Children with chronic myeloid leukemia (CML) tend to present with higher white blood counts and larger spleens than adults with CML, suggesting that the biology of pediatric and adult CML may differ. To investigate whether pediatric and adult CML have unique molecular characteristics, we studied the transcriptomic signature of pediatric and adult CML CD34+ cells and healthy pediatric and adult CD34+ control cells. Using high-throughput RNA sequencing, we found 567 genes (207 up- and 360 downregulated) differentially expressed in pediatric CML CD34+ cells compared to pediatric healthy CD34+ cells. Sex- And Age - Specific Migratory Strategies Of Blue Whales In The Northeast Pacific, Christina Blevins

Sequential subsampling of blue whale baleen plates can yield information of individual migratory plasticity of this endangered species in the northeast Pacific Ocean. We measured δ13C and δ15N isotope values along baleen from thirteen whales of different age and sex. Results showed sex/age-specific migratory strategies. Adult females exhibited relatively stable cyclical movements between temperate and subtropical regions. Adult males exhibited two strategies, most remained within temperate latitudes, whereas two migrated to subtropical latitudes. Movement patterns in juveniles were erratic. These patterns are potentially driven by energetic requirements of females, intraspecific competitions among adult males, and inexperience in locating prey in …

Abandoned uranium mines across the U.S. disproportionately affect indigenous communities who suffer numerous intergenerational health consequences from chronic exposure to toxic metal mixtures. The Jackpile-Paguate Uranium Mine on Laguna Pueblo in west-central New Mexico was designated a Superfund Site in 2013 and exposure risks from inhalation of toxic metals in airborne particulate matter are of concern for the local tribal communities. Genomic studies of pediatric cancer have primarily focused on specific tumor types or high-risk disease. Here, we used a three-platform sequencing approach, including whole-genome sequencing (WGS), whole-exome sequencing (WES), and RNA sequencing (RNA-seq), to examine tumor and germline genomes from 309 prospectively identified children with newly diagnosed (85%) or relapsed/refractory (15%) cancers, unselected for tumor type. Eighty-six percent of patients harbored diagnostic (53%), prognostic (57%), therapeutically relevant (25%), and/or cancer-predisposing (18%) variants. Inclusion of WGS enabled detection of activating gene fusions and enhancer hijacks (36% and 8% of tumors, respectively), small intragenic deletions (15% of tumors), and mutational signatures revealing …

The objective of this study was to determine whether the polyphosphate metabolism in purple photosynthetic bacteria (PPB) is similar to that of wastewater polyphosphate-accumulating organisms (PAOs) such as Accumulibacter. Dairy lagoon samples were studied in continuous and batch laboratory tests to evaluate whether cyclic light and dark conditions for PPB are similar to aerobic and anaerobic conditions for PAOs in domestic wastewater treatment systems. 16s rRNA gene amplicon sequencing indicated the presence of the purple sulfur bacterium Thiolamprovum (53.92%) in one lagoon source, and Thiodictyon (3.50%) in a second lagoon. Background: Immune responses to vaccination are a known trigger for a new onset of glomerular disease or disease flare in susceptible individuals. Mass immunization against SARS-CoV-2 in the COVID-19 pandemic provides a unique opportunity to study vaccination-associated autoimmune kidney diseases. In the recent literature, there are several patient reports demonstrating a temporal association of SARS-CoV-2 immunization and kidney diseases.

Crosstalk between different receptor tyrosine kinases (RTKs) is thought to drive oncogenic signaling and allow therapeutic escape. EGFR and RON are two such RTKs from different subfamilies, which engage in crosstalk through unknown mechanisms. We combined high-resolution imaging with biochemical and mutational studies to ask how EGFR and RON communicate. EGF stimulation promotes EGFR-dependent phosphorylation of RON, but ligand stimulation of RON does not trigger EGFR phosphorylation - arguing that crosstalk is unidirectional. Nanoscale imaging reveals association of EGFR and RON in common plasma membrane microdomains. Two-color single particle tracking captured formation of complexes between RON and EGF-bound EGFR. BACKGROUND: Signaling pathways mediated by microRNAs (miRNAs) have been identified as one of the mechanisms that regulate stroke progression and recovery. Recent investigations using stroke patient blood and cerebrospinal fluid (CSF) demonstrated disease-specific alterations in miRNA expression. In this study, for the first time, we investigated miRNA expression signatures in freshly removed human stroke brain tissue.

METHODS: Human brain samples were obtained during craniectomy and brain tissue resection in severe stroke patients with life-threatening brain swelling. The tissue samples were subjected to histopathological and immunofluorescence microscopy evaluation, next generation miRNA sequencing (NGS), and bioinformatic analysis.

In this dissertation interferometric lithography is approached in two different ways to address two important constraints of nanopatterning. One approach solves the problem of scaling up interferometric lithography to wafer scale (4 inch or larger) area. Through the second approach we have developed a nanopatterning technique based on interferometric lithography by using an inexpensive (~$100) diode laser as source, making interferometric lithography a very cost-effective technique.

