Soil Health Assessment In Reclaimed Mine Soils, Katie Danielle Stutler
Soil Health Assessment In Reclaimed Mine Soils, Katie Danielle Stutler
Graduate Theses, Dissertations, and Problem Reports (ETD)
West Virginia is one of the top coal-producing states in the nation, which has resulted in over 500,000 acres of highly disturbed, anthropogenic mine soils caused by surface mining. Mine soils are often rocky and contain low organic matter content, low plant-available nutrients, and toxic metals, which could result in poor soil health and low productivity. Soil health has been defined as the continued capacity of soil to function as a living ecosystem that sustains plants, animals, and humans. It is assumed that with time since reclamation, mine soil properties and related functions may change, and soil health will improve …
Development Of Improved Acellular Pertussis Vaccines Through Inclusion Of The Rtx Antigen Or Induction Of Mucosal Immunity, Dylan Tyler Boehm
Development Of Improved Acellular Pertussis Vaccines Through Inclusion Of The Rtx Antigen Or Induction Of Mucosal Immunity, Dylan Tyler Boehm
Graduate Theses, Dissertations, and Problem Reports (ETD)
Pertussis is a human respiratory disease, primarily caused by the Gram-negative pathogen Bordetella pertussis. The infection is most severe and can be life-threatening in young children and infants where it manifests as a series of paroxysmal coughs. The disease is more commonly known as whooping cough, due to the whoop omitted during a massive inspiratory effort to bring air back into the lungs. Pertussis is a toxin-mediated disease that persists due to an early release of toxins that allow that bacteria to evade the cells of the innate immune response. The inhibition of the host response continues as toxin activity …
Influence Of Climate Change And Prescribed Fire On Habitat Suitability And Abundance Of The High-Elevation Endemic Cow Knob Salamander (Plethodon Punctatus), Carl David Jacobsen
Influence Of Climate Change And Prescribed Fire On Habitat Suitability And Abundance Of The High-Elevation Endemic Cow Knob Salamander (Plethodon Punctatus), Carl David Jacobsen
Graduate Theses, Dissertations, and Problem Reports (ETD)
Amphibians are facing global declines due to climate change, loss and degradation of habitat, invasive species, and disease. The Appalachian region of the eastern USA is a global biodiversity hotspot for salamanders, which are considered keystone species that influence nutrient dynamics in terrestrial and aquatic food webs. There are high rates of salamander endemism in the Appalachian region, with many species restricted to isolated, high elevation areas. The Cow Knob Salamander (Plethodon punctatus) is one such species. It is only found at elevations >675 m (most populations are above 900 m) on Shenandoah Mountain, North Mountain, and Nathaniel …
Role Of Sox9 In Cancer Stem Cell Regulation In Lung Cancer Chemoresistance And Particle-Induced Carcinogenesis, Maria Voronkova
Role Of Sox9 In Cancer Stem Cell Regulation In Lung Cancer Chemoresistance And Particle-Induced Carcinogenesis, Maria Voronkova
Graduate Theses, Dissertations, and Problem Reports (ETD)
Lung cancer is a leading cause of cancer-related death worldwide. Every year it claims more lives than other common malignancies, breast, prostate, and colorectal cancers, combined. And despite recent improvements in disease diagnostics and treatment, drug resistance and tumor relapse remain major contributors to low patient survival. Both issues have been largely attributed to the existence of cancer stem-like cells (CSCs) or tumor initiating cells (TICs). CSCs are a small cell population within heterogeneous tumors that is inherently resistant to apoptosis, chemo- and radiation therapy. Thus, it is critical to elucidate CSC-specific signaling pathways and identify early biomarkers for therapeutic …

Reconciling The Dissonance Between Historic Preservation And Virtual Reality Through A Place-Based Virtual Heritage System., Danny J. Bonenberger
Reconciling The Dissonance Between Historic Preservation And Virtual Reality Through A Place-Based Virtual Heritage System., Danny J. Bonenberger
Graduate Theses, Dissertations, and Problem Reports (ETD)
This study explores a problematic disconnect associated with virtual heritage and the immersive 3D computer modeling of cultural heritage. The products of virtual heritage often fail to adhere to long-standing principles and recent international conventions associated with historic preservation, heritage recording, designation, and interpretation. By drawing upon the geographic concepts of space, landscape, and place, along with advances in Geographic Information Systems, first-person serious games, and head-mounted Virtual Reality platforms this study envisions, designs, implements, and evaluates a virtual heritage system that seeks to reconcile the dissonance between Virtual Reality and historic preservation. Finally, the dissertation examines the contributions and …

