Monoclonal antibodies used in serological procedures have the potential to recognize and identify one single antigenic determinant on a virus that is not shared by closely related viruses. This procedure would have the advantage of reducing non-specific background and would allow differentiation between a mixture of viruses. In this investigation, a monoclonal antibody to reovirus type 1 was produced and compared with polyclonal antibody for immunoassay detection of virus. The tests compared were the enzyme-linked immunosorbent assay (ELISA), the enzyme-linked fluorescent assay (ELFA), and the immunofluorescent cell count assay. The present light and electron microscope study revealed that the in vivo pattern of development of A. aggregata resembles that of the in vitro development of A. apis, the only other member of the Ascospheraceae to be studied in detail by Spiltoir in 1955. As well as documenting the similarities between the two species, this study also revealed several distinct differences between the two species and answered several developmental questions left unanswered by Spiltoir's study. Nongerminated and germinating ascospores were seen within the gut lumen of the feeding larvae. After germination minimal hyphal development occurred within the gut lumen. The …

Murine Rotavirus Infection: Characterization Of The Disease And Influence Of Host Malnutrition And Dietary Protein, Roger Lee Noble

A possible correlation between malnutrition and the increased incidence and severity of rotavirus-associated gastroenteritis in human infants was investigated using a mouse model originally described by others, but which was further defined and characterized as . a part of these studies. Two- or three-day-old specific-pathogen free Swiss Webster mice were orally infected with a 10% homogenate of intestines from mice infected with murine rotavirus. In the resulting disease, gross signs (diarrhea, alteration in intestinal appearance, failure to gain weight) and histologic changes typical of rotaviral gastroenteritis were manifest. AF1312/TS, an antifertility agent synthesized by Silvestrini and associates in Italy, has the potential to be developed as a contraceptive in human males. This chemical's action is extremely specific, affecting initially, only the elements of the seminiferous epithelium. The drug's action is reversible, and dose, and species specific. This study was undertaken to observe the general morphplogical changes in the testis and epididymis of the mouse due to oral dosing with AF1312/TS. A study was conducted to examine the effects of neutral and sex-specific terminology on sex stereotyping in regard to two primary questions: 1) whether or not the use of sex-neutral terminology alters subjects ' associations to particular words, and 2) whether, in the absence of gender identification, subjects make traditional sex-role assumptions about neutral terms. A third questions examined potential differences in male and female subjects' responses to neutral and sex-specific terminology.

Using a semantic differential technique, 40 male and 40 female volunteer subjects described a number of occupations and roles identified by sex-specific or sex-unspecified labels. Descriptions of those …

This study was designed to determine the contributions of mothers and fathers to infant social development. Nine 10-month-olds, 12 fourteen-month-olds, and 12 eighteen-month-olds were observed with their mothers and fathers in a laboratory situation. Parent-infant interactions were videotaped during three different episodes: Mother-infant dyad, father-infant dyad, and mother-father-infant triad. Findings revealed different interaction patterns as a result of the ages of infants and the interaction situation. Older infants and their parents engaged in more verbal behavior (responsive talk, social speech, and story reading) than younger infants and their parents. The primary purpose of this research was to gain insight into what childhood was like in turn-of-the-century Mormon polygamous families. This purpose was executed through two main avenues: basic empirical data and descriptive accounts. This type of research was crucial inasmuch as previous research and commentaries dealt with adult relations but little was known about children in this complex Mormon family structure.

From April of 1981 to June of 1982 a Clean Lakes Program Phase 1 Diagnostic Feasibility Study was conducted on Bear Lake, a 282 km2 (109m2) fresh-water lake located in southeastern Idaho and northeastern Utah. A portion of this Clean Lakes Program required an itemized inventory of all nonpoint sources affecting the lake's water quality.

A 16 month lake tributary monitoring program, which included the spring runoff periods of 1981 and 1982, provided data from which nutrient loading values (kg/day and kg/yr) were calculated. Analysis of these data, along with land use percentages within the Bear Lake …

Several aspects of kokanee (Onchorhynchus nerka) ecology were studied in the fluctuating, 80ha Porcupine Reservoir, Utah in order to determine optimum management strategies. In 1981, escapement was enumerated and estimates of egg deposition were made. An estimate of the resulting fry recruitment was also attempted and was successful up to the arrival of spring runoff. In 1982, escapement was enumerated and estimates of egg deposition were made, with subsequent estimates of egg deposition and percent survival to the advanced eyed stage. Age and growth rates were determined and survival rates estimated.

A review of the literature indicated a need for nutrition education materials to be current with prevailing scientific knowledge, using advanced instructional technology. Individuals need help in applying nutrition principles to their dietary practices.

A nutrition education program utilizing the nutrient density concept and the dietary guidelines for use on videodisc with microcomputer interface was developed and evaluated. The evaluation of the program did not prove to be effective statistically but learners did show an interest in the program and had positive feedback in regard to the information presented and the technology used.

Three separate experiments were conducted to investigate the effects of heat treatment on meat heme and nonheme iron bioavailability in anemic rats. Male weanling Sprague-Drawley rats were first made anemic, then subsequently fed with experimental diets. In the first experiment, dietary iron sources were raw or autoclaved meat mixed with ferrous sulfate to give the ratios of iron from meat and ferrous sulfate equal to 100:0, 75:25, 50:50, 25:75, and 0:100 respectively. The effect of iron absorption on subsequent hemoglobin regeneration was investigated by repletion and subsequent analysis of an anemic weanling rat model. Primary assay procedure in these studies was the Regeneration Efficiency method.

