We compared 2 methods of direct gene delivery into live rainbow trout Oncorhynchus mykiss tissue, with the final goal of developing DNA-based vaccines for bacterial diseases in salmonids. The introduction of plasmid constructs containing the luciferase and β-galactosidase reporter genes was achieved either by direct injection or by particle bombardment with DNA-coated gold microparticles. Luciferase expression was observed in homogenates of trout flank muscle and skin 2 d after injection of 10 to 100 pg of DNA per fish or bombardment of 1 pm gold particles coated with 5 to 25 pg DNA per fish at helium pressures ranging from …

This document describes the major ecotourism attractions of the Ruteng Nature Recreation Park in Flores, which was established in August 1993. Development of the Park began in 1995 with assistance from the Indonesia Biodiversity Conservation Project (IBCP) funded by the Asian Development Bank. I served as the Team Leader of the IBCP in Flores (1995-1996). My major tasks were supervising the completion of a 25 year management plan for the Park, and providing technical assistance during the first year of implementation of the new management plan. An avirulent, streptomycin-resistant Salmonella typhimurium strain, SL5319, and its lipopolysaccharide (LPS)-deficient mutant strain, SL5325, differ in their ability to colonize the large intestines of streptomycin-treated mice. When fed to mice independently, the strains colonize equally well, but when fed together, the LPS-deficient mutant is outcompeted by the wild-type strain during establishment in the gut (J.J. Nevola, B.A.D. Stocker, D.C. Laux, and P.S. Cohen, Infect. Immun. 50:152-159, 1985). In the present study, the spatial distribution in the intestinal mucosal layer of the two strains was visualized by specific hybridization to bacterial rRNA in histological sections of mouse colon and cecum. Factors Affecting Interannual Movements Of Snowy Plovers, Peter W. C. Paton, Thomas C. Edwards Jr.

We studied the interannual movements of 361 individually color-banded adult Snowy Plovers (Charadrius alexandrinus nivosus) at Great Salt Lake, Utah from 1990 to 1993. In northern Utah, Snowy Plovers nested in a dynamic environment; suitable breeding habitat declined by 50% at two study areas in four years. Male Snowy Plovers were more site faithful than females; 40% of males exhibited fidelity compared with 26% of females (P < 0.01). However, as the amount of available suitable nesting habitat declined, male site fidelity diminished, whereas female fidelity remained relatively constant. We found strong evidence that female site fidelity was affected by nesting success in the previous year. Females that nested unsuccessfully were less likely than successful females to exhibit site fidelity the following year; males did not exhibit this nest-success bias. In addition, unsuccessful females breeding at sites with high densities of nests tended to disperse the following year, whereas male site fidelity did not appear to be affected by either a study site's overall nesting success the previous year or a study site's nest density the previous year. Female avoidance of areas with high densities of nests may be an antipredator strategy. Snowy Plovers in northern Utah have biparental incubation duties, but only males care for broods. Familiarity with brood-rearing areas was one plausible explanation for male-biased fidelity. However, we could not eliminate an alternative hypothesis that both focal study sites represented scarce breeding areas due to the presence of freshwater, and male Snowy Plovers preferred to use the same areas rather than disperse. We propose that more landscape-level studies are needed to address questions concerning local and regional movement patterns.

Purification And Characterization Of Diacylglycerol Pyrophosphate Phosphatase From Saccharomyces Cerevisiae, Wen-I Wu, Yongsheng Liu, Bettina Riedel, Josef B. Wissing, Anthony S. Fischl, George M. Carman

Diacylglycerol pyrophosphate (DGPP) phosphatase is a novel membrane-associated enzyme that catalyzes the dephosphorylation of the β phosphate of DGPP to yield phosphatidate and P. DGPP phosphatase was purified 33,333-fold from Saccharomyces cerevisiae by a procedure that included Triton X-100 solubilization of microsomal membranes followed by chromatography with DE53, Affi-Gel Blue, hydroxylapatite, and Mono Q. The procedure resulted in the isolation of an apparent homogeneous protein with a subunit molecular mass of 34 kDa. DGPP phosphatase activity was associated with the 34-kDa protein. DGPP phosphatase had a broad pH optimum between 6.0 and 8.5 and was dependent on Triton X-100 for …

The decline of most Mexican fisheries has occurred so rapidly that few Mexicans and Americans alike fully appreciate the loss. The major focus of this paper will examine the false impression that Mexico is engaging in marine conservation; in reality, its conservation efforts are responses to after-the-fact actions brought on by world critics and as a vehicle to pacify conservationists. The Mexican government can pass a law but lacks the resources to administer and enforce the law and subsequently manage its fisheries in an effective manner. This study examined the potential of social conflict related to private and public rights, to constrain aquaculture on Cape Cod. While the ability of social conflict to constrain aquaculture has been demonstrated in a few cases, its current potential to constrain aquaculture on Cape Cod had not been established prior to this study. This study was based on information gathered from town records and public hearings related to proposed aquaculture ventures. In addition, town officials were interviewed to obtain additional information related to cases. The principal objective of this thesis was to descriptively and empirically assess shellfish consumption hazards in the state of Rhode Island and to evaluate the use of a shellfish hazard warning label to modify unsafe consumptive behaviors. To accomplish this task, three research problems were examined using a survey instrument. The survey instrument was a questionnaire which included both closed and open-ended questions. The survey questions were designed to identify four primary research variables; shellfish consumptive behaviors, consumer knowledge of shellfish hazards and related government advisories, perceptions of risk regarding personal shellfish consumption, and response to warning label information. Calculations and measurements of nitrogen in groundwater, stream flow and atmospheric deposition were used to see if the cumulative impacts of management decisions over a fifteen year period of time resulted in measurable changes in nitrogen loading to six of the salt ponds on the south shore of Rhode Island. The changes in the source and transmission of nitrogen were tested by comparing calculated and measured nitrogen data from 1980 and 1995. The calculated method included a nitrogen budget based on land-use and literature values for nitrogen loss to groundwater. A large number of marine species migrate within and without national jurisdictions and a small number occur entirely outside any national jurisdiction. Thus, the species migrating through waters under the jurisdiction of coastal states to high seas beyond the jurisdiction of any single state are defined as highly migratory species (HMS). The passage of the Wallop-Breaux Amendments to the Federal Aid in Sportfish Restoration Program (FASRP), in 1984, significantly increased the funding available to states participating in the Program. Subsequently, the FASRP became the largest single source of funding for state marine artificial reef development activities. In recent years, there has been concern among program constituents that FASRP funds are not being utilized as intended. An examination is made of the use of broad bandwidth high-frequency acoustic scattering to infer remotely the spatial structure of the temperature field of a thermally driven buoyant plume. Application of the far-field Born approximation results in a linear relationship between the transfer function of the scattering process, G, the ratio of received to transmitted pressure, to the spatial Fourier transform of the temperature field, φ (K), where K = ks – ki is the Bragg wave-number vector.
