Synthesis And Characterization Of Tethered Poly(N-Isopropylacrylamide) For Quantitative Cell Adhesion Studies, Phanindhar J. Shivapooja Jul 2011

Synthesis And Characterization Of Tethered Poly(N-Isopropylacrylamide) For Quantitative Cell Adhesion Studies, Phanindhar J. Shivapooja

Chemical and Biological Engineering ETDs

Environmentally responsive (or smart') polymers represent a new approach for controlling biofouling release. Poly(N-isopropylacrylamide) (PNIPAAm) is a thermo responsive polymer that can potentially be used to control biofouling, as it exhibits a change in its relative hydrophobicity above and below its lower critical solution temperature (LCST ~ 32\uf0b0C). This thesis reports on grafted PNIPAAm brushes, synthesized using a relatively simple and rapid method which allows atom transfer radical polymerization (ATRP) in presence of air. In this study the structure, purity and thermally responsive behavior of grafted brushes have been characterized using X-ray photoelectron spectroscopy, time-of-flight secondary ion mass spectroscopy, Fourier …

Ionic Self-Assembly Of Porphyrin Micro-Nano Structures For Solar Hydrogen Evolution, Yongming Tian Jul 2011

Ionic Self-Assembly Of Porphyrin Micro-Nano Structures For Solar Hydrogen Evolution, Yongming Tian

Chemical and Biological Engineering ETDs

This research project develops self-assembled porphyrin micro-nano structures and their metal nanocomposites for efficient solar H2 evolution. The project is composed of three inter-related tasks. The first task is to develop the synthetic methods for preparing the binary porphyrin micro-nano structures by ionic self-assembly under various aqueous solution conditions. Although the porphyrin molecules have been widely studied in artificial photosynthesis, the porphyrin monomers absorb light in a limited range of the solar spectrum and are unstable in solution. In using ionic selfassembly of two oppositely charged porphyrin molecules, we successfully synthesized the more robust micro/nano clover-like structures to replace the …

Antibacterial Activity Of Conjugated Electrolytes, Thomas S. Corbitt Jun 2011

Antibacterial Activity Of Conjugated Electrolytes, Thomas S. Corbitt

Chemistry and Chemical Biology ETDs

A series of water soluble, cationic, anionic and zwitterionic conjugated polyelectrolytes (CPEs) with main chains based on an arylene ethynylene repeat unit structure with tetraalkylammonium and/or alkylsulfonate side groups were found to exhibit significant antibacterial activity. A number of these complexes also showed pronounced light-induced modulation of their effectiveness, with either biocidal enhancement or suppression, depending on such properties as singlet oxygen sensitization potential and lipophilicity. These collected studies examine the biocidal activity of the CPEs, as determined by Confocal Laser Scanning Microscopy (CLSM) and Flow Cytometry, correlating this activity with the photophysical properties of the polymers. We demonstrate biocidal …

Using Polymer Ligand Films For Rapid Radiochemical Analyses, Crystal L. Tulley-Cordova May 2011

Using Polymer Ligand Films For Rapid Radiochemical Analyses, Crystal L. Tulley-Cordova

Water Resources Professional Project Reports

Existing procedures for radiochemical analysis of environmental samples involves extensive laboratory preparation requiring chemical separations that are time consuming, and labor intensive to remove interferences and impurities associated with the sample matrix. Technology developed in this project reduces turnaround time, labor and supplies needed to examine radionuclides in water samples. The thin film technique can be used as a screening method. A solution including an extractive ligand and polymer are used to create a thin film to concentrate radionuclides from solution. In this project, several ligands, polymers, solvents, thin film types, and extraction procedures were examined. The polymer ligand thin …

Induction Of Mucosal Immunity Using Virus-Like Particle Based Vaccines, Zoe Hunter May 2011

Induction Of Mucosal Immunity Using Virus-Like Particle Based Vaccines, Zoe Hunter

Biomedical Sciences ETDs

Many viral structural proteins are capable of spontaneously self-assembling into structures that resemble virus particles. These structures, called virus-like particles (VLPs), have multivalent, highly repetitive structures which are capable of inducing robust and enduring immune responses, and, therefore, can serve as the basis for effective vaccines. VLPs can be used as stand-alone vaccines targeting the viruses from which they are derived and also as platforms for presenting heterologous antigens; their ability to evoke strong antibody responses against even poorly immunogenic targets makes them an attractive model for future vaccine and drug delivery vehicle designs. Our lab developed display technologies that …

