The effects of different pH's (4 .5 , 5.5, 6.5 and 7.5) and temperatures (30-80 •c) on the functional properties and microstructure of chicken breast salt soluble protein (SSP) in 0.6 M NaCI were investigated. Protein solubility decreased from 98% at pH 6.5 to less than I% at pH 4.3 and below. Salt soluble protein at pH 4.5 produced a discontinuous gel which could not be measured by back extrusion and had an expressible moisture above 80% at all heating temperatures . Macromolecular assemblies of proteins and hydrocolloids in low-fat dairy products contribute to the structure and texture of these foods. Immunolocalization techniques were used to identify B-lactoglobulin, casein, bovine whey proteins, and egg albumin in low-fat frozen desserts, reduced fat process cheese, and salad dressings. SimpJesse18 I 00 protein particles were examined and characterized by these methods and compared to naturally occurring protein structures in the low-fat foods. Hydrocolloid identification in low-fat salad dressing was accomplished by complexing cellulase and hemicellulase with colloidal gold and applying the probe as a pre-embedding step, on sections of embedded specimens or on material dried …

The microstructure of "Xixona turron" , a typical Spanish confectionery product, made with toasted almonds, sugar and honey, was elucidated, in order to confirm the nature of those physical changes occurring in its manufacturing process, which has not been previously studied. The effect of the major components of "Xixona turron" (oil, proteins and sugar) on its structural c!Jange was established. The initial oil suspension of sugar syrup drops and proteins undergoes a phase inversion , becoming an oil /water emulsion when hot, and a solid-like structure, when the aqueous sugar continuous phase solidifies at room temperature. The distribution of aromatic constituents, including lignin , in the leaf cell walls of "Coastal" hermudagrass (Cynodon dacrylon (L.) Pers.) was investigated using scanning ultraviolet (UV) microspectrophotometry. Leaf blade sections and individual tissue types were scanned at three wavelengths representing the absorbance maxima (318, 287 and 250 nm) of aromatic constituents present in bermudagrass leaves. The measured absorbance data were printed in a geometric arrangement to produce an image of the distribution and amount of aromatic constituents among and within cell wall types which vary in digestibility . The effects of five chemical agents on the stability, texture and microstructure of cooked comminuted meat batters were studied. B-mercaptoethanol (/3-ME), hydrogen peroxide (H20~, ethylenediaminetetraacetic acid (EDT A), urea, and polyoxyetbylene sorbitan monooleate (fween 80) were used. The reduction of disulphide bonds by 8-ME or the oxidation of free sulphydryls by H20 2 prior to cooking did not affect liquid and fat losses during cooking as compared to the control (2.5% NaCI). However, texture profile analysis (fPA) showed that treatment with H20 2 resulted in a harder, more cohesive product than the control and ,8-ME treatments because a more fine-stranded, …

Soil Chemistry And Nutrition Of North American Red Spruce-Fir Stands: Evidence For Recent Change, J. D. Joslin, J. M. Kelly, H. Van Miegroet
Watershed Sciences Faculty Publications
One set of hypotheses offered to explain the decline of red spruce (Picea rubens Sarg.) in eastern North America focuses on the effect of acidic deposition on soil chemistry changes that may affect nutrient availability and root function. Long-term soils data suggest that soil acidification has occurred in some spruce stands over the past 50 yr, with plant uptake and cation leaching both contributing to the loss of cations. Studies of tree ring chemistry also have indicated changes in Ca/Al and Mg/Al ratios in red spruce wood, suggesting increases in the ionic strength of soil solution. Irrigation studies using strong …

This review is concerned with the functional and structural changes occurring in the aging heart. These changes were investigated in an integrated fashion in Fischer 344 rats at 4, 12, 20, and 29 months after birth. Mean arterial pressure, left ventricular systolic and end-diastolic pressures, as well as stroke volume remained substantially constant up to 20 months. At 29 months, however, end-diastolic pressure was significantly increased, and dP/dt and stroke volume were depressed. Focal areas of interstitial and replacement fibrosis were markedly increased at 20 and 29 months, mostly in the subendocardial region of the ventricular wall. Also the aggregate …

Microphotometric Quantification Of Crystal Growth In Gels For The Study Of Calcium Oxalate Urolithiasis, Wolfgang Achilles

