Oviposition, Development, And Feeding Of Scymnus (Neopullus) Sinuanodulus (Coleoptera: Coccinellidae): A Predator Of Adelges Tsugae (Homoptera: Adelgidae), Wenhua Lu, Michael E. Montgomery

The hemlock woolly adelgid, Adelges tsugae Annand, is a major threat to hemlocks in the eastern United States. As part of efforts to control this pest, Scymnus (Neopullus) sinuanodulus Yu et Yao, a potential predator, was collected from hemlocks in Yunnan, China. Three shipments were imported during 1996 and 1997 to a quarantine laboratory to study the beetle’s biology. Beetles began oviposition immediately after transferal from 5°C in the spring, ceased oviposition by June, and laid the same number of eggs, whether egg laying began in February or in April. Two-year-old beetles laid as many eggs per year …

β-Glucuronidase Assay For The Detection Of Escherichia Coli In Foods, Seref Tagi

Enzyme assays were developed for the measurement of β-D-glucuronide (GUS) as a rapid detection method for E. coli in food. A fluorescent assay utilizing 4-methylumbelliferyl-β-D-glucuronide as substrate was optimized and tested with growth cultures of E. coli. A relationship between E. coli and the level of GUS activity was established. A chemiluminescent assay using a 1,2-dioxetene derivative as substrate was compared to the fluorescent assay, and was found to be 10x more sensitive. Anti-E. coli GUS antibodies were covalently immobilized on magnetic beads and used in a 30 min immunocapture method. GUS was recovered from E. coli grown at 37°C …

The purpose of this study was to advance understanding of the self-change process in recovery from anorexia nervosa (AN) and bulimia nervosa (BN) through: (a) development of a staging instrument for recovery from BN; (b) development of TTM-based measures, including decisional balance, self-efficacy, and processes of change; (c) invariance testing of these measures across AN and BN subsamples; and (d) characteristic description of individuals at each stage of change. In connection with the CDC National Conference on Pfiesteria, a multidisciplinary panel evaluated Pfiesteria-related research. The panel set out what was known and what was not known about adverse effects of the organism on estuarine ecology, fish, and human health; assessed the methods used in Pfiesteria research; and offered suggestions to address data gaps. The panel's expertise covered dinoflagellate ecology; fish pathology and toxicology; laboratory measurement of toxins, epidemiology, and neurology. The panel evaluated peer-reviewed and non-peer-reviewed literature available through June 2000 in a systematic conceptual framework that moved from the source of exposure, through exposure research and dose, …

We examined the effects of food concentration and temperature on nucleic acids and protein content of Calanus finmarchicus in order to evaluate the use of RNA as a growth rate index for this species. We measured RNA, DNA, and protein content of copepods reared from egg to adult stage in 5 combinations of food and temperature conditions (25 to 500 µg C l-1, 4 to 12°C). At 8°C, DNA, RNA and protein content and RNA:DNA differed among food treatments during Stages N6 through to adult female. Staphylococcus epidermidis is a major cause of infections associated with indwelling medical devices. Biofilm production is an important virulence attribute in the pathogenesis of device-related infections. Therefore, elimination of these biofilms is an ideal treatment. Salicylate (5 mM) combined with 1 μg of vancomycin per ml inhibited biofilm formation by S. epidermidis (RP62A) by ≥99.9%. When biofilm-coated polystyrene beads were exposed to 5 mM sodium salicylate and 4 μg of vancomycin per ml (one-half the minimum biofilm eradication concentration), there was a >99.9% reduction in viable count.

Honey amylase was evaluated for potential to degrade food starch and cause viscosity loss. Honey was assayed for amylase activity with diastase number (DN). A viscosity assay, developed with unmodified waxy maize starch, measured rate of viscosity decrease (RVD) by honey amylase. The relationship between DN and RVD was linear (R2 = 0.98). Modified waxy maize starches showed resistance to honey amylase. Heat treatment at 85 °C reduced the amylase activity in honey, but confirmed enzyme heat resistance. Optimum pH for honey amylase was confirmed at pH 5.3-5.6. Lower pH was effective at decreasing enzyme activity in both pure starch …

This study describes the New England Otter Trawl Fishery between 1982 and 1998 and the potential economic impacts of regulations on the Northeastern Multispecies Groundfish Fishery (MGF). The study uses primary data collected by the author and secondary data obtained from the National Marine Fisheries Service (NMFS). This research required extensive econometric analysis of both value landed and cost data. The landed value is influenced by the vessel's physical characteristics, port of landing, seasonal choices of fishing gear-species combinations, days at sea and distance from shore, and the captain's skill (also referred to as the "skipper effect"). Alginate, carrageenan, soy protein concentrate (SPC), sodium tripolyphosphate (STPP), and sorbitol were chosen as potential cryoprotectants. Their effects on physicochemical and sensory properties of frozen red hake (Urophycis chuss) and Alaska pollock (Theragra chalcogramma ) mince at ˆ’20 °C for 17 weeks were investigated. The mince was mixed with 0.3% STPP, 4% sorbitol and either 0.2-0.4% alginate, 0.2-0.4% iota-carrageenan, or 4% SPC. Changes in texture, microstructure, and profiles of proteins, free amino acids and lipids of fresh and frozen mince were evaluated. Development rates, nitrogen- and carbon-specific growth rates, size, and condition were determined for the copepod Calanus finmarchicus reared at 3 temperatures (4, 8, and 12°C) at non-limiting food concentrations and 2 limiting food concentrations at 8°C in the laboratory. Development rates were equiproportional, but not isochronal. Naupliar stage durations were similar, except for non-feeding stages, which were of short duration, and the first feeding stage, which was prolonged, while copepodite stage durations increased with increasing stage of development. Under limiting food concentrations at 8°C, development rates were prolonged but similar relative patterns in stage durations were observed. Body size (length …
