Patients with autosomal polycystic kidney disease (ADPKD) have an increased risk and worse outcomes after acute myocardial infarction, but the mechanism for this is unknown. Polycystin 2 (PC2), one of the two main proteins mutated in ADPKD, is a calcium permeant channel that localizes to the ER/SR. In non-cardiac cells, PC2 regulates the unfolded protein response (UPR) pathway under ER stress. However, whether PC2 is cardioprotective against ER stress is unknown. We found that PC2 expression increased in cardiomyocytes from human ischemic hearts and infarcted murine hearts. Cardiomyocyte-specific PC2 KO (PC2 KO) mice had worsened cardiac function after myocardial infarction …

Cells interpret biochemical and mechanical signals through dynamic regulation of the cytoskeleton and adhesion complexes, enabling key processes such as shape adaptation, force generation and migration. As these signals vary in both time and space, they can be challenging to resolve using traditional bulk assays. To overcome this limitation, we employ light-based optogenetic tools to reversibly modulate biochemical signals and mechanical equilibrium within cells. Specifically, we use the iLID optogenetic approach to locally activate signaling and manipulate contractility within the cytoskeleton for the purpose of investigating the role of RhoA and contractility on septin organization and adhesion protein localization. The 2025 Georgia English Language Arts standards indicate that teachers will be moving away from a monomodal approach to literacy learning to include students’ construction and evaluation of multimodal texts in grades 9-12. As such, this qualitative case study explored 39 Honors American Literature students' experiences with constructing multimodal arguments. The study sought to understand students’ perceptions and attitudes of creating an argumentative multimodal composition, including their attitudes toward each stage of the composing process: planning, drafting/designing, revising/editing, and publishing. Poor sleep quality and circadian rhythm disruption are commonly reported among cancer survivors and may contribute to worsened health outcomes, including increased fatigue, psychological distress, and reduced quality of life. A global majority of steams are non-perennial, drying at some point in time and space. Despite the vital roles that fungi and other microbes play in stream ecosystems, most research on stream microbiomes comes from perennial (continuously flowing) streams. The present series of studies addresses this research gap by examining microbial community composition and function in non-perennial streams, with an emphasis on fungi. This dissertation investigates how, and if, utopianism is being taught in college classrooms across the United States. Long considered impractical, overly idealistic, and/or politically dangerous, utopianism has been largely marginalized in political science and American culture. However, this project finds that there are instructors teaching utopianism and they are using the concept to challenge students to think differently about governance, justice, and institutional design. Drawing from political theory literature, course syllabi submitted by faculty from different U.S. universities, interviews with instructors, and personal case studies of my own courses at Central State University and Manchester University, I explore how utopian …

Adverse Childhood Experiences (ACEs) have been associated with many neurodevelopmental and affective disorders, including attention deficit hyperactivity disorder (ADHD) and generalized anxiety disorder (GAD), with more exposures increasing negative risks. Sex and genetic background are biological variables involved in adverse psychiatric outcomes due to ACE exposure(s). Few preclinical studies have properly powered each sex to determine the interaction between sex and genetics on stress-impacting behavior, especially in early life. This dissertation investigates the postcolonial condition of Nigerian woman through a critical philosophical engagement with feminist theory, focusing particularly on Simone de Beauvoir’s existentialism and Irigaray’s ethics of sexual difference. It interrogates the historical processes through which gender became the basis of hierarchy in the Nigerian society, arguing that colonial and patriarchal override precolonial forms of relationality—marked by dual-sex relationship and age-based hierarchies—into rigid, male-dominated structures of authority and representation. Drawing also on Oyèrónkẹ́ Oyěwùmí’s critique of western gender epistemologies, the work contextualizes the Nigerian woman’s experience within a broader matrix of colonial imposition, cultural displacement, and epistemic violence. This dissertation explores how the Russo-Ukrainian war has transformed the lives, identities, and resistance strategies of Ukrainian women. Drawing on in-depth, semi-structured interviews with forty-five Ukrainian women conducted between March 2022 and February 2023, it analyzes how everyday survival, political thought, and civic activism have changed during the war. Using a modified grounded theory approach and a feminist lens, this study examines women’s psychosocial experiences, their evolving political consciousness, and their active roles in humanitarian, cultural, and cyber resistance.Findings reveal that the war simultaneously reinforced and disrupted traditional gender expectations. Tourette Syndrome (TS) is a neurodevelopmental disorder characterized by motor and phonic tics that is frequently associated with functional impairments. The present study examined the relationship between motor and phonic tic severity, comorbid conditions, academic accommodations, and the social, emotional, and school functioning of children with TS. Caregiver data were collected on tic severity, comorbid conditions, social, emotional, and school functioning, academic accommodations, and