This paper gives a review on an efficient microtechnique (Gel Crystallization Method; GCM) which quantifies crystal growth in gel matrices, and its application to research into urinary stone formation. Relative crystal growth rates (Vcr) of calcium oxalate hydrates (CaOx) were determined by simultaneous multiple measurements in 96-well microplates using computer controlled microscopic photometry. Efficiency: 120 kinetic measurements/h; imprecision: < 2% at standard conditions. The method was applied to study the effects of diverse small-ionic and macromolecular constituents of human urine on the crystal growth rate of CaOx. The 'inhibitory activity' of urine could be evaluated by comparison of the Vcr measured in native and corresponding artificial samples. Macromolecules, at physiological concentration, played only a minor role as inhibitors in the model under regard. The GCM was employed to study the in-vivo effects of different therapeutic measures on CaOx growth in urine of normal volunteers. While the application of alkali citrate resulted in a > 70% decline of Vcr, no beneficial effects could be found with three magnesium compounds, and a dietary fiber preparation. In a study including 20 male recurrent CaOx stone formers and 29 well-matched controls, among all parameters under investigation, the Vcr showed the …

Strategies for managing wildlife damage may be divided into 3 broad categories: direct manipulation of populations, manipulation of behavioral or ecological traits of pest species, and manipulation of environmental features. For each of these categories, I review the importance of ecological considerations in determining the effectiveness of management strategies. Proper incorporation of ecological information is important to the success of management strategies in all 3 categories. I predict that future demands will increase for ecologically-based strategies that require minimal intervention, and for integration of management strategies that simultaneously address problems posed by both vertebrate and invertebrate pests. Data are presented on scanning electron microscopy (SEM) on small intestinal biopsies of children with chronic diarrhea. In particular, there were 230 patients aged 3 months to 13 years with the following diagnoses: chronic nonspecific diarrhea, cow's milk protein intolerance, soy protein intolerance, giardiasis, cystic fibrosis, gluten-sensitive enteropathy, isolated lactase deficiency, isolated sucrase-isomaltase lactase deficiency, microvillus inclusion disease, rotavirus ententis, protracted diarrhea of infancy, chylomicron retention disease, visceral myopathy and villous asthenia.
Examination of biopsied intestinal mucosa by SEM has yielded important new information and insights on structural pathology and ultrastructural topography. Many of the observed changes helped to better …

The aims of this review are: 1. to provide a bibliography of the publications that have used the corrosion casting technique; 2. to describe the advantages and limitations of the methodology; 3. to illustrate possible applications in the field of medicine, and 4. to highlight the significance of this method in the teaching of medical students. Thus, this paper is primarily focused on the scanning electron microscopical examination of vascular corrosion casts.
The unsurpassed three-dimensionality of the corrosion casting technique compared to any other means stands out in particular. This can be especially useful when complex vascular-anatomical relationships are present. …

The major objective of this dissertation was to describe some of the factors that may be important in the establishment of vesicular-arbuscular (VA) mycorrhizal fungi following a disturbance. The specific focus of the dissertation is primarily on the interaction of the western harvester ant and VA mycorrhizal fungi and how this interaction affects the dispersion and establishment of these mutualistic endophytes. Methods were developed to test the use of polyclonal antibodies on field and greenhouse samples. This approach proved viable and allowed me to differentiate between both the extramatrical hyphae and spores of introduced and native VA mycorrhizal fungi. I …

Data presented in this thesis were collected on a population of Yellow-eyed Juncos (Junco phaeonotus) in the Chiricahua Mountains of southeastern Arizona, from 1985 to 1990. Severe drought and an associated reduction in the food supply occurred at the study site in 1989 and 1990. This provided the opportunity to document the effects of stressful environmental conditions on the breeding biology of the juncos. My field work covered the 1989 season, and for comparative purposes these thesis data are combined with data previously collected by Dr. Kimberly A. Sullivan and Dr. Wesley W. Weathers. Use of living organisms as a pest control measure is the essence of biocontrol. This study involves evaluation of the potential biocontrol agents for two organisms: a weedy plant species and a fungal plant pathogen. The weed is dyer's woad (Isatis tinctoria) which is rapidly encroaching on all lands in the Intermountain West. The fungal plant pathogen is Phoma betae, a seed- and soilborne pathogen of sugar beets. Puccinia thlaspeos, a native rust pathogen, causes systemic infection on dyer's woad, resulting in severe deformations of stems and leaves and prevention of seed production. It occurs in …

This study was designed to investigate the hypothalamic-pituitary-testicular axis of the primary infertile dark male mink. The specific objectives were (1) to describe the morphology and localization of the neurons containing gonadotropin-releasing hormone (GnRH) and its precursor (proGnRH) within the preoptic-basal hypothalamus (POA-BH) of the sexually mature dark male mink, (2) to ascertain any differences in the number or the distribution of GnRH- and proGnRH-containing neurons between the fertile and infertile dark male mink, (3) to determine if GnRH and proGnRH are produced in insufficient amounts by the hypothalami of primary infertile dark male mink, and (4) to describe the …
