Characterization Of The Heat Shock Response And Identification Of Heat Shock Protein Antigens Of Borrelia Burgdorferi, Margaret M. Carreiro, David C. Laux, David R. Nelson

The heat shock response of Borrelia burgdorferi B31 cells was characterized with regard to the heat shock proteins (Hsps) produced. Five to seven Hsps were detected by sodium dodecyl sulfate-gel electrophoresis and fluorography of proteins from cells labeled with [35S]methionine after shifts from 33°C to 37 or 40°C or from 20°C to 33, 37, or 40°C. Analysis of [35S]methionine-labeled Hsps by two-dimensional electrophoresis and autoradiography revealed 12 Hsps. Western immunoblot analysis with antisera to highly conserved Escherichia coli and Mycobacterium tuberculosis Hsps revealed a single 72-kilodalton (kDa) protein band that reacted with antibodies to E. coli …

This thesis presents an examination of the fish auction system in the Azores and three New England ports; Boston, New Bedford and Portland. On the island of Sao Miguel, the village of Rabo de Peixe was selected as a case study where a survey of local fishermen and dealers was conducted. Data obtained through interviews provided some insights into the socio-economic status of local fishermen and how they perceive infrastructure improvements, government support and changes in the auction system. Nude bathing in the coastal zone has become a challenging issue for coastal managers. This thesis focuses on the conflicts involving public uses of a beach on a national wildlife refuge. The beach is nesting habitat for the piping plover (Charadrius melodus), a species listed as threatened on the federal Endangered Species List. The beach has also been traditionally used by nude bathers. The impact of the Capital Construction Fund, a tax deferral program utilized for the construction of fishing vessels, is examined in the context of Rhode Island's fishing industry. It was believed that the Capital Construction Fund contributed to overcapitalization, was an incentive to purchase a vessel, and was mainly used by individuals targeting underutilized species. Indicators of capitalization such as number, size, and ages of vessels owned were gathered from the owners of vessels home-ported in Rhode Island during personal interviews. Sediment cores have long been used to formulate the chronology of environmental contamination. This paper will investigate the utility of sediment cores in determining the chronology of trace metal contamination in Apponaug Cove, Rhode Island. A meter-long sediment core taken from the Cove was analyzed for chromium, copper, lead, zinc and nickel as well as organic pollutants coprostanol, polyaromatic hydrocarbons, benzotriazole and clorobenzotriazole. The core represents a close to 200 year record of sediments in Apponaug Cove. The cultural/economic changes in the Watershed in addition to environmental laws composed in the State and Federal levels which may have correlated with …

The pharmacokinetics of oral fluoroquinolone antibiotics in normal volunteers have been studied extensively; however, limited patient data exist. Enoxacin steady-state pharmacokinetics and bioavailability were determined following repeated 400-mg intravenous (i.v.) and oral dosing by using compartmental and noncompartmental methods in 10 elderly (mean age, 73.8 years) men with complicated urinary tract infections. Average peak enoxacin concentrations following i.v. and oral dosing were 8.15 and 5.45 mg/liter, respectively. Mean values for major pharmacokinetic parameters (noncompartmental) were similar following i.v. and oral administration, respectively: area under the concentration-time curve from 0 to 12 h, 47.6 and 41.0 mg.h/liter; volume of distribution or …

Significance Of "Extravascular" Protein Binding For Antimicrobial Pharmacodynamics In An In Vitro Capillary Model Of Infection, Michael N. Dudley, Blaser, Deborah Gilbert, Stephen H. Zinner

The effect of protein binding in an "extravascular" space on antimicrobial pharmacodynamics was studied in an in vitro capillary model of infection. Simulated 500-mg oral doses of dicloxacillin (~ 96% bound) or cephalexin (< 5% bound) were administered every 6 h for four doses. A 10-fold-higher dose of dicloxacillin was also studied to determine the effect of drug concentration on the reduction of bacterial killing in the presence of protein. Staphylococcus aureus ATCC 25923 was inoculated into peripheral chambers filled with either Mueller-Hinton broth or Mueller-Hinton broth plus 25% human serum. Serial samples for bacterial counts were collected over 24 h. The presence of serum in the chambers significantly reduced bacterial killing by dicloxacillin but not by cephalexin during the first 6 h (two-way analysis of variance, F = 6.04, P < 0.05) but not at 24 h. Reduction of dicloxacillin activity in serum-containing chambers persisted with the higher dose. These data suggest that despite attaining higher total drug concentrations in protein-containing extravascular spaces with highly bound drugs, protein binding reduces bactericidal activity during the early stages of treatment in this model.

Stable oxygen and carbon isotope ratios from modern African freshwater molluscs and water samples were examined to ascertain the reliability of such information as a proxy in the study of paleoclimates and paleoenvironments.

In this study, the isotope ratios of the freshwater molluscan shell samples were determined from both the apertures and at interval of accretionary growth of individual shells collected at monthly intervals, from four sites. These ratios were examined to ascertain the extent to which they correlate with temperature, rainfall, water isotope ratios and other ambient conditions.

Hopelessness was explored from a developmental perspective. The investigation focused primarily on the relative importance of structural vs. functional variables in determining levels of hopelessness in children and adolescents. The two secondary aims of this study were to further clarify the concept of hopelessness, and to assess the construct validity of the Hopelessness Scale for Children (HSC: Kazdin et al., 1983).

Sixty-four children and adolescents scoring either "high" or "low" on the HSC were interviewed. Thirty-two of the subjects were children, 6-10 years of age. One half of these subjects were clinically involved. The adolescent sample also consisted of thirty-two …

A multitrait-multimethod paradigm was used to assess the stability, discriminant validity, and convergent validity of two tasks of executive functioning within a sample of 96 normal preschoolers. Memory served as the discriminant construct against which the executive-function construct was compared, and each construct was measured verbally and nonverbally. A matrix of cross-validated, Pearson product-moment correlation coefficients was computed to evaluate the psychometric properties of the executive-functioning tasks. Results did not support the construct validity of the selected tasks, as measures of executive functioning in this preschool population. The purpose of this study was to examine the effects of both field of study and gender on locus of control orientation among graduate students at the University of Rhode Island at Kingston.

This study solicited participation from graduate students enrolled at the University of Rhode Island, spring semester, 1990 excluding those enrolled in the Graduate School of Oceanography, School of Business and in courses offered by the Director of the College of Continuing Education (CCE) Special Programs Division were excluded from the study. Lexical acquisition ability was studied in fourth grade children through use of a paired associate design. Reading achievement predicted ability to learn words more highly than did other factors, including estimated I.Q., and short-term memory did not. Examination of two subgroups of skilled and less skilled readers indicated that less skilled readers had more difficulty in acquiring new words. Less skilled readers made more errors and required more trials than did skilled readers. Less skilled readers also achieved lower scores on measures of short and long term recognition of the word's referents. No differences in rate of forgetting over time …
