Investigation Of Orthohantavirus Genetics In Rodent Reservoirs And Clinical Samples, Samuel Melvin Goodfellow
Investigation Of Orthohantavirus Genetics In Rodent Reservoirs And Clinical Samples, Samuel Melvin Goodfellow
Biomedical Sciences ETDs
Orthohantaviruses are negative-sense, single stranded RNA viruses with trisegmented genomes that can cause severe disease in humans and are carried by several host reservoirs throughout the world. In the United States, Sin Nombre orthohantavirus (SNV) is the primary cause of hantavirus cardiopulmonary syndrome (HCPS) with a fatality rate of ~36% and the highest cases occuring in the southwest region. The primary SNV host reservoir is thought to be the western deer mouse, Peromyscus maniculatus, however it has been shown that other rodents can carry different orthohantaviruses. We designed a pan-orthohantavirus detection tool to survey several small mammal populations throughout New …
High P-T Single Crystal Elasticity Of Zircon And Its Implications For Determining Metamorphic P-T Conditions In Nature, Arlacee Luu
Earth and Planetary Sciences ETDs
Understanding the natural petrologic and tectonic processes, such as slab subduction, requires accurate determination of the pressure (P) and temperature (T) conditions of these processes. Recent development of elastic thermobarometry based on the difference between the elastic evolution of mineral inclusions and their host minerals allows constraining the formation P-T conditions of the inclusion/host system without assuming thermodynamic equilibrium, which is typically needed for the widely used chemistry-based thermobarometry in metamorphic petrology. However, the reliability of elastic thermobarometry heavily depends on the accurate determination of the high P-T single-crystal elastic properties of relevant minerals. In this study, we performed the …
Testing The Niche Center Hypothesis In The Fossil Record Of Atlantic Bivalves, Rhiannon Z. Nolan
Testing The Niche Center Hypothesis In The Fossil Record Of Atlantic Bivalves, Rhiannon Z. Nolan
Earth and Planetary Sciences ETDs
Paleoecological analyses of six shallow marine bivalves were conducted to test the Abundant Center Hypothesis using data from the fossil record of the Pleistocene through modern day. This hypothesis predicts the highest abundance of a species is at the center of the geographic or environmental range, decreasing toward the edges. In geographic space, distances to a centerline within a geographic range were variably correlated with population abundances, and some species displayed a sharp drop-off in abundance as distance increased. In environmental space, bivalve species showed moderate correlations between abundance and centrality when measured using cumulative data across the last 2.8 …

