Extracellular Matrix Vesicles In Endochondral Bone Development And In Healing After Injury, J. Sela, Z. Schwartz, L. D. Swain, D. Amir, B. D. Boyan

Matrix vesicles are fundamental components of initial mineral formation in endochondral and primary ossification. Regulation of their structure and function is multi-faceted and involves genomic and nongenomic pathways. Their function in mineralization may include matrix maturation and conveying cellular signals to the extracellular matrix as well. Intravesicular hydroxyapatite crystal formation is associated with primary mineralization; subsequent crystal formation, particularly at later stages, appears to be mediated by other mechanisms.

The purpose of this study was to examine the ultrastructural relationships of cholesterol crystals and apatite deposits in human atherosclerotic lesions. Segments of human aortic atherosclerotic lesions were obtained at autopsy , fixed in glutaraldehyde and dehydrated without using any organic solvents. The aortic segments were coated with carbon and subjected to various scanning electron microscope analyses. These included secondary electron imaging, back scattering of primary electrons, energy dispersive X-ray analysis of selected spots followed by area mapping of calcium and phosphorus , and cathodoluminescence.

This report documents the Tiger Team Assessment of the Naval Petroleum and Oil Shale Reserves in Colordao, Utah, and Wyoming (NPOSR-CUW). NPOSR-CUW consists of Naval Petroleum Reserve Number 3 (NPR-3) located near Casper, Wyoming; Naval Oil Shale Reserve Number 1 (NOSR-1) and Naval Oil Shale Reserve Number 3 (NOSR-3) located near Rifle, Colorado; and Naval Oil Shale Reserve Number 2 (NOSR-2) located near Vernal, Utah, which was not examined as part of this assessment.

Effects Of Seeder Design And Seed Placement On Seedling Size And Cull Rates At Western Forest Nurseries, United States Department Of Agriculture, Forest Service

Nursery plots sown with the Summit Precision Seeder and the Oyjord Seeder were compared with plots oversown and hand thinned to desired spacings. We studied 26 seedlots of nine conifer species at five Forest Service, U.S. Department of Agriculture, nurseries. Results varied with nursery, species, and seed characteristics. Thinned plots produced less clumping and fewer gaps between seedlings, but showed little or no difference in seedling size, nor in the number of acceptable seedlings.

This Final Environmental Impact Statement (FEIS) documents the analysis conducted for the Stanley Basin Cattle and Horse Allotment Management Plan. The allotment management plan involves National Forest System lands within the Sawtooth and Challis National Forests. The analysis of the current condition of the Stanley Basin Allotment has found that the existing grazing system does not comply with the direction, standards and guidelines of the Sawtooth and Challis Forest Land and Resource Management Plans (FLRMP). The proposed action of this Final Environmental Impact Statement is to bring management of the allotment into compliance with the FLRMPs. The FEIS describes five …

Three-dimensional structures have recently been determined by electron crystallography at a resolution high enough to determine atomic arrangements in both protein and mineral specimens. The different nature of these two types of specimens produces some very significant differences in the way data is obtained and processed, although the principles are the same. The sensitivity of proteins to damage by the electron beam limits the signal-to-noise ratio in the image and the resolution to which data can be extracted from the image. Cryo-electron microscopy of single biological particles poses new challenges to digital image processing due to the low signal-to-noise ratio of the data. New tools have been devised to deal with important aspects of 3-D reconstruction following the random-conical data collection scheme: (a) a new shift-invariant function has been derived, which promises to facilitate alignment and classification of single particle projections; (b) a new method of orientation search is proposed, which makes it possible to relate random-conical data sets to one another prior to reconstruction; and (c) the foundation is laid for a 3-D variance estimation which utilizes the oversampling of …

The three-dimensional visualization of the 400 micron thick, transparent, in situ cornea is described to demonstrate the use of confocal light microscopy for noninvasive imaging of living cells and thick tissues in their normal, vital conditions. Specimen preparation and physiological stability, as well as light attenuation corrections are critical to data acquisition. The technique to provide mechanical stability of the specimen during the duration of the image acquisition is explained. A laser scanning confocal light microscope (LSCM) was used to obtain optical serial sections from rabbit eyes that were freshly removed and placed in a physiological Ringer's solution. This study …

