Colorectal cancer (CRC) is the third leading cause of cancer-related deaths within the United States. A variety of factors including exposure to environmental carcinogens, such as benzo[a]pyrene (B[a]P), dysbiosis of the gut microbiome and hereditary influences, including APC mutations, have been shown to lead to an increased risk of developing colorectal cancer. Pseudomonas aeruginosa is a Gram-negative bacillus able to colonize a wide variety of environments. In the human host, P. aeruginosa can establish an acute infection or persist and create a chronic infection. P. aeruginosa is able to establish a niche and persist in human hosts by using a wide array of virulence factors used for: movement, killing host cells, and evading immune cells and antibiotics. The overarching goals of the current work are to fill key gaps in the current understanding of alcohol consumption and the risk of metastasis to the liver. Considering the evidence this research group has compiled confirming that the hepatic matrisome responds dynamically to injury, an altered extracellular matrix (ECM) profile appears to be a key feature of pre-fibrotic inflammatory injury in the liver. This group has demonstrated that the hepatic ECM responds dynamically to alcohol exposure, in particular, sensitizing the liver to LPS-induced inflammatory damage. Loss of function mutation in CD11b has been associated with incidence of systemic lupus erythematosus (SLE), a disease driven by B cell production of pathogenic autoimmune antibody. Our previous work has revealed the ability of CD11b to regulate B cell receptor (BCR) signaling and control autoimmune disease in mice. However, how CD11b regulates the immune response under normal conditions remains unknown. Through the use of a CD11b knockout model, we demonstrated that CD11b-deficient mice have an elevated antigen-specific humoral response in a Th2 type immunization. Microbotryum lychnidis-dioicae causes anther smut fungus in its host plant, Silene latifolia. The goal of this work is to identify and characterize the virulence determinants of this pathogen to better understand the molecular basis behind this host-pathogen interaction. This work studied for the first time the key effectors in the mechanism of infection by this fungal species. Using, yeast two-hybrid screens, I have identified the host plant interaction partners for the effector, MVLG_01732. A second effector MVLG_05720, interacts with other fungal proteins that appear to facilitate the fungal establishment and colonization during the infection. Our findings indicate that a …

Familial hypercholesterolemia (FH) is a common genetic disease and has been studied with the aim of finding a curative measure for decades. FH is caused by mutations in the low-density lipoprotein receptor (LDLR) resulting in defects in LDL-cholesterol (LDL-C)-receptor mediated endocytosis and development of premature cardiovascular disease (CVD). Here I describe the use of a corrected and non-corrected LDLR FH cell model to investigate receptor-mediated endocytosis and statin effects. For the past few decades, researchers in mathematics education have been exploring the concept of pedagogical content knowledge (PCK)—or knowledge related to teaching content—and applying it to various areas of mathematics, such as algebra. Research related to teacher knowledge of algebra is critical because researchers (e.g., Hill, Rowan, & Ball, 2005) have found correlations between some types of teacher knowledge and student achievement in mathematics; students from around the world are outperforming U.S. students on international assessments of mathematics, including algebra (Organization for Economic Cooperation and Development, 2014, 2016); and algebra plays an integral role in the K-12 mathematics curriculum …

Conquerors Or Cowards: The Role Of The Kentucky Mounted Militia In The Indian Wars From 1768 To 1841., Joel Anderson

The thesis argues that Kentuckians developed the myth that the Kentucky Mounted Volunteers were the most effective troops to fight Native American warriors in the Northwest Indian War of 1790 to 1794 and the War of 1812. The idea that these troops were the best fighters originated in the decades following the War of 1812 as Kentuckians generated a communal history. Residents of the state listened to orators mythologize the successes of mounted Kentuckians in battle, while remembering the foot militia for their sacrifices rather than their shortcomings.

A recent study demonstrated an association between the Porphyromonas gingivalis (Pg)infection and the progression of ESCC (Gao et al., 2016). However, how Pg infection affects the nature of ESCC remains unknown. We examined the effects of Pg on the proliferation and chemotherapy drug-induced apoptosis of ESCC cells and elucidated the signaling molecules involved in these processes. Method: Apoptosis and cell proliferation were tested using Flow cytometry and Tetrazolium salt based proliferation assays respectively. Western Blot was utilized to detect the protein expression. This thesis examines the history of two North American Indigenous groups, those belonging to the Great Plains and the Arctic, and observes how settler-colonial influence determined the collection and curation of arts and artifacts in these areas. This art includes a mention of pre-Colombian works, but focuses predominantly on works being made after “first-contact” through the contemporary ear. The paper addresses the effect imperialist history has had on the development of Indigenous art markets, and how institutions such as museums may address them through ethical practices, and efforts to decolonize museum spaces.

