Prostate Cancer is the second most deadly cancer in men, after lung cancer. The need for new and effective therapeutics that can constrain prostate cancer progression are challenged by the lack of suitable delivery strategies that target prostate cancer tissue. To study CT20p as potential chemotherapeutic agent in the treatment of prostate cancer we proposed the use of targetable hyperbranched polyester (HBPE) based nanoparticles as delivery system. Folic acid was conjugated to the nanocarrier to improve the selectivity of the nanoparticle towards specific cell surface targets in prostate cancer cell lines. Photopollution present along heavily developed coastlines is known to have a significant negative impact on nesting sea turtle populations by damaging nesting habitat quality, but few studies have explored this relationship on broad spatial scales. The purpose of this study was to characterize the onshore light field along a developed stretch of South Carolina coastline and to quantify how photopollution intensity affects regional loggerhead sea turtle (Carretta carretta) nesting density using historical nesting data. Impact Of Development On Freshwater Turtle Populations Of Coastal South Carolina: An Examination Of Land Use And Road Density, Andre Dominguez

Previous studies (Haxon 2000; Steen and Gibbs 2004; Marchand and Litvaitis 2004; Aresco 2005) have demonstrated that urbanization and development have disproportionally adverse affects on female turtles. In order to test this paradigm I caught 301 turtles (primarily Trachemys scripta scripta) in ten ponds (5 impacted and 5 unimpacted) across coastal South Carolina. Not only do coastal South Carolina turtles not follow the same male biased pattern of many other studies, there was an overall female bias (154:95). The distribution and foraging behavior of predatory salt marsh fishes is shaped by the movements of prey, many of whom seek shelter in shallow and tributary creeks. I hypothesized that the distribution of piscivorous fishes in marsh creek channels would differ with proximity to the mouths of intertidal creeks and with tidal stage. Custom-built trotlines baited with live minnows were deployed during four discrete tidal stages at two microhabitats in main creek channels: (1) adjacent to intertidal creek mouths, and (2) along straight banks in North Inlet estuary, South Carolina. Stable isotope analyses of multiple tissue types have been used to reconstruct life histories of individuals from past populations. This thesis presents the life history reconstructions of a unique population recovered from a 16th to 18th century cathedral crypt located in Vilnius, Lithuania. The sample consists of 23 individuals (21 males, one possible female, and one juvenile). Stable carbon, nitrogen, and oxygen isotope analyses were performed on bone and dentin collagen, and on bone and enamel hydroxyapatite, resulting in eight isotope values per individual, providing both childhood and adult dietary and migration information. We previously demonstrated that embryonic stem (ES) cells over-expressing tissue inhibitor of metalloproteinase-1 (TIMP-1) have increased potential to engraft and differentiate into cardiac myocytes following transplantation into the infarcted heart. However, the ability of TIMP-1 to activate endogenous stem cells and enhance their differentiation into cardiac regenerative cell types is still unknown. We postulate that TIMP-1 may additionally activate a stem cell population that enhances cardiac cell type differentiation in the infarcted myocardium. Hantaviruses, family Bunyaviridae, are present throughout the globe in a variety of mouse, rat, mole, vole, shrew, or bat species. Hantaviruses persist for the lifetime of the animal reservoir, while causing no signs or symptoms of disease. Only the rodent-borne hantaviruses cause disease in humans. In contrast, a “spillover” infection of a hantavirus into a nonreservoir rodent species results in an asymptomatic acute infection. We and others in the field are interested in understanding the biology of these virus-host interactions and mechanisms that underlie these three very different outcomes. Complete Congenital Stationary Night Blindness (cCSNB) is a rare hereditary retinal disorder characterized by abnormal night vision. cCSNB is caused by postsynaptic defects in On bipolar cells (BCs) and is identified by the presence of an electroretinogram (ERG) with a normal a-wave, corresponding to photoreceptor function, and the absence of a b-wave, corresponding to a failure of On BC function. Legionella pneumophila is a facultative intracellular pathogen that infects a wide array of protozoan hosts and human alveolar macrophages. L. pneumophila is dependent on a functional Dot/Icm type IVB secretion system that translocates bacterial effector proteins into the host cell cytosol. L. pneumophila genomes encode more than 250 effector proteins, many of which inhibit host cellular processes to form a favorable niche termed the Legionella-containing vacuole (LCV). Polychlorinated biphenyls (PCBs) are persistent environmental toxicants, present in 100% of US adults and dose-dependently associated with nonalcoholic fatty liver disease (NAFLD) in epidemiologic studies. PCBs are predicted to interact with receptors previously implicated in xenobiotic/energy metabolism and NAFLD. These receptors include the aryl hydrocarbon receptor (AhR), pregnane xenobiotic receptor (PXR), constitutive androstane receptor (CAR), peroxisome proliferator-activated receptors (PPARs), liver-X-receptor and farnesoid-X-receptor. The work presented in this dissertation outlines the role of stem cells in the remodeling of adipose tissue under conditions of nutrient excess. Obesity-induced variations in adipose tissue stem cell distribution were uncovered by analysis of the stromal vascular fraction isolated from mice fed a high fat diet for several weeks. Bone marrow cell transplantation was used to determine the derivation of progenitor cells found in adipose tissue; and supplementation of depleted progenitor cell populations achieved via direct cell transplantation, helped to determine the contribution of these progenitor cells to the remodeling process. This response takes place by means of regulated signaling transduction which elicits changes in gene expression, biochemical, and metabolic properties. Adult muscle is composed by alignment of multinucleated, post mitotic muscle fibers formed during embryonic and neonatal development. The type IIa sodium-phosphate cotransporter (Npt2a) is expressed in the apical membrane of the renal proximal tubule and is responsible for the reabsorption of the majority of the filtered load of phosphate. Parathyroid hormone (PTH) is secreted by the parathyroid gland in response to a decrease in serum calcium or an increase in serum phosphorus, and acutely induces phosphaturia through the rapid stimulation of endocytosis of Npt2a and its subsequent lysosomal degradation. Chronic PTH stimulation leads to sustained phosphaturia, but the mechanisms for the chronic regulation of Npt2a by PTH remain unclear. We hypothesize that PTH decreases Npt2a mRNA levels …

