Studies Of Chemical Fixation Effects In Human Cell Lines Using Raman Microspectroscopy, Aidan Meade, Colin Clarke, Florence Draux, Ganesh Sockalingum, Michel Manfait, Fiona Lyng, Hugh Byrne

Articles

The in vitro study of cellular species using Raman spectroscopy has proven a powerful non-invasive modality for the analysis of cell constituents and processes. This work uses micro-Raman spectroscopy to study the chemical fixation mechanism in three human cell lines (normal skin, normal bronchial epithelium, and lung adenocarcinoma) employing fixatives that preferentially preserve proteins (formalin), and nucleic acids (Carnoy’s fixative and methanol–acetic acid). Spectral differences between the mean live cell spectra and fixed cell spectra together with principal components analysis (PCA), and clustering techniques were used to analyse and interpret the spectral changes. The results indicate that fixation in formalin …

Optical Properties Of Photopolymer Layers Doped With Aluminophosphate Nanocrystals, Elsa Leite, Tzvetanka Babeva, E Ng, Vincent Toal, Svetlana Mintova, Izabela Naydenova

Articles

The optical properties of photopolymer layers consisting of an acrylamide–based matrix and microporous aluminophosphate nanocrystals of AEI- type are investigated. The compatibility of the photopolymer doped with the nanoparticles is studied. The surface and volume properties of the layers with different levels of doping with microporous nanocrystals are characterized. The effective refractive indices and absorption coefficients of the doped photopolymer layers are determined and used to calculate the refractive index and porosity of pure AEI nanoparticles used as dopants. Volume transmission gratings were recorded in the doped photopolymer layers at different spatial frequencies. By spatial monitoring of the characteristic Raman …

Medium Mediated Effects Increase Cell Killing In A Human Keratinocyte Cell Line Exposed To Solar Simulated Radiation, Fiona Lyng, Alanna Maguire, Brian Morrissey, James E. Walsh

Articles

Purpose: The objective of this study is to investigate whether cell culture medium is 40 a biologically relevant exposure medium that can be employed in non-ionising photobiological investigations. 42 Methods: The effect of solar simulated irradiation on cell culture medium and its ability to elicit cell death was studied. The role of reactive oxygen species (ROS), cell 44 secreted factors, and the contribution of individual components of the medium were investigated. 46 Results: Cell death was found to be primarily mediated through the formation of ROS via riboflavin photosensitisation and degradation in the cell culture medium. Phenol 48 red was …

Ultrasound-Assisted Swnts Dispersion: Effects Of Sonication Parameters And Solvent Properties, Qiaohuan Cheng, Sourabhi Debnath, Elizabeth Gregan, Hugh Byrne Jan 2010

Ultrasound-Assisted Swnts Dispersion: Effects Of Sonication Parameters And Solvent Properties, Qiaohuan Cheng, Sourabhi Debnath, Elizabeth Gregan, Hugh Byrne

Articles

Ultrasonication is widely used for preparing Single-Walled Carbon Nanotube (SWNT) dispersions in different solvent media and it has been shown to play a critical role in dispersing and debundling SWNTs. The strong shear force that can exfoliate the SWNT bundles during sonication comes from cavitation, which entails a process of bubble formation, growth, and collapse. The efficiency of the cavitation process is closely correlated to many solvent parameters, including vapor pressure, viscosity, and surface tension, as well as the sonication frequency, intensity, and time. In this study, SWNTs were dispersed in a range of organic solvents assisted by tip sonication. …

A Systematic Study Of The Dispersion Of Swnts In Organic Solvents, Qiaohuan Cheng, Sourabhi Debnath, Luke O'Neill, Theresa Hedderman, Elizabeth Gregan, Hugh Byrne

Articles

Dispersions of as-produced HiPco single-walled carbon nanotubes (SWNTs) in a series of organic solvents were prepared by dilution with the aid of tip sonication. Mild centrifugation (~ 945 g) was carried out to remove large bundles. Atomic force microscopy (AFM) studies revealed that the bundle size decreased as the dispersion was diluted. By measuring the UV-vis-NIR absorption before and after centrifugation as a function of the concentration, the dispersion limit of SWNTs in each solvent can be determined. Correlations between the dispersion limit and solvent solubility parameters, including the Hildebrand solubility parameter and three dimensional Hansen solubility parameters, are explored, …

