Protein aggregation and inclusion body formation are hallmarks of neurodegenerative diseases such as Alzheimer's, Parkinson's, Huntington's, and amyotrophic lateral sclerosis (ALS). These neurodegenerative diseases share a common pathology in that all include accumulation of insoluble protein aggregates in the brain. TAR-DNA-binding protein (TDP-43) is the major component found in the pathological inclusions of two of these diseases, ALS and frontotemporal lobar degeneration with ubiquitin-positive inclusions (FTLD-U). This thesis focuses upon the biophysical basis for TDP-43 aggregation in S. cerevisiae. Current in vitro evidence indicates that TDP-43 is a natively dimeric protein and that binding to RNA inhibits aggregation. Corresponding …

Recent emphasis and attention by thinkers, media pundits, and politicians on terrorism requires new, critical evaluation of the processes by which terrorism is understood. By investigating the concept of biopolitics, as developed specifically through Michel Foucault and Giorgio Agamben, new insights into the interactions between terrorism, politics, and religion can emerge. Most notably, the attempts to explain terror as simply an economic problem, an excessive form of violence, and/or as religious fervency gone awry rely on embedded biopolitical concepts. Lactoferrin (LF) is an iron-binding glycoprotein of molecular mass ca. 80 kDa that is predominantly found in mammalian body fluids. Lactoferrin is a multifunctional protein that has a wide range of properties such as antibacterial, antiviral, anti-inflammatory, and anti-allergic functions. Tyrosine residues in the protein play a part in many lactoferrin functions. Protein tyrosine nitration modification represents an oxidative and nitrosative stress process which can be caused by the exposure of proteins to oxidants from air pollution or disease. Categorization is an essential survival skill that we engage in daily. A multitude of behavioral and neuropsychological evidence support the existence of multiple learning systems involved in category learning. COmpetition between Verbal and Implicit Systems (COVIS) theory provides a neuropsychological basis for the existence of an explicit and implicit learning system involved in the learning of category rules. COVIS provides a convincing account of asymptotic performance in human category learning. Emergent narrative, a phenomenon of unexpected contextual stories arising through play, has been researched in the field of game studies since 1999. However, that discussion largely lies in the realm of theoretical stories which are generated by either the system or the player. The purpose of this dissertation is to deepen our understanding of emergent narrative by examining real-world examples of the phenomenon. Background: Exposure therapy (EXP) is a first-line intervention for combat-related PTSD. EXP works by repeatedly exposing the patient to the feared stimuli, situation, or physical sensations in the absence of actual danger until the stimuli no longer evoke maladaptive responses. Over the past decade, multiple technologies have been introduced to augment the EXP process by presenting multi-sensory cues (e.g., sights, smells, sounds) to increase patients' sense of presence. Human activities generate surplus nutrients which may lead to algal bloom events in water resources along with serious ecological problems and thus substantial economic losses. Particularly, harmful algal blooms (HABs) represent toxic cyanobacterial blooms which produce cyanotoxins. The primary concerns of HABs are the exposures to a wide variety of cyanotoxins via ingestion of contaminated drinking water, inhalation during recreational activities, and consumption of contaminated fish and shellfish. In order to explore how to better utilize simulated odors for live training, a study of 180 United States Military Academy at West Point cadets was undertaken to determine whether pre-exposure to a simulated malodor may result in an amelioration of performance issues, as well as improving performance of a complex task. Exposure to malodors has long been shown to increase stress and escape behavior, and reduce performance of complex tasks, in addition to degrading other human factor areas. The study of anatomy is complex and difficult for students in both graduate and undergraduate education. Researchers have attempted to improve anatomical education with the inclusion of three-dimensional visualization, with the prevailing finding that 3D is beneficial to students. However, there is limited research on the relative efficacy of different 3D modalities, including monoscopic, stereoscopic, and autostereoscopic displays. Medical simulations provide hands-on training at various levels of medical expertise. Yet these simulators fail to accurately mimic the look, feel and behavior of human tissue. Applying measured mechanical properties from human cadaver tissues promises to improve the fidelity of simulated tissue behaviors when subjected to medical procedures. Samples of human parietal pleura were tested under uniaxial tension to failure and measured characteristics were replicated in synthetic pleura. Context specific parameters