addition, it was hypothesized that impulsivity, venturesomeness, authentic pride, and hubristic pride would moderate these relationships. The purpose of this quantitative study was to investigate Brain Based Learning (BBL) techniques in teaching science. Participants included 216 K-12, full-time, regular education teachers from 8 Northeast Tennessee school systems who taught at least 1 science class. Specifically this research was guided by 7 research questions on teachers’ perceptions and practices in teaching science.

Data were collected by a survey that consisted of 82 statements where teachers rated their level of agreement and was distributed online via Survey Monkey. Autism spectrum disorder (ASD) now affects 1 in 68 children in the United States. Disorders within this spectrum share hallmark deficits in verbal and nonverbal communication, repetitive behavior, and social interaction. The cause of ASD is still unknown. Even though hundreds of genetic abnormalities have been identified in ASD, these markers account for less than 1% of all ASD cases. In our studies we found that stabilized expression of HIF-1α in heart led to better recovery of function and less tissue death after 30 minutes of global ischemia, via mechanisms that preserve the mitochondrial polarization. Our group previously showed that HIF-1α conferred ischemic tolerance by allowing cardiomyocytes to use fumarate as an alternative terminal electron acceptor to sustain anaerobic mitochondrial polarization. The source of fumarate was identified as the purine nucleotide cycle (PNC). Here we discovered that HIF-1α upregulates AMP deaminase 2 (AMPD2), the entry point to the PNC. Abscisic acid (ABA) induces RD22, responsive to dehydration stress gene. Salicylic acid (SA) has been the focus of research due to its role against pathogens and abiotic stress. Interaction between ABA and SA signaling pathways is still poorly understood. SA-Binding Protein 2 (SABP2) converts methyl salicylate to SA. An attempt was made to identify proteins that interact with SABP2 using a yeast 2-hybrid screening. Several interactors were identified. One of them, SA-Binding Protein 2 Interacting Protein-355 (SBIP-355), showed high homology to RD22. Bioinformatic approaches showed that SBIP-355 contains a BURP domain. While DNA profiling makes it possible to individualize biological stains, the identification of the stain itself can present forensic serologists with a significant challenge. Current antibody- and enzyme activity-based assays used by forensic practitioners for biological stain identification yield only presumptive results. Positive results with non-target body fluids or cross-reactivity with non-human sources has also been well documented. The role of Melanocortin 2 Receptor (MC2R) in adrenal/interrenal glucocorticoid secretion has been well documented in many organisms. Studies in mammals have shown that in the adrenal gland two melanocortin receptors and two melanocortin receptor accessory proteins are expressed: MC2R, MC5R, MRAP, and MRAP2. The MRAPs have an opposite effect on the cell surface expression of MC2R and MC5R. In mammals, MRAP aids MC2R but inhibits MC5R cell surface expression. This research investigates the Cherokee Freedmen, who are people of African-American descent and peoples of mixed African-American and Native American descent, who were once enslaved by Cherokees in the Cherokee Nation, and who have articulated their identity and tribal citizenship status as Cherokee Natives in the Cherokee Nation. Upon emancipation, the Cherokee Nation adopted the Cherokee Freedmen as equal citizens in the Cherokee Nation under article nine from the Treaty of 1866, but this changed after citizens were recorded on the Dawes Rolls based upon blood quantum. Human arylamine N-acetyltransferase 1 (NAT1) is a well-known phase II metabolic enzyme that has been associated with carcinogenesis. Its role in the biotransformation of aromatic and heterocyclic amine carcinogens has been investigated for many years, but more recent investigations focus on a possible endogenous role of human NAT1 in cancer initiation and progression. Background: Spontaneous locomotor recovery in spinal rats has been attributed to animals moving freely in-cage. Environmental enrichment has been shown to increase in-cage movement and functional recovery subsequently. Anxiety has been shown to decrease overnight activity in rats. Methods: Rats were double-housed in medium cages (MC) or single-housed in tiny sized cages (TC). Slotted dividers allowed for partial isolation in TC. Overnight activity was monitored bi-weekly. The open field test and BBB’s were taken weekly. Gait analysis was performed at weeks six and eight. Results: MC showed higher overnight activity and improved gait overtime. The focus of this dissertation is to develop statistical methods, under the framework of penalized regressions, to handle three different problems. The first research topic is to address missing data problem for variable selection models including elastic net (ENet) method and sparse partial least squares (SPLS). I proposed a multiple imputation (MI) based weighted ENet (MI-WENet) method based on the stacked MI data and a weighting scheme for each observation. Numerical simulations were implemented to examine the performance of the MIWENet method, and compare it with competing alternatives.