Host cells respond to bacterial lipid A through Toll-like receptor 4 (TLR4). Activation of TLR4 by lipid A triggers a response that involves two main adaptor proteins, MyD88 and TRIF. MyD88-dependent gene expression is associated with proinflammatory protein production, while TRIF-dependent gene expression is essential for optimal activation of adaptive immunity by antigen-presenting cells. Detoxified, monophosphoryl lipid A agonists (MPLA or synthetic MLA) were previously suggested to elicit TRIF-biased TLR4 signaling; that is, induction of weaker MyD88-associated gene expression but relatively intact TRIF-dependent gene expression when compared to fully active diphosphoryl lipid A (lipid A). Invasive species are one of the leading causes of biodiversity loss and their presence can significantly impact the structure and function of native ecosystems. In this dissertation, the impacts of the invasive honeysuckle shrub, Lonicera maackii, on exotic earthworm populations, leaf litter decay responses, and inorganic and organic nutrient pools in an urban woodland park are assessed. Chapter 1 provides a brief review of honeysuckle's effects on forest ecosystems. Detrusor-sphincter dyssynergia after chronic spinal cord injury (SCI) leads to over-distention, high bladder pressures and vesico-ureteric reflux into the kidney, resulting in repeated infections and pyelonephritis. Given the importance/limited knowledge of SCI effects on the upper urinary tract, this study assessed expression of two proteins in the kidneys of contused (T9) male rats relative to controls that are indicative of tissue stress/damage (TGFβ, CD11b). In tumor cells, glutamine metabolism provides anaplerotic carbon for the TCA cycle, fatty acid synthesis, and precursors for the production of nucleotides and glutathione. This metabolic alteration is primarily driven by disruptions in oncogenic or tumor suppressor function and results in glutamine dependency for tumor cell survival. Troglitazone, a PPARγ agonist, has been reported to alter glutamine metabolism. This thesis project addresses whether troglitazone treatment could disrupt glutamine metabolism in glutamine-dependent tumor cells. Results obtained with troglitazone treatment include: dose-dependent inhibition of cell proliferation, glutamine uptake, glutaminolytic protein expression, steady-state ATP levels, and glutamine carbon flux into the TCA cycle. …

The embryonic program “epithelial-mesenchymal transition” (EMT) is thought to trigger cancer invasion by allowing tumor cell propagation. This thesis examines ways in which rural mountain women of Andean Peru and southern Appalachia use their lived histories and food knowledge in ways that counter Cartesian epistemologies regarding national and international food systems. Using women’s fiction and cookbooks, this thesis examines how voice and narrative reclaim women’s spaces within food landscapes. Further, this thesis examines women’s non-profits and grassroots organizations to illustrate the ways in which rural mountain women expand upon their lived histories in ways that contribute to tangible solutions to poverty and hunger in rural mountainous communities.
