The cystophorous hemiurid cercariae, Cercaria vaullegeardi and C. calliostomae, are recorded from the digestive gland of Gibbula umbilicalis and the gonad of calliostoma ziziphinum respectively. The encysted forms are described at ultrastructural level for the first time, and developmental stages redescribed. Specialisation of the anterior region and birth pore of the daughter sporocyst in c. vaullegeardi, and migration of the redia in C. calliostomae, ensure that in neither species does the cercaria, hindered by a bulky immotile cystophorous tail, itself have to migrate through blood vessels or tissues to the site of emergence from the molluscan host. Dogfish leucocytes were characterised by histochemistry, light and electron microscopical methods and qualitative phagocytic studies. This enabled the recognition of the major phagocytic cells and their role in the immune response of the fish was further evaluated. In addition to in vitro and in vivo studies of phagocytosis, the phagocytic cells were examined for certain surface properties such as the presence of Fe and C3 receptors. Attempts were also made to demonstrate delayed hypersensitivity by in VIVO skin tests and in vitro migration inhibition tests. All the major leucocytic elements found in mammals were demonstrated in dogfish blood together with …

A survey of Myxosporidia in teleosts from the South-Western. coast of England demonstrated the widespread occurrence of these parasites. Infection in selected fish species was related to age, sex and seasonal migration of the host. Structural studies of Myxobolus exiguus and Myxidium incurvatum were undertaken in order to investigate sporogenesis, parasite development and the host-parasite interface. Of particular note was the intimate association between parasite and host cells, the structure of the surface layers in Myxobolus exiguus and Myxidium incurvatum, and the occurrence of epithelioid cells in developmental stages of Myxobolus exiguus. Some of the enzymes involved in the biosynthesis of uric acid have been investigated in two groups of patients: hyperuricaemic and gouty individuals. A group of normal controls has been used for comparison. Human erythrocyte phosphoribosylpyrophosphate synthetase (2.7.6.1.) was found to be much more stable in all samples than has been reported. Kinetic parameters for this enzyme, using a modified assay, were identical in individuals from the three clinical groups. Inhibition studies using a range of inhibitors of widely differing chemical structure showed uniformity in characteristics amongst gouty or healthy individuals. Some inhibitors revealed possible genetic variants of the enzyme …

This dissertation. reports the first detailed study of the possible pathogenic mechanisms and virulence determinants of the fish pathogen V. anguillarum carried out in association with histo-pathological studies using electron microscopy and electrocardiography In addition, possible infection route (s) were investigated and the effect of environmental variables on pathogenesis examined. Pathogenicity was found to be species specific with strains which were avirulent in eels being highly virulent in grey mullet. Following injection of virulent bacteria into eels all tissues were rapidly colonised, death being characterised by severe haemorrhagic septicaemia. In contrast, avirulent strains were rapidly eliminated from the eel host …

This thesis is concerned with lysine vasopressin's (LVP's) behavioural activity. Chapter One describes vasopressin's synthesis, secretion, metabolism, pressor, antidiuretic and putative corticotrophic functions with emphasis on behaviourally significant aspects. Chapter Two reviews behavioural data showing that manipulations of endogenous vasopressin levels alter subsequent avoidance performance. Although these data have predominantly been interpreted in favour of vasopressin altering memory formation (consolidation) results reported from an experiment combining response prevention trials and vasopressin injections failed to support the consolidation hypotheis. Chapter Three reviews the response prevention literature and confirms the feasibility of using prevention trials with automated shuttle box training (Experiment One). …

The cellular responses of Crenimugil labrosus and Pleuronectes platessa to tissue parasites have been investigated using metacercariae of Cryptocotyle lingua and Rhipidocotyle johnstonei, and the myxosporidjan Myxobolus exiguous. The study was based on fish naturally infected with the parasites, and in addition experimental infections of C. lingua were established in grey mullet. The most intense response was induced by R. -johnstonel in plaice, and was composed mainly of cells of the macrophage series, which included epithelioid and giant cells. Fibroblasts and collagen deposition formed the periphery of the lesion which was Interpreted as a granuloma. Eggs released by this progenetic …

