Molecular Imaging To Evaluate Tumor Heterogeneity And Guide Treatment Of Breast Cancer, Yun Lu
All ETDs from UAB
Molecular imaging plays an important role in predicting and evaluating treatment response and revealing the underlying biological mechanisms in the cancer field. Especially, PET imaging serves as a valuable tool for comprehending the tumor microenvironment (TME), allowing us to track tumor metabolism, proliferation, receptor expression, and immune infiltration. In this comprehensive study, three distinct aspects of cancer research were explored using molecular imaging techniques. The first study aimed to evaluate the accuracy of the fluorescence ubiquitination cell cycle indicator (FUCCI) system with fluorescence in vivo imaging compared to 3'-Deoxy-3'-[18F]fluorothymidine ([18F]-FLT) positron emission tomography (PET)/computed tomography (CT) in assessing cancer cell proliferation. …
Lateral Ridge Augmentation Using Allograft Bone Particles Hydrated With Or Without Recombinant Human Platelet-Derived Growth Factor: A Clinical And Histological Randomized Controlled Trial, Lauren Miele
All ETDs from UAB
Lateral ridge augmentation is a common procedure to increase the width of bone in preparation for a dental implant. A certain thickness of bone is necessary for implant health, making ridge augmentation procedures necessary for many patients. Research into the use of growth factors in ridge augmentation is ongoing, but few studies have examined the use of recombinant human platelet-derived growth factor homodimer (rhPDGF-BB) in ridge augmentation. This study examines histologic, clinical, and patient reported outcomes when horizontal ridge augmentation is completed using freeze dried bone allograft hydrated in rhPDGF-BB or saline and a resorbable crosslinked collagen membrane. Histologic results …

Exercise-Driven Epigenetic And Transcriptomic Changes In An Experimental Model Of Temporal Lobe Epilepsy, Silvienne C. Sint Jago
All ETDs from UAB
Temporal lobe epilepsy (TLE) is a neurological disease marked by recurring unprovoked seizures originating from temporal lobe structures and is frequently associated with cognitive deficits. Exercise holds promise as a therapeutic approach to alleviate seizures and cognitive impairments in TLE. However, the exact cellular and molecular mechanisms by which exercise influences pathways related to seizure activity and cognition in the context of TLE still need to be understood. Neuronal network formation relies on the regulation of gene expression, which can lead to either an increase or a decrease in the production of essential proteins involved in neuronal signaling. Genes are …
Pathophysiological Metabolic Alteration In Lung Injury And Fibrosis, Samuel Robert Smith
Pathophysiological Metabolic Alteration In Lung Injury And Fibrosis, Samuel Robert Smith
All ETDs from UAB
Lung injury and lung repair are two sides of the same coin, for where injury occurs, repair must follow. Lung fibrosis occurs when this repair process is dysfunctional and becomes trapped in a chronically incomplete state. The studies described here examine each side of this coin to ask the question of whether metabolism has a role in the pathophysiology of acute lung damage following severe trauma or in the chronic progression of idiopathic pulmonary fibrosis (IPF). In the first half of this work, we investigated the impact that severe lung injury and infection exert on the flux of metabolites in …
Neuropeptide Y Overexpression In The Presence Of P23h Rhodopsin Mutation, Jean Sun
Neuropeptide Y Overexpression In The Presence Of P23h Rhodopsin Mutation, Jean Sun
All ETDs from UAB
Neuropeptide Y (NPY), a 36-amino acid peptide, has been found to be the most abundant neuropeptide in the central nervous system, and it is widespread across mammalian species. NPY plays a dual role as a neurotransmitter and neuromodulator in the nervous system. NPY functions as a multifaceted neuromodulator through binding NPY receptors (NPYRs). NYP interacts with NPYRs and regulates various activities, such as cell growth, neurogenesis, neuroprotection, mitochondrial behaviors, food intake, anxiety, and addiction formation. In terms of NPY’s neuroprotective potential, NPY inhibits neuronal death signaling, thus preventing apoptosis, and displays anti-inflammatory and anti-oxidative effects. Additionally, it influences metabolism by …