were then collected and compared between human pleura and the new synthetics. The magnitude of the overall settlement depends on several variables such as the Compression Index, Cc, and Recompression Index, Cr, which are determined by a consolidation test; however, the test is time consuming and labor intensive. This dissertation examines the pervasiveness of storytelling in theme parks and establishes the theme park as a distinct narrative medium. It traces the characteristics of theme park storytelling, how it has changed over time, and what makes the medium unique. This was accomplished using a mixed methods approach drawing data from interviews with creative professionals, archival research, fieldwork, and an analysis of more than eight hundred narrative attractions. I also consider the experiences of past generations of American black bodies and how their history has molded my world. This thesis examines the commemoration of Indian soldiers who died during the First World War by the Imperial War Graves Commission, Britain's official government body overseeing all imperial commemoration efforts. For the soldiers of the Indian Army their war experience was split between the Western Front in Europe and Mesopotamia in modern-day Iraq. They were also far more ethnically, religiously, and lingually diverse than their British and Dominion counterparts. This digital public history project explores one of the oldest and longest running of Florida's roadside attractions, Weeki Wachee Springs, during the years considered to be the park's heyday, the 1950s through the mid-1970s. With the 75th anniversary of the park approaching in 2022 and preliminary discussions of a new or expanded mermaid museum, there is a growing need to document the experiences of aging former employees and preserve park-related ephemera from that period. Virtual humans serve as role-players in social skills training environments simulating situational face-to-face conversations. Previous research indicates that virtual humans in instructional roles can increase a learner's engagement and motivation towards the training. Left unaddressed is if the learner is looking at the virtual human as one would in a human-to-human, face-to-face interaction. Density-dependent growth is often observed in stream-dwelling Brown Trout Salmo trutta populations. In Spearfish Creek, South Dakota, biomass of adult Brown Trout (>200 mm) is about three times greater than that reported for similar Black Hills streams while mean length of adult fish is about 30% less. Here, we evaluate large-scale density reduction as a management tool for improving growth rate of stream-dwelling Brown Trout. We compared age-specific growth of wild Brown Trout in stream sections receiving 50% reductions in fish abundance. This thesis examines public parks as New Common Pool Resources through a case study of the Dakota Nature Park in Brookings, South Dakota. I identify the formalization and bureaucratization processes experienced by the governing body of the park. These processes occurred as a capped landfill was repurposed and collaboratively managed to serve the community by providing native, natural space and affordable recreational opportunities. Two evaluations were conducted to evaluate the biological and economic production efficiency of retaining replacement females in a straight-bred Angus herd (HR) as opposed to purchasing crossbred replacements for a terminal sire production system (PR) where maximum maternal and individual heterosis is achieved. The first study was a comparison of the HR system versus PR system. It was modeled after a 100 hd, straightbred Angus cowherd that raises their own replacement females. There are only two natural ways by which molecular nitrogen can be available to support life, either by free-living or by symbiotic nitrogen fixing bacteria. The best studied diazotrophs form symbiotic associations with plants, primarily legumes and certain tree species, but little is known about how non-leguminous plants such as grasses obtain nitrogen in their environment. Brassica carinata is an oilseed crop that requires a relatively short growing season and produces high crop yields. It is a great source for aviation biofuel, however it produces a large amount of waste that may be used as a protein supplement for cattle. We tested the effects of solvent- or mechanically-extracted carinata meal as a protein supplement to ad libitum ground or long-stem corn residue (i.e., corn plant left over from grain harvest) had on cow performance and digestibility. Six experiments were conducted for this dissertation research to evaluate strategies to improve growth performance and health of calves. Growth performance, nutrient utilization, and health effects were evaluated when calves were offered water treated with reverse osmosis system (RW), fed starter pellets supplemented with microbially-enhanced soy protein (MSP) or supplemented condensed whey solubles (CWS). The first two experiments were conducted to determine drinking preference, growth performance, and health of dairy calves offered RW or by a municipal water treatment plant (MW), compared with local untreated well water (WW).
