A minitransposon mutant of Salmonella enterica serovar Typhimurium SR-11, SR-11 Fad-, is unable to utilize gluconeogenic substrates as carbon sources and is avirulent and immunogenic when administered perorally to BALB/c mice (M. J. Utley et al., FEMS Microbiol. Lett., 163:129–134, 1998). Here, evidence is presented that the mutation in SR-11 Fad- that renders the strain avirulent is in the cra gene, which encodes the Cra protein, a regulator of central carbon metabolism.

Historically, comparison groups of children from low socioeconomic status (SES) have done less well on intelligence tests than their more well-to-do counterparts. This study was undertaken to explore the contribution of verbal ability to general intelligence and to consider the possibility of developmental change in the relative contributions of the verbal and performance scales. The analyses used verbal, performance, and full scale scores of a subgroup of the standardization sample of the Wechsler Intelligence Scale for Children - Third Edition (WISC-III). This study examined the relationship between at-birth risk factors and three year child developmental outcome. The purpose of this study was to design a multifactor risk screening system to identify those children who are at risk for developmental delay and behavior problems in Rhode Island. The sample was drawn from three separate datasets: the Rhode Island Universal Newborn Screening (Level One) database (N=27,282), the Early Intervention (El) database (N=l ,793), and the Maternal Lifestyle Study Providence/Teen samples (N= 311). Type 2 diabetes is a serious metabolic disorder occurring in over 90% of the people in the United States diagnosed with diabetes. The standard treatment for type 2 diabetes requires individuals to self-manage a diet, exercise, and self-monitoring of blood glucose (SMBG) to control blood glucose levels and reduce the onset of serious complications. Although there is a plethora of literature addressing the difficulty individuals have to maintain these three behaviors, few studies exist describing how these behaviors are sustained. Warfarin is widely used oral anticoagulant and its pharrnacokinetic (PK) and pharrnacodynamic (PD) properties have been extensively studied. It has a narrow therapeutic index and displays poor quality of treatment due to its complex pharmacology and wide inter and intraindividual variability in the dose-response relationship.

This study developed an integrated PK-PD model using STELLA® to describe the dose concentration- effect relationship for warfarin. This model used previously reported\ population PK and PD models and parameter values to generate dose-response data. Blood Glucose Estimation Accuracy In Children With Diabetes: An Investigation Of Repeated Practice With Latent Growth Curve Modeling, Julie Ann Wagner

Blood glucose (BG) estimation is an important part of maintaining euglycemia in individuals with diabetes. Interventions to increase accuracy were designed without taking into account the natural effects of practice. The current study investigated the effects of naturalistic practice on BG estimation accuracy. Latent growth curve modeling (LGCM) was used to analyze data from 43 children with insulin dependent diabetes. LGCM consists of two stages. In the first stage, a growth curve was fit to the repeated measures of each individual in the sample. Results indicated that the true growth trajectory was curvilinear, and best represented by a quadratic function. …

The ability of Escherichia coli to survive stress during growth in different environments is, in large part, dependent onrpoS and the genes that comprise the rpoSregulon. E. coli BJ4 and an isogenic BJ4 rpoSmutant were used to examine the influence of the rpoS gene on E. coli colonization of the streptomycin-treated mouse large intestine. Colonization experiments in which the wild-typeE. coli BJ4 and its rpoS mutant were fed individually as well as simultaneously to mice suggested that E. coli BJ4 does not face prolonged periods of nutrient starvation in the mouse large intestine and that the …

Green-Water Rearing And Delayed Weaning Improve Growth And Survival Of Summer Flounder, David A. Bengtson, Lindsay Lydon, Jeffrey D. Ainley

The advent of an aquaculture industry for summer flounder Paralichthys dentatus requires that optimal methods be identified for hatchery production. Two experiments were conducted to test strategies for larval rearing and for weaning newly metamorphosed juveniles from live to artificial diets. Rearing of larvae in ‘‘green water’’ (with algae added) resulted in better survival (76.1 6 6.5%) from days 5–42 after hatching than did rearing in ‘‘clear water’’ (no algae added; 27.8 6 13.6%), although no differences in growth were apparent. A membrane concentration system consisting of tubular polysulphone ultrafiltration (UF) and polyamide reverse osmosis (RO) was evaluated for concentrating key water soluble flavor compounds from lobster extracts. Major flavor‐giving compounds in the extract were glutamic acid, glycine, arginine, uridine 5′‐monophosphate (UMP), succninic acid and glucose. Factors affecting performance of the UF/RO systems, such as flow rate, feed solid level, temperature and pressure, on permeate flux and solids rejection were measured. The optimum UF conditions were 1.5% feed solid level, 15 L/min feed flow rate, 50°C feed temperature and 1 MPa log mean transmembrane pressure. The RO system retained all dissolved …

The effect of gastrointestinal mucus on protease activity in Vibrio anguillarum was investigated. Protease activity was measured by using an azocasein hydrolysis assay. Cells grown to stationary phase in mucus (200 mg of mucus protein/ml) exhibited ninefold-greater protease activity than cells grown in Luria-Bertani broth plus 2% NaCl (LB20). Protease induction was examined with cells grown in LB20 and resuspended in mucus, LB20, nine-salts solution (NSS [a carbon-, nitrogen-, and phosphorus-free salt solution]), or marine minimal medium (3M) (~109 CFU/ml). Induction of protease activity occurred 60 to 90 min after addition of mucus and was ≥ 70-fold greater than …

The growth physiology of Escherichia coli during colonization of the intestinal tract was studied with four animal models: the streptomycin-treated mouse carrying a reduced microflora, the monoassociated mouse with no other microflora than the introduced strain, the conventionalized streptomycin-treated mouse, and the conventionalized monoassociated mouse harboring a full microflora. A 23S rRNA fluorescent oligonucleotide probe was used for hybridization to whole E. coli cells fixed directly after being taken from the animals, and the respective growth rates of E. coli BJ4 in the four animal models were estimated by correlating the cellular concentrations of ribosomes with the growth rate of …

Exposure of batch cultures of Tetraselmis chuii to three levels (0.020, 0.126 and 0.217 m W/cm2) of artificial UV-B caused a depression of exponential growth corresponding to UV-B dose. Three UV-B experiments were conducted. Cultures were maintained at 25 °C on a 18L:6D photoperiod for the first two experiments and at 12L: 12D for the last experiment. Growth was measured daily by measuring optical density at a standard wavelength of 750 nm. This was compared periodically with hemacytometer cell counts. Cell morphology (projected cell area, length, width, perimeter, and roundness) was determined by microscopic image analysis, and showed …