Pk/Pd Modelling Of Comb-Shaped Pegylated Salmon Calcitonin Conjugates Of Differing Molecular Weights, Sinead Ryan, Jesus Maria Frias, David H. Haddleton, David J. Brayden

Articles

Salmon calcitonin (sCT) was conjugated via cysteine-1 to novel combed-shaped end-functionalised poly(PEG) methyl ether methacrylate) (sCT-P) comb-shaped polymers, to yield conjugates of total molecular weights (MW) inclusive of sCT: 6.5, 9.5, 23 and 40 kDa. The conjugates were characterised by HPLC and their in vitro and in vivo bioactivity was measured by cAMP assay on human T47D cells and following intravenous (i.v.) injection to rats, respectively. Stability against endopeptidases, rat serum and liver homogenates was assessed. There were linear and exponential relationships between conjugate MW with potency and efficacy respectively, however the largest MW conjugate still retained 70% of E …

Purification And Properties Of Amycolatopsis Mediterranei Dsm 43304 Lipase And Its Potential In Flavour Ester Synthesis, Dharmendra Dheeman, Gary Henehan, Jesus Maria Frias

Articles

An extracellular thermostable lipase from Amycolatopsis mediterranei DSM 43304 has been purified to homogeneity using ammonium sulphate precipitation followed by anion exchange chromatography and hydrophobic interaction chromatography. This protocol resulted in 398 fold purification with 36% final recovery. The purified A. mediterranei DSM 43304 lipase (AML) has an apparent molecular mass of 33 kDa. The N-terminal sequence, AANPYERGPDPTTASIEATR, showed highest similarity to a lipase from Streptomyces exfoliatus. The values of and for p-nitrophenyl palmitate (p-NPP) under optimal temperature (60°C) and pH (8.0) conditions were 0.10 ± 0.01 mM and 2.53 ± 0.06 mmol/minmg, respectively. The purified …

Media Literacy And Communication Rights: Ethical Individualism In The New Media Environment, Brian O'Neill Jan 2010

Media Literacy And Communication Rights: Ethical Individualism In The New Media Environment, Brian O'Neill

Articles

The dominant discourse of media literacy policy espouses an ethical individualism within the digital media environment in which the source of moral values and principles, and the basis of ethical evaluation, is the individual. In this perspective, even vulnerable citizens such as children and young people, who tend to be in the vanguard of new media adoption, are required to negotiate the risks and opportunities of the online world with diminishing degrees of institutional support from trusted information sources. Noticeably absent from this discourse is any consideration of the notion of communication rights. Examining an alternative conceptualization of media literacy …

Current Policy Developments In European Media Literacy, Brian O'Neill Jan 2010

Current Policy Developments In European Media Literacy, Brian O'Neill

Articles

In August of 2009, the European Commission issued a formal recommendation on media literacy adding to a series of initiatives underway since 2006 to build a comprehensive European media literacy policy (Commission of the European Communities 2009). Media literacy is now a well-established concept referring to the ability to critically understand and use media in a variety of contexts (see Aufderheide 1997). Recognising that the way people use media is changing and that the volume of information received is enormous, the Commission notes that all citizens need to have the ability to access, analyse and evaluate images, sounds and texts …

Child Sexual Abuse, Links To Later Sexual Exploitation/High-Risk Sexual Behavior, And Prevention/Treatment Programs, Kevin Lalor, Rosaleen Mcelvaney Jan 2010

Child Sexual Abuse, Links To Later Sexual Exploitation/High-Risk Sexual Behavior, And Prevention/Treatment Programs, Kevin Lalor, Rosaleen Mcelvaney

Articles

This paper reviews the literature on the nature and incidence of child sexual abuse, explores the link between child sexual abuse and later sexual exploitation, and reviews the literature on prevention strategies and effective interventions in child sexual abuse services. Our understanding of the international epidemiology of child sexual abuse is considerably greater than it was just 10 years ago, and studies from around the world are examined. Childhood sexual abuse can involve a wide number of psychological sequelae, including low self-esteem, anxiety, and depression. Numerous studies have noted that child sexual abuse victims are vulnerable to later sexual revictimization, …

The Construction Of Locative Situations: Locative Media And The Situationist International, Recuperation Or Redux?, Conor Mcgarrigle Jan 2010

The Construction Of Locative Situations: Locative Media And The Situationist International, Recuperation Or Redux?, Conor Mcgarrigle

