Tall fescue (Lolium arundinaceum), an agronomically important forage grass, is typically associated with a mutualistic asexual fungus Neotyphodium coenophialum. Endophytic fungal growth is confined to the intercellular spaces of the plant and induces no symptoms. The endophyte partner enhances its host's fitness by improving nutrient acquisition as well as by providing protection from various abiotic and biotic stresses. However, grass-endophyte partnerships are detrimental to grazing livestock, due to the production of an array of fungal alkaloids. To investigate this interaction at the molecular level, suppressive subtractive hybridization was used to identify differentially expressed genes from the symbiosis. In total, twenty-nine …

Older adults are at a higher risk for numerous oral and systemic diseases. Oral conditions experienced by the elderly are believed to be cumulative. The purpose of this study was to determine if a relationship exists between early childhood dental care practices/experiences and perceived oral health needs of persons 60 years of age and older who are participants of three West Virginia facilities for the elderly (N = 90). Individuals were interviewed utilizing a two-part 21-item survey. Data analyses, including analysis of variance and tests of chi square, were conducted using the JMP program. The primary purpose of this study was to examine the career opportunities for graduates completing a Bachelor of Science degree in Animal and Veterinary Sciences at West Virginia University as measured by the first and current careers of alumni. The target population consisted of all alumni that graduated between the years 1991 and 2001. Over 61.6% of the West Virginia University Animal and Veterinary Science graduates were satisfied with both their first career selection as well as their current career. Uric acid is an important antioxidant due to its role as a free radical scavenger, and methods to elevate its plasma concentration may be important in animal health. In a preliminary study, the effect of dietary protein on plasma uric acid (PUA) was determined in 3-wk-old chicks. Twenty-four broiler chicks were randomly assigned to 4 diets: low protein (LP) containing 10% casein, medium protein (MP) containing 20% casein, high protein (HP) containing 45% casein and a commercial diet (COD) for 2-wks. Soil series occurring above 1020 m in elevation in the Monongahela National Forest in West Virginia were sampled and analyzed for soil organic and soil microbial biomass carbon as well as various fertility parameters. Additionally, vegetative biomass estimates were made at the sampling sites. The soil series sampled (established and tentative) are frigid, well and moderately-well drained inceptisols and spodosols, covered by northern hardwood or spuce-dominated forests. A longtitudinal study was performed in prepubertal heifer calves to determine when the individual components of the hypothalamic-pituitary-ovarian axis were capable of functioning. In study 1, the hypothalamus and anterior pituitary of prepubertal dairy heifers were challenged with exogenous hormones (estradiol and GnRH, respectively) to determine at what age gonadotropin surges could be induced. Two-, four-, six-, and eight-month-old heifer calves were challenged with GnRH or estradiol. Blood was sampled from the jugular vein every 15 minutes for 6 hours or hourly for 30 hours, for the two treatments, respectively. Heifers in each age group released LH in response to …

The study was conducted to investigate and model phosphorus sorption in soils and to study the effectiveness of acid mine drainage (AMD) floc in controlling phosphorus (P) loss to surface runoff from soils. Phosphorus sorption capacity of surface and subsurface horizons of four benchmark soil series - Berks, Huntington, Lindside, and Monongahela - of West Virginia was measured. The soils of these four series were loaded with increasing amounts of P up to 60% of their P retention capacity. Myostatin, belonging to the TGF-beta family, negatively regulates skeletal muscle growth. The current study was conducted to assess the efficacy of in ovo administration of exogenous myostatin antagonist (MA) to enhance skeletal muscle growth and improve feed efficiency of broilers.;Growth hormone secretion is under the control of a pair of hypothalamic factors, growth hormone releasing hormone and somatostatin. The growth hormone secretagogue receptor (GHSR) and its endogenous ligand represent a novel third mechanism regulating the release of growth hormone. The heat shock proteins (hsps) are believed to provide protection against a variety of insults such as elevated temperature, heavy metals and chemical agents. The goal of this study was to determine if the immortalized HK-2 human proximal tubule cell line could provide a model system to study the stress response of the proximal tubule cell. Heat stress, elevated temperature at 42.5°C for 1 hr, caused a marked increase only in hsp 70 mRNA and protein, but not that of hsp 27 or hsp 60 mRNA and protein. Bacterial infections associated with mastitis reduce pregnancies in cattle. Effects on pregnancy and incidence of mastitis in sheep were investigated after immunization with peptidoglycan-polysaccharide (PG-PS) and killed cells from Strep. pyogenes. Rambouillet (n = 100) and mixed breed (n = 18) ewe lambs were immunized (∼d42 and 22 before breeding) with PG-PS (30mugPG/kg/bw) or killed cells or were not immunized (Control, n = 117). IgG antibodies were detected by ELISA. Ewes were bred at synchronized estrus. All immunized and half of non-immunized ewes were challenged with PG-PS (60mugPG/kg/bw) d5 after breeding. Two first to second order streams (Kent Run and Munnell Hollow) at Ryerson Station State Park, Greene County, Pennsylvania, were compared as to water parameters (dissolved oxygen, hardness, temperature, pH, and current velocity), species diversity, and abundance of both immature and adult insects. Samples were taken biweekly for one year from May, 1999 through April, 2000 for a total of 26 sampling periods. Data were analyzed using a repeated measures ANOVA. Kent Run had lower dissolved oxygen, higher pH and hardness, lower temperatures, and higher current velocity. A total of 137 taxa were collected at both streams. Of the total …

