Alterations In Synaptic Structure And Tau Phosphorylation In Alheimer's Disease, Courtney K. Walker
Alterations In Synaptic Structure And Tau Phosphorylation In Alheimer's Disease, Courtney K. Walker
All ETDs from UAB
Alzheimer’s disease (AD) is the most common cause of dementia in individuals over the age of 65. Of the two hallmark pathologies of AD, tau neurofibrillary tangles (NFTs) correlate more strongly with cognitive decline than do Aβ plaques. NFTs form in the entorhinal cortex (EC) and appear in anatomically connected regions over time as disease progresses. This tau spreading may occur via synaptic connections. Most excitatory synapses occur on dendritic spines, which house the postsynaptic machinery. Spines are actin-rich, plastic structures that change their size and shape in response to synaptic activity in a process that is dependent on actin …
The Adoption Of New Cancer Drugs In Non-Academic Cancer Centers, Warren M. Smedley
The Adoption Of New Cancer Drugs In Non-Academic Cancer Centers, Warren M. Smedley
All ETDs from UAB
Advancements in medical knowledge are producing significant improvements in medical technology, developing breakthrough drug therapies, advancing the practice of medicine, and improving outcomes for cancer patients. However, there are delays in getting new and improved therapies incorporated into routine clinical use, depriving cancer patients of valuable and potentially lifesaving treatments. The purpose of this study was to examine the rate of adoption of new cancer drug discoveries into routine clinical practice in non-academic cancer centers. The process of adoption takes a period of time to embrace new therapies, due in part to a prescriber’s need to gain greater comfort with …

Renal Lactate Dehydrogenase A: Underlying Mechanisms And Inflammatory Roles In Acute Kidney Injury, Gunars Osis
All ETDs from UAB
Kidneys are physiologically vital organs responsible for blood homeostasis preservation through fluid balance maintenance, blood pressure regulation, hormone production, and toxic compound removal. Acute kidney injury (AKI) is a global condition affecting more than 13 million people yearly, and is characterized by a rapid decline in renal filtration capacity, consequent rise in serum creatinine and nitrogenous waste levels, and is associated with an elevated risk of morbidity in the form of progressive chronic kidney disease, kidney failure, and mortality. AKI-induced renal epithelial cell death forces surviving tubular cells to undergo de-differentiation, rapid proliferation to replace lost cells, and re-differentiation to …
Relationship Between Neighborhood Deprivation And Epigenetic Age Acceleration With Pain In Adults With Musculoskeletal Pain, Pamela Jackson
Relationship Between Neighborhood Deprivation And Epigenetic Age Acceleration With Pain In Adults With Musculoskeletal Pain, Pamela Jackson
All ETDs from UAB
Residents of high-deprivation neighborhoods shoulder a greater burden of age-related health conditions, including experiencing worse chronic musculoskeletal pain outcomes, compared to residents of more affluent neighborhoods. Advancing age is a risk factor for chronic health conditions, which epigenetic modifications may explain. Epigenetic age acceleration occurs when an individual’s epigenetic age is older than their chronological age, and that discordance has been identified as a strong predictor of age-related conditions. Epigenetic age acceleration has also been correlated with neighborhood deprivation. However, the mechanisms for neighborhood disparities in chronic musculoskeletal pain remain unclear. The purpose of this dissertation study was to examine …
Hedgehog Signaling Rewires M2 Macrophage And Regulatory T Cell Metabolism Within The Tumor Microenvironment Of Breast Cancer, Dominique C. Hinshaw
Hedgehog Signaling Rewires M2 Macrophage And Regulatory T Cell Metabolism Within The Tumor Microenvironment Of Breast Cancer, Dominique C. Hinshaw
All ETDs from UAB
Current treatments for breast cancer offer little relief to patients with metastatic disease. A contributory factor to tumor growth and metastasis is aberrant up-regulation of developmental signaling pathways such as the Hedgehog (Hh) signaling pathway. This pathway is aberrantly activated in breast cancer and contributes to tumor progression and metastasis. The success of these events is highly influenced by the surrounding immune microenvironment. Macrophages exist along a spectrum of M1 (tumor-eradicating) to M2 (tumor-promoting), while regulatory T cells (Tregs) are an immunosuppressive cell type. The predominant functions of M2 macrophages and Tregs include dampening the immune response and promoting wound …
Comprehensive Characterization Of Structural Variations Using Long-Read Sequencing Data, Yu Chen
Comprehensive Characterization Of Structural Variations Using Long-Read Sequencing Data, Yu Chen
All ETDs from UAB
Structural variants (SVs) contribute to genomic diversity and play pathogenic roles in a wide range of genetic disorders. Accurate characterization of SVs is critical for genomic research and studies of disease mechanisms. The rapid development of Third-Generation Sequencing (TGS) technologies has largely increased sequencing read length compared to Next-Generation Sequencing (NGS), bringing both great potentials and challenges in SV discovery through alignment-based and assembly-based approaches. In order to take full advantage of TGS data, I have developed a suite of bioinformatics tools focusing on comprehensive characterization of SVs. For the alignment-based SV discovery, I have developed DeBreak to identify SVs …