Articles

A trend exists within locative media art of invoking the practices of the Situationist International (SI) as an art historical and theoretical background to contemporary practices. It is claimed that locative media seeks to re-enchant urban space though the application of locative technologies to develop novel and experimental methods for navigating, exploring and experiencing the city. To this end, SI concepts such as psychogeography and the techniques of detournement and the de ́rive (drift) have exerted considerable influence on locative media practices, but questions arise as to whether this constitutes a valid contemporary appropriation or a recuperative co-option, serving to …

Threshold Lives: Exploring The Liminal Consumption Of Tweens, Kevina Cody, Katrina Lawlor, Pauline Maclaren Jan 2010

Threshold Lives: Exploring The Liminal Consumption Of Tweens, Kevina Cody, Katrina Lawlor, Pauline Maclaren

Articles

The intervening spaces of socio-cultural organisation have proved sources of fascination and powerful theory development in the fields of sociology, psychology and anthropology (e.g. Douglas, 1966; Van Gennep, 1961; Freud, 1950, Foucault, 1977). Consumer culture research has hitherto acknowledged the potency of studying transitional phenomena, spaces and places and the interactions of varied ontologies with the consumption experiences of the individuals or group in flux and evolution (e.g. Davies and Fitchett, 2004; Maldonado and Tansuhaj, 1999; Gentry, 1997; Schouten, 1991). But what of those whose reality is the threshold between two ordered and defined worlds, the centre of nowhere? This …

The Invisible Hand Of Preacquired Account Marketing, Prentiss Cox Jan 2010

The Invisible Hand Of Preacquired Account Marketing, Prentiss Cox

Articles

Preacquired account marketing is a sales practice that allows companies to charge consumers for services they do not know they ordered and do not use. The practice depends on a seller's ability to access a consumer's financial account without the consumer directly providing her account number and other access information to that seller. This flips the power dynamic in the solicitation process by shifting the burden to the consumer to stop the seller from accessing her account, rather than requiring the seller to ask the consumer for her account information before her account can be charged. This is possible because …

Framing Justice: Media, Bias, And Legal Decisionmaking, Perry L. Moriearty Jan 2010

Framing Justice: Media, Bias, And Legal Decisionmaking, Perry L. Moriearty

Articles

During the 1990s, the news media saturated the American public with stories and images of glassy-eyed, teenaged “superpredators,” who allegedly killed and maimed for sport. These violent, dark and “morally impoverished” youth were running wild in our city streets was the message, and unless we did something, they would destroy the very moral fabric of our society. Drawing on recent social science studies, which demonstrate that the graphic and racialized content of crime news coverage can increase consumers’ cognitive bias in imperceptible, but determinative ways, I argue that exposure to the “superpredator” narrative may have had a discernable impact on …

Regulating Insurance Sales Or Selling Insurance Regulation?: Against Regulatory Competition In Insurance, Daniel Schwarcz Jan 2010

Regulating Insurance Sales Or Selling Insurance Regulation?: Against Regulatory Competition In Insurance, Daniel Schwarcz

Articles

In both corporate and banking law, firms are empowered to select from a limited menu of options the regulatory regimes that will govern them. Two recent proposals would reform the regulation of property, casualty and life insurance markets by empowering insurers to make similar choices among multiple regulators. This Article argues that such regulatory competition is undesirable. Insurers operating in such a regime would tend to choose the least intrusive regulators, irrespective of whether doing so benefited consumers, third-parties, or even the collective interests of insurers themselves. The resulting decrease in regulatory scrutiny would, in fact, harm insurance markets and …

Justification Norms Under Uncertainty: A Preliminary Inquiry, Claire Hill Jan 2010

Justification Norms Under Uncertainty: A Preliminary Inquiry, Claire Hill

Articles

People making decisions under uncertainty may need to justify those decisions to their reputational community. This Essay considers when and how the potential need to justify might lead a decision-maker to employ a methodology better suited to yielding a justifiable choice that may not be the best choice. When a decision involves uncertainty, the possible outcomes and probabilities are not known. A broad consensus about a methodology that produces a good decision often may not exist. But norms will often arise as to acceptable methodologies - that is, methodologies that will be accepted as justifiable if justification is needed. The …