To evaluate the productivity of a rotational versus continuous grazing system for beef production within a buffer grazing system, thirty crossbred beef cows and their calves were randomly assigned to one of two grazing systems replicated twice over three years of the experiment. Each replication of the rotational system was stocked with 8 cow/calf pairs while the continuous system was stocked with 7 cow/calf pairs. The grazing season was divided into 3 periods to aid in analysis. Period 1 was further divided into grazing cycles consisting of 4 weeks each. Pasture growth during cycle I was found to be higher …

While there is evidence in the literature of herbal supplement education in other disciplines, inclusion in dental hygiene curricula is not reported. The purpose of this study was to assess the prevalence and extent of herbal supplement education in the 256 accredited dental hygiene programs in the United States. A survey was sent to program directors. Data analyses, including frequencies, cumulative frequencies, percentages, analyses of variance, and test of Chi Square were conducted using JMP program. Surveys revealed 63% of respondents (n = 160) included herbal supplement education and primarily incorporated it in the pharmacology course (67%). Statistically significant differences …

This analysis examines the sensitivity of state intergovernmental aid, per capita, to measures of resources, infrastructure, social services, social problems and demographics in Northern and Southern local governments in 1982 and 1992. Time-periods chosen are based on the potential for domestic policies born through new federalism to harm local governments through lesser amounts of available state aid. North and South are compared to account for possible regional differences. Regardless of region and time however, redistribution efforts to local governments with higher levels of need and lower levels of resources are not particularly good. Comparison Of Broiler Breeder Production And Fertility In A Colony Cage System With Two Different Floors Versus A Slat-Floor System, Joseph Wayne Massey

Two separate trials were conducted to compare egg production, egg weight, shell thickness, albumin height, fertility, and mortality of naturally mated broiler breeders maintained on either a slat and litter floor or in two tiered colony cages. In the first trial there was no difference (P > 0.05) in egg production between the two treatments. There was also no significant difference in hen mortality. Higher (P < 0.05) egg shell quality, albumin height, and shell thickness were observed in the cage maintained hens. Lower fertility (P < 0.05) was observed in the cage maintained hens. In the second trial production parameters were compared in naturally mated broiler breeders maintained on slat and litter floor, in colony cages on the original slats, and in colony cages on a modified slats. The modified slats were constructed from plastic and had a two piece design which produced a slight cushioning on the top of the slats. Egg production and mortality were not different (P > 0.05) among the three treatments. Eggshell thickness, albumin height, and shell thickness were lower (P > 0.05) in the litter and slat floor but not between the two cage treatments (P > 0.05). Fertility was higher (P …