Early embryonic loss in cattle is detrimental to reproduction thus, the profitability of both the beef and dairy industries. Therefore, an important aspect of study is to find techniques that beef or dairy producers could use to decrease early embryonic loss Galectin-1, the protein that was investigated in this research is important in modulating the maternal immune system in rodent and human early pregnancies by promoting fetomaternal immune tolerance. The role of galectin-1 within the reproductive tissues has not been thoroughly investigated in cattle. Galectin-1 is located in the caruncular and intercaruncular regions of the bovine endometrium, specifically the maternal …
A Study Of Gastrointestinal Pathophysiology, Emily Minor
A Study Of Gastrointestinal Pathophysiology, Emily Minor
Graduate Theses, Dissertations, and Problem Reports (ETD)
Though the duodenum is the primary site of iron absorption, the colon also contains iron transporters and participates in iron absorption. The purpose of this study was to characterize colonic iron absorption in control patients and those with ulcerative colitis (UC), a disease that frequently results in iron deficiency. Using RT-qPCR and western blot analysis, we found that colonic expression of the apical iron importer, divalent metal transporter 1 (DMT1), is equal to duodenal expression. However, colonic expression of ferroportin 1 (FPN1), an iron exporter, is only about one-quarter that of the duodenum. Patients with active ulcerative colitis were found …
Avian Occupancy Dynamics And Canada Warbler (Cardellina Canadensis) Environmental Correlates On A Central Appalachian Elevation Gradient In West Virginia, Gordon W. Dimmig
Avian Occupancy Dynamics And Canada Warbler (Cardellina Canadensis) Environmental Correlates On A Central Appalachian Elevation Gradient In West Virginia, Gordon W. Dimmig
Graduate Theses, Dissertations, and Problem Reports (ETD)
Mountains are important areas for avian diversity and conservation since they have steep elevational and environmental gradients. In the central Appalachian Mountains, several songbird species have trailing edge populations that are restricted to high elevations. Climate change and other factors can cause species distributions to change, which is of particular concern for birds in restricted trailing edge populations. Additionally, the Canada Warbler (Cardellina canadensis) breeds throughout the Appalachian Mountains and is species of conservation concern due to range-wide population declines. For this project, I used avian point count data from the central Appalachian Mountains to: (1) evaluate the …

Comparing Abundance Estimates From Closed Population Mark-Recapture Models Of Endangered Adult Atlantic Sturgeon, J E. Kahn, C Hager, J C. Watterson, N Mathies, K J. Hartman
Faculty & Staff Scholarship
Atlantic sturgeon Acipenser oxyrinchus oxyrinchus were listed as 5 distinct population segments under the US Endangered Species Act in 2012. At that time, only 2 abundance estimates of the Atlantic sturgeon population were available: one from commercial fisheries landings in the Hudson River ending in 1995 and one from mark-recapture research in the Altamaha River, Georgia, in 2004 and 2005. In 2013, we verified spawning in the York River, Virginia, system and initiated a multiple-year mark-recapture study focusing on spawning-run abundance. We used a Schumacher-Eschmeyer model and Program CAPTURE to produce estimates of annual spawning abundances from 2013 to 2018. …
Neural Processing Of Communication Signals: The Extent Of Sender–Receiver Matching Varies Across Species Of Apteronotus, Kathryne M. Allen, Gary Marsat
Neural Processing Of Communication Signals: The Extent Of Sender–Receiver Matching Varies Across Species Of Apteronotus, Kathryne M. Allen, Gary Marsat
Faculty & Staff Scholarship
As communication signal properties change, through genetic drift or selective pressure, the sensory systems that receive these signals must also adapt to maintain sensitivity and adaptability in an array of contexts. Shedding light on this process helps us to understand how sensory codes are tailored to specific tasks. In a species of weakly electric fish, Apteronotus albifrons, we examined the unique neurophysiological properties that support the encoding of electrosensory communication signals that the animal encounters in social exchanges. We compare our findings to the known coding properties of the closely related species Apteronotus leptorhynchus to establish how these animals …