Despite extensive research and aggressive therapies, glioblastoma (GBM) remains a central nervous system malignancy with poor prognosis. The varied histopathology of GBM suggests a landscape of differing microenvironments and clonal expansions, which may influence metabolism, driving tumor progression. Indeed, GBM metabolic plasticity in response to differing nutrient supply within these microenvironments has emerged as a key driver of aggressiveness. Additionally, emergent biophysical and biochemical interactions in the tumor microenvironment (TME) are offering new perspectives on GBM metabolism. Classifying people with alcohol use disorder into homogenous groups based on observed characteristics (i.e., phenotypes) could match individuals to specific treatments. The reward-relief phenotype classifies individuals based on whether they drink to enhance rewarding experiences and/or to relieve negative states. The current study is a secondary data analysis of a community sample of non-treatment seeking heavy drinkers (n = 189) and two randomized clinical trial samples of individuals with alcohol use disorder (n = 1726, 1383) that aimed to determine if the reward-relief phenotype is identified across samples, is stable over time, and predicts long-term alcohol consumption and consequences. Comparative phylogeography has historically been defined as the study of how genetic variation of co-distributed species has been shaped by biogeographical history. This is a mature field of study out of which several techniques have been developed directed at identifying the role ecology and geography in diversification patterns across time. Opioid addiction is a serious and persistent global health issue. The federal Centers for Disease Control and Prevention (CDC) reported that between 1999 and 2016, more than 630,000 people in the United States died of an overdose of a prescription opioid or illicit drug (CDC 2018). Extant research has suggested that for nearly a century, New Mexico has experienced some of the highest rates of prescription and illicit opioid death in the nation (Goldstein and Herrera, 1995; Landon, 2003; Shah et al., 2008). The first paper looks at the value people place on clean air in Mexico City using an alternative air pollution measure to the typical ground measurement. Research and policy addressing air pollution remains hampered by a lack of ground level air quality data. This study attempts to overcome this issue using aerosol optical depth from satellite imagery as a proxy for air pollution. Quantifying microbial biomass is important for understanding microbial contribution to nutrient cycling and population flux. Widely accepted biomass estimation methods are sufficient for well-studied and high biomass systems, though are often not sensitive enough for systems with extremely low biomass. The McMurdo Dry Valleys of Antarctica are characterized as an extremely harsh environment with severe restrictions on life, with perhaps some of the lowest biomass in the world. Coccidioidomycosis, caused by soil-dwelling species of Coccidioides, is endemic to the Southwestern United States. It can present as a mild respiratory illness to life-threatening disseminated infections.Multi-gene phylogenetics and tracing patient histories revealed the presence of both Coccidioides species in New Mexico, and Native Americans may represent an unrecognized risk group. A PCR method developed distinguishes Coccidioides species by targeting regions of the mitochondrial genome. Recent advancements in molecular genetics solidifies lungs are not sterile. The Reward Positivity (RewP) is a positive deflection in the EEG after receiving a reward. Recent evidence suggests that the RewP is modulated by both reward probability aside from affective liking. Here we examined the sensitivity of the RewP to affective and alcohol images in individuals with alcohol use disorder (AUD). We recruited 55 participants (AUD =28, Control= 27). Participants completed an image rating task and a reinforcement learning task with picture feedback (puppy or alcohol images over a green or red screen). BACKGROUND: Problems continue to exist with the reporting quality and risk of bias in search methods and strategies in systematic reviews and related review types. Peer reviewers who are not familiar with what is required to transparently and fully report a search may not be prepared to review the search components of systematic reviews, nor may they know what is likely to introduce bias into a search. Sin Nombre orthohantavirus (SNV), a negative-sense, single-stranded RNA virus that is carried and transmitted by the North American deer mouse Peromyscus maniculatus, can cause infection in humans through inhalation of aerosolized excreta from infected rodents. This infection can lead to hantavirus cardiopulmonary syndrome (HCPS), which has an ∼36% case-fatality rate. We used reverse transcriptase quantitative PCR (RT-qPCR) to confirm SNV infection in a patient and identified SNV in lung tissues in wild-caught rodents from potential sites of exposure. Using viral whole-genome sequencing (WGS), we identified the likely site of transmission and discovered SNV in multiple rodent species not previously known …

Rapid detection of emerging pathogens, environmental monitoring of waterways, and cancer treatments that operate on a cell by cell basis are all technological advances which could be achieved with DNA-based molecular computers. DNA encodes the building blocks of life and researchers have already shown that it can be used as a powerful tool for us to build nanoscale devices that can interact with the molecular world. However, DNA-based molecular devices are susceptible to degradation by natural defense mechanisms called nucleases. Olfactory sensory neurons (OSNs) directly contact the environment and are exposed to pathogens, such as viruses. When OSNs detect a virus, they coordinate antiviral immune responses locally to stop virus progression into the brain, termed the central nervous system (CNS). For example, in COVID-19 patients the SARS-CoV-2 virus replicates in the olfactory epithelium resulting in loss of olfaction, yet viral detection in the CNS is rare. Monitoring the population size of an animal group can inform researchers of habitat use, potential changes in habitat and the resulting behavioral adaptation, individual health, and the effectiveness of conservation efforts. Arboreal primates are difficult to monitor as their habitat is often poorly accessible and most primate species have some degree of camouflage, making them hard to observe in and below the tree canopy. The Sanapaná language is spoken by around 1000 people in the Paraguayan Chaco region (Crevels 2012). Only about a third of the members of the ethnic Sanapaná group still speak the language, and in five of the seven Sanapaná communities, children now learn Spanish or Paraguayan Guaraní as their native language. This makes Sanapaná one of the many languages that are in danger of disappearing.
