Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is an enveloped, positive-sense, single-stranded RNA virus of the genus Betacoronavirus. Its genome is composed of four structural proteins known as spike (S), envelope (E), membrane (M), and nucleocapsid (N), of which E, M, and N are integrated into the viral envelope. The S glycoprotein, which protrudes from the surface of mature virions as a spike, is essential for virus attachment, fusion, and entry into the host cell. While the relationship between the spike protein of SARS-CoV-2 and the angiotensin- converting enzyme 2 (ACE2) receptor has been readily established, the S1 subunit also …

The Giant Axon Of The Squid: A Simple System For Axonal Transport Studies, Joseph A. Degiorgis, Marcus Jang, Elaine L. Bearer

The squid giant axon has a long history of being a superb experimental system in which to investigate a wide range of questions concerning intracellular transport. In this protocol we describe the method used for dissecting the axon to preserve its viability in vitro, and the technique for injecting exogenous materials into the living axon. Now that the squid genome is emerging, and the CRISPR/cas9 system has been successfully applied to knock-out squid genes, the giant axon will resume its place in the scientific pantheon of powerful experimental systems in which to address biological questions pertaining to all eukaryotes.

Native Americans are the least represented population in science fields. In recent years, undergraduate and graduate level summer research programs that aimed to increase the number of Native Americans in science have made some progress. As new programs are designed, key characteristics that address science self-efficacy and science identity and provide supports for Native American students' commitment to a scientific career should be considered. In this study, we used sequential mixed methods to investigate the potential of culturally tailored internship programs on Native American persistence in science. Nitrogen vacancy (NV) centers have been used extensively for the measurement of magnetic fields in applications of geology, biology, medicine, and fundamental physics. In regard to fundamental physics, NV centers offer a unique opportunity to test potential velocity dependant spin-spin interactions as well as velocity-spin interactions at the micron scale. In regards to magnetic materials, NV centers offer a platform to investigate magnetic properties of nanoparticles in an individual, but highly parallelized measurement. On November 3, 2020 the U.S. Fish and Wildlife Service (“FWS”)1 issued “Removing the Gray Wolf (Canis Lupus) From the List of Endangered and Threatened Wildlife” (“Final Rule”) delisting the gray wolf in forty-eight states, except for the Mexican gray wolf in the Southwest.2 This Final Rule returned gray wolf management to the states. Wolf delisting was a last-minute gift from the Trump administration to conservative voters, particularly hunters, trappers, and livestock owners right before the November 2020 presidential election. The production of personalized cancer vaccines made from autologous tumour cells could benefit from mechanisms that enhance immunogenicity. Here we show that cancer vaccines can be made via the cryogenic silicification of tumour cells, which preserves tumour antigens within nanoscopic layers of silica, followed by the decoration of the silicified surface with pathogen-associated molecular patterns. These pathogen-mimicking cells activate dendritic cells and enhance the internalization, processing and presentation of tumour antigens to T cells. In syngeneic mice with high-grade ovarian cancer, a cell-line-based silicified cancer vaccine supported the polarization of CD4+ T cells towards the T-helper-1 phenotype in the …

Ulcerative Colitis (UC) is a chronic inflammatory disease of the intestinal tract for which a definitive etiology is yet unknown. Both genetic and environmental factors have been implicated in the development of UC. Recently, single cell RNA sequencing (scRNA-seq) technology revealed cell subpopulations contributing to the pathogenesis of UC and brought new insight into the pathways that connect genome to pathology. This review describes key scRNA-seq findings in two major studies by Broad Institute and University of Oxford, investigating the transcriptomic landscape of epithelial cells in UC. Ken Collins has lived with his brain injury for 45 years and has worked with people with brain injuries since 1983. He is an advocate of developing home and community-based services for people with brain injuries. In 1986-88 he was a VISTA Volunteer and helped develop the first transitional living program for people with brain injuries in the U.S. Ken is currently employed as Executive Director, Hozho Center for Personal Enhancement in Gallup, New Mexico where he supervises staff and provides independent living services to Veterans, Navajo's and Zuni's with brain injuries in McKinley County.

