Dectin-1A is a C-type lectin innate immunoreceptor that recognizes β-(1,3;1,6)-glucan, a structural component of Candida species cell walls. β-Glucans can adopt solution structures ranging from random coil to insoluble fiber due to tertiary (helical) and quaternary structure. Fungal β-glucans of medium and high molecular weight are highly structured, but low molecular weight glucan is much less structured. Despite similar affinity for Dectin-1, the ability of glucans to induce Dectin-1A-mediated signaling correlates with degree of structure. BACKGROUND: Prenatal alcohol exposure (PAE) impacts the neurodevelopment of the fetus, including the infant's ability to self-regulate. Heart rate variability (HRV), that is, the beat-to-beat variability in heart rate, is a non-invasive measurement that can indicate autonomic nervous system (ANS) function/dysfunction.

METHODS: The study consisted of a subset of our ENRICH-2 cohort: 80 participants (32 PAE and 48 Controls) who had completed three visits during pregnancy. The participants completed a comprehensive assessment of PAE and other substances throughout pregnancy and assessments for stress, anxiety, and depression in the third trimester. Artificial intelligence (AI) presents an opportunity in anatomic pathology to provide quantitative objective support to a traditionally subjective discipline, thereby enhancing clinical workflows and enriching diagnostic capabilities. AI requires access to digitized pathology materials, which, at present, are most commonly generated from the glass slide using whole-slide imaging. Models are developed collaboratively or sourced externally, and best practices suggest validation with internal datasets most closely resembling the data expected in practice. BACKGROUND: COVID-19 convalescent plasma (CCP) is an important therapeutic option for outpatients at high risk of hospitalization from SARS-CoV-2 infection. We assessed the safety of outpatient CCP transfusions administered during clinical trials.

STUDY DESIGN AND METHODS: We analyzed data pertaining to transfusion-related reactions from two randomized controlled trials in the U.S. that evaluated the efficacy of CCP versus control plasma in various ambulatory settings. Multivariable logistic regression was used to assess whether CCP was associated with transfusion reactions, after adjusting for potential confounders.

Within this dissertation, two distinct classes of metal complexes have been discussed that bear significance in the field of solar energy conversion, photo redox catalysis and memory devices. The first class of the metal complexes is a series of photochromic Ruthenium polypyridyl N-heterocyclic carbene tethered sulfoxide complexes that are reported to undergo unusually slow isomerization from S-bonded to O-bonded and unusually fast thermal reversion from O-bonded to S-bonded configuration. The role of NHC in the excited state dynamics of these complexes have been discussed in detail in Chapter 3.

Smoke from wildland fires has been shown to produce neuroinflammation in preclinical models, characterized by neural infiltrations of neutrophils and monocytes, as well as altered neurovascular endothelial phenotypes. Humans show an attention bias toward emotional versus neutral information, which is considered an adaptive pattern of information processing. Deviations from this pattern have been observed in children with socially withdrawn behaviors, with most research being conducted in controlled settings among children from urban areas. OBJECTIVE: To develop initial American College of Rheumatology (ACR) guidelines on the use of exercise, rehabilitation, diet, and additional interventions in conjunction with disease-modifying antirheumatic drugs (DMARDs) as part of an integrative management approach for people with rheumatoid arthritis (RA).

METHODS: An interprofessional guideline development group constructed clinically relevant Population, Intervention, Comparator, and Outcome (PICO) questions. A literature review team then completed a systematic literature review and applied the Grading of Recommendations Assessment, Development and Evaluation (GRADE) approach to rate the certainty of evidence. The purpose of this study is to assess immigrational trauma as it relates to dental phobia in accessing care from undocumented individuals. Immigrating from a country to the United States presents with unique challenges. These challenges include language barriers, traumatic experiences and cultural factors. These individuals also have perceived notions regarding their treatment with a physician or dentist as their immigration status may or may not be revealed during their care. This dissertation investigates the notion of a paradigm and its related morphological method through the lens of the philosophy of Ludwig Wittgenstein (1889–1951). At once exegetical and philosophical, it is a project that aims to trace the progression of Wittgenstein’s thought, from his early to his mature phase, in order to arrive at a twofold conclusion regarding the ontological status of paradigms and a closely related approach to logic grounded in morphology.