This dissertation examined the contributing factors to the success of high achieving African American male high school students who attend an all-male, predominantly Black, college-preparatory academy. The study took place in an urban, Midwestern school district and highlighted the lived experience and perspectives of students who have at least a 3.0 grade point average. Background. Vinyl chloride (VC) is an abundant environmental contaminant that causes steatohepatitis at high exposure levels. We have shown previously that low concentrations of VC exacerbate high fat diet (HFD)-induced liver injury in mice. The mechanisms involved in the progression of liver injury caused by VC and HFD include oxidative stress, inflammation, metabolic and mitochondrial dysfunction. Mitochondrial aldehyde dehydrogenase 2 (ALDH2) serves as a key line of defense against most reactive aldehydes, including lipid aldehydes (e.g., 4-HNE) and chloroacetaldehyde (VC metabolite). There are thirteen known endogenous EGF-like ligands. We previously reported that Betacellulin (BTC) increases ligand-mediated corneal wound healing more than Epidermal Growth Factor (EGF) [Peterson et al. (2014) IOVS 55(5):2870-80], although the molecular reason for this is unknown. Despite being better at promoting wound healing via enhanced cell migration, BTC has reduced receptor affinity and weaker induction of EGFR phosphorylation. These data indicate that BTC’s response is not due to enhanced affinity or EGFR-kinase activity. Receptor phosphorylation and proximity ligation assays indicate that BTC treatment significantly increases ErbB3 phosphorylation and EGFR:ErbB3 heterodimers. BTC traffics EGFR at a faster rate than …

Hexavalent chromium [Cr(VI)] is a widespread environmental and occupational carcinogen. The mechanism of carcinogenesis remains poorly understood, but chromosome instability (CIN) is the dominant theory. How numerical CIN arises is unclear, but it correlates with centrosome amplification. Both phenotypes are hallmarks of cancers, early events in carcinogenesis, and have been shown to occur after Cr(VI) exposure to human skin and lung fibroblasts. In this study, we investigate numerical CIN and centrosome amplification in whale cells. Whales are our closest marine relatives, have long lifespans, and are exposed to environmental Cr(VI). Invasive plants can alter pollination dynamics in invaded communities by disrupting patterns of pollinator visitation, pollen transfer dynamics (conspecific [CP] and heterospecific [HP]), and reproductive success. The direction of invasive effects (competitive, neutral, and facilitative) may be partially determined by spatial scale and species’ floral traits. Here, we investigated pollinator visitation, CP and HP receipt, and pollen tube growth for species in a C. arvense present community and non-present community at two scales. SABP2-interacting protein 470 (SIP470), a non-specific lipid transfer protein (nsLTP), was discovered in a yeast two-hybrid screening using SABP2 as bait and tobacco leaf proteins as prey. SABP2 is an important enzyme in systemic acquired resistance that converts salicylic acid to methyl salicylate. Localization studies are an important aspect to understanding the biological function of proteins. nsLTPs are generally considered apoplastic proteins and has been localized intracellularly and extracellularly. Transient expression shows highest expression of SIP470-eGFP at 2 days post infiltration into Nicotiana benthamiana. Confocal microscopy showed localization near the periphery of the cell. This is an ethnographic case study of a musical community in Catalonia centered around the performance of bluegrass and old-time music. By using Victor and Edith Turners’ ideas of normative communitas, this paper identifies an aesthetic communitas model which describes a community centered around a performative genre. Through participant observation in the 16th Annual Al Ras Bluegrass and Old Time Music Festival and interviews with local musicians, fans, venue owners, and luthiers, the ethnographic narrative details the characteristics of the aesthetic communitas in Catalonia and searches for associations of Appalachia that accompany the cross-cultural manifestation of bluegrass and old-time music …

Excessive alcohol consumption is the primary contributing factor in the development of alcoholic liver diseases (ALD). Nicotine contained in tobacco is a major addictive alkaloid, which enhances the effects of ALDs. The major enzyme involved in nicotine metabolism is cytochrome P450 2A5 (CYP2A5) which is produced in the liver. Alcohol can stimulate the CYP2A5 enzyme. We utilized cyp2a5-/- knockout mice in this research to examine the effects of CYP2A5.

