Evolutionary Genomics Of Dynamic Sex Chromosomes In The Salicaceae, Ran Zhou Jan 2020

Evolutionary Genomics Of Dynamic Sex Chromosomes In The Salicaceae, Ran Zhou

Graduate Theses, Dissertations, and Problem Reports (ETD)

Identifying the sex-determination region (SDR) and other genomic features of sex chromosomes are of great importance in the studies of the evolution of sex. However, the process of accurately identifying the size and location of the SDR is often difficult, even when a genomic sequence is available. This usually is hindered by large repetitive elements and a lack of recombination in the SDR. In this thesis, I assemble sex chromosomes with whole genomic sequencing data, identify SDRs and explore their genomic features in two sister species from the Salicaceae family. I also develop an interpretation of the lability of the …

Neuroimaging Of Real-World Audio-Visual Sensory Integration In High-Functioning Autism, Paula J. Webster Jan 2020

Neuroimaging Of Real-World Audio-Visual Sensory Integration In High-Functioning Autism, Paula J. Webster

Graduate Theses, Dissertations, and Problem Reports (ETD)

Sensory processing differences are a prevalent aspect of autism spectrum disorder (ASD) that may contribute to core deficits of ASD such as repetitive behaviors as well as comorbidities including anxiety disorders. The ability to integrate information among our senses is required to comprehend the world around us and is crucial for the development of language, motor skills, and social communication. Prior studies have shown that individuals with autism differ from individuals without autism when presented with simple, non-natural audio-visual stimuli such as basic shapes accompanied by pure tones. Because the human brain processes non-natural and natural stimuli differently, more recent …

Interpretations Of Bicoherence In Space & Lab Plasma Dynamics, Gregory Allen Riggs Jan 2020

Interpretations Of Bicoherence In Space & Lab Plasma Dynamics, Gregory Allen Riggs

Graduate Theses, Dissertations, and Problem Reports (ETD)

The application of bicoherence analysis to plasma research, particularly in non-linear, coupled-wave regimes, has thus far been significantly belied by poor resolution in time, and/or outright destruction of frequency information. Though the typical power spectrum cloaks the phase-coherency between frequencies, Fourier transforms of higher-order convolutions provide an n-dimensional spectrum which is adept at elucidating n-wave phase coherence. As such, this investigation focuses on the utility of the normalized bispectrum for detection of wave-wave coupling in general, with emphasis on distinct implications within the scope of non-linear plasma physics. Interpretations of bicoherent features are given for time series from …

Two Approaches To Analyzing Institutions’ Spending Responses To Campus Sexual Assault, Amanda M. Rose

Graduate Theses, Dissertations, and Problem Reports (ETD)

Despite occupying a growing portion in contemporary U.S. higher education institutions’ annual budgets, empirical research on sexual assault costs is limited. The purpose of this study is to look at whether incidences of sexual violence on campus are associated with increased costs in policies and programs geared toward prevention of these crimes. Informing the analysis is a theoretical framework consisting of the revenue theory of costs and positional arms race theory. This study examines data from both IPEDS and the U.S. Department of Education Crime and Safety website over a ten year period, 2006-2015, for a set of over 2,300 …

Utilizing Conservation Genetics As A Strategy For Recovering The Endangered Candy Darter (Etheostoma Osburni) In West Virginia, Brin E. Kessinger

Graduate Theses, Dissertations, and Problem Reports (ETD)

The Candy Darter (Etheostoma osburni) is a small freshwater fish native to the New River drainage in West Virginia and Virginia that was listed as endangered in November 2018. It has been extirpated from much of its historic range in West Virginia, restricting it to the Gauley and Greenbrier river drainages. In addition to extirpations, the species is threatened by introgressive hybridization with the invasive Variegate Darter (E. variatum). Previous research primarily focused on hybridization, but population genetic analyses were limited. Population genetic analyses aim to identify distinct populations through genetic structure and characterize the levels …

Role Of Ciliary Proteins Adp Ribosylation Factor Like Gtpase 13b (Arl13b) And Bardet-Biedl Syndrome-8 (Bbs8) In Photoreceptor Outer Segment Morphogenesis, Maintenance, And Viability, Tanya L. Dilan

Graduate Theses, Dissertations, and Problem Reports (ETD)

Photoreceptor neurons are modified primary cilia with an extended ciliary compartment known as the outer segment (OS). The mechanisms behind the elaboration of photoreceptor cilia, OS morphogenesis, and maintenance remain poorly understood. In this work, we focused on dissecting the role of two ciliary proteins, the small GTPase ADP-ribosylation factor-like GTPase 13B (ARL13B) and Bardet-Biedl Syndrome-8 (BBS8) in the context of photoreceptor biology. Both BBS8 and ARL13B are linked to defects in ciliogenesis (cilia development) and Retinitis Pigmentosa (vision loss). ARL13B is implicated in regulating ciliary length, and BBS8 is part of the Bardet-Biedl Syndrome complex (BBSome); the BBSome is …

Alternative Media And Mulches In Organic Vegetable Production, Heather R. Griffith Jan 2020

Alternative Media And Mulches In Organic Vegetable Production, Heather R. Griffith

Graduate Theses, Dissertations, and Problem Reports (ETD)

Reducing or eliminating waste from organic vegetable production can conserve money as well as resources. Conventional greenhouse production of vegetable transplants often relies on abundant fertigation which produces large amounts of polluted runoff, and the field production of organic vegetables frequently relies on the use of polyethylene mulch. A greenhouse study was conducted to determine if organic transplants can be successfully produced in a greenhouse under reduced soil moisture in order to reduce leaching of nutrients and potential water pollution. To test this, we compared the effect of reduced irrigation volumes to the standard practice of irrigating with 120% of …