Synthetic Biology Enabled Modification Of Bone Cells And Impact On Bone Mineral Resorption, Dulce C. Hayes
Biomedical Sciences ETDs
Development of a cell culture based bone model can contribute to biomedical research by producing a reproducible model system that mimics a bone remodeling disorder and reduces the use of animals in research studies. Synthetic biology was used to engineer osteoblast cell lines to alter a key protein in the RANKL pathway in efforts to control the bone remodeling process. Osteoblast cell lines, K7M2 and MC3T3-E1 Subclone 4 were engineered to express osteoprotegerin (OPG) and inhibit OPG expression, respectively. The upregulation of OPG expression was confirmed with Real Time PCR and Quantikine® Immunoassay. To identify the functionality of the engineered …
Cholesterol Regulation Of Egfr-Dependent Vasoconstriction In Chronic Hypoxia-Induced Pulmonary Hypertension, Rosstin Ahmadian
Cholesterol Regulation Of Egfr-Dependent Vasoconstriction In Chronic Hypoxia-Induced Pulmonary Hypertension, Rosstin Ahmadian
Biomedical Sciences ETDs
Pulmonary arterial constriction and remodeling resulting from chronic hypoxia (CH) lead to increased pulmonary vascular resistance and resultant pulmonary hypertension in patients with chronic obstructive pulmonary diseases, sleep apnea, or in residents at high altitude. Recent studies from our laboratory have demonstrated that CH augments vasoconstrictor reactivity to a variety of stimuli, including pulmonary arterial smooth muscle cell (PASMC) stretch, membrane depolarization, and stimulation of G protein-coupled receptors. This effect of CH is mediated by a switch in PASMC signaling from Ca2+-dependent mechanisms towards a novel epidermal growth factor receptor (EGFR)-NADPH oxidase 2 (NOX2)-Rho kinase Ca2+ sensitization pathway that occurs …
Determining The Existence And Regulation Of Microlipophagy In Primary Brown And White Adipocytes, Sara C. Arenas De Leon
Determining The Existence And Regulation Of Microlipophagy In Primary Brown And White Adipocytes, Sara C. Arenas De Leon
Biomedical Sciences ETDs
Lipids are one of life’s four main macromolecules and provide essential functions to cells.
The degradation of lipid droplets and mobilization of lipids occurs through lipolysis. Emerging evidence demonstrates evidence of a selective form of autophagy in lipolysis. The process of microlipophagy has only recently been described, and many features of its regulation are still poorly understood. Our recent study showed that inhibiting mammalian target of rapamycin complex 1 (mTORC1) pathway by depletion of Raptor in adipocytes led to an influx of lysosomes and accumulation of lipid droplets within lysosome. Here, we expand on these previous findings. We were able …
Hydrothermal Liquefaction Of Wastewater Sludges For Energy Resource Recovery, Carl L. Abadam
Hydrothermal Liquefaction Of Wastewater Sludges For Energy Resource Recovery, Carl L. Abadam
Civil Engineering ETDs
This study aims to identify how variabilities in wastewater process configurations impact the biocrude yields from sludges processed and stabilized by hydrothermal liquefaction. Clean and renewable energy sources are vital to ensure a sustainable future. However, the current consumption of fossil fuels limits the ability to achieve this goal. Hydrothermal liquefaction (HTL) is a thermochemical process that converts wet biomass to valuable biocrude oil and biochar while reducing solid masses. Wastewater sludges were sampled across three facilities with varying influent design capacities and biological treatment configurations. Lipids extracted via solvent extraction and biocrudes produced from HTL were determined gravimetrically. Relative …
Performance Evaluation Of E-Ticketing Technologies On Asphalt Paving Projects In New Mexico, Gena C. Robertson
Performance Evaluation Of E-Ticketing Technologies On Asphalt Paving Projects In New Mexico, Gena C. Robertson
Civil Engineering ETDs
The collection and processing of paper tickets for construction material delivery is an inefficient and antiquated practice that exposes inspectors on highway paving projects to substantial safety risks. An alternative to the collection of paper tickets is e-Ticketing, which uses web-based systems to deliver similar information. This research documented a current paper ticketing process and compared it to an e-Ticketing process that was implemented on several pilot projects. Data were obtained using stakeholder meetings, personnel interviews, and surveys. When compared to a paper ticketing system, e-Ticketing was shown to have fewer opportunities for error, require fewer decision points, and provide …

Civil Engineering ETDs
Hazardous metals mobilized from natural deposits and mining legacy are environmental and human health concerns. The waters of our partner Native American communities affected by uranium and gold mining legacy exceed the EPA MCL of 10 µg/L arsenic (As). Bioremediation is a novel technology that uses native resources to remediate metals in the soil and groundwater. Plant-fungi symbiosis for bioremediation is widely studied, but the role of endophytic fungi in metal uptake is not well understood. The objectives of this study are to 1) determine As uptake by symbiotic endophyte Cadophora sp. and little bluestem grass (Schizachyrium scoparium) …
Hydraulic Habitat Suitability For The Rio Grande Silvery Minnow At The Los Chavez Irrigation Outfall, New Mexico, Haley A. Ormsbee
Hydraulic Habitat Suitability For The Rio Grande Silvery Minnow At The Los Chavez Irrigation Outfall, New Mexico, Haley A. Ormsbee
Civil Engineering ETDs
It is essential to manage the Middle Rio Grande for aquatic species to preserve the native biodiversity of the river. During periods of low flow, endangered species such as the Rio Grande Silvery Minnow (RGSM) may get trapped in a drying reach without the ability to escape from isolated pools. Very little is known about how areas of refuge, such as irrigational outfall channels, buffer RGSM from stream flow intermittency. Hence, there is a crucial need to quantify available habitat for RGSM in areas of refuge. This study aims to make connections between flow regimes and physical-habitat conditions to help …
Identifying The Required Ecological Flow For Sediment Movement In The Río Chama: Field Data Informed Visual Spatial Model, Cristian Kremer
Identifying The Required Ecological Flow For Sediment Movement In The Río Chama: Field Data Informed Visual Spatial Model, Cristian Kremer
Civil Engineering ETDs
The continuous alteration of flow regimes in rivers has led to negative impacts on habitats and biotic communities, causing irreversible effects on river biodiversity. Implementing e-flows in management strategies can repair historical mismanagement, prevent further degradation, and restore ecosystem services. In the case of the Rio Chama, damming has altered its hydrological regime, affecting sediment transport. Experts recommend specific e-flow conditions for the Rio Chama to improve ecological conditions, in particular a bankfull discharge of 70 m3s-1 every 2 years for sediment flushing and channel maintenance. The Río Gallina, a tributary of the Rio Chama, plays a …