Cadmium is present in food and groundwater. Tobacco smoking and occupational exposure are also major sources for cadmium. Cadmium is primarily accumulated in liver, a major organ metabolizing exogenous chemicals. Chemical metabolism may cause detoxification, but it can also cause bio-activation resulting in liver damage. Cytochrome P450s (CYP) are major liver metabolism enzymes, and cadmium chloride (CdCl2) can induce CYP2A5 in mice. We examined the effect of CYP2A5 on CdCl2-induced liver injury using CYP2A5-knockout (cyp2a5-/-) mice. This dissertation describes how poly-chlorinated biphenyls (PCBs) exacerbate the pathogenesis of non-alcoholic fatty liver disease (NAFLD). While PCBs were banned in 1979, they still persist in contaminated biota, including food, and are detected in human plasma and adipose. The body burden of PCBs is associated with elevation of liver enzymes and necrosis markers in humans, characteristic of NAFLD. PCB exposure in high-fat diet fed mice leads to steatohepatitis that recapitulate the findings seen in exposed humans. The global estimate of people diagnosed with NAFLD is up to 1 in 4 people, unrelated to dietary or genetic factors. The hepatic mechanisms …

SABP2 (Salicylic Acid Binding Protein 2) plays a vital role in the salicylic acid signaling pathway of plants both regarding basal resistance and systemic acquired resistance against pathogen infection. SIP-428 (SABP2 Interacting Protein-428) is a Silent information regulator 2 (SIR2) like deacetylase enzyme that physically interacts with SABP2 in a yeast two-hybrid interaction and confirmed independently by a GST pull-down assay. We demonstrated that SIP- 428 is an NAD+ dependent SIR2 deacetylase enzyme. Transgenic tobacco plants silenced in SIP- 428 expression via RNAi showed enhanced basal resistance to microbial pathogens. Projections from the Nucleus Accumbens (NAc) to the Ventral Pallidum (VP) play a critical role in motivation and reward. Rewards and reward-associated cues are thought to alter this pathway by suppressing GABA release to the VP, however, the role of the NAc to VP pathway has never been investigated with regard to nicotine self-administration. We hypothesized that increasing GABAergic signaling from the NAc to the VP would decrease the reinforcing effects of nicotine. Chlamydia trachomatiscauses a bacterial sexually transmitted infection, Chlamydia, that is often chronic and casues reproductive complications in women. We hypothesized that Chlamydia infection increases local acetylcholine (ACh) production, which regulates the host’s inflammatory response to the infection. Female mice infected with C. muridarumwere sacrificed at days 3, 9, 15, and 21 post-infection, genital tract tissues harvested, and immunohistochemistry performed to enumerate ACh-producing cells. The initial and maintenance effects of a Daily Behavior Report Card (DBRC) intervention and fading procedure on Academic Engaged Time (AET) of a 6-year-old boy with Developmental Delay and Attention Deficits were evaluated with a reversal design. Following an initial baseline, the DBRC was implemented and then completely removed followed by reapplication of the DBRC. Organisms adapt to their environments by adjusting their biochemistry and physiology; such adaptation is limited by resource availability and physiological constraints. The freshwater crustacean Daphnia magna inhabits a wide range of environments and must survive and reproduce within a range of temperatures. One limit to low-temperature adaptation is thought to be the availability of unsaturated fatty acids necessary to maintain proper fluidity of cellular membranes. Animal and human wastes introduce pathogens into rivers and streams, creating human health and economic burdens. While direct monitoring for pathogens is possible, it is impractical due to the sporadic distribution of pathogens, cost to identify, and health risks to laboratory workers. To overcome these issues, fecal indicator organisms are used to estimate the presence of pathogens. Although fecal indicators generally protect public health, they fall short in their utility because of difficulties in public health risk characterization, inconsistent correlations with pathogens, weak source identification, and their potential to persist in environments with no point sources of fecal pollution. Rhodococcushas been recognized as a potential antibiotic producer. Recently, a strain of Rhodococcussp. MTM3W5.2 was isolated from a soil sample collected in Morristown, Tennessee and was found to produce an inhibitory compound which is active against other related species. The purpose of this research is to extract, purify and analyze the active metabolite. The compound was extracted from RM broth cultures and purified by preliminary fractionation of crude extract through a Sephadex LH-20 column. Further purification was completed using semi-preparative reversed phase column chromatography. Final purification was obtained using multiple rounds of an analytical C18 HPLC column. Based …
