The UROtsa cell line is a unique model of human urothelium with considerable utility in toxicology-based research. UROtsa cells maintained in serum-free medium have several morphological features of transitional epithelium of the urinary bladder including a multilayered appearance and apically located umbrella-like cells that exhibit tight junctions. These cells also have a basal expression pattern of heat shock protein (hsp) and metallothionein (MT) genes similar to that observed in in situ urothelium. Integrated biomarkers of physiologic and genetic damage in sperm are needed to identify factors that affect male fertility and influence a father's risk of producing abnormal progeny. Offspring studies show a preferential paternal origin for heritable structural chromosomal abnormalities, presenting the need for methods to efficiently detect structural chromosomal abnormalities in human sperm. This dissertation breaks new conceptual ground toward the understanding of paternally transmitted genetic disease by adapting FISH technology to detect human sperm that carry chromosomal breaks and partial chromosomal duplications and deletions and by integrating state-of-the-art biomarkers of semen quality including computer-assisted sperm analysis (CASA). Differential Reinforcement Of Fixed-Interval Interresponse Times: Effects On Choice., Tammy Renee Wade

Eight rats were exposed to a concurrent-chains procedure. Half of the rats chose between a fixed-interval schedule that required low rates of responding in the paced terminal link and a simple fixed-interval schedule in the unpaced terminal link. The other half chose between a fixed-interval schedule that required high rates of responding and a simple fixed-interval schedule. Interresponse-time criteria were based on the percentiles of a baseline distribution of interresponse times obtained for each rat. The criteria increased in stringency across conditions. Consistent changes in response rates as the result of the interresponse-time criteria were not obtained. Therefore, preference between …

Located sixty miles above Philadelphia, the Lehigh Valley follows the seventy-mile path of the Lehigh River as it descends from the Pocono Mountains south through the Blue Mountains into the rich agricultural lands of the Appalachian Great Valley. With anthracite deposits in its northern reaches, rich agricultural lands in its middle section, and commercial connections to Philadelphia at its mouth, nineteenth-century industrialists saw the river as a potential transportation route. From 1817 to 1829, the Lehigh Coal and Navigation Company developed a system of slack water navigation along the river. With the opening of the channel in 1829, extractive industries …

In chick embryo hepatocytes, activation of acetyl-CoA carboxylase-α (ACCα) transcription by 3,5,3′-triiodothyronine (T3) is mediated by a cis-acting regulatory unit (−101 to −71 bp) that binds the nuclear T3 receptor (TR) and sterol regulatory element-binding protein-1 (SREBP-1). SREBP-1 directly interacts with TR on the ACCα gene to enhance T3-induced transcription. Here, we show that treating hepatocytes with T3 or insulin stimulates a 4-fold increase in the concentration of the mature, active form of SREBP-1. When T3 and insulin are added together, a 7-fold increase in the mature SREBP-1 concentration is observed. Time course studies indicate that the T3-induced increase in …

Metals have been indicated in causing generation of reactive oxygen species (ROS) and cellular injury. The overall objective of the present study was to investigate ROS generation from metals and the related effects on biological systems. The results obtained from the present studies have demonstrated that: (a) Cobalt is capable of generating ·OH via a Co(I)-mediated Fenton-like reaction. (b) The ·OH radical is generated by reaction of H2O2 with Cr(V), which is produced by enzymatic one-electron reduction of Cr(VI). The effects of weight; high (HW) or low (LW), and diet; high-energy (HE) or low-energy (LE), on the fertility of 118 yearling Angus, Hereford, and Angus x Hereford beef heifers were investigated. Heifers were blocked by body weight, and randomly assigned to diets for 47 and 42 days in years, 1 and 2, respectively. Estrus was synchronized with MGA (8 d, 0.5mg/hd/d), PGF2α (25 mg i.m.), and estradiol benzoate (400 µg i.m, 48 h after PGF2α) regimen. Heifers were artificially inseminated between 78-84 h after the PGF2α injection and then exposed to a bull for the second service. Overall pregnancy …