Development Of The Navigator: A Lagrangian Smart Sensing System To Characterize Aquatic Ecosystems., Aashish Sanjay Khandelwal
Civil Engineering ETDs
Most freshwater aquatic studies rely on Eulerian monitoring, i.e., water quality and quantity are monitored using grab samples or semi-continuous sensors deployed at fixed cross-sections. While Eulerian monitoring is practical, it provides a limited understanding of spatial and temporal heterogeneity and their effects on environmental processes. This dissertation summarizes the design and application of The Navigator, an alternative Lagrangian monitoring system that offers cost-effective solutions for in-situ, real-time data collection in lotic and lentic freshwater ecosystems such as streams, rivers, ponds, and reservoirs. The Navigator features a suite of methods – an autonomous surface vehicle (ASV) with GPS and LTE …

Civil Engineering ETDs
The Los Lagos region of Chile is experiencing ecological and socioeconomic challenges because of climate change. Despite efforts by the agricultural sector to understand and manage water resources, the lack of basic water quantification data affects their ability to effectively plan and implement water management strategies. This research aims to address this critical issue through three key objectives. First, characterize soil properties related to texture and structure in a micro-basin of the Los Lagos region. Secondly, it aims to investigate the hydrologic budget of the micro-basin with an emphasis on soil moisture using the Soil Moisture Routing model (SMR). Lastly, …
Effects Of Acute And Chronic Gut Injury On Secretory Lineage Differentiation, Roger Atanga
Effects Of Acute And Chronic Gut Injury On Secretory Lineage Differentiation, Roger Atanga
Biomedical Sciences ETDs
Acute and chronic injury to the gut has several detrimental consequences. However, dysregulation of secretory lineage cells is a common observation. To model acute gut injury, we use non-fissile uranium bearing dust (UBD), an environmental toxicant prevalent in communities adjacent abandoned uranium mines, majority of which are located in the Southwestern United States. Repeated exposure to environmental toxins and heavy metals has been obscurely linked to intestinal inflammation, increased susceptibility to pathogen-induced diseases, and higher incidences of colorectal cancer. However, there is no molecular evidence that non-fissile uranium can cause damage to the intestinal epithelia. Human colonoids exposed to UBD …

Heterochiral Dna Nanotechnology For Biomedical Applications, Tracy L. Mallette
Biomedical Engineering ETDs
In the past 30 years, there have been major advancements on how to treat and diagnose disease because of the improvement and increase in accessibility of sequencing technology. Nucleic acid-based therapeutics can manipulate protein expression. Likewise, pathogens can be identified and detected with single nucleotide specificity. However, the underlying oligonucleotide technology requires protection against natural defense systems that have evolved to destroy foreign nucleic acids. Many chemical modifications that can protect nucleotides also have significant cytotoxic side effects and must be carefully designed into the strands. A novel way to protect against nuclease-mediated degradation is through the use of mirror-image, …