grades. Overall, social, emotional, and school functioning fell within the moderate range, with school functioning positively associated with social and emotional functioning. The sustainability of America's National Park System increasingly relies on public–private partnerships that leverage private philanthropy. While the National Park Foundation provides national support, local Park Partner nonprofit organizations, like the Gateway Arch Park Foundation, are key to community-level fundraising and advocacy. This qualitative study of the St. Louis Arch grounds renovation examines how this public–private partnership structure influences donor motivation, trust, and engagement. The present study investigates the relationship between mathematics teachers’ qualifications and their students’ performance on standardized assessments, using data from a Midwestern urban school district’s ACT mathematics scores. Findings are discussed within the broader context of Science, Technology, Engineering, and Mathematics (STEM) education and national efforts to enhance competitiveness among industrialized nations. Engineered antibodies (Abs) have found applications in disparate fields such as enzyme catalysis and medicinal therapeutics because of their exquisite binding affinity and specificity. To engineer antibodies for these purposes, a firm understanding of the underlying mechanisms that control Ab affinity and specificity is needed, so these properties can be tightly controlled and adapted to the application at hand. This study utilizes Plato’s conception of purification as a diagnostic tool to examine contemporary issues of police brutality and political violence. Within this context, I aim to treat the multiple tensions that arise with the question of living a good life. What does it mean to live and what does it mean to live a good life within constraints that magnify fear, anxiety, and despair? I argue that philosophical purification involves ‘aporia’ (perplexity; difficulty; confusion; loss; or puzzlement) in both a literal and generative sense. Spina bifida (SB) is a common congenital birth defect with multiple and varied medical, physical, and psychological impacts. The objective of these studies is to examine youth and parent condition-related knowledge in the context of SB and associations with executive function, quality of life, and medical self-management. Collectively, these studies examine both qualitative and quantitative multi-informant data from youth with SB, their parents, and their teachers. Reactive sulfur species (RSS) such as hydrogen sulfide (H2S) and hydropersulfides (RSSH) are crucial mediators in a variety of physiological and pathological processes. Their regulatory roles span from maintaining cellular redox balance to influencing signaling pathways critical for homeostasis and stress responses. This dissertation investigates H2S and RSSH as biologically important molecules and explores the development of boron-dipyrromethene (BODIPY)-based photocages for its controlled, light-triggered release. Fish schools exhibit a collective behavior and self-organization that is mediated by hydrodynamic interactions between individual fish. However, the long-time evolution of hydrodynamically interacting collectives is challenging to investigate due to the persistent influence of long-lived vortical structures, and the high-resolution requirements of direct numerical simulation at large Reynolds numbers. Reduced-order models have therefore played an important role in theoretical investigations of collectives of swimming bodies. Thompson-Miller & Picca (2016) and Feimster (2023) argued that enslaved African American women, and even free African Americans after the Civil War were victims of non-consensual sexual activity and rape. The NISVS report revealed that before the age of 18 years old, 35.5% of African American women experienced contact sexual violence, 29.5% experienced non-contact unwanted sexual encounters, and 20.7% experienced rape (Basile et al., 2022). The issue of gun control is a hot topic in the field of American politics. It is a huge issue in the field of political science and in American public policy. In 2023, there were 18,874 firearm deaths in the United States, excluding suicides. The highest was in 2021 with the firearm related deaths at 21,068. Rett syndrome (RTT) is a rare neurodevelopmental disorder caused by mutations in the gene coding for methyl-CpG binding protein 2 (MeCP2). Currently, there is no cure for RTT and few effective treatment options to address the social, cognitive, motor, and respiratory symptoms that patients face. With the goal of identifying novel targets, our lab performed RNA-sequencing experiments in human autopsy samples and found decreased levels of the M1 muscarinic acetylcholine receptor. Cardiomyopathies, such as hypertrophic and dilated subtypes, are leading causes of heart failure and are often driven by genetic mutations that disrupt the structure and function of the cardiac sarcomere. While much of the research has focused on ventricular involvement, the role of atrial sarcomere dysfunction remains unexplored. The regulation of contractile function by sarcomeric proteins in the atria remains poorly understood, especially the impact of protein stoichiometry among myosin binding proteins. RNA biology is a rapidly expanding field, with many avenues of discovery among marine organisms. In this study, we analyze both mollusks and sawfish. First, we perform an evaluation of sRNA biogenesis pathways throughout Mollusca, with emphasis on the easter oyster, Crassostrea virginica. Understanding these molecules prescribes RNAi-based gene silencing approaches, benefiting genetic investigation and biotechnology. Similar to other animal groups, mollusks have conserved microRNAs (miRNAs), with some shared with ecdysozoans and deuterostomes; however, there is no evidence of an endogenous small interfering RNA (siRNA) pathway.
