Although pain responding and emotional states such as fear are highly correlated and appear to interact, the causal relations between these variables have yet to be fully defined. This study sought to further illuminate these relations by investigating how exposure to one of these states (i.e., pain or fear) affects responding to the other. The order in which participants experienced fearful or painful stimuli was manipulated, as well as fear level (i.e., "low" & "high"). Measures of behavioral escape, heart rate, and verbal reports were obtained throughout four experimental trials, as well as rankings of aversiveness for all stimuli encountered. …

This present study was designed to examine the relation between cardiovascular reactivity to stress and the behaviors of individuals classified as either "extraverts" or "introverts." Although experimental psychophysiological studies have been conducted to examine the relation between extraversion and physiological arousal, little is known about the ways in which extraversion and cardiovascular reactivity to stress are related. According to the optimal arousal theory, both extraverts and introverts would be expected to be more reactive to a social challenge than to a mundane non-social task, with introverts exhibiting greater reactivity to both tasks than extraverts. Specific encapsidation of viral RNA is an essential step of retroviral replication cycle. Extensive biochemical and genetic analyses suggest that majority of retroviruses employ similar mechanisms for specific RNA packaging. Better understanding of mechanisms of this process may help to develop new anti-retroviral drugs against such serious pathogens as human immunodeficiency virus types 1 and 2.;This study concerns with specific packaging of RNA by two simple retroviruses, murine leukemia virus (MLV) and spleen necrosis virus (SNV). Previous studies demonstrated that quinidine causes G1/G0 cell cycle arrest and inhibition of proliferation in MCF-7 human breast cancer cell line (Woodfork, K. et al., 1995). The goal of studies reported here was to understand the molecular mechanisms of c-myc gene regulation by quinidine. C-myc is one of the most common oncogene aberrations in breast cancers (Deming, S. L. et al., 2000). C-myc functions include regulation of cell cycle, proliferation, differentiation, and apoptosis. Ohio River electrofishing surveys identified embayments as preferred macrohabitat of subadult largemouth bass. Catch rates within macrohabitats were low ranging from 0.00--2.73 subadult bass/hour. Embayment electrofishing CPUE was higher than tributary or main channel except in spring, when tributary CPUE was highest.;Core use areas were calculated from biotelemetry data and ranged from 88--59,400 m2, representing 0.02--89.00% of available study location macrohabitat. Subadult bass movement was limited to nursery embayments or tributaries with infrequent movement to nearby main channel areas. Bass preferred a mixed microhabitat consisting of a combination of vegetation and woody debris. This mixed microhabitat was less common in …

Frequency of detection was monitored in three flocks of turkeys from May, 2000 to March 2001. The effect of time was considered for hens in flocks 1 and 2, and the effect of time, gender, and litter (fresh or used) was determined for flock 3. Poults, poult-box liners, waterers, and fecal droppings were monitered throughout production for the presence of Campylobacter using Campy-Cefex agar incubated at 42°C under microaerophilic conditions (85% N2, 10% CO2, and 5% O2). Peak colonization occurred near 3 weeks of production. Frequency of Campylobacter isolation from bird sources paralleled isolation from waterers. Frequency of detection from …

Mountaintop surface mining for coal has been practiced in West Virginia for over three decades, but its environmental impacts have become increasingly scrutinized in recent years. Therefore, this study evaluated soil development on different aged mined lands. Four different ages (2, 7, 11, and 23 years) of minesoils and two different slope classes, nearly level to gently sloping and steep to very steep, along with three contiguous native soils on a reclaimed mountaintop removal mine in Logan County, West Virginia were described and sampled. The Deckers Creek drainage basin covers approximately 16,600 hectares (62.9 square miles) in Monongalia and Preston Counties of West Virginia. Throughout the twentieth century, this watershed has received high levels of pollution in several forms. A 1974 study examined the water chemistry in Deckers Creek over a six-month period at 29 sample points. Water samples from the same sites were collected and analyzed monthly from March 1999 through October 2000 for acidity, alkalinity, total iron, aluminum, manganese, and other metals, as well as for fecal coliform and fecal streptococcus concentrations. In porcine ovaries, endothelin-1 (ET-1) may have an autocrine/paracrine role in follicular development. Endothelin-converting enzyme-1 (ECE-1) converts big endothelin-1 to ET-1. In situ hybridization was used to determine the ovarian compartment that synthesizes ECE-1. ECE-1 mRNA was detected in granulosa cells of large (10 mm) and medium (3 mm) antral follicles. This study, in conjunction with previous studies suggests a novel autocrine role of ET-1 as a possible local ovarian factor of follicular development. ET-1 may play a role during prostaglandin (PG) F2 alpha-induced luteal regression in the cow. Effects of PGF2 alpha treatment were examined during early and mid-luteal …