The (Im)Mobility Of Being: The Anti-Black And Colonial Roots Of Contemporary Policing, Korey Tillman
The (Im)Mobility Of Being: The Anti-Black And Colonial Roots Of Contemporary Policing, Korey Tillman
Sociology ETDs
Popular debates over policing often revolve around spectacular forms of police violence that grab national media attention. In this study, I examine the everydayness of contemporary U.S. policing and the antiblack and colonial roots that sustains its many forms. Drawing on three years of ethnographic fieldwork with a grassroots abolitionist organization and homeless service provider, and 23 interviews with Black adults across the U.S., I show how mundane policing serves as sites of racialization where the category of human is contested. I offer stops as an analytic to observe how these mundane forms of policing occur in places such as …
Disrupting Whiteness In Education Organizations: Community Testimonios Of Strategy, Resistance, And Perseverance In Educational Justice Organizing, Florence E. Castillo
Disrupting Whiteness In Education Organizations: Community Testimonios Of Strategy, Resistance, And Perseverance In Educational Justice Organizing, Florence E. Castillo
Sociology ETDs
Through this project, I utilize multiple approaches to disrupt whiteness. I make a case for centering Indigenous and non-Western methods, such as testimonio in sociological studies, to disrupt the academic whiteness of knowledge creation and validation. Whiteness itself is amorphous and looks different depending on circumstances, location, who holds power, and context (Hughey 2016). Utilizing a theory of racialized organizations, I shed light on how whiteness is normalized in education institutions through race-neutral, everyday actions (Ray 2019). This further disrupts whiteness by rendering it visible. Finally, I highlight the different modes of resistance that educational activists use to disrupt whiteness. …

Cornelia de Lange Syndrome (CdLS) is a rare, dominantly inherited multisystem developmental disorder characterized by highly variable manifestations of growth and developmental delays, upper limb involvement, hypertrichosis, cardiac, gastrointestinal, craniofacial, and other systemic features. Pathogenic variants in genes encoding cohesin complex structural subunits and regulatory proteins (NIPBL, SMC1A, SMC3, HDAC8, and RAD21) are the major pathogenic contributors to CdLS. Heterozygous or hemizygous variants in the genes encoding these five proteins have been found to be contributory to CdLS, with variants in NIPBL accounting for the majority (>60%) of cases, and the only gene identified to date that results in …

Examining The Measurement Invariance Of The Revised Sociosexual Orientation Inventory Among Hispanic And Non-Hispanic White College Women, Christina Nicole Gillezeau
Psychology ETDs
I examined whether the Revised Sociosexual Orientation Inventory (SOI-R) was invariant between Hispanic/Latina and Non-Hispanic White (NHW) undergraduate women. I performed confirmatory factor analysis to assess model fit and used increasingly restrictive models to test invariance. I included 208 Hispanic/Latina women and 190 NHW women. The CFI and RMSEA model fit statistics (CFI=0.98, RMSEA=0.06 (90% confidence interval=0.04-0.08, p-value=0.16) showed that the model fit well. I constrained factor loadings to be equal in both groups to test metric invariance. I observed non-significant differences in model fit (deltaX2 (9)=88.67, p>0.95, deltaCFI=0.003, deltaRMSEA=-0.011). I also constrained intercepts to be equal to …

Combining Semi-Continuous And Discrete Water Quality Data To Characterize Solute Transport, Biogeochemical Processes, And Wildfire Disturbances In Fluvial Systems, Justin Rae Nichols
Civil Engineering ETDs
In the past decade, high-frequency water quality sondes have become more abundant in watersheds across North America and Europe and are gaining a foothold in Asia and South America. In this dissertation, three relevant topics associated with high-frequency data are investigated, i.e., the impact of winter’s precipitation on surface water quality and stream metabolism, the longitudinal propagation of wildfire disturbances through a fluvial network, and the use of machine learning with high-frequency data to estimate fluvial nutrient processing. First, we found that significant snow precipitation can cause surface water anoxia and declines in stream metabolism. Second, our data illustrate that …

