Heart And Sole: The Functional Role Of Fast-Skeletal Myosin Binding Protein-C In Cardiac And Skeletal Muscle, Brian Leei Lin Jan 2016

Heart And Sole: The Functional Role Of Fast-Skeletal Myosin Binding Protein-C In Cardiac And Skeletal Muscle, Brian Leei Lin

Dissertations

The goal of my dissertation was to compare and contrast the function of all three major isoforms of Myosin Binding Protein-C (MyBP-C): slow-skeletal, fast-skeletal, and cardiac (ssMyBP-C, fsMyBP-C, and cMyBP-C, respectively), with a focus on the least characterized isoform, fsMyBP-C. Using a variety of ex vivo, in vitro, and in silico methods, my research demonstrated that the N-terminal region of all MyBP-C isoforms bind to actin and shift tropomyosin, thus activating the thin filament during contraction. Furthermore, each isoform differentially activated the thin filament over isoform-specific ranges of Ca2+: slow-skeletal activates at low Ca2+, fast-skeletal activates at higher Ca2+, and …

The Impact Of Altered T Cell Receptor—Peptide-Major Histocompatibility Complex Interactions On Antigen Recognition And T Cell Function, Timothy T. Spear

Dissertations

Adoptive cell transfer (ACT) using T cell receptor (TCR) gene-modified T cells is an exciting and rapidly evolving field. Numerous basic science and clinical studies have demonstrated various levels of feasibility, safety, and efficacy using TCR-engineered T cells to treat cancer and viral infections. Genomic instability of targeted diseases, however, requires effective and safe TCRs to cross-recognize mutated antigens while minimizing on- or off-target toxicities. Thus, improvements to T cell-based therapeutics mandate a broader understanding of the principles governing antigen recognition. This dissertation addresses critical biologic questions evaluating which parameters are most important in facilitating antigen recognition, and how alterations …

Consequences Of Estrogen Receptor Beta Phosphorylation In The Aged Female Brain And Heart, Elena Pinceti Jan 2016

Consequences Of Estrogen Receptor Beta Phosphorylation In The Aged Female Brain And Heart, Elena Pinceti

Dissertations

Female life expectancy in the United States has steadily increased and now averages 81 years. Meanwhile, the age at which women reach menopause remains constant at 51 years. Both neurological and cardiovascular disorders increase following menopause, affecting women’s quality of life and leading to large health care costs. Estrogens are neuroprotective and cardioprotective for women, however when administered long after menopause, they fail to improve their health.

The effects of estrogen are mediated by Estrogen Receptor α (ERα) and Estrogen Receptor β (ERβ). Evidence supports that ERβ plays an important role in neuroprotection and cardioprotection, but aspects of ERβ molecular …

The Psychological Impact Of Smartphones: The Effect Of Access To One's Smartphone On Psychological Power, Risk Taking, Cheating, And Moral Orientation., Amanda Christine Egan

Dissertations

Egan and Larson (2015) found that access to one’s smartphone resulted in an increase in one’s sense of psychological power. Psychological power is associated with a variety of behavioral outcomes, many of them moral in nature (Keltner, Gruenfeld, & Anderson, 2003). This dissertation attempted to conceptually replicate the findings obtained by Egan and Larson (2015) and to extend them by testing whether smartphone-induced power had moral implications. Specifically, Study 1 tested whether access to one’s smartphone increased psychological power, and in turn promoted risk taking and a shift in moral orientation. Study 2 tested whether access to one’s smartphone increased …

The Amino Terminal Region Of Cardiac Myosin Binding Protein-C Is Necessary For Cardiac Function, Thomas Lawrence Lynch Jan 2016

The Amino Terminal Region Of Cardiac Myosin Binding Protein-C Is Necessary For Cardiac Function, Thomas Lawrence Lynch

Dissertations

Cardiac myosin binding protein-C (cMyBP-C) is a thick filament-associated protein that has been suggested to regulate cardiac contraction via its amino terminal (N’) region. Following ischemic injury to the heart, cMyBP-C is cleaved into a predominant N’ fragment consisting of domains C0 through C1 and the first 17 residues of the M-domain that is referred to as C0-C1f. However, the necessity of the N’-C0-C1f region of cMyBP-C in regulating cardiac function in vivo has not been elucidated. I hypothesized that the N’-C0-C1f region of cMyBP-C is critical for normal cardiac function in vivo. To test this hypothesis, transgenic (TG) mice …