Evaluation Of Salmonella Enteritidis And Escherichia Coli O157:H7 Volatile Organic Compound Analysis For Food Safety Monitoring: A Preliminary Study, Jessica E. Lemley

Graduate Theses, Dissertations, and Problem Reports (ETD)

Food borne illness has the potential to impact every point of production from producers to consumers. Food product recalls are common due to the presence of food borne illness causing bacteria. Detection of volatile organic compounds (VOC) have the potential to reduce the number of illnesses and recalls through application of additional monitoring of food products like those most commonly recalled. The objective of this research was to 1) explore the use of closed-loop headspace analysis coupled with gas chromatography-mass spectrometry (GC-MS) on food borne illness causing bacteria and 2) examine the VOC profile of Escherichia coli serotype O157:H7 in …

The Host Gatekeeper: Using The Flagellar Pathway To Understand Symbiont Host Adaptation, Adam R. Pollio Jan 2020

The Host Gatekeeper: Using The Flagellar Pathway To Understand Symbiont Host Adaptation, Adam R. Pollio

Graduate Theses, Dissertations, and Problem Reports (ETD)

The acquisition of microbial partners is a strategy used by a diverse group of arthropods to overcome ecological barriers that might normally make certain niches uninhabitable. The unique phylogenetic opportunities attainable from the natural experiment of the Sodalis-allied clade allow for better understanding of how molecular structures evolve through time. Here, we focus on the evolution of the flagellar synthesis pathway, due to its complexity and ability to diverge in response to ecological pressures. We used this molecular pathway and natural experiment to show that normal evolutionary outcomes associated with symbiosis (i.e., genome reduction) do not explain the predicted conservation …

Allopurinol Alters The Expression Of Cytokines And Mediators Of Immune Cells In The Systemic 2 Tissue By Decreasing Plasma Urate., Lundrim S. Marku

Graduate Theses, Dissertations, and Problem Reports (ETD)

Diminished levels of urate have been linked to oxidative stress in birds and mammals. Urate, a major antioxidant that lowers reactive oxygen/nitrogen species (ROS/RNS), is thought to ameliorate the effects. The relationship between reduced urate, the immune system and the pathogenesis of the intestine, the liver or the brain has not been well characterized in avians. Moreover, recent studies have proposed that an increased permeability in the intestine due to some insult can induce inflammation in peripheral organs such as the brain. Allopurinol, a relatively toxic purine analogue that serves as a xanthine oxidase inhibitor, reduces urate levels which can …

Clincial Translational Science Research Of The Functional Role Of Large Conductance Potassium Channels In Selective Destruction Of Triple Negative Breast Cancer Cells, Gina Sizemore

Graduate Theses, Dissertations, and Problem Reports (ETD)

ABSTRACT

The preliminary background that puts this research into context is threefold; it is the aggressive nature of triple negative breast cancer (TNBC), the complexity surrounding its pathology, and the significant lack of targeted treatment for this disease. To clarify the focus of my research, I have concentrated on identifying a targeted treatment for TNBC. In the process, I have identified cycles of reciprocity between treatment, clinical diagnosis, staging, and pathology that will be addressed in smaller papers. However, the weight of this work is in the discovery of a novel target for triple negative breast cancer. The value of …

Legal Remedies To Address Stigma-Based Health Inequalities In The United States: Opportunities And Challenges, Valarie K. Blake, Mark L. Hatzenbuehler Jun 2019

Legal Remedies To Address Stigma-Based Health Inequalities In The United States: Opportunities And Challenges, Valarie K. Blake, Mark L. Hatzenbuehler

Faculty & Staff Scholarship

Stigma is an established driver of population-level health outcomes. Antidiscrimination laws can generate or alleviate stigma and, thus, are a critical component in the study of improving population health.

Currently, antidiscrimination laws are often underenforced and are sometimes conceptualized by courts and lawmakers in ways that are too narrow to fully reach all forms of stigma and all individuals who are stigmatized.

To remedy these limitations, we propose the creation of a new population-level surveillance system of antidiscrimination law and its enforcement, a central body to enforce antidiscrimination laws, as well as a collaborative research initiative to enhance the study …

Faculty & Staff Scholarship

Rates of adverse cardiovascular events have increased among middle-aged adults. Elevated ceramides have been proposed as a risk factor for cardiovascular events. Diet quality and weight status are inversely associated with several traditional risk factors; however, the relationship to ceramides is less clear. This study aimed to determine associations of adiposity and diet quality with circulating ceramides in middle-aged adults (n = 96). Diet quality was estimated using the Healthy Eating Index 2015 (HEI-2015). Serum ceramide concentrations were determined by liquid chromatography–mass spectrometry. A ceramide risk score was determined based on ceramides C16:0, C18:0, and C24:1 and their ratios to …

Mammary Tumor And Mastectomy Synergistically Promote Neuroinflammation In A Breast Cancer Survivor Model, Kathryn M. Emmer, William H. Walker Ii, Ning Zhang, A. Courtney Devries

Clinical and Translational Science Institute

Understanding why breast cancer survivors are at an increased risk for cognitive and affective disorders is essential for developing targeted treatment plans and improving quality of life. Microglia priming results in chronic neuroinflammation and can contribute to neuronal degeneration and dysfunction, thereby offering a potential mechanism for altered brain function that persists after tumor removal. This study examined whether mammary tumors alter microglia and augment the inflammatory profile and behavior of mice. To test this, non-metastatic mammary tumor cells (67NR) were injected orthotopically into the mammary glands of BALB/c mice, allowed to grow for 16 days, and then the tumors …