Cognitive reserve refers to the cerebral plasticity perspective in which premorbid buffer factors, explained as enriching life experiences, protect against the negative outcomes of neurodegenerative and neuropsychiatric diseases (Stern, 2009; Harrison et al., 2015). Previous research has observed significant group differences regarding neuroanatomical and psychosocial variables between incarcerated boys who have and have not previously committed a homicide, resulting in successful postdictive classification (Cope et al., 2014). However, no study to date has investigated whether similar group differences characterize future homicide offenders. Following the methodology of Cope et al. (2014), the current study aimed to identify baseline neural, clinical, and environmental deficits (collected in a sample of n = 242 incarcerated juvenile offenders) associated with future homicidal behavior as adults. Recent evidence suggests corroded trust in scientific institutions, but particularly among political conservatives. The current investigation examined the potential role of trust in conservative media sources by capturing associations between trust in politically-biased media sources, trust in science and scientists, and COVID-19 vaccine hesitancy. A total of 1,117 participants completed an online cross-sectional self-report study. Results revealed that: (1) the less trust respondents had in science and scientists, the greater their overall COVID-19 vaccine hesitancy (and perceived risks and fears of COVID-19 vaccines); and (2) the more respondents trusted conservative-leaning news media sources, the greater their overall COVID-19 vaccine hesitancy …

BACKGROUND: Myeloid cells are critical for iron and immune homeostasis. Ferritin heavy chain (FTH1) is essential for intracellular iron storage. Myeloid FTH1 is important in the pathogenesis of many inflammatory diseases. However, the role of myeloid FTH1 in colitis and colitis-associated cancer has not been determined.

METHODS: Myeloid FTH1 deficient and wild-type mice were treated with dextran sodium sulfate (DSS) or azoxymethane (AOM)-DSS to compare their susceptibility to acute colitis or colitis-associated cancer.

Human papillomaviruses (HPVs) are a significant public health concern due to their widespread transmission, morbidity, and oncogenic potential. Despite efficacious vaccines, millions of unvaccinated individuals and those with existing infections will develop HPV-related diseases for the next two decades and beyond. The continuing burden of HPV-related diseases is exacerbated by the lack of effective therapies or cures for infections, highlighting the need to identify and develop antivirals. This dissertation is a collection of chapters that demonstrate the factors affecting human capital and labor market outcomes of adolescents and the gender differences in the quality of employment of adults in South Africa.

The second chapter evaluates the impact of South Africa’s Child Support Grant, an unconditional means-tested grant provided to age-eligible children, on adolescent nutrition outcomes. The findings suggest that welfare programs can alleviate extreme undernutrition and overnutrition among females during the critical growth period of adolescence.

Oral plaque biofilm has been identified as the primary contributor to dental conditions such as dental caries and periodontal disease that promotes a diverse and pervasive collection of microbial species to inhabit the oral cavity. Disrupting the plaque matrix early could help combat this global disease burden. This study evaluated the effect of IONPs on primary gingival keratinocytes.

In 2022 I investigated niche partitioning among native and invasive freshwater turtles across three sites in the Middle Rio Grande using bulk stable isotope analysis. I found high degrees of niche partitioning at sites in which Red-eared sliders (Trachemys scripta) have yet to establish. However, at sites in which red-eared sliders are now the most abundant species they overlapped greatly in isotopic niche with all native turtles, in particular the painted turtle (Chrysemys picta). Diversification By Dispersal Out Of The Andes Revealed By A Newly Discovered Hummingbird Clade, The ‘True Brilliants’, Marialejandra Castro-Farías

We identified a novel clade with striking biogeographic implications. Unsampled genera, Hylonympha and Sternoclyta, were found to be close to the genus Heliodoxa within the Brilliants. Heliodoxa tend to occur in lower montane habitats, such as the tropical, upper tropical, and subtropical elevational zones of the Andes and the Neotropics. Analysis of geographic and elevational zones showed that genus Heliodoxa and related genera diversified by out-of-Andes dispersal to mountain ranges of Central America, coastal Venezuela, the Pantepui, and southeastern Brazil, since the mid-Miocene. Increased wildfire activity may lead to significant shifts in vegetation dynamics in high elevation mixed conifer forests. These changes raise concerns about the resilience of forests following wildfire, prompting the need for a deeper understanding of post-fire plant community response. Extreme temperatures and severe drought events have led to widespread tree mortality worldwide. In semi-arid regions of the Southwest United States, these events pose a significant threat to piñon-juniper (PJ) woodlands. We studied the effects of piñon and juniper mortality on the growth and physiology of existing saplings in PJ woodlands by analyzing water status, photosynthetic activity, and tissue chemistry to gain insights into these impacts. Juniper saplings exhibited improved water status and water use efficiency in response to overstory mortality, whereas piñon saplings did not. Polyploidy, or whole genome duplication, is a common phenomenon in plants, but the establishment and persistence of mixed-ploidy populations remains a paradox. This dissertation explores factors that contribute to the persistence and establishment of mixed-ploidy populations in nature. The first chapter investigates the role of unreduced gametes in neopolyploid establishment and finds that variability in their formation rate can have a significant impact on polyploid establishment and persistence. The second chapter searches for evidence of soil microbes exhibiting ploidy-specificity, a pre-condition for microbe-mediated niche differentiation, a possible stabilizing mechanism contributing to ploidy coexistence.