To Be Black, Caribbean, And American: Social Connectedness As A Mediator To Racial And Ethnic Socialization And Well-Being Among Afro-Caribbean American Emerging Adults, Gihane Emeline Jeremie-Brink

Dissertations

Racial and ethnic socialization are integral to the functioning and parenting process in ethnic minorities’ families (Brown & Krishnakumar, 2007; Hughes, Rodriguez, Smith, Johnson, Stevenson, & Spicer, 2006). Unfortunately, there is no scholarly consensus with respect to definitions and operations for racial and ethnic socialization which then evidences several conceptual and methodological shortcomings in racial and ethnic socialization research (Brown, 2004). Furthermore, very little empirical research has used these findings in relation to the socialization processes of first- and second-generation Afro-Caribbean emerging adults.

The purpose of this study was to test the roles of both racial socialization and ethnic socialization …

“My Masculinity Is A Little Love Poem To Myself”: Trans*Masculine College Students’ Conceptualizations Of Masculinities, T.J. Jourian

Dissertations

Men and masculinities studies in higher education are gaining prominence within the literature, illuminating how cisgender men understand and grapple with masculinity on college campuses. Additionally, the increased visibility of trans* students has fueled the expanding scholarship and attention to their experiences, often however centering on White gender-conforming trans* students with little if any focus on their multiple and intersecting identities. This phenomenological study seeks to bridge these two areas of literature, by investigating how trans*masculine students understand, define, and adopt a masculine identity, and how that identity is informed by their various intersecting and salient identities. Dominant masculinities function …

Parental Involvement, Socio-Economic Status And Students' Perceptions Of The Classroom Learning Environmnet As Predictors Of 8th Grade Students Academic Achievement: A Structural Equation Model, Allyson Blandin

Dissertations

Problem

Academic achievement among the nation’s youth has been on the decline for decades. The statistics from the National Assessment of Educational Progress (NAEP) paints a bleak picture of the academic performance of more than half of the 4th and 8th grade students being below the required levels of proficiency in reading and mathematics. This is evidence that a problem of low academic achievement exists among certain student groups within the public education system. Therefore, it creates an academic achievement gap, which is reflected in the disparity in the standardized scores between students of color from low socio-economic status with …

Sex Differences In Stroke Recovery: Synaptic Proteins And The Growth Inhibitory Protein Nogo A, Vincent Joseph Borkowski

Dissertations

Ischemic stroke is a major world-wide health problem, resulting in death and disability especially in the older population. A sex difference exists in functional recovery post-stroke, with post-menopausal women having worse functional outcome as compared to age-matched men. Although the mechanisms underlying this sex difference are not entirely clear, it is recommended that any potential therapy for stroke recovery be tested in pre-clinical models including both male and females in order to determine the efficacy of the proposed treatment on the entire population. We have reported a novel therapy to enhance brain plasticity and improve functional recovery after stroke in …

Dissertations

An exponential growth of data traffic that originates on edge devices and a shift toward cloud computing necessitate finding new approaches to optimize file transfers. Whereas compression utilities can improve effective throughput and energy efficiency of file transfers between edge devices and the cloud, finding a best-performing utility for a given file transfer is a challenging task. In this dissertation, we introduce a framework for optimizing file transfers between edge devices and the cloud using compression utilities. The proposed framework involves agents running on edge devices and the cloud that are responsible for selecting an effective transfer mode by considering …

Associative Data Analytics And Its Application To Protein Crystallization Analysis, Imren Dinc Jan 2016

Associative Data Analytics And Its Application To Protein Crystallization Analysis, Imren Dinc

Dissertations

Data analytics focuses on processing raw data to extract important information and involves data acquisition, data analysis, and visualization. To maximize the amount of information obtained, an efficient experimental design method is necessary. Many existing experimental design methods do not consider the previous experimental results when constructing new trial conditions. It is quite difficult to employ supervision in this domain due to limited data size and skewed data distribution. Once the data is collected, proper analysis methods should be developed for making useful decisions regarding the type of data collected (e.g., numerical, time, visual, image analysis). Visualization that encompasses both …