Stable Isotopes Provide Insight Into The Use Of Wildlife Water Developments By Resident And Migrant Birds In The Sonoran Desert Of Arizona, Theresa C. Hyde May 2011

Stable Isotopes Provide Insight Into The Use Of Wildlife Water Developments By Resident And Migrant Birds In The Sonoran Desert Of Arizona, Theresa C. Hyde

Biology ETDs

Several studies have looked at the importance of man-made water resources to wildlife in desert regions. To our knowledge, however, none have attempted to directly quantify their importance to both resident and Neotropical migratory birds. During the spring and summer from 2007-2009, we enriched man-made water developments in the Sonoran Desert on the Kofa National Wildlife Refuge, Arizona. We enriched water developments using deuteriated water and sampled the body water pools of resident and migrant birds to quantify development use. We used a simple two end-point mixing model to estimate the proportion of an individual birds body water pool that …

Divergence In The Ecology Of Two Species Of Gambusia In Secondary Contact, Daniella Marilyn Swenton May 2011

Divergence In The Ecology Of Two Species Of Gambusia In Secondary Contact, Daniella Marilyn Swenton

Biology ETDs

Gambusia nobilis, a federally endangered species, and G. affinis (Poeciliidae) are small, livebearing fishes found in the southwestern U.S. The invasive G. affinis has been introduced globally to control mosquito populations. It is found in some populations of G. nobilis on Bitter Lake National Wildlife Refuge (BLNWR), the field site for this study. It is unclear to what extent the two species have diverged in behavior and ecology and how extensively they have hybridized on BLNWR, thereby threatening the endangered G. nobilis via genetic introgression. In this study I examined divergence in behavior and ecology between two species with four …

From Growth To Extinction: Explored By Life History And Metabolic Theory, Wenyun Zuo May 2011

From Growth To Extinction: Explored By Life History And Metabolic Theory, Wenyun Zuo

Biology ETDs

The laws of energy and material conservation are fundamental principles across various scales and systems. Based on the conservation laws, I derive several theoretical models to understand mechanisms behind the energy budget of ontogenetic growth and the pattern of the late Pleistocene extinction of megafauna in the Americas. First, I present a model, empirically grounded in data from birds and mammals, that correctly predicts how growing animals allocate food energy between synthesis of new biomass and maintenance of existing biomass. Previous energy budget models have typically been based on rates of either food consumption or metabolic energy expenditure. The model …

The Role Of Diffusion And Membrane Topography In The Initiation Of High Affinity Ige Receptor Signaling, Nicholas L. Andrews May 2011

The Role Of Diffusion And Membrane Topography In The Initiation Of High Affinity Ige Receptor Signaling, Nicholas L. Andrews

Biomedical Sciences ETDs

The high affinity IgE receptor (FcεRI) plays a primary role in the pathogenesis of allergic disease and shares significant similarities with the two other multichain immune recognition receptor family members, the B-cell receptor and T-cell receptor. A wealth of information exists in all three of these receptor systems with regard to the signaling cascades occurring subsequent to receptor activation. It is also known that all three require binding of multivalent antigen to initiate signaling. However, very little is known about the precise mechanism by which multivalent antigen binding initializes downstream signaling. It has long been known that, in response to …

Regulation Of Spreading Depression Events In Brain Slices By Astrocyte Metabolism, Jessica Lousie Seidel May 2011

Regulation Of Spreading Depression Events In Brain Slices By Astrocyte Metabolism, Jessica Lousie Seidel

Biomedical Sciences ETDs

Spreading depression (SD) is a severe depolarization of both neurons and astrocytes that can propagate throughout CNS tissue. SD has long been associated with migraine, and recent work has strongly suggested that closely related events are likely to be involved in pathophysiological conditions where they provide a large additional metabolic burden to injured tissues. The major goal of this dissertation was to test the overall hypothesis that manipulation of astrocyte metabolic capacity can modify the initiation and propagation of deleterious spreading depression (SD)-like events. If this were the case, then it may support targeting these cells in a range of …

Snap-25 Is A Component Of A Ubiquitous Snare Complex Required For Evoked Neuroexocytosis In Gabaergic Neurons, Lawrence C. R. Tafoya May 2011