Monocytes And Monocyte-Derived Antigen-Presenting Cells Have Distinct Gene Signatures In Experimental Model Of Multiple Sclerosis, Kelly L. Monaghan, Wen Zheng, Gangqing Hu, Edwin C. K. Wan
Faculty & Staff Scholarship
Multiple sclerosis (MS) is a chronic inflammatory disease mediated by a complex interaction between the autoreactive lymphocytes and the effector myeloid cells within the central nervous system (CNS). In a murine model of MS, experimental autoimmune encephalomyelitis (EAE), Ly6Chi monocytes migrate into the CNS and further differentiate into antigen-presenting cells (APCs) during disease progression. Currently, there is no information about gene signatures that can distinguish between monocytes and the monocyte-derived APCs. We developed a surface marker-based strategy to distinguish between these two cell types during the stage of EAE when the clinical symptoms were most severe, and performed transcriptome analysis …

Exhibit Panels
Which Side Are You On? is a new work by Gina Mamone, co-founder of the West Virginia-based art collective Queer Appalachia. Which Side Are You On? invokes the spirit of Zoe Leonard’s 1992 poem I Want a President, but speaks with the voice of 2018 rural America.
"I want a survivor for Governor. I want a Governor whose home has been raided by ICE. I want the child of a public school teacher for Governor, and I want someone who knows what days to hit what food pantries. I want a Governor who has had experience heating a home …
Discrimination And Behavioral Responses To Communication Signals Compared Across Apteronotids., Danielle Leigh Dillon-Seeger
Discrimination And Behavioral Responses To Communication Signals Compared Across Apteronotids., Danielle Leigh Dillon-Seeger
Graduate Theses, Dissertations, and Problem Reports (ETD)
Sensory systems are often uniquely tailored to encode behaviorally relevant signals and comparative studies across species can thus reveal how evolutionary changes shape sensory functions. The structure of communication signals varies widely between ghost knifefish species. Recent findings suggest that the nervous system co-adapted to the various signal structures observed across species to support different sensory behaviors. The aim for this thesis was to compare the sensory behavior of 3 species of ghost knifefish to contrast their behavioral performance with the known differences in neurophysiology. We hypothesize that for the different signal types and species, the ability to discriminate small …

Fundamental Mechanism Of Time Dependent Failure In Shale, Neel Gupta
Fundamental Mechanism Of Time Dependent Failure In Shale, Neel Gupta
Graduate Theses, Dissertations, and Problem Reports (ETD)
In underground coal mines, mining activity disturbs the natural equilibrium state of in-situ stresses. The induced and in-situ stresses deform the rockmass surrounding the mine openings. Primary roof supports impede the deformation of the rockmass overlying the entries. However, failure can occur in the bolted rockmass, causing fatalities and injuries. The rockmass failure is erratic, and its occurrence often varies from a few days to months and years after the opening of the entry. One of the often-neglected factors in this process is the effect of time on the stability of mine openings, which can often be observed in the …
Determining The Role Of Nudt7 In The Regulation Of Cellular Coa Levels And Metabolism, Stephanie Anne Shumar
Determining The Role Of Nudt7 In The Regulation Of Cellular Coa Levels And Metabolism, Stephanie Anne Shumar
Graduate Theses, Dissertations, and Problem Reports (ETD)
Coenzyme A (CoA) is an essential cofactor required for hundreds of metabolic processes. Because it is such a critical cofactor, CoA levels are tightly regulated. In the fasted state and in diabetic mice, the concentration of CoA increases dramatically in the liver. This phenotype is associated with constitutively low CoA degradation, a process that is emerging as a potentially important mechanism for CoA regulation. Nudt7 and Nudt19 are two mammalian peroxisomal enzymes with CoA-degrading activity, which are highly expressed in the liver and kidney, respectively. Limited information is available on the biochemistry of Nudt7 and Nudt19; the structural basis for …