The purpose of this study was to provide information that might be useful to researchers and extension personnel at West Virginia University in developing and designing educational programs concerning pest management for corn farmers. This research identified current trends in management practices, insect, fungal, and weed problems, and educational needs so that instructional materials can be designed to meet clientele needs. Data were collected via 100 questionnaires mailed to farmers in the top ten corn producing counties in West Virginia. Despite the current recognition of infant procedural pain and distress, there is a dearth of research, both in the assessment and treatment literature, of infant procedural distress. This study examined how specific parent behaviors relate to infant procedural distress and whether this relation is mediated by birth order. Results indicated that females were rated as more distressed than were males, and that birth order did not prove to be a distinguishing factor in infant distress. Finally, while some behaviors appear to have a similar influence on both preschoolers' and infants' behaviors, parents' behavior during infants' distressing procedures were found to …

Birds have few reliable indicators of aging. Pentosidine (Ps), a biomarker of aging, is a product of non-enzymatic glycation, which accumulates in the tissues of an animal over its lifespan. The intent of this study was to determine if comparable change in Ps concentrations could be established in the skin of wild birds of known ages. Skin samples were obtained from the breast of 44 birds of various species. Foot webbing samples were obtained from 24 California Gulls. Chronic, inescapable stress during pregnancy (i.e. prenatal stress) modifies the behavior of the adult offspring. Several behaviors were evaluated in adult prenatally stressed (PS) rats, most notably the acoustic startle response and behavioral inhibition (i.e. freezing). PS rats do not differ in baseline peak startle responding, or in the habituation of the peak startle response when compared to the non-prenatally stressed (CON) rats. However, PS rats have greater peak responses and longer latencies to reach the peak response following systemic doses of the 5-HT 1A agonist, 8-OH-DPAT, compared to CON rats. Researchers have demonstrated that individuals can successfully reduce their heart rate (HR) response to a stressor when provided with heart rate feedback. However, it is unclear whether individuals can transfer HR reduction skills to stressors not used during HR feedback training. The present study used a multiple baseline, single subject design to examine the transfer of HR feedback training among six individuals. Participants were provided with HR feedback training during the presentation of a videogame, a mental arithmetic challenge, and a hand grip task until the participants reduced their HR to within 4 bpm of their resting HR or until …

This study addresses the genotoxicity of oxidative stress. We induced oxidative stress in cultured rat oligodendrocytes by exposure to hydrogen peroxide (H2O2). Intact genomic DNA was isolated by agarose embedding and its fragmentation was assessed by field inversion gel electrophoresis (FIGE). To detect single strand fragmentations, the DNA was digested with S1 nuclease before FIGE. We found that the exposure of oligodendrocytes to pathological concentrations of H2O2, induces a rapid digestion of nuclear chromatin in a pattern consistent with higher order chromatin degradation (HOCD). HOCD, the hallmark of programmed cell death (PCD), proceeds through single strand scissions that accumulate at …

Despite a profound reduction in hemodynamic shear stress, a sustained release of endothelium-derived nitric oxide (NO) limits arteriolar constriction during periods of increased sympathetic nerve activity in the rat small intestine. In this project, we sought to test the hypothesis that adenosine, formed in response to a flow-dependent fall in local PO2, serves as the stimulus to preserve endothelial NO release under these conditions. Sympathetic nerve stimulation induced frequency-dependent arteriolar constrictions and a flow-dependent fall in local PO2. The arteriolar responses to nerve stimulation were enhanced after inhibition of NO synthase with NG-monomethyl-L-arginine (L-NMMA). Under a high (20%) O2 superfusate, …