First-Principles Modeling Of Mechanical Behavior And Thermoelectric Property Under Strain And Pressure, Chia-Min Lin
All ETDs from UAB
Progresses in computer hardware and numerical algorithms in the past decade have made it possible to use first-principles calculations to model material properties, with the prediction results typically in good agreement with experiments. In this dissertation, density functional theory (DFT) calculations enable the studies of shear and tensile deformations. The theoretical ideal shear stress for transition metal osmium is found along the (001)[1–10] shear direction with the maximal shear stress ∼24 GPa at critical strain ∼0.13. DFT results indicated that transition metal borides ReB2 becomes more ductile with enhanced tendency towards metallic bonding under compression. The ReB2 DFT results also …
A Value-Based Framework For Process Alignment In Complex Provisioning Systems, Michael Lipscomb
A Value-Based Framework For Process Alignment In Complex Provisioning Systems, Michael Lipscomb
All ETDs from UAB
Provisioning systems are human-built, process-based systems whose activities are directed toward providing something of value to another entity. The things provided are not limited to any one type but may include, for example, objects, services, and information. Systems under consideration herein are limited to those that are human based, decentralized, and complex. “Human based” means that many of the agents and all the customers of the system are human. “Decentralized” means that agents act beyond central control. Such a system presents different challenges from one in which tasks are delegated from a central authority. In a decentralized system, agents are …

Pkd-1 Signaling Inthe Regulation Of Cancer Stem Cell-Like Features And Cancer Cell Interactions With Endothelial Cells In Breast Cancer And Pancreatic Neuroendocrine Tumor, Yichen Hailey Guo
All ETDs from UAB
Cancer stem cells (CSCs) are a subpopulation cancer cells that play critical roles in tumor progression, recurrence, metastasis, and drug resistance. CSCs are characterized by self-renewal and plasticity. Protein kinase D-1 (PKD-1) signaling regulates CSC/stemness features in both breast cancers and pancreatic neuroendocrine tumors. In PNETs, PKD-1 signaling regulates not only stemness properties of cancer, but also epithelial-mesenchymal transition (EMT). PNETs are typically well vascularized and heterogeneous. Everolimus (an mTOR inhibitor and the 2nd generation derivative of rapamycin) has been approved as a targeted chemotherapy for PNET patients. Unfortunately, nearly all patients will progress on treatment due to drug resistance …
The Role Of Protein Translation And Degradation In Aging And Alzheimer Disease Pathogenesis, Harper S. Kim
The Role Of Protein Translation And Degradation In Aging And Alzheimer Disease Pathogenesis, Harper S. Kim
All ETDs from UAB
Protein translation (PT) is an essential cellular process playing crucial roles in growth and development. PT precipitously declines with age in multiple animal species, including humans. It has been implicitly assumed that elevated PT at young ages is beneficial to health while PT ends up dropping as a passive byproduct of aging. However, whether this holds true and how dynamic fluctuations in PT over time impact aging remain unknown. In Drosophila, we show that a transient PT spike in early-adulthood drives aging by triggering dysfunction in protein homeostasis (proteostasis) at old ages. We propose the early-adulthood spike in PT to …

Determining The Impact Of Immune Dysregulation On Cd4 T Cell Responses To Hiv-1 And Sars-Cov-2, Jacob K. Files
All ETDs from UAB
CD4+ T cells, often referred to as helper T cells, play a crucial role in the formation of antiviral immune responses following infection and vaccination. However, this can be disrupted by various forms of immune dysregulation. Many groups have described how HIV-1 adaptation and immune escape can lead to dysregulated HIV-specific immune responses. Our lab has previously shown that predicted HLA-II associated HIV-1 adaptation leads to dysregulated, poorly immunogenic CD4+ T cell responses in HIV-1 infection. Here, we focus on the impact of this HLA-II associated viral adaptation in the setting of HIV-1 vaccination. We show that HLA-II associated, vaccine-matched …

Although Black students are overrepresented in the special education (SPED) population within the educational system in the United States, the number of Black students identified with dyslexia can be concluded to be significantly lower (Annamma et al., 2018; Farkas et al., 2020; Sullivan & Bal, 2013). With experts positing that dyslexia affects as many as one in five persons, attention to this matter is needed especially regarding the youngest Black students (Hyles & Hoyles, 2010; Moats & Dakin, 2017). The characteristics of dyslexia are manifested in neurological processes surrounding reading abilities, thought processes, and motor skills (Moats & Dakin, 2017). …