Private Ordering And Family Law, Brian Bix Jan 2010

Private Ordering And Family Law, Brian Bix

Articles

Until recently in American family law (and the family law of most countries), private arrangements to alter the legal rules surrounding family status were rarely enforced. There was, of course, “private ordering” of a basic sort: e.g., one chose whether to marry or not, and whom to marry, but once one married, the legally enforceable rules of marriage, the ability to exit through divorce or annulment, the financial obligations upon divorce, and so on, were all set by the state, which might also limit the power of the parties to distribute their own property upon death, as with a spouse’s …

The Trademark Fair Use Reform Act, William Mcgeveran Jan 2010

The Trademark Fair Use Reform Act, William Mcgeveran

Articles

Sweeping assertions of exclusive trademark rights in brand names and images have a pernicious chilling effect on free expression, including fictional portrayals, commentary, political speech, parody, and comparative advertising. Some disputes lead to lawsuits. More often, speakers capitulate to litigation threats, even when the substance of the legal claims they face is very weak. As demonstrated in the author’s previous work, existing trademark “fair use” doctrines are not simple defenses that end suits quickly and cheaply, and many defendants cannot bear the resulting costs of protracted litigation.Observers have proposed a variety of improvements to trademark fair use, but this Article …

State Standards For Nationwide Products Revisited: Federalism, Green Building Codes, And Appliance Efficiency Standards, Alexandra B. Klass Jan 2010

State Standards For Nationwide Products Revisited: Federalism, Green Building Codes, And Appliance Efficiency Standards, Alexandra B. Klass

Articles

This Article considers the federal preemption of state standards for building appliances and places the issue within the ongoing federalism debate over the role of state standards for “nationwide products” such as automobiles, pharmaceuticals, and other consumer products. Notably, residential, commercial, and industrial buildings make up approximately 40 percent of total U.S. energy demand and the same percentage of U.S. carbon dioxide (CO2) emissions, while the appliances within those buildings are responsible for 70 percent of building energy use, making appliance efficiency a central component of any national effort to reduce energy use and greenhouse gas (GHG) emissions. For decades …

Concepts, Categories, And Compliance In The Regulatory State, Kristin Hickman, Claire Hill Jan 2010

Concepts, Categories, And Compliance In The Regulatory State, Kristin Hickman, Claire Hill

Articles

Law is, of course, always a product of its history. But for some regimes, history matters both more and differently than for others. In some instances, the requirements and scope of a regulatory regime’s coverage are sufficiently attenuated from statutory text and purpose that they can only be explained or understood by reference to history. At its (perhaps caricatured) extreme, such a regime is one in which regulated parties expend significant efforts attempting to comply with the law and often succeed in complying at the most minimal level possible, to the point that compliance is perceived as optional and, to …

Returning Home: Women In Post-Conflict Societies, Naomi Cahn, Dina Francesca Haynes, Fionnuala Ní Aoláin Jan 2010

Returning Home: Women In Post-Conflict Societies, Naomi Cahn, Dina Francesca Haynes, Fionnuala Ní Aoláin

Articles

This paper explores the situation of women returning to their homes and communities after their countries have experienced major conflicts. In that context, it assesses the range of barriers and challenges that women face and offers some thinking to addresses and remedy these complex issues. As countries face the transition process, they can begin to measure the conflict’s impact on the population and the civil infrastructure. Not only have people been displaced from their homes, but, typically, health clinics, schools, roads, businesses, and markets have deteriorated substantially. While the focus is on humanitarian aid in the midst of and during …

Macrophage Responses To Cf Pathogens: Jnkmap Kinase Signaling By Burkholderia Cepacia Complex Lipopolysaccharide, Suzanne Mckeon, Siobhan Mcclean, Máire Callaghan Jan 2010

Macrophage Responses To Cf Pathogens: Jnkmap Kinase Signaling By Burkholderia Cepacia Complex Lipopolysaccharide, Suzanne Mckeon, Siobhan Mcclean, Máire Callaghan

Articles

Chronic bacterial colonization of the airways with opportunistic pathogens is the primary cause of morbidity and mortality in cystic fibrosis (CF) patients. Burkholderia cepacia complex (Bcc) organisms pose a particular challenge in CF lung disease, due in part to their ability to trigger a fulminant pneumonia. This study compares the U937 macrophage response to two Bcc species, B. cenocepacia and Burkholderia multivorans, against Pseudomonas aeruginosa and Staphylococcus aureus. The two Bcc strains demonstrated higher levels of U937 macrophage internalization compared with both P. aeruginosa and S. aureus. Both the Bcc strains also stimulated significantly greater levels of tumor necrosis factor-a …