Impact Of Alcohol Intoxication On Pedestrian Risk-Taking Behaviors, Yixin Wang
All ETDs from UAB
In the United States, motor vehicle crashes account for approximately 21.8% of unintentionaldeaths, and 17.2% of these crashes involved the death of a pedestrian in 2020. Two preventable factors that impact pedestrian fatalities are alcohol intoxication and smartphone distraction. Approximately 30% of fatal pedestrian crashes involve a pedestrian with a BAC of at least 0.08, and distraction is a commonly observed risk factor that was shown to impair crossing performance in virtual environments. The present research considers both distraction and intoxication as risk factors for pedestrian safety, examining both risk factors in isolation as well as their interactional effects on …

BACKGROUND: Failure to achieve pregnancy (infertility) affects over 6 million women ages 15–44 in the United States. African American women have more difficulty getting pregnant than White women but are less likely to seek help for infertility. According to the current research, there are several factors (cost, insurance coverage, etc.) that create difficulty for women to seek help to get pregnant. However, little is known about the factors that influence or impede infertility help-seeking among African American women. The purpose of this study was to explore the barriers and facilitators to infertility help-seeking among African American women.METHODS: Factors affecting infertility …
Select Lipid Signaling. Derived From T-Cells Is An Important Contributor To Type 1 Diabetes Development, Tayleur White
Select Lipid Signaling. Derived From T-Cells Is An Important Contributor To Type 1 Diabetes Development, Tayleur White
All ETDs from UAB
We reported that Ca2+-independent phospholipase A2β (iPLA2) contributes to type 1 diabetes (T1D) development; however, the impact of lipid signaling from T-cells is unknown. To address this, we first administered splenocytes from NOD, NOD.iPLA2+/- (NOD.HET), or NOD.iPLA2-/- (NOD.KO) mice to 4-week-old NOD.scid recipients. As expected, T1D onset was rapid in NOD cell recipients and 100% by 12 weeks. However, onset was delayed by 1-3 weeks in NOD.HET or NOD.KO cell recipients and only 60% became diabetic, suggesting a role for T-cell iPLA2. Next, to establish importance of iPLA2 in CD4 or CD8 cells, purified cells were administered to NOD.scid mice. …

Obesity is a prevalent chronic condition determined by social, psychological, behavioral, and biological factors. The prevalence of obesity remains a major public health issue and has continued to rise in the United States and around the world. People who are obese face a higher risk of cardiovascular and metabolic diseases. The fat mass and obesityassociated gene (FTO) is one of the most commonly identified genes in obesity-related research. This three-paper dissertation is aimed at improving understanding of sex and race differences in obesity risk factors in older Americans from genetic (FTO SNPs) and lifestyle (diet and physical activity) perspectives, using …
Targeting The Frataxin Transcriptional Defect As A Therapeutic Approach For Friedreich's Ataxia, Anna Maria Schreiber
Targeting The Frataxin Transcriptional Defect As A Therapeutic Approach For Friedreich's Ataxia, Anna Maria Schreiber
All ETDs from UAB
Friedreich’s ataxia (FRDA) is the most common inherited ataxia worldwide. This autosomal recessive neurodegenerative disorder manifests in the central and peripheral nervous system but also affects the cardiovascular and endocrine systems. No effective treatment is available. FRDA is caused by expansion of GAA repeat tracts in intron 1 of the frataxin (FXN) gene on both alleles. An inverse correlation between FXN levels and GAA repeat length exists. Frataxin is a mitochondrial protein participating in iron-sulfur (Fe-S) cluster formation. Frataxin deficiency results in reduced activity of Fe-S-containing enzymes involved in critical biological processes, such as cellular respiration and DNA repair. Expansion …