Snap-25 Is A Component Of A Ubiquitous Snare Complex Required For Evoked Neuroexocytosis In Gabaergic Neurons, Lawrence C. R. Tafoya

Biomedical Sciences ETDs

The processes of vesicular trafficking and membrane fusion are fundamental to nervous system development and communication among neurons within integrated circuits. The regulated release of several neurotransmitters is dependent on synaptosomal-associated protein 25 kDa (SNAP-25)-containing soluble N-ethylmaleimide-sensitive factor attachment protein receptor (SNARE) core complexes. The requirement for this fusogenic machinery in evoked neurosecretion has been repeatedly demonstrated through the use of mutant mouse models and neurotoxin blockades. However, the existence of a functional role for a SNAP-25-containing SNARE complex has not been shown in the major inhibitory neurotransmitter system of the brain, gamma-aminobutyric acid (GABA). To determine whether SNAP-25 participates …

Live Cell Kinetics Of Erbb Dimerization Reveals Influences Of Activation State And Membrane Organization, Shalini T. Low-Nam May 2011

Live Cell Kinetics Of Erbb Dimerization Reveals Influences Of Activation State And Membrane Organization, Shalini T. Low-Nam

Biomedical Sciences ETDs

The erbB1 receptor regulates cellular programs including proliferation, migration, and differentiation and is the prototypical receptor tyrosine kinase (RTK). The erbB family consists of four homologous transmembrane receptors (erbB1/HER1/EGFR, erbB2/HER2, erbB3/HER3, erbB4). Canonically, ligand binding leads to an extracellular conformational change that promotes the formation of a receptor-mediated back-to-back dimer, asymmetric orientation of the catalytic kinase domains, and downstream transphosphorylation of cytoplasmic tyrosine residues. Exceptions to this paradigm are the orphan erbB2 and the kinase defective erbB3. The erbB receptors are implicated in mechanisms of carcinogenesis and are, thus, important therapeutic targets. Antibodies and small molecule inhibitors have been used …

Carbonylation Of Neuronal Cytoskeletal Proteins And Their Proteolytic Degradation In Acute Experimental Autoimmune Encephalomyelitis, Suzanne Smerjac May 2011

Carbonylation Of Neuronal Cytoskeletal Proteins And Their Proteolytic Degradation In Acute Experimental Autoimmune Encephalomyelitis, Suzanne Smerjac

Biomedical Sciences ETDs

Oxidative stress and its damage resulting in carbonylation of proteins is known to take place in the CNS tissue of multiple sclerosis patients and of mice with experimental autoimmune encephalomyelitis (EAE), and has been implicated in the pathophysiology of these diseases. In order to investigate the possible consequences of this protein damage during acute myelin basic protein (MBP) -directed autoimmune neuroinflammation the first goal of this thesis was to characterize oxidative stress and protein carbonylation, including identification of the major oxidized protein species, in the Lewis rat model of EAE. I discovered that there is significant oxidative stress in the …

Historic And Demographic Changes That Impact The Future Of The Diné And Developing Community-Based Policy, Yolynda Begay May 2011

Historic And Demographic Changes That Impact The Future Of The Diné And Developing Community-Based Policy, Yolynda Begay

Architecture and Planning ETDs

The history and legacy of Native American relationship with the U.S. government has been marred with policies on termination, assimilation, annihilation, and displacement of Indian communities. The Dawes Act, also known as the General Allotment Act of 1887, was the starting point of enrolling tribal members for land allotments (Spruhan, 2006-2007, p. 2). The act required a population survey of individual persons residing on allotted lands. This, in turn, required Indian agents to conduct census rolls (see glossary). The purpose of the Dawes Act was to turn Indian people into Jeffersonian farmers by breaking up communal landholdings and allotting parcels …

Home/Land: Kiowa, New Mexico- A Grassland's Story, Heather R. Yaryan May 2011

Home/Land: Kiowa, New Mexico- A Grassland's Story, Heather R. Yaryan

Architecture and Planning ETDs

The purpose of this thesis was to illustrate how cultural and natural histories inform place-based community planning. Utilizing both cultural and natural histories, the planner may begin to see patterns of habitancy as they correlate to ecological fluctuations. I focused my research on Kiowa, New Mexico the community my family homesteaded in the grasslands of Northeastern New Mexico and took an auto-ethnographic approach to conducting my research. The story of Kiowa is both unique and universal. The intricacies of the land and people are, indeed, woven into a specific place and times. It is the intersection of Place and of …