The PIC system, an integrated package of Fortran programs and subroutines designed to run on the Digital Equipment Corporation VAX family of computers, has been developed for analysis of electron micrographs with emphasis on the particular requirements for structural analysis of biological macromolecules. The substantially improved VAX version of PIC reported here has been developed from an earlier PDP-11 version which was, in turn, developed from a set of IBM 370 programs called MDPP. The perturbation theory of intermolecular forces in conjunction with the supermolecular Møller–Plesset perturbation theory is applied to the analysis of the potential‐energy surfaces of Kr–H2O and Kr–NH3 complexes. The valleylike minimum region on the potential‐energy surface of Kr–H2O ranges from the coplanar geometry with the C2 axis of H2O nearly perpendicular to the O–Kr axis (T structure) to the H‐bond structure in which Kr faces the H atom of H2O. Macromolecular assemblies of proteins and hydrocolloids in low-fat dairy products contribute to the structure and texture of these foods. Immunolocalization techniques were used to identify B-lactoglobulin, casein, bovine whey proteins, and egg albumin in low-fat frozen desserts, reduced fat process cheese, and salad dressings. SimpJesse18 I 00 protein particles were examined and characterized by these methods and compared to naturally occurring protein structures in the low-fat foods. Skim milk of approximately 3% total protein was ultrafiltered to 16.2% protein and 4.1 % lactose, and then diafiltered to reduce lactose to 2.1% and 0.9%. Total protein con ten t was maintained at approximately 17%. A ponion of the skim milk was condensed in a rising film evaporator to 14.7% protein and 15.7% lactose . All concentrates were spray dried at 120 to 125 °C inlet air temperature and 75 to 80 C outlet temperature using a rotary atomizer in a pilot plant spray dryer . One set of hypotheses offered to explain the decline of red spruce (Picea rubens Sarg.) in eastern North America focuses on the effect of acidic deposition on soil chemistry changes that may affect nutrient availability and root function. Long-term soils data suggest that soil acidification has occurred in some spruce stands over the past 50 yr, with plant uptake and cation leaching both contributing to the loss of cations. Studies of tree ring chemistry also have indicated changes in Ca/Al and Mg/Al ratios in red spruce wood, suggesting increases in the ionic strength of soil solution. Irrigation studies using strong …

In 1990, Utah State University (USU) established a U.S. Department of Agriculture, Animal and Plant Health Inspection Service, Animal Damage Control (ADC)-funded center dedicated to research, education, and extension in wildlife damage management. USU' s Program in Wildlife Damage Management is designed to increase appreciation for wildlife damage management among wildlife professionals, provide superior training for future practitioners of wildlife damage management through education and innovative research, and create better public understanding of the role of wildlife damage management in today's society.

Cambial electrical resistance (CER) was assessed over a year-long period, using the Shigometer, for 109 sweetgum trees (Liquidambarstyraciflua) planted along streets in Seattle, Washington. CER decreased through the spring, reached a minimum in summer, increased through the fall and was maximal in mid-winter. The seasonal maximum was 200-600% greater than the summer minimum. CER measurements could not be related to growth parameters such as seasonal shoot increment and diameter growth.

Generally, the embedding technique for optimizing groundwater management has been limited to small scale or steady-state groundwater flow management cases. The embedding approach causes sparse, highly structured constraint matrices which have been sometimes difficult to solve. Embedding applications have been limited by lack of: optimization algorithms able to easily address such matrices for large scale problems, and efficient methods for addressing common nonlinearities and converging to a solution. Computer software, USUEM, addresses these issues by: creation of alternative linear, nonlinear, combined and partitioned formulations of the same problem, implemention of an efficient cycling procedure, and good selection of optimization algorithm …

A management model is developed for maximizing crop yield while avoiding unacceptable pesticide leaching. Utilized constraint equations: maintain a soil moisture volume balance, describe downward pesticide transport, and limit the amount of pesticide reaching groundwater. The reported optimization model is the first which includes unsaturated zone pesticide transport. It is designed to help prevent nonpoint-source contamination of shallow groundwater aquifers. The model computes optimal irrigation amounts for given soil, crop, chemical, and climate data and irrigation frequencies. The model is tested for different irrigation scenarios. The modeling approach is promising as a tool to aid developing environmentally sound agricultural production …

Determining the biological effects of low doses of radiation with high linear energy transfer (LET) is complicated by the stochastic nature of charged-particle interactions. Populations of cells exposed to very low radiation doses contain a few cells which have been hit by a charged particle, while the majority of the cells receive no radiation damage. At somewhat higher doses, a few cells receive two or more events. Because the effects of damage produced by separate events can interact in the cell, we have had to make assumptions about the nature of these interactions in order to interpret the results of …

The microvasculature of human tongue papillae originating from 9 males and 6 females, aged 0.5 to 2 years was studied by scanning electron microscopy (SEM) of vascular corrosion casts and by light microscopy (LM) of India ink injected specimens. All papillae showed a microvasculature characterized by primary, secondary and tertiary capillary loops. In the filiform papillae the loops were generally arranged in a corolla-like pattern with the tertiary loops demonstrating a hair-pin shape. The fungiform papillae showed basically a similar architectural pattern although the loops were somewhat more compact and complex in structure. A small, shallow depression of the tertiary …
