Enhanced Triacylglycerol Production In Chlorella Vulgaris By Flux Balance Analysis On Metabolic Network, Issalapap Kaothong

Chulalongkorn University Theses and Dissertations (Chula ETD)

Triacylglycerol (TAG) is a type of lipid that can be produced by a microalga called Chlorella vulgaris, which is a promising source of biodiesel. However, there is a lack of research and models explaining how to directly produce this lipid. To address this, we propose using linear minimization of metabolic adjustment (LMOMA), which predicts gene manipulation effects based on Flux Balance Analysis (FBA), to enhance TAG production. We conducted single-gene deletion experiments on the genome-scale metabolic model of C. vulgaris UTEX 395 (iCZ843) under different growth conditions: heterotrophy, photoautotrophy, and mixotrophy conditions. We found that the TAG production increased by …

Development Of High Protein Beverage From Hydrolyzed Egg White, Nuntorn Charoensak Jan 2022

Development Of High Protein Beverage From Hydrolyzed Egg White, Nuntorn Charoensak

Chulalongkorn University Theses and Dissertations (Chula ETD)

The primary objective of this study was to examine the impact of enzymatic treatment on pasteurized egg white protein (EWP) and assess its influence on various physicochemical properties and antioxidant activity. Our findings revealed that the addition of SDAY10 enzymes led to a significant increase in lightness (L*), redness (a*), and yellowness (b*) parameters. Furthermore, higher concentrations of the enzyme resulted in a considerable reduction in pH, viscosity, and volume mean diameter (D4,3) of EWP protein compared to the control group (P < 0.05). Additionally, the application of SDAY10 enzyme (0.1-0.3% w/v) for EWP hydrolysis exhibited a noteworthy enhancement in antioxidant activity (measured by DPPH, ABTS, and FRAP assays) compared to lower enzyme concentrations. Notably, the 0.3% concentration of SDAY10 exhibited the highest release of amino acids during in vitro digestion and received the highest sensory evaluation score. Consequently, employing SDAY10 enzymes at a concentration of 0.3% w/v for EWP hydrolysis demonstrates significant potential for improving the physical properties of EWP and enhancing antioxidant activity. However, it is imperative to investigate the sensory evaluation with regards to the bitterness of the product developed using such enzymes.

Objective: Indirect Jagged1 immobilization efficiently activates canonical Notch signaling in hDPSCs. This study aimed to investigate the characteristic of the Jagged1-treated hDPSCs-derived dECM and its biological activity on odonto/osteogenic differentiation SCAPs. Methods: Bioinformatic database of Jagged1-treated hDPSCs was analyzed by NetworkAnalyst. hDPSCs seeded on Jagged1 immobilized surface were maintained with N or OM followed by decellularization procedure, dECM-N or dECM-OM, respectively. SCAPs were reseeded on each dECM with either normal medium or OM. Cell viability was determined by MTT assay. Characteristics of dECMs and SCAPs were evaluated by SEM, EDX, immunofluorescent staining and alcian blue staining. A Multipotential differentiation assay. …

Prediction Of Future Caries In Toddlers Via Salivary Microbiome: A 1-Year Longitudinal Study, Ratcha Raksakmanut

Chulalongkorn University Theses and Dissertations (Chula ETD)

Dental caries in children is the most common chronic oral disease that could disturb their quality of life including their development. Early prevention is a key approach to reducing the prevalence of early childhood caries. However, a reliable caries predictor, as an essential tool for targeted prevention that is important to this approach, is still lacking for infants before caries onset. Therefore, we aimed to develop the caries risk prediction model based on the salivary microbiome of caries-free 1-year-old children to predict caries onset at 1-year follow-up. Using a nested case-control design within a prospective cohort study, 30 saliva samples, …

Chulalongkorn University Theses and Dissertations (Chula ETD)

This study investigates the challenges NGOs face in providing healthcare access to the urban poor during the COVID-19 pandemic, using cases of slum communities in Kampala, the capital city of Uganda. The COVID-19 pandemic has disproportionately affected vulnerable populations, including those living in slums characterized by overcrowding, inadequate infrastructure, and limited access to basic services, especially access to healthcare, a fundamental right of people. During the past years, the pandemic has further restricted the already limited accessibility to healthcare services for the urban poor. The primary research methodology employed in this study is case study research, using in-depth interviews, observations, …

The Infection Dynamics, Co-Infection, Multi-Gene Phylogeny, And Molecular Screening Of Plasmodium Caprae Infection In Goats And Mosquito Species, Hoang Lan Anh Nguyen

Chulalongkorn University Theses and Dissertations (Chula ETD)

Ungulate malaria parasites, including Plasmodium caprae that infects the domestic goats, have received little attention. There are only few studies about the prevalence and mitochondrial genome analyses. This study, therefore, aimed to investigate the natural infection dynamics of P. caprae and its mosquito vectors. Besides, genome analyses based on nuclear genes of P. caprae and other related haemosporidian parasites were also carried out to obtain an in depth understanding about their evolutionary history and origin. A total of 423 goat blood samples were collected during rainy seasons from 2018 to 2021 to assess the malaria infection status. In addition, 1,019 …

Establishment Of Transplantation Platform For Delivering Mouse Induced Pluripotent Stem Cell-Derived Insulin-Producing Cells (Mips-Ipcs) For Diabetes Treatment, Hong Thuan Tran

Chulalongkorn University Theses and Dissertations (Chula ETD)

Pancreatic beta-cell replacement is recognized for feasible type 1 diabetes (T1D) treatment. However, in post-transplantation, the autoimmune destruction incidentally attacks the activity and survival of beta-cells are reported in animal and human. To address these concerns, the generation of immortalized, biocompatible beta-cells, and the engraftment platform are insightfully investigated. The stepwise chemical process was used for in vitro Insulin-producing cells (IPCs) production from mouse gingival fibroblast-induced pluripotent stem cells (mGF-iPSCs). The real-time qRT-PCR, glucose stimulation C-peptide/Insulin secretion, immunostaining, and visible cell methods were examined during IPC differentiation. The encapsulated-IPC beads were loaded into subcutaneous pocket space via transplantation platform. Completed …