This article treats four Supreme Court opinions that have had a lasting impact, largely negative, on public lands and resources. They rest on highly selective statements of fact, and dubious footing with precedent and statutory law. As a quartet they make the protection of natural resources extremely difficult. Resources that, in law, belong to us all. The first case, Southern Utah Wilderness Association, opened up a designated Wilderness Area too off-road vehicle use, where these uses are explicitly prohibited by law. Larger, more powerful wildfires increasingly threaten inhabited areas as well as vital infrastructure, including many installations of the United States Armed Forces. This article first introduces readers to what wildfire is and the unique challenges it creates to the environment, health and, specifically, to the Department of Defense (DoD). Since Roger Williams’ arrival in Narragansett Territory in 1636, and his subsequent settlement of the Providence Plantations, the Narragansett Indian Tribe--the Indigenous people to this land--have faced a series of intergenerational atrocities, including attempted genocides. For generations, these heinous wrongs have not been corrected by state or federal courts, which have often compounded the harms against the Narragansett people. A key question in SARS-CoV-2 infection is why viral loads and patient outcomes vary dramatically across individuals. Because spatial-temporal dynamics of viral spread and immune response are challenging to study in vivo, we developed Spatial Immune Model of Coronavirus (SIMCoV), a scalable computational model that simulates hundreds of millions of lung cells, including respiratory epithelial cells and T cells. SIMCoV replicates viral growth dynamics observed in patients and shows how spatially dispersed infections can lead to increased viral loads. The model also shows how the timing and strength of the T cell response can affect viral persistence, oscillations, and control. …

BACKGROUND: The efficacy of polyclonal high titer convalescent plasma to prevent serious complications of COVID-19 in outpatients with recent onset of illness is uncertain.

METHODS: This multicenter, double-blind randomized controlled trial compared the efficacy and safety of SARS-CoV-2 high titer convalescent plasma to placebo control plasma in symptomatic adults ≥18 years positive for SARS-CoV-2 regardless of risk factors for disease progression or vaccine status. Participants with symptom onset within 8 days were enrolled, then transfused within the subsequent day. HPV is the most common sexually transmitted infection. It is estimated that 80% of sexually active adults will contract an HPV infection. Thus, there is a significant need for low-cost and easily distributable HPV antivirals that can be used alongside vaccines to combat infection. Herein I have identified protamine sulfate (PS) as a potent microbicide against HPV infection. PS was agnostic to genotype and efficacious pre and post HPV exposure. PS application topically reduced HPV internalization by altering epidermal growth factor receptor (EGFR) signaling and suppressed EGFR-stimulated HPV entry. Tetrasphaera polyphosphate accumulating organisms (PAOs) have been found in greater abundance and may remove more P in full-scale enhanced biological phosphorus removal (EBPR) water resource recovery facilities (WRRF) worldwide than the more extensively studied Accumulibacter PAO. Outdoor learning experiences offer schools an initial avenue to address global community issues, including climate change, natural resource depletion, disconnection from nature, and the current global coronavirus pandemic. This narrative inquiry study, grounded in the theoretical work of Vygotsky, Cajete, Brody, and Kahn, seeks to understand how children’s perceptions of their outdoor learning experiences provide insights into their learning processes. Intrinsically disordered proteins (IDPs) play vital roles in cell signaling and regulation by changing conformation dynamically under various stimulus. Thus, it is crucial to understand their physical properties in vitro especially liquid-liquid phase separation (LLPS). Here, we first used UV-vis spectrophotometry to investigate two of IDPs' physical properties – transition temperature (Tt) and coacervate size. This deeper insight allows us to further understand the dynamics of phase growth. We then constructed a binodal curve in the phase diagram by measurement of volume ratios in phase separated IDP solution in glass capillaries.

Mixed-conifer forests play an important role in the Southwestern USA’s regional carbon cycle by sequestering and storing large amounts of atmospheric carbon onto the landscape. Despite this, little is known about how large, stand-replacing fire alters ecosystem carbon sink-source dynamics. In this study, we used eddy covariance to quantify how stand-replacing fire altered forest sink strength, and the sensitivity of carbon uptake to soil and atmospheric drought in a mixed-conifer forest located in The Valles Caldera National Preserve, NM. The resolution limit of optical microscopy can be extended by using Imaging Interferometric Microscopy (IIM), which uses a low numerical aperture (NA) objective lens to achieve resolution equivalent to that of a high-NA objective lens with multiple sub-images. Along with the resolution enhancement challenge, IIM often suffers from poor image quality. In this dissertation, several image quality improvement methods are proposed and verified with simulation and experimental results. Infectious disease is a primary source of mortality for most mammal species, but scientists have little understanding of factors driving variation in infection and immunity between individuals, populations, and species in the wild. Life history theory provides an evolutionary framework for delineating distribution of available energy to competing physiological demands, including growth, reproduction, and maintenance. Early life reproduction should be favored over late life survival, but, in long-lived species, reproductive success is strongly tied to survival to old age. Prenatal alcohol exposure (PAE) is associated with structural and physiological changes that impact the central nervous system and can result in persistent negative consequences in a broad spectrum of cognitive and behavioral domains including deficits in spatial learning and memory and behavioral flexibility. The current study focuses on examining the behavioral and mechanistic consequences of PAE, sex and housing. Rats were tested in the Morris Water Task where they were trained to navigate to a goal location and tested in a pool shift manipulation.