To evaluate the influence of mass of rumen contents on voluntary intake and rumen function, five ruminally cannulated steers (500 kg) were fed a hay diet ad libitum in a 5 x 5 Latin square experiment. Mass of ruminal contents was altered by adding varying weights of modified tennis balls to the rumen before the initiation of each experimental period. Treatments consisted of 50 balls with a specific gravity (SG) of either 1.0, 1.1, 1.2, 1.3 or 1.4 and the total weight of the balls was 7.45 kg, 8.50 kg; 9.25 kg, 10.55; and 11.55 kg, respectively. Osteoporosis is a major health problem in the United States and those that are on energy restricted diets such as anorectic subjects. Thirty Wistar strain, young male rats were fed diets with varying quantities of fat for a period of four weeks (energy intake 67--75% of requirement). The rat tibias' physical properties were measured. Bone mineral composition testing was conducted for calcium, magnesium, and phosphorus, on the tibia shaft. There were no significant differences among the parameters of length, diameter, volume, density, bone bending (elasticity), fracture load, and minerals between experimental diets. Factors affecting pregnancy maintenance between 28 and 67 days of gestation were studied in dairy cows (N = 211). Beginning on day 28 to 37 post breeding, cows were examined by ultrasonography for a viable pregnancy. Pregnant cows were re-examined for pregnancy losses during the late embryonic (≤day 45) and early fetal stages up to day 67. Overall pregnancy loss was 11%, mostly occurring before day 45. Cows with two corpora lutea maintained fewer pregnancies than cows with one corpus luteum. Pregnancy retention was less when concentrations of progesterone were low during days 28 to 37. The embryo apparently died …

Subjects were fifty-six women and their infants who were between the ages of six months and twenty-four months. They were clients of Special Supplemental Nutrition Program for Women Infants and Children (WIC) clinics. The WIC clinics were in the West Virginia counties of Ritchie, Gilmer, Calhoun, Braxton, Mason, Jackson, Pleasants, and Greenbrier where the anemia rates were greater than 10%.;Information concerning WIC participation, prenatal vitamin use, history of breast feeding, infant birth weight, and pre-term delivery were obtained through questionnaires. I sampled benthic macroinvertebrates along a 25.2 km stream continuum in the Central Appalachian Mountains of West Virginia from May---October 2000. Six components of macroinvertebrate community structure and function were measured: density, biomass, richness, community taxa similarity, feeding group composition, and metric composition. I quantified these components to determine if localized variation in alkalinity and canopy cover would modify stream continuum predictions. Localized variation in alkalinity and canopy cover significantly affected macroinvertebrate structure and function and modified stream continuum predictions. Results indicated that alkalinity was the primary determinant of macroinvertebrate structure and function, and that the effects of canopy cover …

Each year, the forest products industry sustains considerable product loss due to a reduction in market value of lumber brought about by microbial discoloration. Study examines communication of brand position in current national brand advertising on the four network stations during primetime programming. Content analysis of a random sample of advertising executions was performed on the premise that evaluation of said executions should be performed on the basis of cues provided by the execution to the viewer. Coded results are provided and analyzed. Brand position, as applied to communication in television commercials, is defined and explored.

Heart disease and obesity are concurring health problems in the state of West Virginia (WV). With obesity levels rising and heart disease remaining the number one cause of mortality in WV, the need for a lifestyle change is prominent. The West Virginia Restaurant Survey attempted to determine the availability of heart-healthy food items that restaurants offer and how the items are promoted in order to determine the need for intervention programs that support heart-healthy food choices. The endodontic resin sealer AH26® has been shown in multiple studies to have mutagenic potential. A unique test designed to identify phenotypic change in antibiotic resistance due to chromosomal mutation was described and performed. A broad literature has presented that there is a direct connection between parent participation in a child’s education and learning. However, very little literature exists on high school Professional Development School (PDS) students and their parent’s participation in their learning. This thesis focused on student and parent perceptions of student learning, respect and success, as well as, how parents were involved in their child’s education. The purpose of the current study was to determine whether a two-session media literacy program, A.R.M.E.D. (Acknowledging and Rejecting the Media's Influence on Eating and Body Image Disturbances), was an effective intervention for individuals who fell at various points on the eating disorder continuum. Ninety-three female college students participated in the study, with 46 experimental participants attending A.R.M.E.D and 47 participants serving as a control group. Between-subjects analyses did not reveal differences between the experimental and control group on each of the seven dependent variables. Chronic renal disease is the progressive loss of renal function that can lead to end-stage renal disease. Recent studies from our laboratory have shown decreased total NO production in patients with renal disease. Animal models have shown decreased total and renal NO production and chronic NOS inhibition in otherwise healthy rats causes renal injury. The overall objective in this set of studies was to investigate intrarenal NO production in various models of CRD and determine if NO deficiency parallels, and perhaps causes renal injury.