The concept of teacher self-disclosure has been studied using a variety of methods. A review of teacher self-disclosure literature found that operationalization of teacher self disclosure had yet to be agreed upon by researchers. This thesis applied a new teacher self-disclosure scale developed primarily for this research. It was hypothesized teacher self-disclosure would be related to (1) the student motives of functional, relational, and participatory, (2) class participation, (3) affect for class, (4) affect for teacher, and (5) relational certainty. A study was conducted in the George Washington National Forest in Virginia and the Monongahela National Forest in West Virginia. Carabid samples were collected over four years and compared to soil characteristics, surface characteristics and meteorological conditions to determine what role these factors play in carabid habitat selection. Opportunity also existed to compare carabid populations before and after the application of microbial insecticides on the sampling sites. Three of the pieces were written for this collection and the other two are revisions of works composed for graduate writing workshops. The stories all deal with people who have some type of personal limitation that causes them to have trouble functioning in the world. Double-observer point counts are a new method of inventorying avian fauna that allow detection probabilities to be calculated for each point or species of bird. These detection rates allow estimation of abundances that are more robust than estimates derived from single-observer counts because detection probabilities factor in birds present but not detected. The combined double-observer (DO) technique involves two observers recording data together on a single data sheet. The primary observer dictates the number of each species that is detected while the secondary observer records this information; the secondary observer also notes birds that the primary observer did not detect. …

The Retinal homeobox (Rx) gene codes for a transcriptional regulator involved in retinal tissue development, and the protein sequence of this transcription factor has been highly conserved throughout evolution. Previous Rx-knock-out and over-expression experiments have identified Rx as an essential component for proper eye formation. Here we used gene sequencing to identify 2 different mutations in the hRx gene of three anophthalmic and microphthalmic patients. The Q147X mutation, located in helix 1 of the homeodomain most likely prevents Rx from binding to DNA, therefore preventing transcriptional regulation by Rx. This study develops a quantitative and objective method of analyzing community vulnerability combined with potential exposure to emission hazards. This extends earlier work concerning Environmental Justice Through Pollution Prevention (EJP2) initiatives for optimal selection of facilities for resource allocation for pollution prevention initiatives. A 1992 EPA report indicated that minority and low-income communities are exposed to higher levels of pollution in their neighborhoods than that of the general population. Environmental Justice is a grassroots derived term for addressing these disproportionate shares of negative environmental impacts, which are placed upon people of minority or low socio-economic status. This research is an exploration of how retail is moving from the mall to the Internet. It applies uses and gratifications analysis of consumer satisfaction and ecommerce Web sites. The study compares obtained and sought after e-commerce consumer gratifications. The participants for the study attended West Virginia University in the spring 2002 semester. The study produced 50 valid responses that found consumers sought after gratifications for product display and navigation features were not met. This research also shows that experienced Internet consumers trust e-commerce Web sites privacy/security features. Implications for companies with e-commerce Web sites are explored.

The purpose of this study was to determine and compare the tobacco use behaviors of dental and dental hygiene students enrolled in U.S. dental schools with their perceptions and performance of tobacco use cessation counseling activities (TUCCA). A survey was mailed to a stratified sampling of accredited dental schools with which dental hygiene programs (n=8) were affiliated. Results were reported in frequencies, cumulative frequencies, and percentages. With use of the JMP program, Chi Square analyses were performed to test for statistical significance. An increase in the incidence of respiratory disease reported in a herbicide producing factory correlates with potential exposure to 3-Amino-5-mercapto-1,2,4-triazole (AMT) and AMT based product 1 (AMT-bp1). In animal studies, no signs of systemic toxicity or irritancy were observed with concentrations up to 25% AMT or 40% AMT-bp1. AMT-bp1 was negative while a dose dependent increase was observed following exposure to AMT in the LLNA. An increase in B220+IgE+ cells was observed at 25% AMT exposure. AMT was negative in the MEST (up to 25%). Following dermal exposure 5 days a week, animals exposed to 25% AMT demonstrated elevated levels …

This thesis defines what an effective academic web site is, and examines the utilization of web sites by the Land Grant colleges’ foreign language departments. A system with the parameters of evaluation was created from studies that observed web users’ perceptions of web sites. Four main factors were analyzed: informativeness, interactivity, entertainment, design and usability. The results showed that departments of foreign language are not projecting a positive image throughout their web sites. Moreover, the web masters of the foreign language departmental sites are not considering entertainment and interactivity features as fundamental elements in the development of academic web sites.