An Assessment Of The Methods Used To Teach Content Skill Sets Required For The Introduction To Agriculture, Food, And Natural Resource Course, Lee I. Wright
Graduate Theses, Dissertations, and Problem Reports (ETD)
Numerous studies have shown that students learn best when theory is applied to hands-on experiences. Traditionally, vocational agriculture, which evolved into agricultural education, has incorporated hands-on learning. For example, students typically raise livestock and crops as part of supervised agricultural experiences; learn to use chain saws, tractors, hay rakes, balers and other equipment found on farms; and basic metal working skills. They also sample soil and water to test for mineral levels and types of pollution that might affect agricultural practices. The purpose of this study, which incorporates a survey of agricultural education teachers in West Virginia, is to determine …

The Role Of Sirt1 In Skeletal Muscle Function And Repair Of Older Mice, Matthew John Myers
Graduate Theses, Dissertations, and Problem Reports (ETD)
Human skeletal muscle is a highly metabolic tissue necessary for mobility and coordination. Responsible for approximately one-fifth of the resting human metabolism, skeletal muscle is also an important regulator of metabolites like glucose and contributes to the regulation of body temperature. Although there is a gradual decline in muscle mass associated with aging, a certain percentage of the population suffer from severe muscle mass and strength deterioration, classified as sarcopenia (5-13% for people aged 60-70 years old, 11-50% for those 80 or older). Sarcopenia is linked to increased morbidity and mortality rates in the elderly population, while annual healthcare costs …
Resurgence Of Academic Responses, Catherine Louise Stephens
Resurgence Of Academic Responses, Catherine Louise Stephens
Graduate Theses, Dissertations, and Problem Reports (ETD)
Resurgence is the recurrence of a previously reinforced response after a more recently reinforced response is placed on extinction. Resurgence may explain the recurrence of socially appropriate behavior, including academic responding, but this had not yet been empirically demonstrated. The aim of this study was to determine if resurgence would occur when a participant solved quadratic equations using multiple methods. Each participant was taught two methods of solving quadratic equations across experimental phases, followed by a phase in which neither method resulted in the correct solution. In the first phase, only simple factoring was reinforced. In the second phase, only …

Alzheimer’s disease (AD) is a neurodegenerative condition afflicting 5.7 million Americans with no effective treatments. The pathology of AD is described by the formation of neurotoxic oligomers formed by the self-assembly of amyloid-β (Aβ). The affinity of oligomers for cellular membranes disrupts calcium homeostasis and is believed to be the underlying cause of neurotoxicity; however, due to the metastability of oligomers, it has been difficult to understand their role in membrane interactions and self-assembly. Here we utilize post translational modifications (PTM) to understand the role of specific amino acids on Aβ40-membrane interactions and self-assembly. We employ ThT, AFM, …

Background
Alzheimer’s disease (AD) is defined by amyloid beta (Aβ) plaques and neurofibrillary tangles and characterized by neurodegeneration and memory loss. The majority of AD patients also have Aβ deposition in cerebral vessels known as cerebral amyloid angiopathy (CAA), microhemorrhages, and vascular co-morbidities, suggesting that cerebrovascular dysfunction contributes to AD etiology. Promoting cerebrovascular resilience may therefore be a promising therapeutic or preventative strategy for AD. Plasma high-density lipoproteins (HDL) have several vasoprotective functions and are associated with reduced AD risk in some epidemiological studies and with reduced Aβ deposition and Aβ-induced inflammation in 3D engineered human cerebral vessels. In mice, …

Water‐borne hormone measurement is a noninvasive method suitable for amphibians of all sizes that are otherwise difficult to sample. For this method, containment‐water is assayed for hormones released by the animal. Originally developed in fish, the method has expanded to amphibians, but requires additional species‐specific validations. We wanted to determine physiological relevance of water‐borne corticosterone in spotted salamanders (Ambystoma maculatum) by comparing concentrations to those taken using established corticosterone sampling methods, such as plasma. Using a mixture of field and laboratory studies, we compared water‐borne corticosterone levels to other traditional methods of sampling corticosterone for spotted salamander larvae, …