All ETDs from UAB
Despite major advances in revascularization and drug therapy for post-myocardial infarction (MI) heart failure (HF), heart transplantation remains the only curative therapy to prevent progressive and debilitating disease. Researchers have long sought a replenishable and expandable source of cardiomyocytes (CMs), the primary functional cell in the heart muscle, for a broad array of applications related to the treatment of HF ranging from direct implantation to high throughput drug testing. The lack of cell cycle activity and cell division in adult CMs prevented the development of an expandable primary cell line. However, with the discovery of high efficiency stem cell differentiation …

Temperature-Dependent Sex Determination (Tsd): Implications For The Biology And Conservation Of The Kemp's Ridley Sea Turtle (Lepidochelys Kempii), Katherine Presz
All ETDs from UAB
The Kemp’s ridley sea turtle (Lepidochelys kempii) has temperature-dependent sex determination (TSD) where the incubation temperature of the egg determines the sex of the hatchling. This has significant implications for the ecology, evolution, and conservation of this species because it can affect sex ratios. The studies presented in this thesis address hatchling sex ratios produced in Kemp’s ridley conservation programs using both natural as well as manipulated incubation regimes. First, a histology-based study was utilized to investigate sex ratios produced at the Padre Island National Seashore from 2002-2019. The results indicate that a female bias has been produced each year …

Socioeconomic Status, Hbb, And Ttr Variants, And Cardiorenal Outcomes: The Regards Study, Onika E. Abrams
All ETDs from UAB
Blacks are disproportionately affected by heart failure (HF) and end-stage renal disease (ESRD). While poorer health outcomes in Blacks have been associated with social determinants of health, there are some biological differences worth noting, namely genetic risk variants for both TTR and HBB. These genetic risk variants are known to confer additional risk in the development of heart failure and end-stage renal disease, respectively. While we understand these genetic variants aren’t fully penetrant, and genetics and are not deterministic, we aimed to examine additional factors and mechanisms responsible for the increased risk in HF and ESRD in Blacks. Neighborhood socioeconomic …

Association Between Regional Covid-19 Vaccine Hesitancy And Reported Positive Covid-19 Cases In The Southern United States, Orielle Caudle
All ETDs from UAB
The use of vaccinations has been shown to be beneficial in pandemics in the past, and in some cases, it has led to elimination of a particular disease (i.e. polio). The COVID-19 vaccination can also serve the same purpose, however, vaccine hesitancy within specific regions of the country may prevent and/or dilute the effectiveness of the vaccine. Vaccine hesitancy can be defined as a delay in acceptance, or refusal despite the availability of vaccine services. Southern regions have consistently had the lower vaccination rates compared to its other regional counterparts, even as the pandemic progressed and death rates increased over …

"To Create A New Image Of Women": The Fight For The Equal Rights Amendment In Birmingham, Alabama, 1972-1982, Christopher J. Bertolini
All ETDs from UAB
This thesis argues that the struggle to ratify the Equal Rights Amendment (ERA) in Birmingham, Alabama, rarely focused on the amendment’s legal merits, but rather became a debate about the nature and validity of gender roles. Generally, opponents of the ERA set the terms of the debate and connected the seemingly benign concept of legal equality between men and women with controversial issues like the conscription of women into the military, women’s freedom from the workplace and supposed right to the economic support of their husbands, religious interpretations of gender roles, and abortion. The struggle was primarily one between middle-class …
The Roles Of Racial Discrimination And Sleep In Cognitive Functioning In Diverse Youth, Queenisha Crichlow
The Roles Of Racial Discrimination And Sleep In Cognitive Functioning In Diverse Youth, Queenisha Crichlow
All ETDs from UAB
Cognitive functioning is an important aspect of human development that is closely related to youth academic outcomes (Caemmerer et al., 2018). Specifically, there is evidence that cognitive abilities such as memory, attention, and self-control are predictive of academic achievement in youth (Peng & Kievit, 2020). Current research suggests that chronic stress reduces cognitive functioning (Korten et al., 2014), but few studies have examined this relationship for race-related stress, specifically racial discrimination. Considering that racial discrimination is a common and reoccurring type of stress in the lives of minority youth (Hughes et al., 2016), it is possible that chronic exposure to …