The Role Of The Bone Marrow Microenvironment In Flt3-Itd Acute Myeloid Leukemia Stem Cell Resistance To Flt3 Inhibition, Nicholas R. Anderson
All ETDs from UAB
Acute myeloid leukemia (AML) is an aggressive, complex hematological malignancy. AML cases with constitutively-activating internal tandem duplications in the Flt3 receptor (Flt3-ITD) represent a distinct subtype of disease with poor prognosis, but unique opportunities for targeted therapy. Flt3-specific tyrosine kinase inhibitors (TKI) have entered clinical practice due to their ability to induce remissions in a large proportion of Flt3-mutant AML patients, but their long-term efficacy remains limited due to the persistence of leukemia stem cells (LSC) in the bone marrow (BM). Herein, we outline the importance of the bone marrow microenvironment (BMM) in harboring LSC through AML development and drug …

A Hybrid Data And Hypothesis-Driven Model For Software Development In Support Of The Machine-Learning Paradigm, Anthony D. Bowman
All ETDs from UAB
istorically, research has often been conducted in a hypothesis-driven manner with software development methodologies created to support those efforts. However, in recent years data-driven approaches to research have seen a dramatic rise in prominence. While software development methodologies such as agile development, extreme programming, and the waterfall model have allowed developers to tackle increasingly complex problems, they were not designed to efficiently support data-driven approaches such as the machine learning paradigm. To address the need to support the different programmatic requirements of both classical, hypothesis-driven as well as data-driven development, novel development strategies are warranted. In this research, we adapted …

Understanding The Protumorigenic Functions Of St6gal1 In Glioblastoma Stemness And Metabolism, Sajina Gc
Understanding The Protumorigenic Functions Of St6gal1 In Glioblastoma Stemness And Metabolism, Sajina Gc
All ETDs from UAB
Glioblastoma (GBM) is a rare but deadly cancer with median survival of just 15 months despite of aggressive treatment. Advancement in novel treatment modalities is hindered by its heterogeneous nature, which includes subsets of neural stem cell-like brain tumor initiating cells (BTICs) that are highly tumorigenic and therapy-resistant. bgalactoside a-2,6-sialyltransferase 1 (ST6Gal1) is elevated in most tumors including in normal and neoplastic stem cells. ST6Gal1 imparts oncogenic phenotypes such as invasion, apoptosis evasion, therapy resistance, TIC maintenance among others via sialylation of critical receptors like, Fas, TNFR1, EGFR and more. Yet ST6Gal1 led regulation of BTIC specific cell surface proteins …

The Evolutionary Ecology Of The Antarctic Red Macroalga Plocamium Sp. In Relation To Its Defensive Secondary Metabolite Diversity, Sabrina Heiser
All ETDs from UAB
Organisms produce chemical defenses to gain a competitive advantage, and to protect themselves from pathogens and predators. There is high inter- and, sometimes, intraspecific diversity in the types and quantities of defensive secondary metabolites. Yet, we do not know much about the mechanisms driving the diversity and its distribution. Macroalgae are important members of ecosystems since they provide living space and are primary producers. To prevent predation, macroalgae make structural or chemical defenses. Chemical defenses in the Antarctic red macroalga Plocamium sp. have extensively been studied. It is widespread along the western Antarctic Peninsula and abundant at sites where it …

Characterization Of Exercise-Associated Skeletal Muscle Secreted Factors Through Aging, Dalton Chase Patterson
All ETDs from UAB
Exercise is a powerful behavioral intervention against Central Nervous System (CNS) aging1–3 and plays an essential role in maintaining healthy neurocognitive function and immune metabolism in the aging brain3. We have developed a new transgenic mouse model that moderately over-expresses transcription factor master regulator of proteostasis, Transcription Factor E-B (TFEB), in skeletal muscle (cTFEB;HSACre mice). Despite living sedentary lifestyles, our model of enhanced skeletal muscle proteostasis shows significantly ameliorated proteotoxicity, increased BDNF levels and neurogenesis while improving neurocognitive function and decreasing neuroinflammation in the aging CNS4,5. These neuroprotective effects are markedly reminiscent of those observed in the aging CNS post-exercise6,7, …