In the ewe, anestrus is caused by increased sensitivity to E2 negative feedback that is mediated by A15 dopaminergic neurons that are only responsive to E2 in anestrus. Experiment 1, tested the hypothesis that synaptic input on A15 neurons varies with season by examining synapsin-positive close contacts on A15 cells using immunofluorescence and confocal microscopy. The number of contacts on dendrites, but not somata, increased during anestrus. There were also corresponding changes in dendritic morphology including an increase in dendritic length, surface area, and number of bifurcations. To demonstrate that alveolar macrophages (AM) from rats exhibit pepT1-like transporter for the uptake of small arginine-containing peptides (ACPs) and utilized these peptides as direct substrates for nitric oxide (NO) production.;Method. A HPLC assay was developed for quantitative measurement of Arg and ACPs in rat plasma and bronchoalveolar lavage (BAL) fluid. A novel portable particulate matter measuring system was designed, developed and qualified. The heavy-duty engine Federal Test Procedure (FTP) requires the use of a full-flow tunnel to measure particulate emissions from diesel fueled engines. However, a full-flow tunnel based constant volume sampler (CVS) system is not only expensive to build and maintain, it also requires a large work space. The portable micro-dilution system that could be used for measuring on-board, in-use emissions from heavy-duty vehicles would cost significantly less. Reverse-transcription polymerase chain reaction (RT-PCR) was performed to measure myostatin, follistatin, activin-B gene expressions during chicken embryonic development. Strong myostatin expression was found in the early chicken embryos (E 0, E 1) and the developmental expression pattern of myostatin mRNA coincided with the periods of primary and secondary muscle fiber formation. Follistatin transcripts followed a linear expression pattern from E 0 to E 20, while activin-B had a quadratic pattern. The ontogeny of myostatin gene expression was nearly identical in satellite cells isolated from pectoralis major (PM) and biceps femoris (BF) muscles of chicken. The objectives of this study were to determine the effect of the crosslink breaker ALT-711 on the concentration of soluble collagen, as well as its effectiveness on meat tenderness.;Final body weights were not affected by treatments. Muscle tenderness was higher (p < 0.05) for TB, SS, BF, ST, and LT (p < 0.10) for A-maturity animals compared to control and ALT-711 treated animals. No consistent ALT-711 effect on shear force measurements was found. Shear force increased linearly with age BF, R2 = 0.94; SM, R2 = 0.81; LL, R 2 = 0.77; SS, R2 = 0.63; LT, R2 = 0.32; TB, R2 = 0.37; ST, R2 = 0.40. Total collagen content was not different (p > 0.10) between A-maturity, control and ALT-711 treated animals. However, the concentration of soluble collagen was higher (P < 0.05) for A-maturity group. Insoluble collagen content was higher (p < 0.05) for longissimus thoracis, biceps femoris, and (p < 0.10) for supraspinatus of control animals. Muscle soluble collagen content decreased linearly as chronological age increased LT R2 = 0.98; LL R 2 = 0.92; TB R2 = 0.99; BF R2 = 0.99; ST R2 = 0.95; SM R2 = 0.99; SS R2 = 0.36. The soluble collagen percentage was higher for ALT-711 treated animals compared to control animals.;ALT-711 tended to increase muscle soluble collagen content of aged cattle. ALT-711 may be an effective age-related collagen crosslink breaker. However, ALT-711 may not be used to improve meat quality of aged cattle.

Rainbow trout, Oncorhynchus mykiss, fillets were brined in 8.7 or 17.4% (w/v) sodium chloride solution, respectively, for 30, 60, 90, or 120 min at 3°C. Increased brine concentration decreased brined pH, myosin solubility, brined and cooked moisture content, and water activity, and increased fillet weight loss after brining, cook yield, actin solubility, water-phase salt content, and shear force. Procrastination has been defined as the choice of a larger, later work requirement over a smaller, sooner work requirement. In one experiment (Mazur, 1996), pigeons chose a later-onset work requirement over an earlier-onset work requirement when work requirements were identical and reinforcement was at an equal delay from the choice point. In Phase I of the present experiment, reinforcement was delivered to pigeons immediately upon completion of either an early- or a late-onset work requirement in the terminal links of a concurrent-chains schedule. The present study was performed to investigate the effects of low ascending concentrations of progesterone on fertility in lactating beef cattle. Cows on the low ascending progesterone treatment received PGF2alpha from days 3 to 4 after estrus. All cows received PGF2alpha on day 9, and estradiol benzoate on day 11. Cows were inseminated artificially 12 hours after estrus. Concentrations of progesterone were lower in treated versus control cows. Pregnancy rate did not differ between treated and control cows. Pregnancy rate was greater in cows that ovulated a follicle from the right (78%) versus left (52%) ovary (p < 0.01). Low ascending progesterone early in the estrous cycle did not decrease fertility to the same extent seen with extended periods of low progesterone.

The effect of three clipping managements on the growth and production of Kentucky bluegrass was investigated at the West Virginia University Livestock Farm. Herbage accumulation, botanical composition, leaf extension rate (LER), leaf appearance rate (LAR), and tiller density were observed. The sward was clipped to three heights, 2.5, 5.1, and 7.6 cm, from a recovery growth of 13 to 15 cm. Nitrogen was applied in split applications twice a year at three rates (0, 80, and 160 kg N ha-1).