Smoking And Perceived Stress: Examining The Impact Of Racial Identity And Risk For Obstructive Sleep Apnea Through A Moderated Mediation Model, Sofia Mildrum Chana
All ETDs from UAB
Background: Cigarette smoking is known to have a negative effect on individuals’ sleep quality, and symptoms of insomnia. Smoking can particularly exacerbate sleep disorders such as obstructive sleep apnea (OSA) by increasing irritation and inflammation of the upper respiratory conducts. Previous research highlights the complex positive association between cigarette smoking and perceived stress. Sleep may be an important aspect intervening in the association between smoking and perceived stress, given that individuals who report high stress also report worse sleep quality and are at greater risk for OSA. However, limited research has examined the indirect association of cigarette smoking and perceived …

Cancer is one of the leading causes of death in the United States. Based on the American Cancer Society statistics, in 2021 1,806,590 new cancer cases and 605,520 cancer deaths are projected to occur in the United States. Amongst all, breast cancer (BC) is one of the most prevalent cancers that occurs in women. Various lifestyle factors such as alcohol intake, smoking, an unhealthy diet and physical inactivity may result in increased rates of BC risks (cancer-transition) by inducing mutations in DNA. Numerous epidemiological studies indicate that a diet rich in green tea polyphenols (GTPs), and/or cruciferous vegetables such as …

Characterization Of The Phenotype And Functional Properties Of Myeloid Subpopulations In Chronic Inflammatory Conditions, Krystle Leandra Ong
All ETDs from UAB
Myeloid cells represent a subset of leukocytes traditionally recognized as first responders to acute inflammatory stimuli. In recent years, there has been a growing appreciation of the role of myeloid cells as critical regulators of the immune system in disease pathogenesis and progression. Chronic inflammatory diseases including viral infections, autoimmune diseases, and malignancies are frequently associated with altered myelopoiesis characterized by a profound shift in the myeloid cell phenotype and function. The work presented in this dissertation provides an insight into dysregulated myelopoiesis and myeloid heterogeneity in multiple myeloma (MM), human immunodeficiency virus-1 (HIV-1), and coronavirus disease-19 (COVID-19). We identify …

The Role Of Nitric Oxide And Cyclic Guanosine Monophosphate Signaling In Arteriovenous Fistula Maturation, Maheshika S. Somarathna
All ETDs from UAB
Vascular access is the single most important component of the hemodialysis procedure and the arteriovenous fistula (AVF) is the most preferred type. However, 60% of AVFs created fail to mature successfully for dialysis use (AVF maturation failure). AVF maturation failure is primarily the result of 1) intimal hyperplasia formation (IH) and 2) poor vascular outward remodeling. Currently, a major unmet clinical need in the field is the lack of effective therapies to treat/prevent AVF maturation failure due to the poor understanding of molecular mechanisms. This proposal is designed to: 1) elucidate the pathologic significance of nitric oxide (NO) and cyclic …

By 2040, the U.S. is projected to have 26.1 million cancer survivors; 73% of these will be over 65 years old. Much is known about life-space mobility (LSM) in patients with non-cancer serious illnesses. Restricted LSM affects every aspect of an older adult’s life; their independence, autonomy, and quality of life (QOL). To date, there are no studies examining LSM in older cancer survivors. Therefore, we examined LSM, factors that affect LSM, and the relationship between LSM and QOL in older cancer survivors using existing data from the University of Alabama at Birmingham (UAB) Study of Aging (SOA) (NIA AG15062). …

The Roles Of Repressive And Activating Epigenetic Factors In Vertebrates Neural And Neural Crest Development, Saeid Mohammad Parast
All ETDs from UAB
The vertebrate nervous system comes from specific regions of the ectoderm that comprises of the neural plate and the neural crest. Although genetic mechanisms governing vertebrate neural development have been investigated in depth, there is a knowledge gap regarding the roles of epigenetic mechanisms in this process. As epigenetic modulators, the COMPASS (also known as SET1/MLL complex) and HP1 proteins are responsible for regulating chromatin accessibility to transcription factors. COMPASS is responsible for deposition of activating histone H3K4 methylation marks at promoters and enhancers and thus creates open chromatin domains. The critical structural and regulatory subunits of COMPASS, Dpy30 and …

The accelerated growth in sequencing technology assisted us to explore the bio-logical systems comprehensively through a top-down approach rather than the conven-tional bottom-up approach. The top-down approach is a part of systems biology that gen-erates massive data that has driven this course of investigation to decipher the system-wide genetic perturbations. To reduce the complexity of humongous datasets, the graph (network)-based techniques facilitate the easy handling, analysis, and interpretation of biological associations. Networks are the graphical representation of nodes (biomole-cules) and edges (associations/interactions). The most common biological networks are correlation-based gene co-expression network (GCN), gene regulatory network (GRN), and protein-protein interaction …