Labor Union Membership Tenure And Midlife Health: A Gendered Perspective, Clifford Ross
All ETDs from UAB
The benefits of labor unions have not gone unstudied. Individuals in labor unions have better access to lower cost/more substantial health insurance plans, higher quality pension plans, and better wages leading to increased lifetime earnings. Even though many of these benefits create important pathways that could lead to better health, unions have been paid little attention in health literature. Additionally, in the modern workplace Mothers are offered lower starting salaries, are perceived as less competent, and face a penalty regarding hiring, promotion, and workplace educational opportunities. Compared to men, women with children see an income gap 20 cents wider than …
The Role Ofbiomechanics In Vascular Endothelial Growth Factor Receptor-2 Signaling, Bronte Miller
The Role Ofbiomechanics In Vascular Endothelial Growth Factor Receptor-2 Signaling, Bronte Miller
All ETDs from UAB
Angiogenesis, or the growth of new vasculature from existing blood vessels, is widely considered a primary hallmark of cancer progression. When a tumor is small, diffusion is sufficient to receive essential nutrients; however, as the tumor grows a vascular supply is needed to deliver oxygen and nutrients into the increasing mass. Several anti-angiogenic cancer therapies target VEGF and its receptor VEGFR-2, which are major promoters of blood vessel development. Unfortunately, many of these cancer treatments fail to completely stop angiogenesis in the tumor microenvironment (TME). Since these therapies focus on the biochemical activation of VEGFR-2 via VEGF ligand binding, we …

Low-Dose Dextromethorphan For The Treatment Of Pain In Fibromyalgia: A Single-Blind Placebo-Controlled Crossover Trial, Stephanie Christina Mueller
All ETDs from UAB
Fibromyalgia (FM) is a debilitating chronic pain condition. Its pathophysiology is largely unknown, which has hindered the development of effective treatments. Central sensitization and neuroinflammation have been forwarded as models of FM pathophysiology, both of which indicate the antitussive drug dextromethorphan (DXM) as a potentially effective treatment. DXM is an N-Methyl-D-aspartic acid (NMDA)-receptor antagonist and microglial modulator with anti-neuroinflammatory properties at low doses. It is currently available for clinical use, but has not been tested as a treatment for FM at low dosages. The current study evaluated the effectiveness of DXM in treating FM-associated symptoms of pain, fatigue, cognitive problems, …

The Association Between Metabolically Healthy Obesity And Coronary Heart Disease Among Regards Study Participants, Kiara N. Aaron
All ETDs from UAB
A major obstacle to the advancement of our understanding of the metabolically healthy obesity (MHO) phenotype is the inconsistent definition of metabolic health and obesity among studies. A harmonized definition of MHO in adults has been proposed based on the diagnosis of obesity (BMI > 30 k/m2) and meeting the cardio-metabolic criteria for triglycerides, high-density lipoprotein (HDL) cholesterol, systolic blood pressure (SBP), diastolic blood pressure (DBP), no antihypertensive treatment, fasting blood glucose, and no drug treatment with glucose-lowering agents. In addition, waist circumference can be associated with increased disease risk when WC is >94 cm in men and >80 cm in …

Klf2 Regulation Of Glycocalyx Expression, Barrier Function, And Thromboinflammation In Human Lung Microvascular Endothelial Cells, Cambria O'Grady
All ETDs from UAB
KLF2 is an important transcription factor for maintaining vascular homeostasis. Its expression in endothelial cells is triggered by shear stress. The endothelial glycocalyx also plays a role in regulating vascular function, and prior work has shown that glycocalyx expression is affected by shear stress as well. However, the direct link between KLF2 and mechanisms that regulate glycocalyx synthesis has not been established. In the present study, we used human lung microvascular endothelial cells (HLMVECs) to observe changes in glycocalyx expression, barrier function, and thromboinflammation caused by KLF2 overexpression. Lentiviral transduction was the method used for KLF2 overexpression, resulting in a …
