Nutritional Aspects Of Cytotoxin Production By Clostridium Difficile, Doreen P. Osgood, Norris P. Wood, Jay F. Sperry

Arginine was the only amino acid used by Clostridium difficile that permitted cytotoxin synthesis in a peptone-based medium. Synthesis of cytotoxin was delayed when glucose was used as the substrate. Addition of rifampin or puromycin to cultures prior to release of cytotoxin inhibited the release of cytotoxin, suggesting that a protein essential for cytotoxin release is synthesized after cytotoxin is synthesized.

Direct Plating Procedure For Enumerating Vibrio Vulnificus In Oysters (Crassostrea Virginica), G. A. Miceli, W. D. Watkins, S. R. Rippey

A procedure for enumerating and identifying Vibrio vulnificus in oysters was developed and evaluated. This method consists of growth on a direct plating medium (VVE medium) for isolating the organism from shellfish tissues, followed by biochemical tests for differentiating and identifying presumptively positive isolates. Densities of V. vulnificus are reliably obtained in 2 to 4 days, and as few as 10 culturable cells per 100 g can be identified. The procedure was evaluated by using a DNA probe technique specific for the cytotoxin-hemolysin gene of V. vulnificus and gas chromatographic analysis of the fatty acid contents of positive isolates. …

The aim of this study was to characterize the Escherichia coli K88-specific receptors in mucus from the small intestines of 35-day-old piglets with the isogenic strains E. coli K-12(pMK005) (K88+) and E. coli K-12(pMK002) (K88-). These strains differed only in that the latter one cannot produce intact K88 fimbriae because of a deletion in the gene coding for the major fimbrial subunit. Adhesion was studied by incubating 3H-labeled bacteria with crude mucus, pronase-treated whole mucus, mucus fractionated by gel filtration, delipidated mucus, or extracted lipids immobilized in microtiter wells. Through discussion of Semisubmersible Exploratory Drilling Platforms and their role as vectors in marine biological introductions, this paper establishes a base of knowledge for future referral. Included in this project are details of semisubmersibles, their unique characteristics and operations, historical movements, past introductions, future operations, and continuing potential for invasions. Agricultural nonpoint source pollution is the pollution coming from agricultural production, which has no clear origin. It comes from a diffuse source, such as a farm field or an animal holding area. While programs and studies probe every facet of point sources and extol the efforts of point source clean-up, the relatively more difficult task of identifying nonpoint sources and the process of proposing to do something about them has been largely ignored. This review and characterization of the U.S. Atlantic sea herring fishery and industry analyzes opportunities for and constraints to developing the herring resource more fully. Chapter 1 describes the herring resource, the existing fishery, and reviews current research into the international transboundary migrations of the Atlantic sea herring. Chapters 2 and 3 describe the development of the herring fishery management plan in the context of the global herring market and the management complications that arise from the transboundary migrations of the stocks. Understanding what motivates public perception and actions is important in assessing public response and forecasting the success of resource management plans. Failure of the policy makers to foresee public reaction can result in expensive delays, bad press and lengthy court battles which at best, increases the cost of implementation and at worst stops resource management projects altogether. The interaction of the herring (Clupea harengus) with the oil and gas industry in the North Sea is reviewed. The population crash of the herring in the early 1970's and the subsequent closing of that important fishery between 1977 and 1983 has drawn considerable attention to the influences, both biotic and abiotic, affecting the recruitment to the North Sea stock. Current accepted theory points to a change in oceanic current and the subsequent decrease in salinity in the North Sea region, resulting in a change in plankton availability for larval herring as the cause of the recruitment failure during the …

In Vitro Postantibiotic Effect Following Repeated Exposure To Imipenem, Temafloxacin, And Tobramycin, Betty J. Mcgrath, Randall Marchbanks, Deborah H. Gilbert, Michael N. Dudley

The postantibiotic effect (PAE) following three consecutive 2-h exposures to imipenem, temafloxacin, and tobramycin was determined in Pseudomonas aeruginosa. A PAE and a bactericidal effect were consistently observed for imipenem following each cycle of drug exposure and regrowth. In contrast, the PAE increased with repeated exposure with temafloxacin (1.8 to > 5 h), but disappeared with tobramycin by the third exposure (0.9 to 0 h). These data show that the in vitro PAE may change within a strain following multiple cycles of drug exposure and bacterial regrowth.

The effect of dose or dose interval on the pharmacodynamics of simulated high-dose intravenous ciprofloxacin therapy on infection due to Pseudomonas aeruginosa and Staphylococcus aureus was studied in an in vitro hollow-fiber model of infection. Simulated doses of 1,200 mg of ciprofloxacin per day as either 400 mg every 8 h or 600 mg every 12 h against P. aeruginosa resulted in selection of ciprofloxacin-resistant bacteria. The results with one test strain that was isolated from a patient prior to administration of intravenous ciprofloxacin demonstrated selection of a gyrA mutant in the model, as had occurred in vivo.