The Impacts Of Transfer Learning For Ungulate Recognition At Sevilleta National Wildlife Refuge, Michael Gurule
Geography ETDs
As camera traps have grown in popularity, their utilization has expanded to numerous fields, including wildlife research, conservation, and ecological studies. The information gathered using this equipment gives researchers a precise and comprehensive understanding about the activities of animals in their natural environments. For this type of data to be useful, camera trap images must be labeled so that the species in the images can be classified and counted. This has typically been done by teams of researchers and volunteers, and it can be said that the process is at best time-consuming. With recent developments in deep learning, the process …

High Altitude Exercise, Autophagy, And Angiogenesis, Quint Nikolas Berkemeier
Health, Exercise, and Sports Sciences ETDs
Environmental hypoxia, as a result of decreased barometric pressure upon ascent to high altitudes (>2500m) presents increased physiological demands compared to low altitudes. Competitive athletes, mountaineers, wildland firefighters, military personnel, miners, and outdoor enthusiasts commonly participate in, or are exposed to, forms of exercise or physical labor at moderate to high altitudes. Interestingly, however, the majority of research on intermittent hypoxic exposure is centered around hematological markers and the skeletal muscle cellular responses to exercise in hypoxic environments remain largely unknown. Two processes that are integral for maintenance of cellular health in skeletal muscle include autophagy and angiogenesis. We …

Comparison Of Random Coupling Model To Stochastic Dyadic Green’S Function When Coupling To A Distributed Port, Evelyn Anne Dohme
Comparison Of Random Coupling Model To Stochastic Dyadic Green’S Function When Coupling To A Distributed Port, Evelyn Anne Dohme
Electrical and Computer Engineering ETDs
The Random Coupling Model (RCM) provides statistical representation of coupling between ports in a highly overmoded resonant cavity based on knowledge of radiation impedances of system ports and cavity properties. A recent variant of RCM, the Stochastic Dyadic Green’s Functions (S-DGF), provides the statistical distribution of fields in the cavity. By using Source Reconstruction Methods (SRM) to approximate equivalent surface currents, I hypothesized that the S-DGF can calculate the fields at their location, enabling a statistical estimate of the coupled power. Using nearfield radiation from simple patch antenna simulations and the SRM to estimate the equivalent fields and account for …

The water mills of New Mexico played a major role in the agricultural economy of the Río Arriba for centuries following the introduction of wheat from the Old World to the Americas. Wheat, in its ground form as flour, was a staple during the Spanish colonial period. To process raw wheat, local grist mills (molinos) were essential infrastructure as were the aceq uias (ditches) that powered them. Situated near the banks of rivers, the internal components of each mill were driven by the gravity force of water from an acequia, itself diverted from the river. Researchers have documented …

The Third Domain Of T Cells In The Evolution Of Novel Immune Cell Types, Kimberly Anne Morrissey
Biology ETDs
αβ and γδ T cells play an essential role in the adaptive immune response and are present in the majority of jawed vertebrates. The T cell receptor (TCR) structure and genetic organization have been generally stable throughout 400 million years of gnathostome evolution. However, the TCRδ chain has displayed degrees of plasticity unseen in other TCR isoforms. One such form appears to be a chimera of a TCR chain with an antibody-like binding domain, known as VHδ and has been found a wide array of vertebrates including cartilaginous fish, coelacanths, amphibians, tuataras, birds, crocodilians, and monotreme mammals. Another, more extreme …

Investigating The Costs And Benefits Of A Nutritional Fungal Endosymbiont, Nolan L. Perryman
Biology ETDs
Research on insect symbiosis is dominated by insect–bacteria models, however, the role of fungal symbionts in insects is poorly understood. Fungi are de novo synthesizers of many essential nutrients, therefore, insect–fungal pairings can permit specialization on nutritionally deprived diets. Here, I address the costs and benefits of symbiosis using a system of beetle–fungal symbiosis. Specifically, I ask 1) when reared on a low-quality diet, does the beetle increase the number of viable symbionts to compensate for a heightened nutritional demand, and 2) Is the conferred fitness benefit of the symbiont only present in low-quality diets? When reared on low-quality diets, …
