Research Reproducibility: Educating for Reproducibility, Pathways to Research Integrity was an interdisciplinary, conference hosted virtually by the University of Florida in December 2020. This event brought together educators, researchers, students, policy makers, and industry representatives from across the globe to explore best practices, innovations, and new ideas for education around reproducibility and replicability. Emphasizing a broad view of rigor and reproducibility, the conference touched on many aspects of introducing learners to transparency, rigorous study design, data science, data management, replications, and more. The classification of myeloid neoplasms and acute leukemias was last updated in 2016 within a collaboration between the World Health Organization (WHO), the Society for Hematopathology, and the European Association for Haematopathology. This collaboration was primarily based on input from a clinical advisory committees (CACs) composed of pathologists, hematologists, oncologists, geneticists, and bioinformaticians from around the world. The recent advances in our understanding of the biology of hematologic malignancies, the experience with the use of the 2016 WHO classification in clinical practice, and the results of clinical trials have indicated the need for further revising and updating the classification. Nepal's political institutions and administrative units were thoroughly restructured in 2015 with the promulgation of the new Constitution. Several rural areas were combined to meet the definition of urban threshold criteria to classify rural areas into urban categories. Accordingly, over 3,900 local political and administrative units were amalgamated into 753 units, of which, 293 units are classified as urban. Within these newly defined urban areas, many natural environments have been converted into impervious surfaces such as paved roads, sidewalks, and building roofs. These impervious surfaces have drastically increased the amount of surface run-offs-often termed as "urban floods"--under increasing precipitation caused …

This film examines a cooperative of the Brazilian Landless Movement (MST) in the South of Brazil, which struggled for access to land and then transitioned to ecological agriculture, or agroecology. This MST cooperative is demonstrating the possibility of an alternative model of flourishing rural life, which provides thriving livelihoods for farmers, produces high quality and low cost food for the region, and rehabilitates the earth. Von Willebrand factor (VWF) level and/or function is altered in von Willebrand disease (VWD), the most common heritable bleeding disorder worldwide. Laboratory assessment of VWF is continually evolving. Historically, the primary method for the assessment of VWF platelet-binding activity was the ristocetin cofactor assay (VWF:RCo). Contemporary alternative measures of VWF platelet-binding activity include VWF:GPIbR (recombinant; using ristocetin), VWF:GPIbM (recombinant; gain-of-function mutant), and monoclonal antibody. Background: The purpose of this study was to evaluate the impact of the COVID-19 pandemic activity restrictions on pediatric supracondylar humerus fracture epidemiology in terms of both mechanism of injury and fracture patterns.
Methods: This was a retrospective chart review of all patients presenting with types II-IV supracondylar humerus fractures between March 15th and June 30th 2020 at the McMaster University Medical Centre. The high-affinity immunoglobulin E (IgE) receptor, FcεRI, is the primary immune receptor found on mast cells and basophils. Signal initiation is classically attributed to phosphorylation of FcεRI β- and γ-subunits by the Src family kinase (SFK) Lyn, followed by the recruitment and activation of the tyrosine kinase Syk. FcεRI signaling is tuned by the balance between Syk-driven positive signaling and the engagement of inhibitory molecules, including SHIP1. Here, we investigate the mechanistic contributions of Lyn, Syk, and SHIP1 to the formation of the FcεRI signalosome. Using Lyn-deficient RBL-2H3 mast cells, we found that another SFK can weakly monophosphorylate the γ-subunit, …

DESCRIPTION: Coronavirus disease 2019 convalescent plasma (CCP) has emerged as a potential treatment of COVID-19. However, meta-analysis data and recommendations are limited. The Association for the Advancement of Blood and Biotherapies (AABB) developed clinical practice guidelines for the appropriate use of CCP.
METHODS: These guidelines are based on 2 living systematic reviews of randomized controlled trials (RCTs) evaluating CCP from 1 January 2019 to 26 January 2022. There were 33 RCTs assessing 21 916 participants. The results were summarized using the GRADE (Grading of Recommendations Assessment, Development and Evaluation) method. Background: Critically ill COVID-19 patients are in a hypercoagulable state with increased risk of thrombotic complications. Rotational thromboelastometry (ROTEM) is a viscoelastic test with the potential to reflect COVID-19-associated hypercoagulability and may therefore be useful to predict thrombotic complications.
Objective: To investigate the potential of ROTEM profiles to predict thrombotic complications in critically ill COVID-19 patients.
Patients/methods: Retrospective multicenter cohort study in 113 adult patients with confirmed COVID-19 infection admitted to the intensive care unit (ICU) of two large teaching hospitals in the United States and in the Netherlands. PURPOSE: This is the first multisite investigation of the validity of scores from the current version of the Medical College Admission Test (MCAT) in clerkship and licensure contexts. It examined the predictive validity of MCAT scores and undergraduate grade point averages (UGPAs) for performance in preclerkship and clerkship courses and on the United States Medical Licensing Examination Step 1 and Step 2 Clinical Knowledge examinations. Acute lymphoblastic leukemia (ALL) is the most common childhood cancer. Here, using whole-genome, exome and transcriptome sequencing of 2,754 childhood patients with ALL, we find that, despite a generally low mutation burden, ALL cases harbor a median of four putative somatic driver alterations per sample, with 376 putative driver genes identified varying in prevalence across ALL subtypes. Most samples harbor at least one rare gene alteration, including 70 putative cancer driver genes associated with ubiquitination, SUMOylation, noncoding transcripts and other functions. Cervical cancer is the fourth most common malignancy in women worldwide. Although infection from human papillomavirus (HPV) has been the leading cause of cervical cancer, HPV-negative cervical cancer accounts for approximately 3-8% of all cases. Previous research studies on cervical cancer have focused on HPV-positive cervical cancer due to its prevalence, resulting in HPV-negative cervical cancer receiving considerably less attention. As a result, HPV-negative cervical cancer is poorly understood. Its etiology remains elusive mainly due to limitations in research methodology such as lack of defined markers and model systems.