The Materiality Of The Self: A Multimodal, Communicative Approach To Identity, Sachi Sekimoto May 2011

The Materiality Of The Self: A Multimodal, Communicative Approach To Identity, Sachi Sekimoto

Communication ETDs

The purpose of this dissertation is to propose a multimodal approach as an alternative way of theorizing and researching identity. The multimodal approach utilizes four modes of interaction--multidirectional interpellation, spatiality, temporality, and corporealit--to explore the processes of interaction and engagement between an individual and his/her social worlds. The multimodal approach focuses on the materiality of lived experience and the process of interaction and engagement between an individual and his/her social worlds through which his or her identity materializes. I apply the multimodal approach to analyze two autobiographical texts in which the authors deal with Asian identity in different cultural and …

Comparison Of Two Riparian Assessment Surveys : Proper Functioning Condition And The New Mexico Watershed Watch Riparian Survey, Katherine A. Smith Apr 2011

Comparison Of Two Riparian Assessment Surveys : Proper Functioning Condition And The New Mexico Watershed Watch Riparian Survey, Katherine A. Smith

Water Resources Professional Project Reports

Riparian areas perform functions which in turn provide economic and environmental benefits. Many of the riparian areas in the Southwest are in degraded states. There is a need to determine the health of these ecosystems, in order to decide which need rehabilitation or restoration. Long term monitoring is necessary to assure projects are progressing towards goals, and prevent further degradation in response to new disturbances. Two riparian assessment surveys currently in use are the Proper Functioning Condition (PFC) and the New Mexico Watershed Watch Riparian Survey (NMWWRS). The PFC tool is a qualitative method utilized by government agencies such as …

Pharmaceutically Active Compounds In Residential And Hospital Effluent, Municipal Wastewater, And The Rio Grande In Albuquerque, New Mexico, Kathryn D. Brown Apr 2011

Pharmaceutically Active Compounds In Residential And Hospital Effluent, Municipal Wastewater, And The Rio Grande In Albuquerque, New Mexico, Kathryn D. Brown

Water Resources Professional Project Reports

This project investigated: 1) the contribution of pharmaceutically active compounds (PhACs) from residential and hospital effluent sources, 2) resultant concentrations of PhACs in the Albuquerque Southside Water Reclamation Plant (SWRP) raw influent and treated effluent, and 3) concentrations of PhACs in the Rio Grande, which receives SWRP effluent. PhACs present in surface waters have been shown to adversely impact organisms (Jobling et al., 1998) and, in the case of antibiotics, perhaps increase resistance to these drugs (Ash, 1999; Eichorst, 1999; Guardabassi et al, 1998; Sternes, 1999). In this study, ten sample sites were identified and samples collected and analyzed for …

Performance Of A Green Roof Lysimeter In An Arid Climate, Rick Young Apr 2011

Performance Of A Green Roof Lysimeter In An Arid Climate, Rick Young

Water Resources Professional Project Reports

Green roofs, also known as living roofs or eco-roofs, are becoming part of American architectural and engineering design as a method of reducing stormwater runoff. Although soil roofs have been in use for as long as adobe bricks, modern green roofs have a larger-scale function. Modern green roofs are used to solve or mitigate a number of architectural and engineering, stormwater management and ecological problems, especially in urban settings with large impervious areas. The effectiveness of green roofs in reducing these problems has been studied primarily in wet to moderately wet climates; however, relatively little research has been performed in …

Integrating External Costs Into Water Utility Asset Management : An Application Of The Threshold Break Rate Method, Megan Marsee Apr 2011

Integrating External Costs Into Water Utility Asset Management : An Application Of The Threshold Break Rate Method, Megan Marsee

Water Resources Professional Project Reports

The goal of an asset management program is to minimize the life-cycle cost of water utility assets, while continuing to provide the service levels expected by utility customers. The life-cycle cost of an asset includes both the utilitys internal costs to maintain the asset, and external costs borne by customers, the community, and the environment when the asset fails. This project demonstrates how to integrate external costs into asset management through an application of the threshold break rate model, a pipe-replacement decision-model that minimizes the life-cycle cost of water mains. The model is employed to determine which six-inch-diameter steel water …