The pathogenesis of familial amyotrophic lateral sclerosis (fALS), a fatal neurodegenerative disease that sometimes results from SOD1 mutations, may involve the peroxidase reaction of the SOD1 protein. The aim of this thesis was to test the hypothesis that the peroxidase reaction of SOD results in SOD-centered peroxyl radical formation. The source of oxygen incorporated into 2-oxohistidine was to have been determined using HPLC/Mass spectrometry. This aim was not accomplished. Islands on the Ohio River contain valuable habitats that are unique and irreplaceable on a national or ecoregion basis. The bone marrow microenvironment composed in part of stromal cells provides the niche in which normal hematopoiesis occurs. Hematopoiesis generates cellular components of the immune system and blood through proliferation and differentiation of multipotent hematopoietic stem cells (HSC). In preparation for bone marrow transplantation, radiation or chemotherapy treatment temporarily disrupts the balance of hematopoiesis as many HSC and progenitor cells are destroyed. Following treatment cessation, transplanted HSC and progenitor cells migrate, or "home", back to the bone marrow microenvironment and initiate productive hematopoiesis. HepG2 cells were used as a model system to study the regulation of glucose-6- phosphate dehydrogenase (G6PD). Regulation of hepatic G6PD expression involves changes in the amount of G6PD pre-mRNA in the nucleus in the absence of changes in the transcription rate of the gene. G6PD expression is increased in HepG2 cells incubated with increasing glucose concentrations. G6PD mRNA is increased 3- to 4- fold by incubation in 25 mM glucose, as compared to 0 mM glucose. Structure, stability, kinetics and mechanisms of oxidation of some important organosulfur compounds (2-aminoethanethiolsulfuric acid, 4-methyl-3-thiosemicarbazide and thionicotinamide) have been investigated by using UV-VIS, NMR, stopped-flow, and qualitative analysis. Results obtained showed that oxidation occurs mainly at the reactive sulfur center of the molecules via successive addition of oxygen atoms. The oxidation of thionicotinamide, an important xenobiotic, gave different oxygenated products, depending on the oxidizing power of the oxidant used. The sulfinic acid of thionicotinamide is stable with mild oxidizing agents while stronger oxidants desulfurize the molecule to yield sulfate as observed with other organosulfur compounds studied. The Myogenic Regulatory Factor family (MRFs) is a family of transcription factors that plays a key role in the development of skeletal muscle. MRFs include Myo D, myf-5, myogenin, and MRF-4. Myo D and myf-5 are genes expressed during myoblast proliferation, whereas myogenin and MRF-4 are expressed during myotube formation and differentiation. Insulin-Like Growth Factor 1 (IGF-1) controls muscle development both embryonically and during postnatal growth and repair by inducing myoblast proliferation and muscle hypertrophy. Patterns of commerce and settlement in colonial America were shaped by watercourses and portages. Road building skills were primitive and boating represented the most effective means of moving bulk freight and passengers. Traders and frontiersmen leading the advance of European settlement into the Appalachian Mountains therefore adopted the shortest crossings between watersheds. Natural conditions in Pennsylvania did not promote an orderly advance westward and produced isolated clusters of settlement around Philadelphia and Pittsburgh, with small and widespread settlements in Northwest Pennsylvania. Population growth, combined with economic growth have been primary causes of urban sprawl and increased pressure on natural resources. Urban development often adversely affects land, water and air quality. Although not considered a threat to the nation's food production, land development and urbanization are critical issues because they can lead to fragmentation of agricultural and forest lands, loss of prime farmland, wildlife habitat and other resources, additional infrastructure costs for communities and regional authorities, and competition for water. Each of the four children was considered individually and holistically through case study methodology, and some comparisons across children were considered. Since the expression of the brain specific metallothionein isoform, MT-3, was shown to be expressed in many, but not all prostate cancers, this gene was transfected into the commonly utilized prostate tumor cell line, PC-3, to study the consequence of MT-3 expression on growth rate and resistance to chemotherapeutic agents. PC-3 cells were stably transfected with MT-3 or MT-1E under control of the constitutive CMV promoter. The levels of MT-3 protein for the 5 overexpressing clones had an average level of 8.3 ng MT-3 per μg protein as assessed by quantitative immunodot blot analysis, and three were picked for further …

The purpose of this study was to identify the predominant self-perceived brain hemispheric cognitive styles and leadership styles of principals of schools identified as a West Virginia School of Excellence and to compare these with principals of schools that have not been identified as a West Virginia School of Excellence.