As repletion or curative assay techniques form the central body of investigative research in iron utilization, the first subject of inquiry was the effect of repletion interval and degree of depletion upon the animals as monitored by physical indicators such as blood volume, growth and hematinic response. Pantothenic acid nutritional status was evaluated in a cohort of twenty-nine Caucasian pregnant women, aged 20-35, during their third trimester of pregnancy, at two-weeks postpartum and at three months postpartum. Seventeen non-pregnant, non-lactating women who had similar demographic backgrounds as the pregnant women were selected as a control group and followed the same experimental schedule as the pregnant women. A fasting blood sample, two days 24-h urine specimens and diet record were obtained for each of two days from each subject at each period.

Process cheese models were prepared by blending acid or rennet casein, milk fat, sodium chloride, 2.5% emulsifying salt and water and heating to 80 C. Acid casein cheese models were subjected to sodium hydroxide conditioning at 65 C in the cooker. Model process cheeses were acidified with lactic acid and treated by addition of undenatured and heat-denatured whey protein, four different emulsifying salts and sodium oxalate.

Meltability and toughness of the model cheese increased to a maximum with increased sodium hydroxide conditioning of acid casein to pH 7.20. These same properties decreased with addition of undenatured and heat-denatured whey protein …

Gardner's syndrome is an autosomal dominant disease presenting multiple colonic polyps with a predisposition for malignant change. In addition to colonic polyp formation by early adolescence, extracolonic lesions appear often prior to polyp formation. One theoretical mechanism for the origin of polyps and malignancies in Gardner's syndrome is a genetic defect in the natural killer cell activity of patients with this disease. Natural killer cells are a subpopulation of lymphocytes that spontaneously lyse tumor cells and virally transformed cells. A study was undertaken to determine the natural killer activity of patients with Gardner's syndrome.

The purpose of this research was to develop and test a group of predictors that could be used by fundraisers to determine potential corporate support for environmental and recreational programs. Local surveys were conducted to determine environmental and recreational needs for Cache Valley and twenty interviews were held with local corporate managers. The information gathered in these interviews and the literature review provided a list of approximately 20 variables which were narrowed down to 10 to predict both the potential for giving and the level of giving.

The purpose of this study was to evaluate the thesis that matching locus of control with type of relaxation training enhances physiological self-control. This was accomplished by comparing the effectiveness of a somatic and a cognitive relaxation method for internal and external locus of control subjects. It was hypothesized that the two techniques vary in degree of cue salience, and that the attentional strategy of internals would interact with the cognitive approach while externals would respond better to the somatic approach.

A series of investigations were undertaken to determine whether physical or emotional stress, catecholamines or naloxone (B-endorphon blocker) would increase serum total cholesterol and LDL and HDL levels. Physical stress given daily over a period of time caused a steady increase in serum total cholesterol and LDL without a significantly altering high density lipoproteins (HDL) or serum triglycerides. Daily injections of epinerphrine in oil caused an increase in both serum total cholesterol and LDL levels while daily injections of norepinephrine did not. Mule deer (Odocoileus hemionus) reproduction and survival on the LaSal Mountains, Utah were studied from 1978-81. Reproductive rates were high, suggesting that summer and fall forage quality was adequate and was not responsible for low recruitment observed in recent years. The combined fetal rate, determined from laparotomies and carcasses, was 1.72 for all females older than 1 year (N = 114). Fetal rates were 1.15 for yearlings (N = 20), 1.80 for prime females (2-7 years, N = 78), and 1.75 for old females (8+ years, N = 16). No evidence of breeding was found among fawns (N …

Studies have been carried out towards the synthesis of bicyclic octadidepsipeptide lanthionine 4, an analog of des-N-tetramethyl triostin A. Starting with a symmetrical lanthionine unit 5 (R1, R3 = Me; R2, R4 = t-butyloxycarbonyl), synthesized from the corresponding cystine by contraction with hexaethylphosphorus triamide, linear octadidepsipeptides 29 and 32 were synthesized.

Attempted cyclizations of 29 and 32 to the bicyclic octapeptide were unsuccessful.

Parts of the Overthrust Belt of western Wyoming and adjoining areas in Utah and Idaho contain geologic formations with significant accumulations of oil and natural gas. Some of these formations, though, yield gas that is contaminated with toxic hydrogen sulfide. As a consequence, the development of these so-called sour-gas reservoirs requires special safety procedures and technologies in order to prevent accidental releases of gas to the atmosphere that could cause adverse occupational and public health effects. As a result of an extended period of abnormally high precipitation, prolonged snow melt, and subsequent high water tales, Utahns, in 1983 suffered from landslides, debris flows, flooding, and a dam failure. On April 30, 1983, the President determined the Spanish Fork slide-dam and Thistle flood covering Utah, Carbon, and Emery Counties was of sufficient seventy and magnitude to issue a Major Disaster Declaration under Public Law 93-288. The incident period was not closed until July 1, 1983. By the time all flooding had ceased, nineteen additional counties (over 75% of Utah) had been included in the Declaration.

Symptoms induced by phytotoxic gases within conifer needles can be differentiated histologically from those caused by other abiotic agents including winter drying, drought, and salt. However, it is not possible to differentiate among symptoms caused by hydrogen fluoride, sulfur dioxide, ethyl mercaptan, and hydrogen sulfide. Phytotoxic gases cause hypertrophy and hyperplasia of vascular parenchyma, endodermis collapse, and intense vascular staining. The other abiotic agents induce mesophyll collapse with little or no observable effects on vascular tissues. Histological analyses should be useful in diagnosis of air pollution-induced injury and damage in coniferous forests.