Mature oaks covering the full range of the indigenous Quercus species, from the oak woods of Dartmoor and its periphery, were sampled each month during the growing season for the presence of cynipid oak galls. The distributions of galls within sites, within trees and within leaves were recorded, and an investigation into the factors which affect them was made. The distributions of galls on oaks were found to be influenced by climatic and topographical factors as well as by the behaviour of the wasps and the taxonomic status of the host oak. Effectiveness in biological chemical environments virtually defines the term 'drug' when applied to any attempt to modify that environment by the introduction of an influence in terms of a specific compound or group of compounds. Interest in the configuration of the molecules involved in such modifications led to the X-ray structure determinations, discussed in the thesis, of the following three compounds: (i) 7-chloro-2-methyl-5-phenyl-3-propyl [2,3-b]-imidazolyl quinoline. Some effects of DDT on the shore crab (Carcinus maenas) were investigated. Parameters examined included the activity of gill ATPases and their effects on ion and ammonia levels of the haemolymph. Histological alterations were observed by light microscopy. In an in vitro study, an homogenate of Carcinus gills was exposed to concentrations of DDT from 10ˉ¹º M to 10-5M. The activity of Na+ , K+ -ATPase was stimulated at all concentrations. The activity of Mg2+ -ATPase was inhibited at all concentrations. Studies were performed on two types of infection of teleost fish where viruses have been observed by electron microscopy: erythrocytic infections in the Atlantic Cod (Gadus morhua) and the Common Blenny (Blennius hpo lis) and lymphcystis disease., Searches were made for new, isolations of these infections Ja British coastal waters and on shores chiefly in the vicinity of Plymouth and Aberystwyth. The epidermis of the eel A. Anguilla L. secretes a mucous layer mainly from its mucous cells although there may also be a contribution from surface epidermal cells . Under certain conditions, the epidermal cells may transform to a secretory cell type that resembles, but is structurally distinct from a mucous cell . This observation has not previously been reported in fish epidermis. Specialised club cells, characteristic of the Anguillidae contain filamentous material that may be secreted beneath the epidermal surface. This previously unobserved phenomenon together with an increase in cell numbers around the lateral region may suggest an additional …

Synthetic plant-growth regulators derived from fluorene-9-carboxylic acid ('morphactins') are introduced and the literature on them is reviewed. Similar herbicidal activity has been found with 9,9-bishydroxymethylfluorene, a compound which can be obtained directly from the hydrocarbon fluorene. There follows a detailed review of the chemistry of fluorene and 9,9- bishydroxymethylfluorene. 9-Substituted and ring-substituted derivatives of this diol have been synthesised and their chemistry and biological activity are discussed. The hydroxymethylation of substituted fluorenes is possible on a small scale in suitable solvents which do not stabilise the corresponding radical anions. This radical anion formation, when it occurs, seriously reduces the yield …

In this work it is shown that if the rates of ionization and electron loss are linear in the electron concentration ( n ), then the stability of the discharge is determined by the rates of dissipative processes even in the case when the stationary regime is determined by volume processes. To study the stability of the positive column of a fast-flowing steady glow discharge, a boundary value problem is solved for perturbations taking into account diffusion, thermal conductivity, and the characteristics of the external electrical circuit. The studies reported herein cover two main areas of research. The distribution of lead through the plant has been monitored using histochemical and quantitative techniques, and physiological responses of the plant to lead contamination have been examined. In seeds the testa prevents lead contamination of the embryo prior to germination. In seedlings root-applied lead is capable of essentially unrestricted movement through the plant, although anomalies in distribution exist. The metal is sequestered during its passage through the plant, and the endodermis offers a partial barrier to lead movement, restricting contamination of the aerial parts of the plant. A number of aspects of the humoral and cellular immune responses of the common dogfish Scyliorhinus canicula L. were examined in order to define the immunological status of the animal in relation to its phylogenetic origins. The dogfish was shown to have the ability to produce specific antibody in response to a variety of soluble and particulate protein antigens. The existence of several non-specific defence mechanisms was demonstrated including a well established reticulo-endothelial system capable of rapidly clearing injected foreign material from the blood and also the bacteriolytic enzyme, lysozyme. Specific antibody from the fish had marked agglutinating properties which …

The ecology of Polydora, epp was reviewed and the infestation of molluscs by the boring, spionid polychaete, Polydora ciliata, was studied in detail. X-radiography showed that, P. ciailt a infested a wide variety of molluscan substrates, but it . occurred in greatest abundance on large, rough-surfaced, epibenthic shells, which were collected from the-sublittoral zone and the level of extreme low water spring tides, of shores sheltered from wave action. P. ciliata was contagiously dispersed on mussel--shells. Monthly radiographs of mussels indicated that maximum settlement of P. ciliata larvae in the river Fowey, 1 occurred in May.
